The Janus kinase/signal transducers and activators of transcription (JAK-STAT) control natural killer (NK) cells development and cytotoxic functions, however, whether long non-coding RNAs (lncRNAs) are involved in this pathway remains unknown. We found that miR155HG was elevated in activated NK cells and promoted their proliferation and effector functions in both NK92 and induced-pluripotent stem cells (iPSCs)-derived NK (iPSC-NK) cells, without reliance on its derived miR-155 and micropeptide P155. Mechanistically, miR155HG bound to miR-6756 and relieved its repression of JAK3 expression, thereby promoting the JAK-STAT pathway and enhancing NK cell proliferation and function. Further investigations disclosed that upon cytokine stimulation, STAT3 directly interacts with miR155HG promoter and induces miR155HG transcription. Collectively, we identify a miR155HG-mediated positive feedback loop of the JAK-STAT signaling. Our study will also provide a power target regarding miR155HG for improving NK cell generation and effector function in the field of NK cell adoptive transfer therapy against cancer, especially iPSC-derived NK cells.

The clinical characteristics, diagnosis and treatment process of a patient with type Ⅰ sialidosis (ST-1) caused by a homozygous mutation in the NEU1 gene who was missed diagnosis for 5 years were retrospectively analyzed to improve the understanding of the disease. A 16-year-old female patient presented with episodic limb shaking for more than 5 years and single generalised tonic-clonic seizure. Electroencephalogram (EEG) tests conducted at external hospital did not show any abnormalities, and head magnetic resonance imaging (MRI) showed general normality. Multiple antiepileptic drugs could not control the attack and the symptoms gradually worsened. After admission, the patient was found to have symptoms of easy wrestling and decreased vision, as well as signs of nystagmus and ataxia. The reexamination of the EEG showed extensive spike-and-slow complexes, and the brain MRI showed cerebellar atrophy. Furthermore, the whole-exome gene testing revealed the c.544A>G homozygous mutation in the NEU1 gene, leading to the diagnosis of ST-1. Levetiracetam tablets and clonazepam were given to improve the patient′s symptoms. During the follow-up, sleep improved compared to before, and myoclonus was significantly reduced. Therefore, patients with recurrent myoclonus, ataxia, and visual impairment without cognitive impairment should be aware of the possibility of sialidosis. Genetic testing plays an important role in the diagnosis of sialidosis.

Objective To analyze the serotypes distribution characteristics of salmonella infection in infants below 2 years old in Dongguan.Methods Feces samples collected in children below 2 years old with diarrhea,caused by salmonella infection,were cultured for salmonella isolates,which were then identified for serotypes.The distribution characteristics of different serotypes of salmonella were analyzed.Results A total of 369 strains of salmonella were isolated,including 36 serotypes.The most common three serotypes were Salmonella typhimurium(S.ty),Salmonella stanley(S.st) and Salmonella enteritidis(S.en),which accounted for 68.7%.In all infants with diarrhea,there were 253 boys and 143 girls,and the male:female ratio was 1.77∶1.The average age of infants was (9.3±2.3) months.In male and female infants,the most serotype was S.ty,accounting for 62.3% and 53.3%,but the least serotypes in male and female infants were S.st(16.8%) and S.en(19.0%) respectively.S.ty was the most common serotype in different age groups.The least serotypes in low-age group(0-5.99 months) and middle-age group(6-11.99 months)were S.st(18.4%) and S.en(14.4%) respectively.In high-age group(12-24 months),the proportion of S.st and S.en were both 21.8%.The diversity of salmonella serotypes in male infants was higher than female infants,and with the increasing of age,the diversity was more obvious.Conclusion There could be unique serotype distribution characteristics of salmonella infection in infants with diarrhea,and the diversity of salmonella serotypes should be paid more attention.

Objective To observe the clinical efficacy of applying Sweat Reduction Formula (SRF) externally in the treatment of cancer-linked hyperhidrosis. Methods 45 tumor patients, who exhibited excessive perspiration, were selected and recruited randomly into an experimental group (Sweat Reduction Formula group) with 24 patients in it and a control group (Placebo group) with 21 patients in it. The experimental group was treated with SRF and the control group was administrated with placebo. Neuropad diagnostic patches were used to observe the period of time that required for any visual changes in color before and after medication. These observations were then matched with the commonly seen signs and symptoms scoring table, to evaluate the changes of symptoms and KPS. Results The color changing time of the experimental group was 14.45±3.91 min. and 19.51±5.30 min. before and after medications respectively. And the changing time in the control group was 13.49±4.96 min. After medication. The results were highly significant with P<0.05. There were also different levels of significant improvements in terms of spontaneous perspiration, night sweating, dry mouth, feverish sensation over hand-foot centers and body, aversion to cold etc after the treatment in the experimental group. Conclusion It would be more objective to evaluate the clinical efficacy of applying SRF on navel to treat cancer-linked hyperhidrosis with neuropad diagnostic patches.
