Aging has emerged as a cutting edge and hotspot in global life science field， with anti-aging and geriatric disease prevention and treatment becoming critical issues urgently demanding solutions in international medical communities. In the face of the challenge of accelerating global population aging， in-depth exploration of aging mechanisms and the development of effective intervention strategies hold significant scientific and clinical value. This study supported by the national key research and development program of China， employed the essence-Qi-spirit theory of Qiluo doctrine as its guiding framework， focusing on the key scientific issue of the core traditional Chinese pathogenesis of aging， namely "depletion of kidney essence， deficiency of primordial Qi， and impairment of body and spirit". The treatment principle of "tonifying the kidney to replenish essence， harmonizing Yin and Yang， warming and invigorating primordial Qi， and nourishing the body and spirit" was established. Centered on holistic aging， systemic aging， and aging-related diseases， the research integrated multidisciplinary research approaches to construct multi-modal aging models and a multi-dimensional evaluation system， and it utilized multi-omics technologies to deeply analyze aging mechanisms. By systematically reviewing historical kidney-tonifying and anti-aging formulas and combining big data with artificial intelligence technologies， an information database of anti-aging traditional Chinese medicine substance was developed to reveal the differences and synergistic effects of various treatment methods and formulas on anti-aging. Based on this treatment method， the research integrated two millennia of kidney-tonifying medicinal experience to develop the innovative anti-aging traditional Chinese medicine， namely Bazhi Bushen capsules. It was validated that this capsule can delay holistic and systemic aging through multiple targets and mechanisms， thereby elucidating the scientific connotation of the essence-Qi-spirit theory of Qiluo doctrine in guiding anti-aging research from multiple dimensions and providing robust support for leveraging the advantages of traditional Chinese medicine to occupy the commanding heights of international anti-aging research.

ObjectiveExploring the formula rules of commonly used traditional Chinese medicines（TCMs） for epidemic treatment from the Qin and Han dynasties to the Qing dynasty through data mining， providing reference for the prevention and control of contemporary epidemics. MethodsThe articles on epidemic treatment in the electronic database of Chinese Medical Code V5.0 were systematically searched， and the contents such as source， dynasty， author， diagnosis， formula name， therapeutic method and efficacy， and composition of medicines from each article that met the inclusion criteria were extracted. Then， an Excel standardized database was established， and Python programs were used for data mining to summarize the frequency of commonly used medicines and perform hierarchical cluster analysis， Pearson correlation analysis， and association rule analysis. ResultsA total of 1 595 formulas were included， involving 558 TCMs. The efficacy of these medicines could be classified into two categories， namely， expeling pathogenic factors and reinforcing healthy Qi. According to the frequency deconstruction analysis， high-frequency medicines were mainly detoxification， Fu-organ dredging， aromatization and promoting blood circulation， followed by the medicines with the effect of treating the lungs， such as clearing the lungs and resolving phlegm， clearing heat and purging the lungs， relieving cough and asthma， and purging the lungs and relieving asthma. And the proportions of acrid-warm herbs and acrid-cold herbs varied in different periods. Hierarchical clustering and correlation analysis both suggested TCMs for expeling pathogenic factors and reinforcing healthy Qi often formed stable combinations with high association degrees. Association rule analysis showed that the core acrid-warm herb was mainly Ephedrae Herba， and the core acrid-cold herb was mainly Forsythiae Fructus， resulting in the core formulas of Maxing Shigantang and Yinqiaosan. ConclusionThroughout history， the prevention and control of epidemics have been based on the principle of "preserving healthy Qi and avoiding toxic Qi"， focusing on the treatment of the causes and characteristics of epidemics through detoxification， Fu-organ dredging， and aromatization， emphasizing the use of Rhei Radix et Rhizoma and other herbs to dredge Fu-organ， eliminate toxins and pathogens， and playing the role of actively intervene with symptomatic medication. And based on the external manifestations of the body's struggle between evil and righteousness， diagnose and treatment according to syndrome differentiation was performed.

