1.Cross-sectional study on aesthetic abdominal wall morphology parameters in East Asian young females

Objective:To investigate the quantitative characteristics of abdominal wall aesthetics in young East Asian women.Methods:A cross-sectional survey study design was adopted. From June to August 2024, the Department of Plastic Surgery of Zhongshan Hospital, Fudan University disseminated volunteer recruitment announcements via multiple channels. Young female volunteers from East Asia aged 22-35 years were stratified enrolled according to the demographic ratios (30∶2∶1) of young women aged 22-35 years in China, Japan, and Repubic of Korea reported by authoritative organizations. These volunteers were unanimously evaluated to have an aesthetically "GOOD" abdominal wall via observations by four plastic surgery medical staff and four individuals without a medical background with meeting other inclusion and exclusion criteria. Three-dimensional (3D) photography was utilized to capture and reconstruct the 3D images of the abdominal walls. Key aesthetic landmarks of the abdominal wall were observed and measured. The main parameters include linearity, proportionality, angles, and areas of the umbilicus, abdominal wall depression, rectus abdominis protrusion, and abdominal wall contour. Statistical software (SPSS 26.0) was employed to calculate the average level of each parameter. Continuous variables conforming to the normal distribution were expressed as Mean±SD. Continuous variables that do not conform to the normal distribution were expressed as M( Q1,Q3), and compared using Mann-Whitney U test. Categorical variables were presented as frequencies and proportions (%). P<0.05 was considered statistically significant. Results:A total of 63 young East Asian women defined having an aesthetically "GOOD" abdominal wall were enrolled with an age of (26.7±7.4) years and body mass index of (19.6±3.9) kg/m 2. Umbilicus: 52.4%(33/63) umbilicus exhibited a vertical elliptical shape with a vertical diameter of 1.8(1.7, 2.1) cm, transverse diameter of 1.2 (1.0, 1.7) cm, depth of 0.4(0.3, 0.7) cm and projected area of 2.1(1.3, 3.0) cm 2; the presence of a bulge at the base of the umbilicus was relatively rare [6 cases (9.5%)], and no cases were observed protrusion beyond the abdominal wall and most umbilicus were at the lower 3/5 level of abdominal wall. Midline depression: the upper umbilical segment was significantly longer than the lower segment [7.8 (6.7, 8.4) cm vs. 3.6 (2.8, 4.1) cm, P<0.01], with an approximate upper-to-lower ratio of 2∶1; the upper segment was also wider [0.6 (0.5, 0.6) cm vs. 0.4 (0.4, 0.5) cm, P<0.01] and deeper than the lower segment [ 0.4(0.3, 0.6)cm vs. 0.1(0.1, 0.3) cm ( P<0.05)]. Semilunar lines: the left side measured a length of 14.5(10.6, 16.4) cm while right side measured 15.6(14.7, 16.1) cm ( P>0.05). Costal margin depression: the left side measured a depth of 0.3(0.2, 0.4) cm and the right side 0.3(0.2, 0.6) cm ( P>0.05) with infrasternal angle(upper abdominal angle) measuring 72.6°(59.8°, 89.6°). Iliac fossa depression: the depth of left side measured 1.0(0.8, 1.3) cm, while right side 1.0(0.8, 1.1) cm ( P>0.05); the left projected area was 16.4 (14.7, 18.3) cm 2 and right side 15.8(13.6, 17.8) cm 2 ( P>0.05). Rectus abdominis eminence: at costal margin level, the left side measured a width of 4.8 (4.1, 5.1) cm and the right side 4.7(4.1, 4.9) cm ( P>0.05); at umbilical level, the left side width was 4.1(3.7, 4.5) cm and the right side 3.8(3.2, 4.7) cm ( P>0.05); the overall projected area was 36.5(33.7, 38.5) cm 2. Outer contour lines: the length of left side was 33.6(31.5, 35.8) cm and the right side 35.7(31.6, 38.5) cm ( P>0.05); the waistline was located at the 2/5 level of the abdominal wall, approximately 2.4(1.7, 3.6) cm higher than the umbilicus; the outer contour of abdominal wall exhibited a morphology reminiscent of swan-neck vase. Conclusion:Using 3D photography technology, the body surface measurements of the abdominal wall in young East Asian women were conducted to quantify its aesthetic characteristics. The abdominal wall deemed aesthetically pleasing typically exhibits a "swan-neck vase" shape, characterized by relatively fixed landmark points, lines, planes, and proportional relationships. These results reflect the current positive and health-oriented aesthetic preferences prevalent among the general public.

