ObjectiveTo explore the mechanism of Huashi Baidu granules in treating influenza A (H1N1) virus infection based on the gut-lung axis. MethodsBALB/c mice were randomly assigned into a blank control group, a model control group, a positive control (oseltamivir phosphate capsules, 19.5 mg·kg-1) group, and a Huashi Baidu granules (17.81 g·kg-1) group. Mice in other groups except the blank control group were instilled nasally by H1N1 viral fluid for the modeling of influenza viral pneumonia and treated with corresponding drugs once daily by gavage for 5 consecutive days. The mouse lung index was calculated, and pathological changes in the lung and colon tissue were observed. Western blot was employed to quantify the expression levels of nucleotide-binding oligomeric domain-like receptor protein 3 (NLRP3), cysteinyl aspartate-specific proteinase -1 (Caspase-1), apoptosis-associated Speck-like protein containing a CARD (ASC), gasdermin D (GSDMD), interleukin-18 (IL-18), IL-1β, and colonic tight junction protein 1 (CLDN1), and Occludin in the lung and colon tissue. Non-targeted metabolomics was employed to analyze the metabolic profiles of the lung tissue and intestinal contents. The 16S rRNA sequencing was performed to analyze the gut microbiota structure in intestinal contents. Enzyme-linked immunosorbent assay (ELISA) was employed to measure the 5-hydroxytryptamine (5-HT) levels in the serum, lung tissue, and colon tissue. ResultsCompared with the blank control group, the model control group showed elevated lung index, marked inflammatory cell infiltration and alveolar structural destruction in the lung tissue, obvious inflammatory cell infiltration and mucosal epithelial damage in the colon tissue, upregulated protein levels of NLRP3 inflammasome pathway proteins NLRP3, Caspase-1, GSDMD, IL-1β, and IL-18 in the lung and colon tissue (P<0.05, P<0.01), downregulated protein levels of CLDN1 and Occludin in the colon tissue (P<0.05, P<0.01). Compared with the model control group, Huashi Baidu Granules reduced the lung index (P<0.01), alleviated pathological damage in the lungs and colon, inhibited the expression of NLRP3, Caspase-1, GSDMD, IL-1β, and IL-18 in lung and colon tissue (P<0.05, P<0.01), and upregulated the expression of CLDN1 and Occludin in the colon tissue (P<0.05, P<0.01). Metabolomics analysis showed that Huashi Baidu Granules downregulated the 5-HT pathway and arachidonic acid metabolism pathway in the lung and colon tissue, while upregulating nucleotide metabolism and TCA cycle. Compared with the blank control group, the model control group exhibited elevated levels of 5-HT in the serum, lung tissue, and colon tissue (P<0.01). The intervention with Huashi Baidu Granules lowered the levels of 5-HT in the serum (P<0.05) and the lung and colon tissue (P<0.01). Huashi Baidu Granules increased the diversity of gut microbiota, enriched beneficial bacteria such as Blautia, and inhibited pathogenic bacteria such as Desulfovibrio. ConclusionHuashi Baidu Granules significantly alleviate lung inflammation and pathological damage in mice infected with influenza A (H1N1) virus, reshape the intestinal microecology, and repair the intestinal barrier. It may exert cross-organ regulation by inhibiting the cascade of "gutogenic 5-HT release-circulatory transport-pulmonary 5-HT receptor activation-NLRP3 inflammasome assembly." It exerts therapeutic effects on influenza viral pneumonia through the gut-lung axis. This study provides a modern biological interpretation for the TCM theories of interior-exterior relationship between the lung and large intestine and simultaneous treatment of the lung and intestine.

