1.Aldehyde Dehydrogenase 2 Gene Mutation May Reduce the Risk of Rupture of Intracranial Aneurysm in Chinese Han Population

Background:
and Purpose Ruptured intracranial aneurysms (RIA) are associated with a mortality rate of up to 40% in the Chinese population, highlighting the critical need for targeted treatment interventions for at-risk individuals. Although the impact of aldehyde dehydrogenase 2 (ALDH2) gene mutations on susceptibility to intracranial aneurysms (IA) is well documented, the potential connection between ALDH2 rs671 single-nucleotide polymorphism (SNP) and RIA remains unexplored. Given the increased prevalence of ALDH2 gene mutations among Chinese Han individuals, it is clinically relevant to investigate the link between ALDH2 rs671 SNP and IA rupture.
Methods:
A prospective study was conducted on 546 patients diagnosed with IA to investigate the association between ALDH2 rs671 SNP and the risk of IA rupture.
Results:
The ALDH2 rs671 SNP (ALDH2*2) was significantly more prevalent in patients with unruptured IA (UIA) than in those with RIA (32.56% vs. 18.58%, P=0.004). Multivariate logistic regression analysis revealed that people with the ALDH2 mutation (ALDH2*1/*2 and ALDH2*2/*2 gene type) had a significantly reduced odds ratio (OR=0.49; 95% confidence level [CI] 0.27–0.88; P=0.018) for RIAs. Age-specific subgroup analysis indicated that the ALDH2 mutation provided a stronger protective effect in individuals aged 60 years and above with IA compared to those under 60 years old (OR=0.38 vs. OR=0.52, both P<0.05).
Conclusion
The incidence of RIA was significantly higher in individuals with a normal ALDH2 gene (ALDH2*1/*1) than in those with an ALDH2 rs671 SNP (ALDH2*1/*2 or ALDH2*2/*2). ALDH2 rs671 SNP may serve as a protective factor against RIA in the Chinese Han population.

ObjectiveAlthough expression of the TEAD1 protein in preadipocytes has been established, its function remains unclear. In this study, we sought to detect transcripts of TEAD1 in chicken and to examine the effects of this protein on the proliferation, migration, apoptosis, and differentiation of immortalized chicken preadipocyte cell lines (ICP1). MethodsThe full-length sequence of the TEAD1 gene was cloned and the two transcripts were subjected to bioinformatics analysis. The subcellsular localization of TEAD1 transcripts was determined based on indirect immunofluorescence. The effects of TEAD1 transcripts overexpression on the proliferation of ICP1 cells were examined by RT-qPCR, CCK-8, and EdU assays; the effects of TEAD1 transcripts on ICP1 cells migration were examined based on the scratch test; and the effects of TEAD1 transcripts overexpression on ICP1 cells apoptosis were analyzed using apoptosis-Hoechst staining and RT-qPCR. The expression of TEAD1 transcripts in different tissues, cells lines, and ICP1 at different periods of differentiation was analyzed by RT-qPCR. The effects of TEAD1 transcripts overexpression on lipid droplet accumulation and adipogenic-related gene expression in ICP1 cells were analyzed based on Oil Red O and BODIPY staining, RT-qPCR, Western blot, and dual-luciferase reporter gene assays. Finally, the content of triglyceride (TG) was measured in TEAD1 overexpressed ICP1 cells. ResultsThe full-length TEAD1 was cloned and two TEAD1 transcripts were identified. The TEAD1-V1 protein was found to be localized primarily in the cell nucleus, whereas the TEAD1-V2 protein is localized in the cell cytoplasm and nucleus. The overexpression of both TEAD1-V1 and TEAD1-V2 significantly inhibited the proliferation of ICP1 cells. Whereas the overexpression of TEAD1-V1 promoted ICP1 cell migration, the overexpression of TEAD1-V2 had no significant effects on ICP1 migration; the overexpression of both TEAD1-V1 and TEAD1-V2 significantly promoted the apoptosis of ICP1 cells. We found that the different transcripts of TEAD1 have similar expression pattern in different tissues and cells lines. During induced preadipocyte differentiation, the expression of these genes initially declined, although subsequently increased. Overexpression of TEAD1-V1 promoted a significant reduction in lipid droplet formation and inhibited C/EBPα expression during the differentiation of ICP1 cells (P<0.05). However, the overexpression of TEAD1-V2 had no significant effect on lipid droplet accumulation or the expression of adipogenic-related proteins (P>0.05). Overexpression of TEAD1-V1 significantly decreased triglyceride content in ICP1 cells (P<0.05), while overexpression of TEAD1-V2 had no effect on triglyceride content in ICP1 cells (P>0.05). ConclusionIn this study, for the first time, identified two TEAD1 transcripts. Overexpressed transcripts TEAD1-V1 and TEAD1-V2 both inhibited the proliferation of chicken preadipocytes and promoted apoptosis of chicken preadipocytes. TEAD1-V1 inhibited the differentiation of preadipocytes and promoted the migration of preadipocytes, while TEAD1-V2 had no effect on the differentiation and migration of preadipocytes.

