1.Clinical and prognostic analysis of nine cases of immune checkpoint inhibitor-related Stevens-Johnson syndrome/toxic epidermal necrolysis

Objective:To summarize clinical characteristics and prognosis of immune checkpoint inhibitor (ICI) -related Stevens-Johnson syndrome (SJS) /toxic epidermal necrolysis (TEN) .Methods:A retrospective analysis was conducted on patients diagnosed with ICI-related SJS/TEN in Zhongda Hospital, Southeast University from January 2018 to October 2023. Data on clinical manifestations, laboratory examinations, treatment and prognosis of the patients were analyzed.Results:A total of 9 patients were diagnosed with ICI-related SJS/TEN, including 8 males and 1 female, with the onset age ranging from 58 to 77 (67.56 ± 7.33) years. ICI were applied to all the 9 patients before onset of SJS/TEN. The latent period was 6 - 261 d, and the median duration was 76 d. All the patients presented with erythema as initial lesions, 3 cases with target lesions, 6 with blisters, and 5 cases with a positive Nikolsky's sign. Oral mucosal damage occurred in 7 cases, eye damage occurred in 6 cases, and urogenital damage occurred in 6 cases. All the 9 cases were treated with systematic glucocorticoids, 7 cases with intravenous immunoglobulin and 7 cases with antibiotics. Eight cases recovered and 1 case died. Among 6 patients followed for an average of 6.92 months, none achieved complete or partial remission, 3 died and 3 experienced disease progression.Conclusions:ICI-related SJS/TEN occurred with a relatively long latent period, and all the cases presented with erythema initially, with mucosal damage present in most of the cases. Management required discontinuation of ICI, and most patients recovered with corticosteroids and intravenous immunoglobulin, however, the prognosis for the primary malignancy remained poor.

3.Experimental research of adipogenic differentiative ability of adipose tissue-derived stromal cells in patients with type 2 diabetes mellitus

Objective To investiagte the adipogenic differentiative ability of adipose tissue-derived stromal cells(ADSCs) between the patients with type 2 diabetes mellitus(T2DM) and healthy persons.Methods The adipose tissues were taken from the adipose tissue in T2DM patients and healthy persons for separating and culturing ADSCs.The cells of third generation were taken for inoculation.The difference in cellular phenotype and growth speed were compared between the two groups.Adding adipogenesis inducing fluid,the adipogenic differentiative situation was observed in the two groups.The oil red O was added on 14 d for conducting the cell staining and observation.The oil red O was extracted by isopropanol,and the cellular absorbances were compared between two groups.Meanwhile,the expression of PPAR-γ,C/EBP-α and C/EBP-β on 14 d of adipogenic differentiation were compared between two groups by using qPCR method.Results The cellular phenotype and growth speed of ADSCs had no statisticat difference between T2DM patients and healthy persons.On 14 d of adipogenic differentiation,the oil red O absorbance value of AD-SCs in T2DM patients was significantly higher than that in the healthy persons,and the expression of PPAR-γ,C/EBP-α and C/EBP-β were significantly higher than those in the healthy persons.Conclusion The adipogenic differentiative ability of ADSCs in T2DM patients is obviously higher than that in healthy persons,which may be one of causes easy to be obese in T2DM patients.
4.Development of nursing needs scale for school-age children with acute leukemia during lumbar puncture

Objective To construct nursing needs scale for school-age children with acute leukemia (AL) during lumbar puncture by using Delphi technique.Methods A certain number of experts were chosen for consulting to form a consulting scale.Two-round consultations were conducted and the results were analyzed.Results The effective response rate of the experts' questionnaire for 2 rounds was 100% (17/17).The specialist group authority coefficient was 0.85.The coordination coefficients for 2 rounds were 0.437 and 0.516 respectively (P < 0.01).The nursing needs scale for school-age children with AL during lumbar puncture comprised of 5 first-level indicators and 30 second-level indicators.Conclusions This scale can scientifically and quickly evaluate the nursing needs of school-age children with AL during lumbar puncture,it provides a research tool to develop individualized interventions with demand as the direction.
5.Research progress of nursing interventions of lumbar puncture in children

This review has summarized the methods and contents of nursing interventions of lumbar puncture in children.The specific intervention strategies have been expounded from four aspects,including supportive psychotherapy,cognitive behavior intervention,music therapy intervention and comprehensive nursing intervention.The evaluations of intervention effects have been summarized and put forward the research shortcomings in current domestic and international research.We should pay attention to the need of children undergoing lunber puncture.This review has pointed out the direction of further research in the future.
6.Research progress of nursing interventions and effect evaluation in bowel preparation before colonoscopy

The specific intervention strategies expounded from four aspects, which included dietary intervention, psychological intervention, drug intervention and exercise intervention. The form and effect evaluation of interventions also were summarized. This review expects to provide evidence-based basis for clinical interventions in the future.
7.Parents′requirement survey of children with acute leukemia during the period of lumbar puncture

Objective To investigate the parents′ requirement of children with acute leukemia during the period of lumbar puncture. Methods A total of 105 parents of children with acute leukemia during the period of lumbar puncture were selected by purposive sampling to complete the questionnaire. Results The total of requirements was 3. 53, in which the top three were ensuring the children safe and comfortable (3. 92 ± 0. 54) score, the information acquisition (3. 79 ± 0. 65) score and receiving support and help (3. 68 ± 0. 71 ) score. The parents had different specific care requirements before and after lumbar puncture. Conclusions Doctors and nurses should pay more attention to the needs of parents, and actively take corresponding measures to meet their different requirements so as to help children to cooperate with lumbar puncture and prevent complications.