Laccases (LACs), belonging to the multicopper oxidase family, are closely associated with various biological functions including lignin synthesis and responses to biotic and abiotic stresses in plants. However, few studies have reported the laccase genes in China rose (Rosa chinensis). Prickles cause difficulties to the management and harvest of R. chinensis and have become a trait concerned in the breeding. To investigate the expression patterns of laccase genes in roses, we cloned a laccase gene from an ancient variety R. chinensis 'Old Blush' and named it RcLAC15. The expression level of RcLAC15 in prickles was significantly higher than those in roots, stems, and leaves. Fifty-eight laccase genes were identified in the genome of R. chinensis, and bioinformatics analysis revealed that RcLAC15 was a homolog of AtLAC15, predicting that RcLAC15 was a stable hydrophilic protein without transmembrane structures. The recombinant expression vector pBI121-proRcLAC15:: GUS was introduced into Arabidopsis, and GUS staining results showed that the RcLAC15 promoter specifically drove GUS gene expression at the edges of Arabidopsis leaves. In summary, RcLAC15 is a gene specifically expressed in the prickles of R. chinensis. This discovery provides a reference for exploring the biological functions of laccase genes in the prickles of R. chinensis.

Journal of International Oncology 2024;51(4):204-209
Objective:To observe the serum levels of miR-497 and miR-383 in patients with esophageal cancer, and to analyze the clinical significance for esophageal cancer. Methods:Esophageal cancer patients admitted to Union Wuhan Red Cross Hospital from July 2018 to February 2020 were collected as the esophageal cancer group ( n=96), which were divided into a recurrence group ( n=29) and a non recurrence group ( n=67) based on follow-up results. The control group included healthy individuals who underwent physical examinations at this hospital during the corresponding period ( n=83), and the benign lesion group included patients with benign esophageal lesions who underwent treatment at same hospital during the corresponding period ( n=78). Real time fluorogenic quantitative PCR was applied to detect serum levels of miR-497 and miR-383 in each group; Pearson method was applied to analyze the correlation between serum levels of miR-497 and miR-383 in esophageal cancer patients; Relationship between serum levels of miR-497, miR-383 and clinical pathological characteristics in esophageal cancer patients was analyzed; Receiver operator characteristic (ROC) curve was plotted to analyze the efficacy of serum miR-497, miR-383, and their combination in predicting the prognosis of esophageal cancer patients. Results:The serum miR-497 levels in the control group, benign lesion group and the esophageal cancer group were 1.01±0.18, 0.86±0.15, 0.77±0.14, respectively, and the serum miR-383 levels were 1.02±0.21, 0.95±0.15, 0.84±0.15, respectively, with statistically significant differences ( F=52.59, P<0.001; F=25.12, P<0.001) ; There were statistically significant differences in serum miR-497 and miR-383 levels between any two of the three groups (all P<0.05). Pearson correlation analysis reveled that serum miR-497 level was positively correlated with miR-383 level in esophageal cancer patients ( r=0.46, P<0.001). There were statistically significant differences in serum miR-497 and miR-383 levels among esophageal cancer patients with different tumor diameters ( t=6.58, P<0.001; t=5.06, P<0.001), lymph node metastases ( t=5.55, P<0.001; t=4.63, P<0.001) and TNM stages ( t=5.00, P<0.001; t=2.75, P<0.001). The serum miR-497 (0.83±0.15 vs. 0.62±0.11, t=6.78, P<0.001) and miR-383 (0.91±0.16 vs. 0.67±0.13, t=7.12, P<0.001) levels in the non recurrence group were both higher than those in the recurrence group. ROC curve showed that the critical value of serum miR-497 for predicting the prognostic situation of esophageal cancer was 0.72, with a sensitivity of 68.97%, a specificity of 68.66%, and an area under the curve (AUC) of 0.756; The critical value of serum miR-383 was 0.84, with a sensitivity of 82.76%, a specificity of 67.16%, and an AUC of 0.827; The sensitivity of the combination of the two was 79.31%, the specificity was 85.07%, and the AUC was 0.899; The combination of the two had a better prediction effect than that of serum miR-497 ( Z=3.31, P=0.001) and miR-383 ( Z=2.51, P=0.012) alone. Conclusion:The serum levels of miR-497 and miR-383 in patients with esophageal cancer are lower than those in healthy individuals and patients with benign lesions, which are related to tumor diameter, lymph node metastasis, TNM stage, and prognosis. The combined detection of the two has certain predictive value for the prognosis of esophageal cancer.

