1.Study on rapid identification of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants with Fourier exchange infrared spectroscopy

Objective:To establish a Fourier transform infrared spectroscopy (FT-IR) method for rapid identification of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants.Methods:The Fourier transform infrared spectra of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants were established, and the second derivative spectral analysis, clustering analysis, principal component analysis, opls-da and cluster independent soft mode classification model were analyzed to explore the difference characteristic peaks of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants.Results:The first order infrared spectrum showed that the peak shape and peak intensity of Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants were different. Clustering analysis, principal component analysis and OPLS-DA results showed that Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants showed good clustering characteristics. SIMCA method was used to construct the model, and the accuracy of the training set and the verification set were 100%, which further verified the feasibility of this method in identifying the authenticity of Cremastrae Pseudobulbus Pleiones Pseudobulbus.Conclusions:The second-order infrared spectroscopy can accurately distinguish the differences between Cremastrae Pseudobulbus Pleiones Pseudobulbus and its adulterants. The method is fast and accurate, and can be used for the authenticity identification of Cremastrae Pseudobulbus Pleiones Pseudobulbus.
2.Feasibility study of"four low"protocol based on body weight in coronary CT angiography

Objective To explore the feasibility of protocol with low tube voltage,low contrast agent dose,low injection rate,and low trigger threshold based on body weight in coronary computed tomography angiography(CCTA).Methods Patients suspected with coronary heart disease who underwent 320 rows of CCTA were prospectively selected.They were divided into 4 groups:<60 kg,60-70 kg,71-80 kg,>80 kg based on different body weights,120 cases for each group,then were randomly divided into experimental group and control group using a random number table method,with 60 patients in each group.Each case in the control group had a tube voltage of 120 kV and a trigger threshold of 240 HU;each case in the experimental group had a tube voltage of 100 kV and a trigger threshold of 190 HU.According to the CCTA application guidelines,the iodine flow rate was used to determine the contrast agent injection rate in the control group,while the injection rate in the experimental group was reduced by 30%.Using an injection time of 12 s,the control group's contrast agent dose was obtained,and that of the experimental group decreased by 30%.Two radiologists evaluated the CT values,contrast-to-noise ratio(CNR),image quality score,and signal-to-noise ratio(SNR)of the middle segment of the right coronary artery,proximal segment of the left anterior descending branch,and proximal segment of the left circumflex branch,as well as compared the effective dose(ED)of volume scanning and threshold monitoring sequences.One-way ANOVA,independent sample t-test,and Mann-Whitney U-test were used to evaluate the differences in each parameter.Results There were no statistically significant differences in general information and subjective image scores between the experimental group and the control group(P>0.05).There were no statistically significant differences in CT values,CNR,and SNR between the experimental group and the control group at different body weights for the middle segment of the right coronary artery,the proximal segment of the left anterior descending branch,and the proximal segment of the left circumflex branch(P>0.05).In terms of radiation dose,the comparison between the control group and the experimental group at the same body weights showed statistically significant differences both in volume scanning and threshold monitoring sequences(P<0.001),and the ED of the experimental group was significantly lower than that of the control group.Conclusion The protocol with low tube voltage,low contrast agent dose,low injection rate,and low trigger threshold determined based on different body weights can achieve satisfactory image quality for CCTA,while significantly reducing the radiation dose of volume scanning and threshold monitoring sequences.

4.Application of indocyanine green coaxial fluorescence projective imaging for sentinel lymph node biopsy in acral malignant melanoma

Objective:To observe the tracer effect of indocyanine green (ICG) surgical coaxial fluorescence projective imaging system (CFPI) and methylene blue (MB) staining method in the sentinel lymph node biopsy (SLNB) of acral malignant melanoma.Methods:Data of patients with acral malignant melanoma requiring SLNB diagnosed in the First Affiliated Hospital of University of Science and Technology of China from March 2020 to October 2022 were retrospectively analyzed. Both ICG and MB were injected intraoperatively to trace the sentinel lymph node (SLN), and SLNB was performed according to fluorescent labeling. Numbers of SLN and metastatic SLN deteted by ICG, MB, ICG combined with MB during operation were recorded. Statistical analysis was performed using Graphpad Prism 6 software, and the detected number of SLN was expressed as Mean±SD, and paired t-test was performed. The detection rate of SLN was expressed by % and χ2 test was used. P<0.05 was considered statistically significant. Results:A total of 21 patients were included, and 62 SLNs were detected, among which 58 (93.5%) were detected by ICG, 41 (66.1%) by MB, and 62 (100.0%) by ICG combined with MB, respectively. The detection rate of ICG was significantly higher than that of MB, with statistical significance ( χ2=22.34, P=0.001). The detected numbers of SLNs by ICG, MB, ICG combined with MB were (2.76±0.23), (1.95±0.21) and (2.95±0.25) /per person, respectively. ICG was significantly more than MB, and the difference was statistically significant ( t=2.60, P=0.013). A total of 10 metastatic SLNs were detected, of which 10 were detected by ICG, 7 by MB, and 10 by ICG combined with MB. Conclusion:Compared with MB, ICG CFPI showed better performance in SLNB, and ICG combined with MB showed better performance than ICG alone.

