Objective To explore the effect of microRNA-22-3p (miR-22-3p) regulating the expression of Kruppel-like factor 6 (KLF6) on the cardiomyocyte-like differentiation of bone marrow mesenchymal stem cell (BMSC). Methods Rat BMSC was isolated and cultured,and the third-generation BMSC was divided into a control group,a 5-azacytidine(5-AZA)group,a mimics-NC group,a miR-22-3p mimics group,a miR-22-3p mimics+pcDNA group,and a miR-22-3p mimics+pcDNA-KLF6 group.Real-time fluorescent quantitative PCR (qRT-PCR) was carried out to determine the expression of miR-22-3p and KLF6 in cells.Immunofluorescence staining was employed to detect the expression of Desmin,cardiac troponin T (cTnT),and connexin 43 (Cx43).Western blotting was employed to determine the protein levels of cTnT,Cx43,Desmin,and KLF6,and flow cytometry to detect the apoptosis of BMSC.The targeting relationship between miR-22-3p and KLF6 was analyzed by dual luciferase reporter gene assay. Results Compared with the control group,5-AZA up-regulated the expression of miR-22-3p (q=7.971,P<0.001),Desmin (q=7.876,P<0.001),cTnT (q=10.272,P<0.001),and Cx43 (q=6.256,P<0.001),increased the apoptosis rate of BMSC (q=12.708,P<0.001),and down-regulated the mRNA (q=20.850,P<0.001) and protein (q=11.080,P<0.001) levels of KLF6.Compared with the 5-AZA group and the mimics-NC group,miR-22-3p mimics up-regulated the expression of miR-22-3p (q=3.591,P<0.001;q=11.650,P<0.001),Desmin (q=5.975,P<0.001;q=13.579,P<0.001),cTnT (q=7.133,P<0.001;q=17.548,P<0.001),and Cx43 (q=4.571,P=0.037;q=11.068,P<0.001),and down-regulated the mRNA (q=7.384,P<0.001;q=28.234,P<0.001) and protein (q=4.594,P=0.036;q=15.945,P<0.001) levels of KLF6.The apoptosis rate of miR-22-3p mimics group was lower than that of 5-AZA group (q=8.216,P<0.001).Compared with the miR-22-3p mimics+pcDNA group,miR-22-3p mimics+pcDNA-KLF6 up-regulated the mRNA(q=23.891,P<0.001) and protein(q=13.378,P<0.001)levels of KLF6,down-regulated the expression of Desmin (q=9.505,P<0.001),cTnT (q=10.985,P<0.001),and Cx43 (q=8.301,P<0.001),and increased the apoptosis rate (q=4.713,P=0.029).The dual luciferase reporter gene experiment demonstrated that KLF6 was a potential target gene of miR-22-3p. Conclusion MiR-22-3p promotes cardiomyocyte-like differentiation of BMSC by inhibiting the expression of KLF6.

4.Establishment of a discrinatory dissolution method for orlistat capule and evalution of the similarity of dissolution curue

Objective To establish a discriminative dissolution test method for orlistat capsules and evaluate the similarity of dissolution curves of 6 domestic enterprises and imported orlistat capsules.Methods The HPLC method was used,the chromatographic column was Thermo Hyersil GOLD C18(150 mm×4.6 mm,5 μm)column,the mobile phase was acetonitrile and water(85∶15),the flow rate was 1.0 mL·min-1,the detection wavelength was 195 nm,the column temperature was 30℃,and the injection volume was 20 μL.The effects of different concentrations of sodium dodecyl sulfate,different concentrations of sodium chloride and different pH dissolution media on the dissolution curve were investigated,and the best dissolution conditions were selected.The similarity of the dissolution curve was evaluated using the dissolution curve similarity factor method.Results Orlistat had good linear relationship within the range of 5.989-179.697 μg·mL-1(r=0.999 8),and its average recovery rate was 100.4%,with an RSD of 1.1%(n=9).The optimal dissolution conditions selected were as follows:the pH 6.0 phosphate buffer solution containing 1.0%sodium dodecyl sulfate and 0.5%sodium chloride was as dissolution medium;the conduct dissolution tests conducted under conditions of paddle method,75 r·min-1 and medium 1 000 mL.Only one domestic enterprise had a similar dissolution curve between the product and the reference formulation,while the other five enterprises had inconsistent dissolution behavior between the product and the reference formulation.Conclusion This measurement method can effectively distinguish the dissolution behavior of products from different enterprises,and has certain reference significance to evaluate the consistency of the quality and efficacy of orlistat capsules in China.
5.Exploration of the thinking and methods in treatment of cancer pain with acupuncture and moxibustion on the base of the fascia theory.