Schizophrenia is a chronic disabling disease of unknown etiology. Its neuroimaging abnormalities have been confirmed by a large number of studies. However, when these structural abnormalities occur and how they develop over time are unclear. The relationship between structural abnormalities and antipsychotic treatment needs to be further explored. Longitudinal studies on the brain structure of clinical high risk for psychosis, first-episode schizophrenia and chronic schizophrenia, exploring the influence of drug therapy on them, are helpful to construct the trajectories of brain structure changes in the whole course of schizophrenia and dynamically track the brain structure changes, so as to further explore the pathophysiology of schizophrenia. This article reviews the longitudinal research progress of brain structure changes in patients with schizophrenia.

Objective:To explore differences of resting brain regional homogeneity (ReHo) between patients with major depressive disorder (MDD) and their siblings. Methods:From January to December 2013, the resting-state functional magnetic resonance imaging (fMRI) data of 87 patients with MDD and 21 healthy siblings were collected.DPABI v5.1 software was used to preprocess the resting-state fMRI data, and ReHo maps of each subject was obtained. A two-sample t-test was used to compare differences between the patients with MDD and their siblings in ReHo values throughout the brain. ReHo values within the significant brain regions were extracted out, and used to calculate Spearman correlation with the total score of 17-items Hamilton depression rating scale(HAMD-17) in the patients with MDD and their siblings respectively. The software of SPSS 20.0 was used for statistical analysis. Results:The patients with MDD exhibited lower ReHo values in the precuneus extending to the posterior cingulate cortex (PCu/PCC) compared with their siblings (cluster-size=126 voxel, cluster-level PFDR=0.033; MNI: x=-4, y=-58, z=38, t=4.30). ReHo values of the PCu/PCC in patient with MDD were positively correlated with the severity of depressive symptoms ( r=0.255, P=0.021). Conclusion:Compared with the siblings, local brain activity of the PCu/PCC in the patients with MDD was decreased, and related to the severity of depressive symptoms. It is helpful to further reveal the intrinsic neural mechanism of MDD.

Chinese Journal of Psychiatry 2023;56(3):227-231
A 51-year-old schizophrenic patient with long-term clozapine treatment developed intestinal obstruction combined with infection. Subsequently, the patient′s clozapine plasma concentration abnormally increased to 3 094.80 μg/L (reference range 350-600 μg/L), accompanied with pulmonary infection. Patient′s symptoms showed significant improvement after clozapine was discontinued, along with intravenous fluid reinfusion and antibiotic treatment. This current article also provided a literature review of 16 reported cases of infection with abnormally elevated clozapine plasma concentration. Clozapine plasma concentration and blood routine should be monitored in clozapine users, and adverse reactions should be treated in time. For clozapine users with infection but normal leukocyte count, multiple bacterial infection indicators such as the neutrophil proportion and C-reactive protein should begiven comprehensive consideration, and antibiotics should be used when appropriate. If clozapine plasma concentration is abnormally high, clozapine can be discontinued and intravenous fluid reinfusion can accelerate clozapine elimination. After the patient′s condition stabilizes, clozapine could be reused.

Chinese Journal of Psychiatry 2023;56(5):395-400
Major depressive disorder is a prevalent and burdensome mental illness that exhibits a high degree of heritability. Despite this, the underlying neural mechanisms that contribute to its development remain unclear. To enhance our comprehension of the pathology and susceptibility mechanisms of major depressive disorder, this study aims to compare the neuroimaging characteristics of patients diagnosed with major depressive disorder and their first-degree relatives. The objective of this study is to conduct a comprehensive analysis and synthesis of brain functional images of individuals diagnosed with major depressive disorder, including those in the acute and remission stages, as well as their first-degree relatives, utilizing various prevalent neuroimaging modalities. Additionally, this study aims to identify potential avenues for future research in this area. This study shows that abnormalities in local brain functional activity and functional interactions of the cortical-limbic-cerebellar circuits are critical in intrinsic neural mechanisms of depression and their first-degree relatives, and this circuits is also a potential target for antidepressants. Future studies are needed to further clarify the role of unbalanced cortical-limbic-cerebellar circuits in the disease process of depression.
