1.Analysis and Verification of Potential Ferroptosis Key Genes in Curcumin Treatment of Osteoarthritis

Objective To explore the molecular mechanism of curcurin in the treatment of Osteoarthritic(OA)based on ferroptosis gene through bioinformatics.Methods Through Gene Expression Omnibus(GEO)retrieval,high-throughput sequencing data of curcumin in the treatment of osteoarthritis were downloaded.Principal component analysis was used to investigate the changes of principal components before and after curcumin treatment.limma algorithm was used to investigate the transcriptome changes before and after curcumin treatment.The change of signal pathway before and after curcumin treatment was investigated by gene ontology enrichment algorithm.GeneMANIA online tool was used to construct protein-protein interaction network and screen hub genes.The effect of curcumin therapy on hub gene expression was verified in vitro by fluorescence quantitative PCR.Results After curcumin treatment,the transcriptome expression and signaling pathway activity of chondrocytes were significantly changed(P<0.05).Protein-protein interaction network showed that GCLC,HMOX1,FTL,FTH1 and SLC7A11 were ferroptosis hub genes.The results of ferrous ion fluorescence probe detection and intracellular Reactive oxygen species(ROS)detection showed that IL-1βsignificantly induced inflammation and increased the accumulation of Fe2+and ROS in chondrocytes compared with the control group.Curcumin treatment significantly alleviated IL-1(3-induced inflammation.The contents of Fe2+and ROS in chondrocytes were significantly decreased.The results of quantitative PCR and western blot showed that curcumin treatment significantly changed the expression level of hub gene(P<0.05).Conclusion Curcumin may affect the ferroptosis process of chondrocytes by mediating up-regulation of GCLC and SLC7A11 genes and down-regulation of HMOX1,FTL and FTH1 genes,so as to have therapeutic effect on osteoarthritis.

3.Retroperitoneal iaparoscopic ureterolithotomy for impacted upper-ureteral calculi: a report of 42 cases

The clinical data of 42 patients with impacted upper-ureteral calculi undergoing retroperitoneal laparoscopicureterolithotomy(RPLU)from January 2008toDecember 2010 were retrospectively analyzed.RPLU was successfully performed in 40 cases,and simultaneous laparoscopic electrocoagulation ablation was also performed in 11 cases complicated with ureteral inflammatory polyps.No massive bleeding occurred during the operations.The stone-free rate 3 d after operation was 100%,and no stone left as reviewed by kidney ureter bladder X-ray (KUB) and B-ultrasonography.During the follow-up of 3 to 24 months (mean 11 months),40 cases showed no stone recurrence and no ureteral stricture in KUB and B-ultrasonography.The hydronephrosis of all RPLU patients relieved.Results indicate that RPLU is a safe and effective minimally invasive operation for treatment of impacted upper-ureteral calculi.

5.Clinical analysis of the blunt renal trauma

Objective:to summarize the experiences of diagnopsis and treatment in the blunt renal trauma,for increasing diag-nostical level and therapeutical results of the renal trauma. Methods:The clinical data of 124 patients with the blunt renal trauma were analysed retrospectively. Results:of 124 cases of the blunt renal trauma,96 were underwent conservative man- agemen(t77.4%),2 were received supra-selectively renal arteral embolization(1.6%),while of 24 cases were administered with surgery(21.4%),14 associated with intra-abdominal injuries. At last,2 died of the renal renal injuries,accounting for 1.6%. Conclusion:Ultrasound type B plus CT scaning are important and effective techniques for diagnosis of the kindy injury. The high-dose intravenous pyelogram remains an excellent tool for assessing renal trauma.
6.Effects of TURP plus TUVP for patients with BPH

Objectives:To investigate the effects of transurethral electrovaporization of the prostate(TUVP)plus transurethral re-section of the prostate(TURP)for patients with benigh prostate hyperplasia(BPH).Methods:The technique of TUVP plus TURP was employed to treat 114 cases of BPH.Results:All patients underwent successful TUVP and TURP procedure intraoperatively,and recovered very well postoperatively.No serious complications were noted.Conclusions:TUVP plus TURP is a feasible and safe technique for patients with BPH.
