1.Applying a virtual reality-based working memory system in assessing executive functioning

Objective:To compare the effectiveness of a new virtual reality (VR) working memory test system with that of the traditional E-prime working memory test system in terms of evaluating executive function in cognitive rehabilitation.Methods:Two experienced rehabilitation therapists tested 30 healthy subjects. The average reaction time and accuracy of the traditional and the VR-based working memory test systems in completing the 2-back task were compared. The usability and user experience of the VR-based working memory test system were also evaluated using the system usability scale (SUS), a simulator discomfort scale (SSQ) and semi-structured interviews.Results:There was no significant difference in the average reaction time and accuracy of the 30 subjects between the two different tests. The SUS score of the VR working memory test was at the " Good-Excellent" level, and the SSQ scores demonstrated that no significant discomfort was observed. In the interviews the VR working memory test system was described as easy to operate and as having a strong sense of immersion.Conclusions:The VR-based working memory test system has good usability and user acceptance, and can provide subjects with an immersive and persona-lized test scenario. Therefore, it can be used as a tool for testing working memory.

3.Current status of hand hygiene practices among primary healthcare workers and its influencing factors

ObjectiveTo investigate the current status and influencing factors of hand hygiene practices among primary healthcare workers, and to provide a reference for improving hand hygiene behavior in primary healthcare institutions. MethodsA self-designed questionnaire was used to conduct a survey among medical staff of seven community health service centers in Jiading District, Shanghai, using a cluster random sampling method. The data collected were subjected to descriptive statistics, reliability and validity tests, correlation analysis, and structural equation model fitting. ResultsA statistical difference in hand hygiene compliance was found among medical staff with varying occupational types and educational levels (P<0.05), and medical staff in clinical departments exhibited higher levels of hand hygiene compliance (P<0.05). In the health belief model, among the core variables, healthcare workers’ perceived benefits, perceived barriers, and self-efficacy had a direct impact on hand hygiene behavior, with direct effect values of 0.341, -0.152, and 0.234, respectively. Meanwhile, cues to action and perceived severity of COVID-19 infection indirectly affected hand hygiene behavior, with an indirect effect value of 0.066 and 0.062, respectively. ConclusionHealthcare workers generally exhibit a high degree of hand hygiene compliance, but there are variations in hand hygiene scores among healthcare workers with different characteristics. Enhancing healthcare worders’ perception of benefits, action cues, and self-efficacy while reducing perceived barriers can effectively improve their hand hygiene behavior.

5.A single center study of colorectal cancer screening for Lynch syndrome.

OBJECTIVE:
To determine the ratio of deficient mismatch repair (dMMR) proteins and Lynch syndrome among patients undergoing colorectal cancer resection.
METHODS:
From June 2014 to May 2016, immunohistochemistry for mismatch repair proteins including mutL homolog 1 (MLH1), mutS homolog 2 (MSH2), mutS homolog 6 (MSH6) and PMS1 homolog 2 (PMS2) were carried out on 207 surgically resected specimens. Samples with lost expression of MMR proteins underwent genetic testing.
RESULTS:
Loss of expression of MMR proteins were found among 21 patients and accounted for 10.14% of the colorectal cancers. dMMR was more common in patients ≤50 years old, or with proximal tumor at splenic flexure and mucinous adenocarcinoma. Ten patients underwent genetic testing, with three pathogenic mutations (MSH6 c.3013C>T, MLH1 c.199G>A and a novel MSH6 c.584delT) and four ambiguous mutations identified. At least 1.4% of the colorectal cancers were diagnosed as Lynch syndrome.
CONCLUSION
Routine screening for Lynch syndrome among patients with colorectal cancer with MMR protein immunohistochemistry as preliminary screening method and MMR gene sequencing as diagnostic method is effective and feasible. It can reduce missed diagnosis of Lynch syndrome and bring lifelong benefit to patients and their families.

6.Effect of hyperbaric oxygen coupled with cisplatin on the apoptosis of Fadu cells in vitro