3.Establishment and application of ultra-fast real-time PCR for Brucella detection

This study was aimed at establishing a method of ultra-fast quantitative PCR for Brucella detection.We used an exogenous recombinant plasmid as the internal reference and targeted the T4SS secretion system,an important Brucella viru-lence factor,to design specific primers and probes.The sensitivity,specificity,and repeatability of this method were evaluated,and a standard curve was constructed.The coincidence rate of detection findings with this method versus quantitative PCR was determined.This method markedly decreased the detection time to only 10 minutes.The standard curve demonstrated a good linear relationship(Y=-3.410 7x+38.357,R2=0.998 5)with a low minimum detection limit of 10 copies/μL.The method exhibited good specificity and did not specifically amplify several common clinical bacteria other than Brucella.The de-tection of three concentrations of positive plasmids yielded coefficients of variation(CVs)of 0.20%to 0.91%,thus demonstra-ting the method's excellent repeatability.Furthermore,140 clinical samples were analyzed concurrently with the fluorescence PCR method,which yielded a 100%compliance rate and consistent results.Our findings indicated that the Brucella ultra-fast quantitative PCR was ultrafast;had high sensitivity,high specificity,and good specificity;and can be used for the clinical de-tection of Brucella and emergency investigation of epidemics.Therefore,this method is valuable for the early diagnosis of Bru-cella.

The Golgi apparatus(GA),an essential membrane organelle in eukaryotes positioned between the endoplasmic reticulum and the plasma membrane,is responsible for transporting and modifying proteins and lipids generated by the endoplasmic reticulum.The homeostasis of the endoplasmic reticulum and GA plays a crucial role in regulating cellular life.When GA is unable to bear the load of protein or lipid processing and transportation,it enters a state of stress.Cells sense this and activate Golgi-related quality control mechanisms.By regulating the structure and function of GA,it provides important protection for the cell or induces programmed cell death,known as the GA stress response.Understanding the mechanisms and outcomes of cellular GA stress is of great significance for exploring Golgi dynamics and its impact on human diseases.This review summarizes the structure and function of GA under physiological conditions,the phenomenon of GA stress under pathological conditions,and the role of GA in infectious diseases,aiming to provide a basis for the clinical study of new strategies for the prevention and treatment of infectious diseases.

6.Clinical Study of MiR-125b-5p/HIF-1α Pathway in Involvement of Vitamin D Deficiency in Pathogenesis of Multiple Myeloma

Objective:To detect the serum levels of 25(OH)D,miR-125b-5p,hypoxia-inducible factor-1α(HIF-1α)and vascular endothelial growth factor A(VEGFA)in patients with multiple myeloma(MM),and explore the role of miR-125b-5p/HIF-1α pathway in the involvement of vitamin D deficiency in the pathogenesis of MM.Methods:Fifty three newly diagnosed/relapsed MM patients admitted to the department of hematology of our hospital from October 2021 to December 2023 were included.Meanwhile,25 healthy individuals matched in gender and age from our hospital's Health Management Center were selected as controls.The serum level of 25(OH)D was monitored by mass spectrometry,the serum level of miR-125b-5p was detected by real-time fluorescence quantitative PCR,and serum levels of HIF-1α and VEGFA were measured by enzyme-linked immunosorbent assay.The levels of 25(OH)D,miR-125b-5p,HIF-1α,and VEGFA were compared between the two groups.According to the level of 25(OH)D,the MM patients were divided into vitamin D deficiency group(<20 ng/ml)and vitamin D non-deficiency group(≥ 20 ng/ml),and the levels of miR-125b-5p,HIF-1α,and VEGFA were compared between the two groups.The correlations between 25(OH)D,miR-125b-5p,HIF-1α and VEGFA were analyzed.The receiver operating characteristic(ROC)curve analysis was used to determine the diagnostic value of25(OH)D combined with miR-125b-5p for newly diagnosed MM.Results:The level of 25(OH)D in MM patients was significantly lower than that in control group(P<0.01).There was no significant difference in 25(OH)D level between newly diagnosed and relapsed MM patients(P>0.05).Compared with the control group,the level of miR-125b-5p was significantly reduced in MM patients(P<0.01),while the levels of HIF-1α and VEGFA were significantly increased(both P<0.001).In MM patients,the miR-125b-5p level in the vitamin D deficiency group was significantly decreased than that in the non-deficiency group(P<0.01),while the levels of HIF-1 α and VEGFA were significantly increased(both P<0.05).In MM patients,25(OH)D was positively correlated with miR-125b-5p,while negatively correlated with HIF-1α and VEGFA(both P<0.05).Moreover,miR-125b-5p was negatively correlated with HIF-1α and VEGFA(both P<0.05).The area under the curve(AUC)for diagnosing MM with 25(OH)D,miR-125b-5p,and their combination were 0.699,0.751,and 0.791,respectively.Conclusion:The incidence of vitamin D deficiency is high in MM patients.Vitamin D deficiency may promote angiogenesis and participate in the occurrence and development of MM by downregulating miR-125b-5p and upregulating HIF-1α and VEGFA expression.
7.Design and application of individually portable oral treatment device field conditions in alpine regions