Objective To investigate the effects and underlying mechanisms of a spray prepared from exosomes derived from M2 macrophages induced by interleukin-4 （IL-4） and tantalum particles （Ta） on the healing of pressure ulcers. Methods Bone marrow-derived macrophages were polarized into M2 macrophages using IL-4 or Ta, and exosomes （Exo-IL-4/Exo-Ta） were extracted. The regulatory effects of Exo-IL-4/Exo-Ta on M1 macrophage phenotypes and fibroblast matrix secretion were evaluated in vitro. Proteomic analysis was conducted to explore the biological processes and regulatory networks associated with Exo-Ta. A rat pressure ulcer model was used to assess the effects of Exo-IL-4/Exo-Ta spray on wound healing rate, inflammatory cell infiltration, and collagen deposition. Results In vitro, Exo-IL-4/Exo-Ta induced the polarization of M1 macrophages to M2 macrophages, reduced the secretion of pro-inflammatory factors, and promoted the expression of anti-inflammatory substances. Additionally, Exo-IL-4/Exo-Ta enhanced the production of collagen and fibronectin in fibroblasts. Proteomic analysis revealed that Exo-Ta primarily participated in biological processes such as energy metabolism and macromolecule biosynthesis. In vivo, Exo-IL-4/Exo-Ta spray accelerated wound healing, reduced inflammatory infiltration, and improved tissue remodeling in the rat pressure ulcer model. Conclusion Exosome sprays derived from M2 macrophages could accelerate pressure ulcer healing by modulating inflammation and promoting tissue regeneration, which demonstrated excellent clinical application potential.

Objective:To investigate the differences in the levels of inactive rhomboid protein (iRhom) 1, iRhom2, and tumor necrosis factor-α (TNF-α) in cervical cancer patients with different prognoses, and to analyze the predictive value of each index for patient's prognosis.Methods:A total of 90 cervical cancer patients who underwent extensive hysterectomy at the People's Hospital of Xinjiang Uygur Autonomous Region from June 2021 to June 2023 were selected as the study objects. Using a propensity score matching method with a caliper value of 0.02, 30 cases with normal cervical biopsy results and matched general clinical data were selected as the controls. Cervical cancer patients were divided into the good prognosis group ( n=69) and the poor prognosis group ( n=21) according to the prognosis. Western blotting was used to detect the protein levels of iRhom1, iRhom2, and TNF-α in tissue samples. The differences in iRhom1, iRhom2, and TNF-α protein levels between cervical cancer tissues, adjacent tissues, normal cervical tissues, and between the good and poor prognosis groups were compared. Multivariate logistic regression was used to analyze the influencing factors of prognosis in cervical cancer patients, and the receiver operator characteristic (ROC) curve was used to evaluate the predictive efficacy of each indicator for prognosis in cervical cancer patients. Results:The levels of iRhom1 protein in cervical cancer tissues, adjacent tissues, and normal cervical tissues were 0.80±0.11, 0.41±0.10, 0.40±0.07, respectively; those of iRhom2 were 0.81±0.12, 0.47±0.10, 0.46±0.05, respectively; and those of TNF-α were 1.15±0.12, 0.58±0.10, 0.56±0.07, respectively. There were statistically significant differences in the levels of iRhom1, iRhom2, and TNF-α protein among the three groups ( F=64.93, P<0.001; F=56.14, P<0.001; F=191.61, P<0.001). There were statistically significant differences in the levels of iRhom1, iRhom2 and TNF-α between cervical cancer tissues and adjacent tissues and normal cervical tissues (all P<0.05). In the poor prognosis group and the good prognosis group, the levels of iRhom1 protein in cervical cancer tissues were 0.90±0.12 and 0.77±0.10, respectively; those of iRhom2 were 0.90±0.10 and 0.79±0.09, respectively; and those of TNF-α were 1.29±0.13 and 1.06±0.10, respectively. There were statistically significant differences in iRhom1, iRhom2, and TNF-α protein levels between the two groups ( t=7.31, P<0.001; t=5.35, P<0.001; t=10.30, P<0.001). Multivariate logistic regression analysis showed that, iRhom1 ( OR=2.29, 95% CI: 1.77-3.71, P<0.001), iRhom2 ( OR=1.51, 95% CI: 1.10-2.71, P<0.001), and TNF-α ( OR=2.10, 95% CI: 1.90-4.44, P<0.001) were all independent influencing factors of the prognosis of cervical cancer patients. The ROC curve indicated that iRhom1 (AUC=0.88, 95% CI: 0.80-0.97), iRhom2 (AUC=0.83, 95% CI: 0.73-0.94), and TNF-α (AUC=0.80, 95% CI: 0.65-0.94) alone and in combination (AUC=0.97, 95% CI: 0.93-1.00) could predict prognosis of cervical cancer patients. Conclusions:The levels of iRhom1, iRhom2 and TNF-α proteins in cervical cancer tissues are higher than those in adjacent tissues and normal cervical tissues, and the levels of these three indexes in cervical cancer tissues with poor prognosis are significantly higher than those in cervical cancer tissues with good prognosis. The levels of iRhom1, iRhom2 and TNF-α protein are all independent factors influencing the prognosis of cervical cancer patients, and the three indicators alone or in combination can predict the prognosis of cervical cancer patients.