Objective To investigate the effect of hyperbaric oxygen (HBO) coupled with cisplatin on the apoptosis of human hypopharyngeal cancer Fadu cells in vitro and also to observe the effect of HBO exposure at different time points on the efficacy of cisplatin.Methods Human hypopharyngeal cancer Fadu cells for culture were bought from the Tumor Research Institute,Shanghai Jiaotong University.MTT assay was applied to observe the effect of different concentrations of cisplatin on the proliferation inhibition of Fadu cells in vitro.In accordance with different treatment,Fadu cells were divided into 6 groups:group C (the control group),group HBO (the HBO group),group CHT (the chemotherapy group),group HBO + CHT (the HBO + chemotherapy group),group CHT + HBO (the chemotherapy + HBO group and group IC (the HBO immediately combined with chemotherapy group).Cells of various groups were detected by flow cytometry 48 hours after experiment to observe changes in apoptosis,and adenosine triphosphate (ATP) changes in the cells of various groups were measured by ATP kit.Results Apoptosis rates of group C,HBO,CHT,HBO + CHT,CT + HBO and IC were respectively(2.00 ±0.17)%,(2.07 ±0.22)%,(8.73 ±0.74)%,(11.9 ±0.68)%,(9.92 ±0.27)% and(10.4 ±0.72)%.No statistical significance could be noticed in the apoptosis rates of group C and HBO(P >0.05).The apoptosis rates of group CHT,HBO + CHT,CHT + HBO and IC were all higher that those of group C and HBO,and statistical significance could be noted when comparisons were made between them(P <0.01).Experiments showed that the apoptosis rate of group HBO + CHT was higher than those of the other groups,with statistical significance(P < 0.05).Suppose the ATP level of group C was 100%,the ATP levels of other groups were respectively 98.7%,83.9%,61.3%,78.5% and 77.1%.There was statistical significance in the ATP levels of group C and HBO(P >0.05).However,the ATP levels of group CHT,HBO + CHT,CHT + HBO and IC were all lower than those of group C and HBO,also with statistical significance (P <0.01).The ATP level of group HBO + CHT was significantly lower than those of the other groups,also with statistical significance(P < 0.05).Conclusions Single HBO exposure had no effect on the apoptosis of hypopharyngeal cancer Fadu cells.HBO coupled with cisplatin could enhance the therapeutic efficacy of cisplatin,promote apoptosis,and the efficacy might be associated with the time points of the combined treatment of HBO.

8.Association of VKORC1 gene -1639G/A polymorphism with atrial fibrillation in ethnic Uygurs and Hans from Xinjiang.

OBJECTIVETo assess the association of VKORC1 gene -1639G/A polymorphism with atrial fibrillation (AF) in ethnic Uygurs and Hans from Xinjiang.
METHODSThe above polymorphism was detected among 100 Uygur and 102 Han AF patients and 103 Uygur and 111 Han subjects that have no AF with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method.
RESULTSA statistically significant difference was detected between the patient and control groups of Uygur origin in terms of genotypic and allelic frequencies (P<0.05). Logistic regression analysis also indicated the -1639G/A polymorphism as an independent risk factor for AF in Uygur population (OR=2.085, 95% CI: 1.067-4.072, P=0.031). No similar statistical difference was found between the patient and control groups of Han origin (P>0.05).
CONCLUSIONThe -1639G/A polymorphism of VKORC1 gene is associated with AF in the Uygur population but not in Hans.

9.Effect of hyperbaric oxygen on osteoporosis in rats with premature ovary failure

Objective To observe effect of hyperbaric oxygen (HBO) intervention on osteoporosis in rats with experimental premature ovary failureMethods Forty female Wister rats (with an age of 6 weeks) were randomly divided into 5 groups:the sham group,the deovary group,the estrin group,the HBO group and the hyperbaric air group,each consisting of 8 animals.With the exception of the sham group,all the other groups had ovariotomy bilaterally following anesthesia.One week later,the animals in the sham group and the deovary group were left there without any treatment,while the animals in the HBO group were exposed to HBO at a pressure of 0.1 MPa,one session a day for a succession of 10 days,then had one week interval every 10 sessions,with a total exposure time of 12 weeks.The animals in the hyperbaric air group were exposed to hyperbaric air at the same pressure and with the same treatment time and conditions.The animals in the estrin group received abdominal injection of estradiol benzoate once a week,for a succession of 12 weeks.After termination of the experiment,the animals of all the groups received lumbar vertebra and femora mineral density,bonc biomechanics and bone morphometry detection.Results After ovariotomy,lumbar vertebra mineral density was (0.087 8 ± 0.007 4) g/cm2 and femora mineral density was (0.114 1 ± 0.006 7) g/cm2.Lumbar vertebra biomechanics (216.32 ± 45.26) N and femora biomechanics (87.50 ± 4.01) N were significantly decreased,and statistical significance could be seen,as compared with those of the sham group [(381.58 ±28.75) N and(120 ± 16.26) N] (P < 0.05).However,the levels of the above indicators did not reduce considerably and there was no significant difference,as compared with those of the sham group (P > 0.05),but there was obviously significant difference in them,when it was compared with that of the hyperbaric air group (P > 0.05).Conclusions HBO could delay the loss of bone mineral density and the reduced property of bone biomechanics in ovarectomized rats,and it had certain protective effect on the occurrence of osteoporosis after menopause.