Objective To design an individually portable oral treatment device to solve the problems of oral diagnosis and treatment under field conditions in alpine regions.Methods The individually portable oral treatment device had a trolley box structure and consisted of an outer box,an inner framework and an operation panel.The outer box was made of low-density polyethylene material and formed by by one-time rotational moulding process;the inner framework integrated a plateau com-pressor,an independent negative-pressure compressor,an integrated control system for programmable logic controller(PLC),an individually portable respiratory synchronized pulsed oxygen supply module for plateau application;there were several curative devices equipped in the operation panel,including a 3-way syringe,a high-speed turbine handpiece,an electric variable-speed handpiece,a water control switch,a light curing machine and an ultrasonic dental cleaning handpiece.Trials were carried out with the test-phase prototype in alpine regions so as to verify the performance of the device.Results Trials proved that the prototype gained advantages in mobility,multifunctionality and pressure supply facilitating continuous operation of power gas source for oral diagnosis and treatment in alpine regions.Conclusion The device developed solves the problems in pressure insufficiency and instability,control system integration,portability and oxygen supply for medical staffs,improves the mobility of oral diagnosis and treatment in alpine regions and enhances the oral support service and equipment effectively.[Chinese Medical Equipment Journal,2025,46(1):108-113]
8.Medicinal properties and mechanisms of p-cymene with mild and warm nature based on deficiency-cold and deficiency-heat syndrome models.

This paper aims to study the effect of p-cymene on mice with deficiency-cold syndrome induced by hydrocortisone and deficiency-heat syndrome induced by dexamethasone and explore the medicinal properties and mechanism of p-cymene with mild and warm nature based on the dominant characteristics of the two-way applicable conditions of mild drugs. A total of 80 KM mice were randomly divided into blank group, deficiency-cold syndrome model group, deficiency-cold syndrome + ginseng group, and deficiency-cold syndrome + low-dose and high-dose p-cymene groups, as well as blank group, deficiency-heat syndrome model group, deficiency-heat syndrome + American ginseng group, and deficiency-heat syndrome + low-dose and high-dose p-cymene groups. Hydrocortisone and dexamethasone solution were intragastrically administered for 14 consecutive days to prepare deficiency-cold syndrome and deficiency-heat syndrome models. Except for the blank group and the model group intragastrically administered with normal saline, the other groups were intragastrically administrated with drugs for 14 days. The levels of cyclic adenosine monophosphate(cAMP), cyclic guanosine monophosphate(cGMP), triiodothyronine(T3), thyroxine(T4), total cholesterol(TC), triglyceride(TG), immunoglobin G(IgG), and immunoglobin M(IgM) in serum, as well as the activity of Na~+-K~+-ATPase in liver tissue were detected. The expression of transient receptor potential melastatin 8(TRPM8), transient receptor potential vanilloid 1(TRPV1), and uncoupling protein 1(UCP1) in brown adipose tissue of deficiency-cold syndrome model after intervention with p-cymene was studied. The results showed that p-cymene could effectively improve the levels of cAMP, cAMP/cGMP, TC, IgM, and IgG in serum and the activity of Na~+-K~+-ATPase in liver tissue of mice with deficiency-cold syndrome and reduce the content of cGMP. The effects on T3, T4, and TG were not statistically significant. At the same time, p-cymene could reduce the levels of cAMP, cAMP/cGMP, and T4 in serum and the activity of Na~+-K~+-ATPase in liver tissue of mice with deficiency-cold syndrome and increase the levels of cGMP, IgM, and IgG, and it had no effect on T3, TC, and TG. In addition, p-cymene could up-regulate the expression of TRPV1 and UCP1 in brown fat of mice with deficiency-cold syndrome and down-regulate the expression of TRPM8. In summary, p-cymene could significantly regulate the syndrome indexes of mice with deficiency-cold syndrome, and some indexes of mice with deficiency-heat syndrome could be improved, but the effects on lipid metabolism and energy metabolism indexes were not obvious, indicating that the regulation effect of p-cymene on deficiency-cold syndrome model was more prominent and that the medicinal properties of p-cymene were mild and warm. The regulation of TRPV1/TRPM8/UCP1 channel expression may be the molecular biological mechanism of p-cymene with mild and warm nature affecting the energy metabolism of the body.