BACKGROUND:Croton cream can activate ERK pathways and have anti-apoptotic effects on neuronal cells.It is not clear whether it synergistically exerts anti-apoptotic effects by inhibiting the activation of JNK and p38 pathways. OBJECTIVE:To explore the effects and mechanisms of croton cream on neuronal damage and apoptosis in the ischemic cortex of rats with cerebral ischemia-reperfusion injury. METHODS:(1)Ninety Sprague-Dawley rats were randomly divided into sham operation group,model group,croton cream low-dose group,croton cream medium-dose group,croton cream high-dose group and nimodipine group,with 15 rats in each group.Except for the sham operation group,animal models of middle cerebral artery occlusion were prepared in rats by the thread method.Rats in the three croton cream groups were given 20,40,and 60 mg/kg croton cream,respectively.Rats in the sham operation and model groups were given the same amount of normal saline,once a day,for 7 consecutive days.The optimal concentration of croton cream,namely the high dose of croton cream,was selected based on neurological deficit score,TTC staining,brain tissue water content,hematoxylin-eosin staining and Nissl staining.(2)Another 120 Sprague-Dawley rats were randomly divided into sham operation group,model group,croton cream group,JNK inhibitor group,croton cream+JNK inhibitor group,p38 MAPK inhibitor group,croton cream+p38 MAPK inhibitor group,and nimodipine group,with 15 rats in each group.Animal models of middle cerebral artery occlusion were prepared using the thread method in all the groups except in the sham operation group.Thirty minutes before modeling,10 μL of SP600125(JNK inhibitor)and 10 μL of SB203580(p38 MAPK inhibitor)were injected into the lateral ventricle of the rats,respectively.Rats in croton cream groups were intragastrically given 60 mg/kg croton cream.Seven days later,the JNK/p38 MAPK signaling pathway,apoptosis-related proteins and cell apoptosis were detected by western blot,TUNEL staining and flow cytometry,respectively. RESULTS AND CONCLUSION:(1)Compared with the sham operation group,neurological deficit score,cerebral water content,cerebral infarction volume and apoptosis rate were significantly increased in the model group(P<0.05),where nerve cells showed scattered distribution.Compared with the model group,neurological deficit score,water content of brain tissue and cerebral infarction volume were significantly decreased in the croton cream medium-dose group,high-dose group and nimodipine group(P<0.05),and the pathological morphology of nerve cells was significantly improved.(2)Compared with the JNK inhibitor group,p-JNK/JNK,p-p38/p38 and Bax expressions in rat brain tissue and the apoptotic rate were significantly decreased in the croton cream+inhibitor groups(P<0.05),while the expression of and Bcl-2 was significantly increased(P<0.05).To conclude,croton cream may inhibit the activation of JNK/p38 MAPK signaling pathway and reduce neuronal apoptosis to achieve neuroprotective effects in rats with cerebral ischemia-reperfusion injury.