There is a commonality between jingjin (muscle region of meridian) and the fascial network for coordinating the balance in the body. The occurrence and the progression of tumor may disrupt the overall coordination between the fascial network and jingjin directly or indirectly, thereby, the impairment of this coordination may result in cancer pain. Rooted on the theory of overall balance of the fascial network, and combined with understanding of pain in jingjin theory, professor HUANG Jin-chang emphasizes the importance of "relaxing the knot" in treatment of cancer pain. It is recommended to select the fascia reaction point as the target point, in accordance with the principle of balance adjustment and apply various acupuncture and moxibustion therapies, such as Fu's subcutaneous needling, small-needle scalpel therapy, fire needling, and moxibustion.

6.Prevalence and risk factors of obesity in children with Diamond-Blackfan anemia.

OBJECTIVES:
To investigate the distribution of body mass index (BMI) and risk factors for obesity in children with Diamond-Blackfan Anemia (DBA).
METHODS:
The children with DBA who attended National Clinical Research Center for Blood Diseases, Institute of Hematology & Blood Diseases Hospital, Chinese Academy of Medical Sciences & Peking Union Medical College, from January 2003 to December 2020 were enrolled as subjects. The related clinical data and treatment regimens were recorded. The height and weight data measured within 1 week before or after follow-up time points were collected to calculate BMI. The risk factors for obesity were determined by multivariate regression analysis in children with DBA.
RESULTS:
A total of 129 children with DBA were enrolled, among whom there were 80 boys (62.0%) and 49 girls (38.0%), with a median age of 49 months (range 3-189 months). The prevalence rate of obesity was 14.7% (19/129). The multivariate logistic regression analysis showed that the absence of ribosomal protein gene mutation was closely associated with obesity in children with DBA (adjusted OR=3.63, 95%CI: 1.16-11.38, adjusted P=0.027). In children with glucocorticoid-dependent DBA, obesity was not associated with age of initiation of glucocorticoid therapy, duration of glucocorticoid therapy, and maintenance dose of glucocorticoids (P>0.05).
CONCLUSIONS
There is a high prevalence rate of obesity in children with DBA, and the absence of ribosomal protein gene mutation is closely associated with obesity in children with DBA.

7.Comparative analysis of metagenomic and 16S rDNA sequencing in gut microbiota of healthy elderly.

Objective: To explore the differences in subsequent analysis between metagenomic and 16Sr DNA sequencing in compositionally characterizing gut microbiota of healthy elderly. Methods: By using a panel study design, five monthly repeated measurements were performed among 76 healthy older people in Jinan City, Shandong Province. Their fecal samples were collected, and genomic DNA was extracted and analyzed through metagenomic and 16Sr DNA sequencing to compare the composition and diversity of gut microbiota. The correlation between species abundance and α diversity was analyzed by Pearson correlation analysis, and the correlation between species abundance and β diversity was determined by Procrustes analysis. Results: The age of 76 participants was (65.07±2.75), and the body mass index was (25.03±2.40) kg/m2. There were 38 males and 38 females. A total of 345 fecal samples were obtained from five monthly repeated measurements . Compared with 16S rDNA sequencing, metagenomic sequencing showed more annotated species at each level. The difference in the number of two sequencing species increased with the decrease of the level. Although there were significant differences in species richness between the two sequencing methods. Their species richness was highly correlated at both phylum (r=0.88, P<0.001) and genus (r=0.77, P<0.001) levels. Bacteroidetes and Firmicutes were the common dominant species. Gut microbiota diversity analysis further showed that there was a significantly positive correlation between α diversity (r=0.70, P<0.001) and β diversities (M2=0.84, P<0.05) in the two groups. Conclusion: The annotation efficiency of metagenomic sequencing is much higher than that of 16S rDNA sequencing. The two sequencing methods are consistent in phylum abundance as well as α diversity.