9.Effects of Sishen Pills and its separated prescriptions on human intestinal flora based on in vitro fermentation model.

Sishen Pills and its separated prescriptions are classic prescriptions of traditional Chinese medicine to treat intestinal diseases. In this study, a high-performance liquid chromatography-electrospray ionization tandem mass spectrometry(HPLC-ESI-MS/MS) technology was used to identify the components of Sishen Pills, Ershen Pills, and Wuweizi Powder. The positive and negative ion sources of electrospray ionization were simultaneously collected by mass spectrometry. A total of 11 effective components were detected in Sishen Pills, with four effective components detected in Ershen Pills and eight effective components detected in Wuweizi Powder, respectively. To explore the effects of Sishen Pills and its separated prescriptions on the human intestinal flora, an in vitro anaerobic fermentation model was established, and the human intestinal flora was incubated with Sishen Pills, Ershen Pills, and Wuweizi Powder in vitro. The 16S rDNA sequencing technology was used to analyze the changes in the intestinal flora. The results showed that compared with the control group, Sishen Pills, and its separated prescriptions could decrease the intestinal flora abundance and increase the Shannon index after fermentation. The abundance of Bifidobacterium was significantly increased in the Sishen Pills and Ershen Pills groups. However, the abundance of Lactobacillus, Weissella, and Pediococcus was significantly increased in the Wuweizi Powder group. After fermentation for 12 h, the pH of the fermentation solution of three kinds of liquids with feces gradually decreased and was lower than that of the control group. The decreasing amplitude in the Wuweizi Powder group was the most obvious. The single-bacteria fermentation experiments further confirmed that Sishen Pills and Wuweizi Powder had inhibitory effects on Escherichia coli, Staphylococcus aureus, and Enterococcus faecalis, and the antibacterial activity of Wuweizi Powder was stronger than that of Sishen Pills. Both Sishen Pills and Ershen Pills could promote the growth of Lactobacillus brevis, and Ershen Pills could promote the growth of Bifidobacterium adolescentis. This study provided a more sufficient theoretical basis for the clinical application of Sishen Pills and its separated prescriptions.

Cervical spine health issues not only seriously affect patients' quality of life but also impose a heavy burden on the social healthcare system. Existing guidelines lack sufficient clinical guidance on lifestyle and work habits, such as exercise, posture, daily routine, and diet, making it difficult to meet practical needs. To address this, relying on the China Association of Chinese Medicine, Wangjing Hospital of China Academy of Chinese Medical Sciences took the lead and joined hands with more than ten institutions to form a multidisciplinary guideline development group. For the first time, the group developed the Guidelines for Cervical Spine Health Intervention based on evidence-based medicine methods, strictly following the standardized procedures outlined in the World Health Organization Handbook for Guideline Development and the Guiding Principles for the Formulation/Revision of Clinical Practice Guidelines in China (2022 Edition). This proposal systematically explains the methods and steps for developing the guideline, aiming to make the guideline development process scientific, standardized, and transparent.
