Background::In vitro fertilization (IVF) has emerged as a transformative solution for infertility. However, achieving favorable live-birth outcomes remains challenging. Current clinical IVF practices in IVF involve the collection of heterogeneous embryo data through diverse methods, including static images and temporal videos. However, traditional embryo selection methods, primarily reliant on visual inspection of morphology, exhibit variability and are contingent on the experience of practitioners. Therefore, an automated system that can evaluate heterogeneous embryo data to predict the final outcomes of live births is highly desirable. Methods::We employed artificial intelligence (AI) for embryo morphological grading, blastocyst embryo selection, aneuploidy prediction, and final live-birth outcome prediction. We developed and validated the AI models using multitask learning for embryo morphological assessment, including pronucleus type on day 1 and the number of blastomeres, asymmetry, and fragmentation of blastomeres on day 3, using 19,201 embryo photographs from 8271 patients. A neural network was trained on embryo and clinical metadata to identify good-quality embryos for implantation on day 3 or day 5, and predict live-birth outcomes. Additionally, a 3D convolutional neural network was trained on 418 time-lapse videos of preimplantation genetic testing (PGT)-based ploidy outcomes for the prediction of aneuploidy and consequent live-birth outcomes.Results::These two approaches enabled us to automatically assess the implantation potential. By combining embryo and maternal metrics in an ensemble AI model, we evaluated live-birth outcomes in a prospective cohort that achieved higher accuracy than experienced embryologists (46.1% vs. 30.7% on day 3, 55.0% vs. 40.7% on day 5). Our results demonstrate the potential for AI-based selection of embryos based on characteristics beyond the observational abilities of human clinicians (area under the curve: 0.769, 95% confidence interval: 0.709–0.820). These findings could potentially provide a noninvasive, high-throughput, and low-cost screening tool to facilitate embryo selection and achieve better outcomes. Conclusions::Our study underscores the AI model’s ability to provide interpretable evidence for clinicians in assisted reproduction, highlighting its potential as a noninvasive, efficient, and cost-effective tool for improved embryo selection and enhanced IVF outcomes. The convergence of cutting-edge technology and reproductive medicine has opened new avenues for addressing infertility challenges and optimizing IVF success rates.

BACKGROUND: Peritonitis is the common detective complications at the end-stage of renal failure patients during continuous ambulatory peritoneal dialysis (CAPD) treatment, which easily repeatedly occurred if not handled properly. Although the plumbing designs of peritoneal dialysis have done a lot of effort and improved in recent years, it still hard to avoid. OBJECTIVE: To explore the relative factors and treatments of infectious peritonitis complicated with CAPD. METHODS: Totally 115 cases received CAPD were selected, including 55 males and 60 females, aged (56.2?.7) years. 68 cases of which were suffered infectious peritonitis, aged (58.5?.3) years. All received a (30.3?.5)-month CAPD treatment. The relative factors such as the nutrition situation, whether complicated with diabetes mellitus or not, culture background, domestic environment, whether operating by special person and so on were investigated, and the relationship between those factors and infectious peritonitis were analyzed. The pathogen method was used to check pathogenic bacteria, and those patients were treated with irrigation of abdominal cavity and antibiotics in two ways, that is, systemic and intra-abdominalcavity. Hemodialysis instead of CAPD was used in those infectious peritonitis patients.RESULTS AND CONCLUSION: Those patients with good nutrition situation, higher culture background, better domestic environment, and operating by special person had low chances to infect peritonitis. And those patients with diabetes mellitus had high chances to infect peritonitis. Treatments of abdominal cavity and anti-infection in two ways-systemic and intra-abdominal cavities were effective to those infectious peritonitis patients. Gram-positive bacteria were the most common pathogen, which were susceptive to vancomycin and cefazolin. If the pathogen was gram-negative bacteria, etimicin sulfate, amikacin, and imipenem cilastatin could be selected. Infectious peritonitis complicated with CAPD affected by many factors. CAPD patients should be provided better nutrition and domestic situation. Also patients should operate in aseptic ways. Doctors should choose proper patients who can operate CAPD right.

AIM: To observe the effects of puerarin on blood lipid and expression of aorta laminin B_1 mRNA in streptozotocin-induced diabetic rats. METHODS: Diabetic nephropathy rats were induced by intraperitoneal injection of STZ, and the experimental rats were divided into normal control group, model group, and puerarin group. During and after the treatment for 12 weeks, the general state, blood suger(BS), triglyceride(TC), cholesterol, low density lipoprotein(LDL-C), high density lipoprotein(HDL-C), glycosylated hemoglobin(HbA1c) and glycosylated low density lipoprotein(G-LDL) were detected. Aorta alteration of tissue morphology was observed by H.E staining, and the expressions of laminin B1 mRNA were determined by in situ hybridization analysis. RESULTS: Diabetes mellitus and aorta lesion occurred in the two model groups. Puerarin could improve the general state, decrease the level of triglyceride(P

Objective To observe the effect of puerarin on the ICAM-1 gene expression in streptozotocin-Induced diabetic nephropathy rats. Methods Diabetic nephropathy rats were induced by intraperitoneal injection of STZ.Rats were allocated into normal control group, model group, and puerarin group. The puerarin treatment lasted for 16 weeks.During and after the treatment, the general health conditions, blood glucose levels, blood urea nitrogen, serum creatinine, urinary albumin excretion rate of 24-hours,and clearance rate of creatinine of the rats were observed. The ICAM-1mRNA expressions were also determined by in situ hybridization analysis in the kidney tissue of the rats. Results Diabetes mellitus and renal function lesion occurred in the model group and puerarin group.Puerarin could improve the general health of the rats, decrease blood urea nitrogen, serum creatinine, and urinary albumin excretion rate of 24-hours;and increase the clearance rate of creatinine. The ICAM-1mRNA expressions in the kidney were significantly inhibited by puerarin treatment. Conclusion Puerarin has a function of renal protection.

Objective To study the effects of rAAV2/1-Acrp30 on sICAM-1 and sVCAM-1 level as well as NF-?B expression in GK rats with diabetic arteriosclerosis so as to explore the effect of adiponectin on diabetic macroangiopathy. Methods A total number of 30 atherosclerotic GK rat models were randomly divided into three groups:① Model group one (M1):hind limb intramuscular injection of normal saline; ② Model group two (M2):hind limb intramuscular injection of vacuity virus rAAV2/1; ③ Treatment group (T):hind limb intramuscular injection of rAAV2/1-Acrp30 at a dose of 1?1012mg/L. After 8 weeks' treatment, the rats were killed, and serum sVCAM-1 and sICAM-1 level as well as aortic NF-?Bp65mRNA expression were measured in each group. Aortic NF-?Bp65mRNA expression was measured by RT-PCR.Results Compared with those in control model groups (M1 and M2),sVCAM-1 and sICAM-1 levels were decreased significantly in the treatment (T) group (P
