1.Reliability and validity test of the speech,spatial qualities of hearing scale in the Chinese language

Objective To investigate the reliability and validity of the Chinese version of speech,spatial and qualities of hearing scale(SSQ).Methods The SSQ was translated into Chinese using the translation and back-translation method.Self-assessment with the scale was conducted for 50 adults with normal hearing and 28 adults with hearing impairment who had bilateral hearing aids fitted.Two weeks after the initial assessment,10 individuals with bilateral hearing aids fitted were randomly selected to be re-assessed using the same scale.The Cronbach's αcoefficients of the scores of each dimension and the total score of the scale was used to evaluate the internal consis-tency of the scale,and the correlation coefficient of the results of the two assessments was used to test the test-retest reliability of the scale.The expert evaluation method was adopted to test the content validity of the scale,and the correlation coefficients of the scores of each dimension and the total score were used to test the construct validity of the scale.The score differences of each item and each dimension between adults with normal hearing and adults with bilateral hearing aids fitted were compared to test the discriminant validity.Results Valid responses were obtained from all 78 participants.① The test-retest reliability coefficients of the total scale and the three dimensions were all greater than 0.750(P<0.01),indicating excellent test-retest reliability.The Cronbach's α coefficients of the total scale and the three dimensions were all greater than 0.700,suggesting excellent internal consistency.② All four ex-perts indicated that the content of each item in the three dimensions of the SSQ scale was in line with the measure-ment requirements,showing excellent content validity.The correlation coefficients between the scores of the three dimensions and the total scale were between 0.809 and 0.890,indicating a high degree of correlation.There were statistically significant differences in the scores of the three dimensions between the normal group and the group with bilateral hearing aids fitted(P<0.05),and the overall discriminant validity of the scale was excellent.Conclusion The Chinese version of the SSQ has good reliability and validity,and can be used as a subjective auditory assessment in clinical practice.

3.Efficiency of Rituximab for the treatment of the initial episode of steroid-sensitive nephrotic syndrome in children

Objective:To evaluate the efficacy and safety of Rituximab (RTX) combined with short-term use of glucocorticoids in the treatment of the initial episode of steroid-sensitive nephrotic syndrome (SSNS).Methods:A retrospective case summary.A total of 30 children with SSNS treated in the Department of Intrarenal Rheumatism and Immunology, Xuzhou Children′s Hospital from December 2021 to March 2023, were enrolled in this study.They were given a standard dose of RTX (375 mg/m 2) and glucocorticoids for a short term.The patients were followed up for 1 year, and the general condition, changes in CD19 + B lymphocytes expression after RTX treatment, the relapse rate, the median relapse-free survival and adverse reactions of RTX were recorded.Kaplan-Meier method was used to analyze the relapse-free survival time. Results:CD19 + B lymphocytes were depleted 2 weeks after RTX treatment, and the median time required for CD19 + B lymphocytes reconstitution was 5 months after RTX treatment.The 1-year relapse-free rate was 90.00%, and the median relapse-free survival was 11 months.The glucocorticoid discontinuation rate was 93.33% in 3 months after RTX treatment, and 100% in 6 and 12 months after RTX treatment.Adverse reactions included infusion reactions in 7 cases (23.33%), neutropenia/leukopenia in 5 cases (16.67%), hypoimmunoglobulinemia in 10 cases (33.33%), infections in 4 cases.One case was complicated with severe influenza A virus infection after CD19 + B lymphocytes reconstruction. Conclusions:A standard dose of RTX can significantly reduce the relapse frequency, maintain long-term remission of proteinuria, and reduce the dose of glucocorticoids in patients at the initial episode of SSNS.Thus, it is worthy of clinical promotion.
4.Analysis of the Main Chemical Components and Seasonal Differences of Mussels