Objective To analyze the combined effect of body mass index(BMI)and age with cancer occurrence among a hypertensive population in Minhang District,Shanghai.Methods Participants of this study were 212 394 hypertensive patients without cancer in Minhang District,Shanghai,registered in the electronic health information system from 2007 to 2015.Age and BMI were included as smoothing functions in the generalized additive Cox proportional risk model.The bivariate response model was constructed to visualize results using surface plots and to comprehensively analyze the association of BMI and age with the risk of cancer occurrence.Results A total of 22 141 participants developed cancer by Dec 31,2018.The association between age and the risk of cancer incidence showed an overall linear trend while the association between BMI and the risk of cancer incidence showed an overall"U"shape.BMI at about 26 kg/m2 showed the lowest risk of cancer incidence.The risk of cancer occurrence increased with increasing age in people with different BMIs.The associations between BMI and the risk of cancer incidence were different at different age groups:there was no significant association between BMI and the risk of cancer incidence in the young people(20-44 years).While in the middle-aged and older people aged over 45 years,BMI was associated with the risk of cancer incidence in a"U"shape.The lowest risk of cancer incidence was around the BMI of 26 kg/m2.Conclusion BMI among the population with hypertension should be controlled in a reasonable range,especially in the middle-aged and older population,to prevent cancer occurrence.
7.Results of surveillance of foodborne diseases in Karamay City

Objective:
To analyze the epidemiological and etiological characteristics of foodborne diseases in Karamay City, Xinjiang Uygur Autonomous Region from 2018 to 2022, so as to provide insights into formulation of foodborne disease control measures.
Methods:
Active surveillance data of foodborne disease cases in Karamay City from 2018 to 2022 were collected from Foodborne Disease Monitoring and Reporting System. Population distribution, temporal distribution, spatial distribution, etiological characteristics and history of suspicious food exposure of foodborne diseases were descriptively analyzed.
Results:
A total of 715 cases with foodborne diseases were reported in Karamay City from 2018 to 2022, and 672 samples were detected, with a sampling rate of 93.99% and a positive rate of 4.17%. Positive rates of detected pathogens showed an increasing trend from 2018 to 2022 (P<0.05). There were 352 men (49.23%) and 363 women (50.77%), 272 cases (38.04%) were at ages of 20 to 39 years, and 225 cases (31.47%) were workers. The detection of foodborne diseases was concentrated during the period between November and December (312 cases, 43.64%), followed by the period between July and August (209 cases, 29.23%). The main clinical symptoms were digestive system symptoms (700 cases, 97.90%). Norovirus were detected (28 cases), while Salmonella, Shigella, Vibrio parahaemolyticus and diarrheagenic Escherichia coli were not detected. The suspicious food exposure was predominantly meat and meat products (211 cases, 29.51%), and the suspicious food exposure place was predominantly at food service establishments (350 cases, 48.95%).
Conclusions
Foodborne diseases were highly prevalent during the period between November and December and between July and August in Karamay City from 2018 to 2022. Residents at ages of 20 to 39 years and workers are high-risk populations. Meat and meat products are the main suspicious food products and dining places are the main exposed places.

Objective The purpose of this article is to summarize and review the current status of the construction of clinical research evaluation systems in domestic public hospitals,identify existing problems in the evaluation system,and propose development strategies and suggestions.Methods Retrieved relevant articles,dissertations and policies from the past five years(2018-2022),screened the titles,viewed the full texts of 52 selected papers and their references,and summarized them.Results The"five-only"indicators have long been an important indicator for evaluating clinical research in public hospitals,but in today's scientific research environment and policy environment,the"five-only"evaluation system has revealed its utilitarian draw-backs and gradually evolved into a hindrance to scientific research.It is urgent to break through the"five-only"orientation and establish a clinical research evaluation system oriented towards"transforming and applying transformation of scientific research achievements".Conclusion The evaluation system for clinical research should break the previous"five-only"evaluation model based on quantity-oriented scientific research evaluation.We can draw on the framework of the research output,influence,and environment indicators in the UK's REF Excellence Framework model,combine the American APT system and the Chinese STEM indicator dimensions,explore multi-outcome evaluation,integrate developmental indicators,and continuously improve the indica-tor system and application methods in practice to promote the development of clinical research in public hospitals.
9.Analysis on positioning error of different body position fixation methods guided by cone beam CT images in breast cancer radiotherapy

