4.Comparison of the efficacy of LARS ligament and autogenous hamstring tendon plus high-strength suture in posterior cruciate ligament reconstruction

Objective To compare the clinical efficacy of ligament advanced reinforcement system(LARS)and autoge-nous hamstring tendon plus high-strength suture in arthroscopic reconstruction of posterior cruciate ligament(PCL).Methods A total of 96 patients with simple PCL injury treated with arthroscopic posterior cruciate ligament reconstructive surgery admit-ted to our hospital between August 2019 and December 2021 were selected for complete follow-up.There were 78 males and 18 females,40 cases of left leg and 56 cases of right leg,the aged from 20 to 56 years old with an average of(32.50±8.68)years old.The transplants were divided into two groups:LARS group(52 cases)and autogenous hamstring tendon plus high-strength suture group(44 cases).In the LARS group,there were 42 males and 10 females;with an average age of(31.84±8.62)years old;body mass index(BMI)was(24.73±2.29)kg·m-2;7 mm LARS was used to reconstruct PCL.In the autologous tendon group,there were 36 males and 8 females,with an average age of(33.06±8.99)years old,BMI was(23.52±2.16)kg·m-2,and the PCL was reconstructed with four strands of hamstring tendons and three pieces of Ethibond suture.All patients underwent functional rehabilitation guided exercise and were followed up regularly after surgery to objectively evaluate the stability of the knee joint by KT-1000 measurement of knee relaxation,and subjective evaluation of knee function by Lysholm score,Tegner score,and International Knee Documentation Council(IKDC)score.Data from preoperative,3,6,and 12 month follow-up were collected and analyzed by SPSS software to compare postoperative recovery and ligament relaxation between the two groups of patients.Results Ninety-six patients were followed up for 12 months.KT-1000 measurement of knee joint in autoge-nous tendon group and LARS group before operation[(10.73±1.46)points vs(10.55±1.53)points],6 months after operation[(3.02±0.75)points vs(2.35±0.60)points],12 months after operation[(3.77±1.76)points vs(2.44±0.60)points].There was significant difference between the two groups at 6 and 12 months after operation(P<0.05),but there was no significant dif-ference between the two groups at 3 months after operation(P>0.05).In the autogenous tendon group and LARS group,before operation and 12 months after operation,total Lysholm score[(40.6±16.8),(91.25±6.35)points vs(51.92±18.52),(92.35±5.30)points],Tegner score[(1.8±0.7),(5.8±0.6)points vs(1.7±0.8)、(5.7±0.7)points]and total IKDC score[(54.50±6.33),(83.90±3.93)points vs(54.40±4.24),(83.62±3.64)points],the differences were statistically significant(P<0.05),indicating that the knee function of the two groups was improved after surgery.At 3 and 6 months after operation in the autoge-nous tendon group and LARS group,the total Lysholm score[(65.86±11.54),(74.60±6.46)points vs(73.46±6.42),(86.73±4.62)points],Tegner score[(2.5±0.6),(3.5±0.5)points vs(4.3±0.7),(5.0±1.4)points],the total scores of IKDC[(55.78±2.68),(70.62±4.74)points vs(65.31±4.60),(79.71±2.93)points].The difference between two groups was statistically sig-nificant(P<0.05).The results showed that the function of the knee joint in the LARS group was better than that the autologous tendon group.However,at 12 months after the operation,there was no significant difference in the score of knee joint function between the two groups(P>0.05).The results showed that the stability of LARS group was better than that of autologous tendon group.Conclusion Both the autogenous hamstring tendon plus high-strength suture and LARS reconstruction can significantly improve the knee function and stability,with satisfactory postoperative results.Howervr the LARS provides superior postopera-tive stability.

