1.Telocytes-Mediated Effects and Mechanisms of Anointing and Massage Therapy Using Oligopeptide-Herbal Medicine Composite Against Obesity in Rats

Objective To investigate the weight loss efficacy and mechanisms of oligopeptide-herbal medicine composite,and to provide new approaches for obesity treatment.Methods Twenty-three SPF female SD rats were randomly divided into control group(n=3)and modeling group(n=20).The control group was fed an ordinary diet for 6 consecutive weeks,and the modeling group was fed a high-fat diet for 6 consecutive weeks to establish a simple obesity rat model.After successful modeling,the modeling group was randomly divided into model group(n=10)and treatment group(n=10).The treatment group started anoint-and-massage therapy with oligopeptide-herbal medicine composite(3 g per rat per time,once a day,20 min each time),which was recorded as day 1.The control group and model group were not treated with oligopeptide-herbal medicine composite but received the same massage,and continued to be fed ordinary feed and high-fat feed respectively.Ten rats in the model group and ten in the treatment group were each divided into 3 groups,with 3,3,and 4 rats in each group.On days 3,6,and 9 after treatment,the rats in the three groups were weighed and cardiac blood collection was performed after isoflurane respiratory anesthesia.After euthanasia by cardiac bloodletting,abdominal subcutaneous adipose tissue(aSAT)and perirenal white adipose tissue(pWAT)samples were collected and weighed.Serum triglycerides(TG)and high-density lipoprotein cholesterol(HDL-C)levels were measured.Image J software was used to measure aSAT thickness and the diameter and area of perirenal white adipocytes and abdominal subcutaneous adipocytes.Immunofluorescence technique was used to observe the number of telocytes(TCs),cell junctions and exosomes per unit area of aSAT.Transmission electron microscopy was used to measure the length of telopodes(Tps)of dermal and subcutaneous TCs in abdominal skin,and to observe the distribution of exosome vesicles,rough endoplasmic reticulum,mitochondria,and cell junctions.Results Compared with the control group,the body weight of rats in the modeling group increased significantly(P<0.05).Compared with the control group,rats in the model group showed a significant increase in serum TG level,a significant decrease in HDL-C level,and a significant increase in pWAT mass(all P<0.05).Compared with the model group,the treatment group showed a significantly increased rate of body weight reduction,significantly decreased pWAT mass,significantly decreased serum TG level,and significantly increased HDL-C level(all P<0.05).Compared with the model group,the treatment group showed significant reductions in aSAT thickness,as well as cell diameter and area in both aSAT and pWAT(all P<0.05).Compared with the model group,the treatment group showed significantly increased number of TCs per unit area of aSAT,number of exosomes from TCs in aSAT,and Tps length(all P<0.05).The treatment group showed an increasing trend in the numbers of mitochondria,rough endoplasmic reticulum,and cell junctions in TCs.Conclusion Oligopeptide-herbal medicine composite applied via anoint-and-massage therapy effectively improves obesity-related symptoms such as dyslipidemia and fat accumulation in obese rats by regulating TCs and their intercellular communication.

3.Discrimination and Analysis of Authenticity of Dalbergiae Odoriferae Lignum in Market Based on Powder Color Digitalization

OBJECTIVE To establish a discriminant analysis of a mathematical model for distinguishing genuine and fake Dalbergiae Odoriferae Lignum based on the digitization of powder color. METHODS The 39 samples of Dalbergiae Odoriferae Lignum sold in the market were collected, and the L*,a*, b*, E*ab values of Dalbergiae Odoriferae Lignum powder were measured by using a color difference meter, and the authenticity of the Dalbergiae Odoriferae Lignum sold in the market was identified by using microscopic and thin layer methods, and a discriminant analysis model for the authenticity of Dalbergiae Odoriferae Lignum was established through statistical analysis. RESULTS A mathematical model for distinguishing true and false color difference of Dalbergiae Odoriferae Lignum was established by measuring the color difference value of Dalbergiae Odoriferae Lignum powder in the market. The results of distinguishing authenticity and falsehood were consistent with those of ordinary discrimination. CONCLUSION The method of rapid identification of true and false Dalbergiae Odoriferae Lignum by color difference value is effective, which can provide reference and basis for color difference identification and analysis of Chinese herbal medicines.
4.Determination of Chlorogenic Acid and Rutin in Tibetan Medicine Nettle by HPLC

Objective:To establish a method to determine the content of chlorogenic acid and rutin in nettle to assess the quality of nettle.Methods:An HPLC method was used with the following chromatographic conditions:CAPCELL PAK C18 column (250 mm × 4.6 mm, 5 μm) was used with the mixture of 0.4%phosphoric acid and methanol as the mobile phase with gradient elution , the de-tection wavelength was set at 340 nm, the flow rate was 1.0 ml· min-1 and the sample size was 10μl.Results:Chlorogenic acid with-in the range of 2.360-23.600μg · ml-1 showed a good linear relationship (r=0.999 9), and rutin within the range of 6.208-62.080μg · ml-1 showed a good linear relationship (r=0.999 9).The recovery of rutin and chlorogenic acid was 99.20% and 100.40%with RSD of 1.2%and 1.1%(n=6), respectively.Conclusion:The method is fast, effective, simple, accurate and reproducible , which can be used to quantitatively analyze chlorogenic acid and rutin in nettle .
5.Study on Processing Technology of Wine Gansu Salvia

Objective With fat-soluble ingredients and water-soluble components as indicators,to investigate the processing technology of wine Gansu Salvia by orthogonal test.Method With fat-soluble ingredients and water-soluble components determined by HPLC,the processing technology of wine Gansu Salvia was optimized by orthogonal test.Result The transfer rate of fat-soluble ingredients with water extraction was 2.4%,with little significance,while it had a significant impact on salvianolic acid B.The best process condition was 10% yellow rice wine,added 20% wine,fried in 80 ℃ for 4 min.Conclusion It was reasonable to choose the best process condition for wine Gansu Salvia.
6.Studies on the Quality of Traditional Chinese Medicinal Herbs Qianhu Growing in Gansu

A comparative study on the histological and morphological characteristics and the intrinsic quality of the roots of Huabei Qianhu(Peucedanum harry-smithii),Shaomao Bei Qiau hun (P.harry-smithii var.subglabrum ) and Baihua Qianhu (P.praeruptorum ) was carried out. Results revcaled that the former two Qianhu produced in Gansu are similar to Baihua Qian hu in their main ingredients.Thus the two Qianhu are worthy for further research and develo pm ent.At the same time, it was observed that Baihua Qianhu Produced in Gansu is of iuferior quality and the content of EtOH extraet of its root is slightly lower than that from elsewhere in China.