Objective To investigate the difference of translational error and rotational error in the ra-diotherapeutic positioning after breast cancer modified radical surgery by using the two fixation methods of the cervicothoracic thermoplastic membrane and body thermoplastic membrane based on conical beam CT (CBCT) images.Methods A total of 82 patients with radiotherapy after breast cancer modified radical sur-gery admitted and treated in this hospital from January 2022 to September 2023 were selected as the study subjects and divided into the cervicothoracic membrane group (using the cervicothoracic thermoplastic mem-brane,n=52) and body membrane group (using the body thermoplastic membrane,n=30) according to the radiotherapeutic fixation methods.All patients weekly conducted the CGCT scanning in the first time radio-therapy and radiotherapeutic period.The verification image conducted the retification with the planned image. The translational errors in the left and right direction (X),head and foot direction (Y) and abdominal and dor-sal direction (Z) were recorded,as well as the rotation errors around the axis under the two fixing methods,including the rotation error of the X-axis rotation direction (Rx),Y-axis rotation direction (Ry),and Z-axis rotation direction (Rz).The differences in positioning errors were compared between the two groups. Results There was no significant difference in X,Y,Z,Rx,Ry and Rx positioning error between the two groups after the first treatment,1-week treatment and 2-week treatment (P>0.05).After 3-week treatment,the positioning errors in X[0.21(0.12,0.27)mm vs. 0.22(0.20,0.35)mm],Y[0.20(0.11,0.24)mm vs. 0.25(0.16,0.37)mm],Z[0.15(0.08,0.25)mm vs. 0.20(0.15,0.29)mm],Rx[0.57(0.22,1.10)° vs. 1.00 (0.70,1.50)°],Ry[0.50(0.30,1.20)° vs. 1.10(0.60,1.40)°]and Rz[0.30(0.20,0.80)° vs. 0.90(0.40,1.50)°]in the cervicothoracic membrane group were smaller compared with the body membrane group (P<0.05).After 4-week treatment,the positioning errors in X[0.19(0.12,0.27)mm vs. 0.25(0.21,0.31)mm],Y[0.21(0.11,0.27)mm vs. 0.26(0.22,0.32)mm],Z[0.12(0.05,0.28)mm vs. 0.22(0.13,0.35)mm],Rx[0.80(0.49,1.10)°vs. 1.20(0.80,1.80)°],Ry[0.55(0.20,1.12)°vs. 1.10(0.80,1.30)°]and Rz[0.61 (0.29,1.10)°vs. 1.10(0.80,1.40)°]in the cervicothoracic membrane group were also smaller compared with the body membrane group (P<0.05).Conclusion The thecervicothoracic thermoplastic membrane could re-duce the positioning error in the breast cancer radiotherapy compared with the body thermoplastic membrane.
10.Efficacy of autofluorescence point-spectral analysis combined with the immune colloidal gold technique for the detection of ectopic microscopic parathyroid glands to guide surgery for secondary hyperparathyroidism

Objective To evaluate the intraoperative identification of ectopic parathyroid tissue in the central neck region using autofluorescence point-spectral analysis(AFPSA)combined with immune colloidal gold technique(ICGT),for guiding total parathyroidectomy(TPTX)or clean parathyroidectomy(CPTX)in the management of secondary hyperparathyroidism(SHPT).Methods Retrospectively collected and compared the clinical data of 64 patients with SHPT from October 2019 to June 2023.In the observation group,TPTX was performed as the initial procedure in 36 cases,followed by sampling of suspicious targets using AFPSA in the central neck area and subsequent detection through ICGT.CPTX was then conducted if a positive result was obtained.On the other hand,the control group consisted of 28 cases where only TPTX was performed without any additional tests during surgery.The surgical data,parathyroid hormone(PTH)levels,blood calcium levels,blood phosphorus levels,alkaline phosphatase(ALP)levels,regression of clinical symptoms,changes in parathyroid function and occurrence of hypocalcemia were compared between these two groups.Results In the observation group,there were 9 cases of AFPSA-ICGT positivity,including 2 left-sided cases,4 right-sided cases,and 3 thymic cases;among these posi-tive cases,there were a total of 10 locations with mildly hyperplastic or nonhyperplastic microscopic parathyroid tissue.The difference in the number of total parathyroid glands removed(including ectopic)between the two groups was statistically significant(P<0.05).At both 3 and 6 months postoperatively,ALP levels in the observation group were significantly lower than those in the control group(P<0.01 and P<0.001 respectively);at 6 months postoperatively,differences in PTH and blood phosphorus levels between the two groups were also statistically significant(P<0.05 and P<0.001 respectively).Joint bone pain and skin itching recurred in some patients within the control group at six months after surgery(P<0.05),whereas recurrence of SHPT was less frequent within the observation group compared to controls(P<0.05);however,no statistically significant differences were observed regarding postoperative hypoparathyroidism or hyperparathyroidism as well as hypocalcemia between either groups.Conclusion The AFPSA-ICGT intraoperative test can be utilized to guide surgery for SHPT,enabling accurate and efficient identification as well as safe targeting of parathyroid tissues that may not exhibit obvious hyperplasia in the central cervical region.