MiR-192 is a kind of microRNA,which is widely expressed in various tissues,and plays an important role in biological processes such as cell differentiation,apoptosis,and proliferation,and is regulated by multiple factors,including transcription factors and epigenetic modifications.The expression of miR-192 varies in different tissues and diseases,and it plays a role in inhibiting tumor growth and promoting cell apoptosis and metastasis in tumors.MiR-192 is considered as a crucial tumor gene,which plays an important role in lung cancer,breast cancer,liver cancer,and colorectal cancer.The expression changes of miR-192 can be used as a potential diagnostic and prognostic markers,which provides certain reference value for the treatment and prevention of these tumors.This article explores the roles of miR-192 in tumor occurrence,progression,and treatment,revealing its potential as a target for tumor treatment.
6.Analysis of key genes in the development from colon adeno-ma to carcinoma through high-throughput RNA sequencing

Objective:To analyze and compare the difference of gene expression profiles in normal colon tissues,colon adenoma and carcinoma tissues by RNA sequencing technology,and re-veal the key genes and potential mechanisms in the development from colon adenoma to carcinoma.Methods:RNA sequencing analysis was carried out on normal colon tissues,colon adenomas and carcinoma tissues of the same patient,and differential genes that were significantly expressed in colon cancer and not significantly expressed in adenoma tissues were obtained,and the GO and KEGG function enrichment analysis was performed.Results:There are 4307 differential genes that are significantly expressed in colon cancer and not significantly expressed in adenoma.The GO and KEGG function enrichment analysis of these genes found that they were mainly enriched in bi-ological processes such as biological process regulation,cell process regulation,protein binding and cancer pathway,PI3K Akt signal pathway MAPK signal pathway.Conclusion:There are many genes involved in the development process from colon adenoma to carcinoma.These genes have the potential to become therapeutic targets for colorectal cancer,providing a new direction for fol-low-up research on colorectal cancer.
7.Effects of Didang Decoction-containing serum on high glucose-induced injury of rat glomerular endothelial cells

AIM To investigate the protective effects of Didang Decoction-containing serum on rat glomerular endothelial cells(RGECs)following high glucose injury.METHODS Rats were given distilled water or Didang Decoction by gavage to prepare the blank serum or Didang Decoction-containing serum.CCK8 method was used to screen the glucose concentration for the modeling and serum concentration of the drug.The RGECs were divided into the blank group,the model group,Didang Decoction group(10%Didang Decoction medicated serum),Dapagliflozin group(normal serum+2 μmol/L Dapagliflozin)and Didang Decoction+Dapagliflozin group(10%Didang Decoction medicated serum+2 μmol/L Dapagliflozin).24 hrs after the drug treatment,the RGECs had their mRNA and protein expressions of Nrf2,HO-1 and NQO-1 detected by RT-qPCR method and Western blot method;their Nrf2 fluorescence expression detected by immunofluorescence method;and their SOD activity and MDA level detected by colorimetric method.RESULTS Compared with the blank group,the model group displayed decreased mRNA and protein expressions of Nrf2,HO-1 and NQO-1 as well as the activity of SOD(P<0.01),and increased MDA level(P<0.01).Compared with the model group,the groups intervened with the drugs showed increased mRNA and protein expressions of Nrf2,HO-1 and NQO-1 as well as the activity of SOD(P<0.05,P<0.01),and decreased MDA level(P<0.01).CONCLUSION Didang Decoction-containing serum can protect RGECs from high glucose injury through activating the Nrf2 signaling pathway.
8.Rigid-body inverse dynamics modelling and analysis of 6RSS parallel bio-inspired masticatory robot