OBJECTIVE To analyze the differences in chemical composition of mussels from Shengsi,Zhoushan in different har-vest months,and to compare the differences in the composition of mussels from different origins,so as to provide ideas for the compre-hensive utilization of its resources.METHODS Ultra-high performance liquid chromatography-triple quadrupole mass spectrometry(UHPLC-QqQ-MS)and spectrophotometry were used to analyze and evaluate the contents of nucleosides,amino acids,water-soluble proteins,total sugars and polysaccharides in mussels from Shengsi,Zhoushan in 12 months of the year and from 5 different origins;principal component analysis(PCA),partial least squares discriminant method(PLS-DA)and TOPSIS method were used to compre-hensively evaluate mussels.RESULTS A total of 16 amino acids and 11 nucleosides were detected in mussels.The average content of total amino acids in Shengsi mussels throughout the year was 4 851.74 μg·g-1,the average content of total nucleosides was 921.40 μg·g-1,and the average contents of water-soluble protein,polysaccharides,and total sugars were 51.32 mg·g-1,74.39 μg·g-1,and 417.22 mg·g-1,respectively.The nucleosides of Shengsi mussels were the highest in March and April,the ami-no acids and water-soluble proteins were the highest in April and May,and the polysaccharides and total sugars were the highest in Oc-tober and November.The PCA and PLS-DA results of the chemical components of different resources showed that there were great differences in Shengsi mussel samples harvested in different seasons.The entropy weight TOPSIS analysis showed that the comprehen-sive scores of Shengsi mussels in March and April(S7 and S8)were better than those in other areas,and the comprehensive score of Shengsi mussels in April(S8)was the highest,which was determined to be the best harvesting month.CONCLUSION The comparison results of multi-type resource chemistry show that there are great differences in the chemical compo-sition of Shengsi mussels in different harvesting seasons;the quality of Shengsi mussels is better than that of other areas;April is the best harvesting season,providing a reference for the quality evaluation and comprehensive development and utilization of mussels.

7.Research progress on congenital heart disease-related TBX5 mutations

Congenital heart diseases (CHDs) seriously affect human health, and genetic mutation is closely related to the occurrence of CHDs. In the process of assisted reproduction, the use of methods such as polar body biopsy, blastomere biopsy and blastocyst biopsy, combined with single-cell DNA amplification and high-throughput sequencing technology, can accurately detect pathogenic mutations associated with single-gene genetic diseases in pre-implantation embryos. T-box transcription factor 5 (TBX5) is an important transcription factor for heart and upper limb development. It is reported 227 TBX5 mutations are related to CHDs. Preimplantation genetic testing for monogenic disease (PGT-M) to detect CHD-related TBX5 mutations in embryos is of great significance for preventing the occurrence of CHDs. In this review, we systematically summarized the TBX5 mutation information, and analyzed the deleterious effects of TBX5 missense mutations based on familial genetic data, animal models, induced pluripotent stem cell disease models and prediction software to provide references for the detection of CHD-related TBX5 mutations using PGT-M.
8.A cohort study of atherogenic index of plasma and the risk of cardiovascular disease in community elderly hypertensive population of Kunshan area

Objective To investigate the relationship between plasma atherosclerosis index (AIP) and the risk of cardiovascular disease in community elderly hypertensive patients in Kunshan area. Methods A total of 21, 226 elderly hypertensive patients who underwent annual health examinations in community were enrolled in this study. Demographic information such as baseline age, gender, smoking history, hypertension, diabetes, and history of cardiovascular disease were collected. Patients' height, body weight, body mass index (BMI), waist circumference, blood pressure, fasting blood glucose, and blood lipids were measured. Lipid indicators including AIP, lipid composite index (LCI), Castelli risk index-Ⅰ (CRI-Ⅰ), and Castelli risk index-Ⅱ(CRI-Ⅱ) were recorded. The primary endpoint was the first occurrence of cardiovascular disease. The multivariate Cox proportional hazards model was used to explore the relationship between baseline AIP level and cardiovascular disease risk in hypertensive patients. Results After a median follow-up period of 5.62 (3.53, 7.16) years, 3, 179 patients occurred cardiovascular disease events. Cox regression analysis showed that a higher baseline AIP level was positively correlated with cardiovascular disease risk without adjusting for confounding factors (HR=1.16; 95%CI, 1.02 to 1.32). After further adjustment for age, gender, BMI, waist circumference, blood pressure, blood glucose, smoking history, and diabetes history, the AIP level remained closely related to cardiovascular disease risk. Restricted cubic spline analysis suggested an overall linear relationship between AIP and cardiovascular disease risk. 9.The prevalence and related risk factors of hyperuricemia in Bozidum Kinghiz township of Xinjiang Aksu region

