BACKGROUND:The cryopreservation technology enables tissues/cells to be stored for a long time in a low-temperature environment while maintaining the integrity of their activity and function, which is of great significance for the construction of cell therapy, tissue engineering and biological sample banks.Cryoprotective agents often contain dimethyl sulfoxide and serum. To avoid the toxic side effects of dimethyl sulfoxide, the complexity of serum components and immune responses, although some finished cryoprotective agents have been marketed, they are faced with many difficulties such as high cost and limited application. Therefore, there is an urgent need to develop a cryoprotective agent with clear components and the ability to solve the above problems.OBJECTIVE:To evaluate the effects of a novel cryoprotectant on cryopreservation efficiency of different tissue and cell sources.METHODS:By applying the novel cryoprotectant as an experimental group with the commercially available and widely used cryoprotectant(control group)to umbilical cord Wharton's jelly tissue, umbilical cord mesenchymal stem cells, umbilical cord blood/peripheral blood mononuclear cells,NK and CIK cells, comparative analyses were conducted in terms of cell morphology, number, viability, surface markers, differentiation potential, and cell-killing toxicity assay before cryopreservation and after resuscitation thawing. We confirmed the cryopreservation effect of the new cryoprotectant and its potential application value.RESULTS AND CONCLUSION:(1)The novel cryoprotectant facilitated the normal growth of cryopreserved Wharton's jelly tissue upon recovery, exhibiting mesenchymal stem cell morphology. No significant differences were observed between the experimental and control groups in terms of cell recovery rate, surface markers, and differentiation potential.(2)There was no significant difference in the number and viability of cells between the experimental group and the control group after cryopreservation of cord blood/peripheral blood mononuclear cells, and the cryo-resuscitated cell numbers and viability of derived NK cells/CIK cells did not show significant difference between the experimental and control groups.(3)For NK cells derived and differentiated from cord blood/peripheral blood mononuclear cells, there was no significant difference in the proportion of CD56+CD16+cell subpopulations between the experimental group and the control group. For CIK cells derived and differentiated from cord blood/peripheral blood mononuclear cells, there was no significant difference in the proportions of CD3+CD8+and CD3+CD56+cell subpopulations between the experimental group and the control group.(4)In terms of cytotoxicity testing, when the effective-target ratio of immune cells and melanoma cell line Mel624 was 20:1,whether it was NK cells/CIK cells derived from cord blood or peripheral blood mononuclear cells, there was no significant difference in the tumoricidal activity of cells between the experimental group and the control group. These findings suggest that the novel cryoprotectant can replace existing commercially available and widely used cryoprotectants, and is applicable to Wharton's jelly tissue, umbilical cord mesenchymal stem cells, umbilical cord blood/peripheral blood mononuclear cells,as well as NK and CIK cells, providing a solid technical foundation for the scaling, standardization, and commercialization of universal cryoprotectants.

Objective To investigate the clinical efficacy of recombinant human granulocyte-macrophage colony-stimulating factor(rhGM-CSF)combined with bifidobacterium in the treatment of oral mucositis after chemotherapy. Methods A total of 60 post-chemotherapy patients with oral mucositis admitted to Ganzhou Cancer Hospital of Jiangxi Province from January 2023 to December 2024 were selected as subjects, the patients were divided into observation group(n=30)and control group(n=30)by using a randomized digital table method. The control group received rhGM-CSF mouthwash treatment, while the observation group was additionally administered bifidobacterium-lactobacillus triple live capsules orally.Clinical efficacy, symptom scores, inflammatory factor levels, oral microbiota indicators, and treatment safety were evaluated after 2 weeks of treatment. Results After treatment, the total effective rate in observation group was significantly higher than that in control group(P<0.05).Post-treatment evaluations showed that patients in observation group exhibited lower scores for edema, congestion, and ulceration, along with reduced pain visual analog scores and decreased levels of inflammatory markers including Toll-like receptor 4,lectin-3,and interleukin-8,the difference were statistically significant(P<0.05).After treatment, the oral microecological flora of patients in observation group, including lactic acid bacteria and bifidobacterium, were higher than that of control group, while porphyromonas gingivalis and fusetella were lower than that of control group, the difference were statistically significant(P<0.05).Conclusion The clinical effect of rhGM-CSF combined with bifidobacterium in the treatment of oral mucositis after chemotherapy was significant, which could reduce various symptoms of patients, inhibit inflammatory factors, improve oral microecology, and have good safety.

Chinese Journal of Geriatrics 2015;34(8):889-892
Objective To investigate the effect of Hydroxychloroquine plus Leflunomide on the lipid profile and disease activity index in patients with rheumatoid arthritis (RA).Methods A total of 140 elderly patients with RA who met the American College of Rheumatology (ACR) criteria were treated with Hydroxychloroquine (Group A,n =70) or leflunomide combined with hydroxychloroquine (Group B,n=70),and 100 healthy volunteers were selected as controls. The lipid profile was compared between the patient groups and control group.Lipid levels,Disease Activity Score 28 (DAS28),erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP) were measured before treatment and 6 months after treatment between Group A and Group B.Results Compared with the control group,RA patients exhibited higher levels of total cholesterol (TC,P<0.05),low-density lipoprotein cholesterol (LDL-C,P<0.05) and triglycerides (TG,P<0.01),and lower levels of high-density lipoprotein cholesterol (HDL-C,P<0.05).After 6 months of treatment, the disease activity index (DAS28,P<0.01),the duration of morning stiffness (P<0.01),ESR,CRP,swollen joint count (SJC) and tender joint count (TJC) decreased in RA patients, compared with controls (P<0.01 or 0.05).HDL-C levels increased and LDL-C levels decreased in Groups A and B after treatment, compared with pretreatment (P<0.05 for all). After treatment,TC levels decreased in Group B,compared with Group A (P< 0.05),while TG levels had no significant difference between the two groups (P>0.05).TC and LDL-C levels,ESR,CRP,DAS28,SJC and TJC were lower in Group B than in Group A (all P<0.05).Conclusions Hydroxychloroquine in combination with Leflunomide therapy can not only control inflammation but also improve the lipid profile, and is more effective than single-drug therapy. The combination may reduce the risk of atherosclerosis and cardiovascular events in patients with RA.
7.Diagnostic Problems of Hyperkinetic Disorders -Literature Analysis
Chinese Mental Health Journal 1992;0(01):-
There is great progress in the study of the diagnosis of hyperkinetic disorders in China within the latest ten years, but it is not enough. Three features are referred by reanalyzing 281 articles dating from 1997 to 2004 collected by the CJFD: 1) Several editions of diagnostic criteria and diagnostic methods are simultaneously used; 2) Comparing with the rate of utilization of the new edition, the old editions of diagnostic criteria constitute a small percentage ; 3) Diagnostic criteria and diagnostic tools have some shortcomings. Four recommendations maybe help to solve these problems:1) Make parents and teachers advance their acknowledgement of hyperkinetic disorders; 2) Develop and improve the diagnostic criteria and diagnostic tools through more researches; 3) Use advanced scientific apparatuses more widely; 4) Establish a perfect diagnostic system according to our culture.