6.Draft Genome Assembly and Annotation for Cutaneotrichosporon dermatis NICC30027, an Oleaginous Yeast Capable of Simultaneous Glucose and Xylose Assimilation

The identification of oleaginous yeast species capable of simultaneously utilizing xylose and glucose as substrates to generate value-added biological products is an area of key economic interest. We have previously demonstrated that the Cutaneotrichosporon dermatisNICC30027 yeast strain is capable of simultaneously assimilating both xylose and glucose, resulting in considerable lipid accumulation. However, as no high-quality genome sequencing data or associated annotations for this strain are available at present, it remains challenging to study the metabolic mechanisms underlying this phenotype. Herein, we report a 39,305,439 bp draft genome assembly for C. dermatis NICC30027 comprised of 37 scaffolds, with 60.15% GC content. Within this genome, we identified 524 tRNAs, 142 sRNAs, 53 miRNAs, 28 snRNAs, and eight rRNA clusters. Moreover, repeat sequences totaling 1,032,129 bp in length were identified (2.63% of the genome), as were 14,238 unigenes that were 1,789.35 bp in length on average (64.82% of the genome). The NCBI non-redundant protein sequences (NR) database was employed to successfully annotate 11,795 of these unigenes, while 3,621 and 11,902 were annotated with the Swiss-Prot and TrEMBL databases, respectively. Unigenes were additionally subjected to pathway enrichment analyses using the Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), Cluster of Orthologous Groups of proteins (COG), Clusters of orthologous groups for eukaryotic complete genomes (KOG), and Non-supervised Orthologous Groups (eggNOG) databases.Together, these results provide a foundation for future studies aimed at clarifying the mechanistic basis for the ability of C. dermatis NICC30027 to simultaneously utilize glucose and xylose to synthesize lipids.
7.Multimodal imaging characteristics of central serous chorioretinopathy converted to polypoidal choroidal vasculopathy and pachychoroidal neovascularization

Objective To investigate the multimodal imaging characteristics of central serous chorioretinopathy (CSC) converted to polypoidal choroidal vasculopathy (PCV) and pachychoroidal neovascularization(PNV).Methods A retrospective case series study was adopted.The clinical data of 91 eyes from 79 patients with CSC who received treatment from June 2009 to September 2017 in Shenzhen Eye Hospital were analyzed.Color fundus photography,fundus fluorescein angiography (FFA),spectral domain optical coherence tomography (SD-OCT) and indocyanine angiography (ICGA) were performed in the patients.The eyes with recurrence or maintained serous retinal pigment epithelium (RPE) after treatment were examined once more.Results In 91 eyes,11 eyes of 9 patients converted to PNV.Late stage of FFA showed multifocal hyperfluorecnece in macular area.ICGA showed plaque hyperflurorescence overlying dilated choroidal vessel.OCT revealed irregular flap shallow RPE detachment with retinal neurosensory detachment.OCTA revealed mass-like enlarged choroidal vessel between RPE and Bruch membrane.Nine eyes of 7 patients with CSC converted to PCV,and late stage of FFA showed serous-hemorrhage pigment epithelium detachment(PED) and multifocal enlargement hyperfluorecnece.ICGA showed branch vessels network (BVN) with small polyps.OCT revealed PED like thumb with double-layer sign and retinal neurosensory detachment.OCTA en-face revealed BVN with small polyps at choroidal capillaries layer.The baselines BCVA of PCV and PNV (LogMAR) was 0.282±0.220 and 0.413±0.190,respectively with a significant difference between them (t =0.037,P < 0.05).Subfoveal choroidal thickness (SFCT) of CSC,PCV and PNV patients was (373.61 ±65.11),(296.22 ±30.24) and (328.63 ±76.18) μm,with siginificant differences among them (F =3.48,both at P<0.05).SFCT of PNV patients was thicker than that of PCV patients,with a significant difference between the two groups (t =2.91,P<0.05).Conclusions Chronic or recurrence CSC is probably to develop to PNV,and the PNV with serous or hemorrhage PED is probably to switch to PCV.Multimodal imaging examination is helpful to the diagnosis of PNV and PCV.
8.The clinical significance of Th17 cells in patients with myelodysplastic syndrome and iron overload