Objective:To investigate the prevalence of hyperuricemia (HUA) in Bozidun Kirghiz township of Xinjiang Aksu region, and to explore the risk factors for the occurrence of HUA in the local area.Methods:A cross-sectional survey study was conducted by randomly selecting 9 villages in Bozidun Kirgiz Township by the whole-group sampling method and questionnaire were distributed to the households. The questionnaire included: demographic information, history of past illness, personal history, and all subjects were measured for height, weight, blood pressure, abdominal circumference, etc. The diagnostic of HUA if the serum uric acid (SUA) level >420 μmol/L in men or >360 μmol/L in women. The incidences of HUA in different age, sex, food type and life style behavior were analyzed. T test, non-parametric test and Chi-square test were used to analyze the differences among the groups, and logistic regression was used to analyze the risk factors. Results:①A total of 2 138 subjects were surveyed, among which 68 patients were with HUA, the prevalence of HUA in Bozidun Kirghiz township, Aksu region in the general population was 3.18%(68/2 138); the prevalence rate in men was 4.60%(45/978), 45 patients were identified; and the prevalence rate in women was 1.98%(23/1 160), 23 patients were identified. The peak age of HUA in male and female patients was 51~60 years old. ②The prevalence of HUA was lower in those who consumed dairy products ( χ2=6.91, P=0.017), nuts ( χ2=8.43, P=0.038) and eggs ( χ2=7.38, P=0.023), and lower in those who consumed more. Different intake of cereals ( χ2=0.87, P=0.647), meat( χ2=0.82, P=0.662), vegetables and fruits( χ2=5.22, P=0.073) had no effect on the prevalence of HUA.③In terms of different life behaviors, the prevalence of HUA in men who had been smoking was higher than those who had never smoked (57.78%, 28.89%, 13.33%, χ2=8.16, P=0.017). In the relationship between drinking and HUA, the prevalence rates of male always drinking, ever drinking and never drinking were 80.00%, 11.11% and 3.89%, respectively, the difference was statistically significant ( χ2=6.67, P=0.038). ④Multi-factor logistic regression analysis showed that high BMI, old age, high TG, increased Cr and increased WBC were risk factors for the occurrence of HUA [ OR(95% CI)=1.13(1.04, 1.23), 1.03(1.00,1.05),1.39(1.00, 1.93), 1.03(1.02, 1.05), 1.27(1.07, 1.49), all P<0.05]. Conclusion:The prevalence of HUA in Bozidun Kirgiz township in Aksu prefecture of Xinjiang is lower than that in other areas with continental climate. High BMI, old age, high TG, increased Cr and increased WBC count are risk factors for the development of HUA .
10.Association between variability of triglyceride glucose index and risk of type 2 diabetes

Objective:To explore the association between the variability of triglyceride glucose index (TyG) and the risk of type 2 diabetes mellitus(T2DM).Methods:This study was a retrospective cohort study. A total of 22 929 community-dwelling elderly (aged≥60 years) who received annual health check-ups in Kunshan city of Suzhou Municipality during 2014 to 2021 were enrolled in the study. Fasting triglycerides and blood glucose were measured during annual physical check-ups and the TyG was calculated, the standard deviation of TyG measurements in three consecutive physical check-ups was used as the indicatior of TyG long-term variability. According to the quartile of TyG long-term variability, the study subjects were divided into four groups, namely Q 1 (0-0.14), Q 2 (>0.14-0.22), Q 3 (>0.22-0.33), Q 4 (>0.33-1.90). The outcome variable was the occurrence of T2DM. The relationship between TyG variability and T2DM incidence was analyzed by multivariate Cox regression. Results:In the study cohort 11 518 (50.2%) were females and the mean age was (67.42±5.35) years. By the end of follow-up, 2 934 cases of new T2DM were diagnosed, with an oveall incidence rate of 12.8%. After adjusting for multiple confounders and average TyG, long-term variability of TyG was significantly associated with T2DM risk ( HR=1.83, 95% CI: 1.51-2.20). The risk of T2DM in Q 4 group was significantly higher than that in Q 1 group ( HR=1.33, 95% CI: 1.19-1.47). Kaplan-Meier survival curve showed that long-term variability of TyG was significantly correlated with the cumulative risk of T2DM incidence ( P<0.001). Conclusions:TyG variability is an independent risk factor for T2DM, suggesting that attention should be paid not only to specific time-point TyG levels but also to TyG fluctuation for early identification of T2DM risk.