Objective To carry out rigid-body inverse dynamics modelling and analysis of a self-designed 6RSS parallel bio-inspired masticatory robot.Methods Firstly,the functions of kinematic variables including translational/rotational velocities and accelerations were derived for rigid-body inverse dynamics modelling.Secondly,the rigid-body inverse dynamics model was established with the Newton-Euler's law.Finally,the chewing motion trajectories of the oral health volunteers were tracked and numerical calculations were carried out in the case where the robot was subjected to a chewing reaction force.Results Numerical calculations showed that the driving torque and the constraint force of the robot peaked when the chewing reaction force was at its maximum.Conclusion The external force has a large impact on the inverse dynamics of the robot,and theoretical references are provided for the motion control and optimal design of the robot.[Chinese Medical Equipment Journal,2024,45(3):16-22]
9.Effect and significance of Wenyang Qudu formula on serum inflammatory factors and immune index in patients with severe infections

Objective To explore effect of Wenyang Qudu formula on serum inflammatory factors and immune index in patients with severe infections.Methods A total of 86 severe infection patients admitted to the Third People's Hospital of Henan Province from January to December 2023 were selected as the research subjects.According to the patient file order,odd numbers were the study group,and even numbers were the control group,with 43 cases in each group.The control group was treated with cefoperazone sulbactam sodium,while the study group was treated with Wenyang Qudu formula in addition to the control group[drug composition:Prepared aconite(first decocted)30 g,Poria cocos 30 g,White peony 15 g,Red peony 15 g,Stir fried atractylodes macrocephala 30 g,Dried ginger 9 g,Roasted licorice 9 g,Cassia twig 15 g,Semen lepidii 15 g,Dragon's bone 15 g,Raw oyster 15 g,Codonopsis pilosula 12 g,Angelica sinensis 12 g,Asarum 3 g,Schisandra chinensis 6 g,and Jujube 12 g].Brew in water,and took one dose daily,once in the morning and once in the evening,for a continuous period of 7 days.The differences in the scores of traditional Chinese medicine symptoms such as fever,dyspnea,frequent urination,urgency,and degree of sputum production,serum levels of interleukin-10(IL-10),C-reactive protein(CRP),eosinophils(EOS),and immune function indicators[immunoglobulin E(IgE),CD3+,CD4+,CD8+,CD4+/CD8+]were compared between two groups after treatment,and observed the occurrence of adverse reactions.Results After treatment,the traditional Chinese medicine symptom scores(fever,dyspnea,frequent urination and urgency,degree of sputum production),as well as IL-10,CRP,EOS levels,IgE,and CD8+ were significantly reduced in both groups compared to before treatment,CD3+,CD4+,and CD4+/CD8+ were significantly increased compared to before treatment.In addition,the study group had significantly lower scores of fever,dyspnea,frequent urination and urgency,degree of sputum production,IL-10,CRP,EOS levels,IgE,and CD8+ compared to the control group(fever score:1.36±0.30 vs.2.57±0.46,dyspnea score:1.22±0.31 vs.2.26±0.75,urinary frequency and urgency score:1.30±0.39 vs.2.33±0.82,degree of sputum production:1.19±0.77 vs.2.51±0.85,IL-10(ng/L):9.03±1.67 vs.10.51±2.40,CRP(mg/L):4.68±1.33 vs.7.82±2.53,EOS(×109/L):0.30±0.04 vs.0.46±0.10,IgE(mg/L):104.62±10.73 vs.135.68±14.64,CD8+:0.228±0.016 vs.0.258±0.020,all P<0.05],the levels of CD3+,CD4+,and CD4+/CD8+ were significantly higher than those in the control group(CD3+:0.636±0.044 vs.0.567±0.055,CD4+:0.537±0.054 vs.0.397±0.045,CD4+/CD8+:1.76±0.51 vs.0.55±0.39,all P<0.05].After treatment,it was discovered that the study group had not experienced any adverse reactions,while the control group had 1 case of nausea and vomiting and 1 case of chest tightness.There was no statistically significant difference in the incidence of adverse reactions between the study group and the control group[0(0/43)vs.0.05%(2/43),P>0.05].Conclusion The Wenyang Qudu formula can reduce the serum factor levels of IL-10,CRP,and EOS in critically infected patients,and improve immune function with good safety.