Objective To study the significance of Th17 cells in patients with myelodysplastic syndrome (MDS) and iron overload.Methods A total of 77 patients with MDS admitted to Guangzhou First People's Hospital were enrolled from January 2017 to December 2018,who were divided into iron overload group (37 cases) with serum ferritin (SF) over 1000 μg/L and non-ferrous overload group(40 cases).CD4+T cells in peripheral blood (PB) and bone marrow (BM) were sorted by flow cytometry.The ratio of Th17 cells and cells with abnormal karyotype were compared.IL-17 and IL-6 protein and RNA expression were detected by ELISA and quantitative real-time PCR(qRT-PCR).Results The proportions of Th17 cells in PB and BM in iron overload group were significantly higher than those in non-iron overload group [(41.06± 0.96)% vs.(26.80± 1.21)%;(47.39± 1.60)% vs.(34.29± 1.03)%;P<0.01].The Th17 positive cells with abnormal karyotype in iron overload group were more than those in non-iron overload group[(4.96±0.53)% vs.(3.67±0.12)% in PB;(10.06±1.67)% vs.(4.36±0.43)% in BM;P<0.01].Similarly,the protein levels as well as mRNA expression of IL-6 and IL-17 in patients with iron overload were significantly higher than those in non-iron overload group (P<0.01 both in PB and BM).Conclusions As hematopoietic regulators secreted by Th17 cells,the expression of IL-6 and IL-17 in MDS patients with iron overload are elevated.This may predict the influence of these factors to the differentiation of Th 17 cells.
9.Relationship between the severity of iron overload in peripheral blood CD4+T cells and the key transcription factors of RORrt, IRF8, STAT3 for cell proliferation and differentiation in elderly patients with myelodysplastic syndrome

Objective To investigate the effects RORrt (RORrt),IRF8 (IRF8) and STAT3 (STAT3) in peripheral blood CD4+T cells on the cell proliferation and differentiation in elderly patients with iron-overload myelodysplastic syndrome(MDS).Methods A prospective case-control study was conducted.Twenty-two elderly hospitalized patients(12 males and 10 females)aged 60-78 years with iron-overload MDS from Jan.2017 to Dec.2018 were enrolled and considered as the observation group.Twenty MDS patients without iron overload hospitalized in the same period were selected as the non-iron overload group,and 26 healthy elderly people were considered as the healthy control group.Peripheral blood monocytes(PBM)were prepared and resident CD4+T cells were sorted by flow cytometry.The mRNA and protein expression levels of transcription factors of RORrt,p-STAT3 and IRF8 were detected by quantitative real time polymerase chain reaction(qRT-PCR)and Western blotting.Results In peripheral blood CD4+T cells,the mRNA expression level of RORrt and p-STAT3 were higher and that of IRF8 was lower in the iron-overload group than in the non-iron overload group and the healthy control group(42.634± 18.613 vs.21.289 ± 15.158 and 22.520 ± 9.896;29.710±9.689 vs.12.355±4.681 and 9.818±3.845;19.293±8.258 vs.23.785±12.498 and 69.619±23.768,P<0.01).In peripheral blood CD4+T cells,the protein expression level of RORrt and p-STAT3 was higher,and that of IRF8 was lower in the iron overload group than in the non-iron overload group and healthy control group(P<0.01).Conclusions The abnormalities of the mRNA and protein expression levels of transcription factors of RORrt,IRF8 and p-STAT3 in CD4+ T cells play a fundamental role in the pathogenesis of iron overload MDS in elderly patients.
10.Setup errors between head-neck-shoulders system and trunk immobilization system in radiotherapy for esophageal cancer

Objective To compare the setup errors of two position fixation, head-neck-shoulders immobilization system and trunk immobilization system in radiotherapy for esophageal cancer, and to provide technical guidance for the fixed modes and determination of plan target volume (PTV) in the intensity modulated radiation therapy (IMRT) of esophageal cancer. Methods 97 thoracic esophageal cancer patients treated with radiotherapy were collected and divided into two groups: head-neck-shoulders group (51 patients immobilized with head-neck-shoulders immobilization system) and trunk group (the other 46 patients immobilized with trunk immobilization system). Before the weekly radiotherapy, all the patients received cone-beam CT (CBCT) scan to get the setup errors of X axis (left and right), Y axis (head and foot) and Z axis (front and back) on line. Results In the head-neck-shoulders group, the errors in X, Y and Z direction were (0.333 ±0.400) cm, (0.333 ±0.291) cm and (0.238 ±0.256) cm, respectively. In trunk group, the errors were (0.327±0.255) cm, (0.582±0.501) cm and (0.189±0.154) cm, respectively. There were statistically significant differences in the setup errors in Y axis and Z axis between the head-neck-shoulders group and the trunk group (P< 0.05). Conclusions In the radiotherapy for thoracic esophageal carcinoma, there is no difference in the X axis setup error between head-neck-shoulders system and trunk immobilization system. The Y axis setup error of head-neck-shoulders group is less than that of the trunk group. The Z axis setup error of trunk group is less than the head-neck-shoulders group.