ObjectiveThe purpose of this study was to analyze the correlation between SUVmax on primary prostate lesion of two-phase 68Ga-PSMA-11 PET/CT and the clinicopathological features of patients with preliminary diagnosis of prostate cancer. MethodsPET/CT images of 19 patients with prostate cancer first diagnosed in our hospital from November 2017 to September 2020, aged 69.0±7.4 years, were collected. All patients underwent 68Ga-PSMA-11 PET/CT dual-phase examination, and radical resection of prostate cancer was performed within two weeks. Semi-quantitative parameters of 68Ga-PSMA-11 PET/CT on primary prostate cancer, SUVmax1 in the standard phase imaging, SUVmax2 in the delayed phase imaging and SUVmax3 (difference between SUVmax2 and SUVmax1), were measured by automatic segmentation algorithm method. Clinical and pathological data of each patient were collected. Mann-Whitney U test and ROC curve analysis were used to compare SUVmax and the clinicopathological characteristics of patients in two independent samples. P<0.05 was considered as statistically significant difference. ResultsThe differences of SUVmax1, SUVmax2 and SUVmax3 between GS≤7 group and >7 group were statistically significant(P<0.05), and the areas under the ROC curves were 0.845, 0.845 and 0.893, respectively. SUVmax1, SUVmax2 and SUVmax3 were significantly different between ISUP grade ≤2 group and >2 group (P<0.05), and the areas under the ROC curve were 0.773, 0.784 and 0.852, respectively. The difference of SUVmax3 between T stage ≤T2 group and >T2 group was statistically significant (P<0.05), while the difference of SUVmax1 and SUVmax2 between T stage ≤T2 group and >T2 group was not statistically significant (P>0.05). The difference of SUVmax3 between D’ Amico low-medium-risk group and high-risk group was statistically significant (P<0.05), while the difference of SUVmax1 and SUVmax2 between D’ Amico low-medium-risk group and high-risk group was not statistically significant (P>0.05). There was no significant difference in SUVmax1, SUVmax2 and SUVmax3 between total PSA ≤20 ng/mL group and >20 ng/mL group (P>0.05). SUVmax1, SUVmax2 and SUVmax3 were significantly different between positive nerve invasion group and negative group (P<0.05), and the areas under the ROC curves were 0.872, 0.923 and 0.974, respectively. ConclusionsSUVmax2 and SUVmax3 were positively correlated with multiple clinical and pathological features of prostate cancer. Compared with standard phase imaging, dual-phase imaging of 68Ga-PSMA-11 PET/CT may provide more favorable value in predicting the clinical and pathological features of prostate cancer patients before surgery.
10. Design mentality of establishing performance distribution system based on homogeneous subsidy base

Performance management is a key direction of the reform of the national medical and health system, and it is also an important starting point for promoting the development of hospital connotation. In view of the contradiction among the current hospital performance distribution system, this paper proposes a design mentality of performance distribution system based on homogenization subsidy base, and expounds the basic connotation and cognitive basis of homogenization subsidy base. The key elements of the design of the performance distribution system are discussed from four aspects: pre-adjusted total control, coefficientized personnel attendance, same performance appraisal, and homogenization subsidy base. At the same time, this paper also analyzes the characteristics of the performance distribution system based on the homogenization subsidy base. The application prospects of the performance distribution system are prospected from three aspects: the national medical reform requirements, the discipline development discipline and the system's own characteristics.
