1.Research advances in methods for determining the activity of molecular glue drugs

Molecular glue drugs exert pharmacological effects by inducing or stabilizing protein-protein interactions. Evaluating their activity not only requires assessing their binding to proteins, but also involves the evaluation of multiple aspects such as ternary complex formation, interface stability, cooperativity, and intracellular function. This review summarizes the commonly used methods and applications for evaluating the activity of molecular glues from three aspects: in vitro quantitative detection, cellular validation, and theoretical calculations and simulations; and proposes a hierarchical approach to activity evaluation based on the actual needs for efficiency in drug development, aiming to provide some reference for the study of molecular glue drug activity and the selection of evaluation methods.
2.Relationship between mammographic features and molecular subtypes in invasive breast cancer patients

Objective: To assess the relationship between mammographic features and molecular subtypes in invasive breast cancer pa-tients. Methods: We performed a retrospective review of mammographic images from a total of 182 histopathologically diagnosed-breast cancer patients treated in Tianjin Medical University General Hospital and Tianjin Medical University Cancer Institute and Hospi-tal between January 2016 and March 2016. Mammographic features were assessed according to the Breast Imaging Reporting and Da-ta System(BI-RADS). Molecular subtypes were classified according to the St Gallen International Expert Consensus. Statistical analysis was performed to assess the correlation between mammographic features and molecular subtypes of breast cancer. Results: There was a relationship between mammographic calcifications and molecular subtypes of invasive breast cancer (V=0.221, P<0.05). In com-parison with Luminal A (12/20), Luminal B (80/132), and HER-2 enriched (4/10) breast cancers, most triple-negative breast cancers (18/20) were non-calcified lesions (P<0.05). The proportion of spiculated masses was higher in Luminal breast cancer (30/85) than in non-Luminal breast cancer (1/16) (P<0.05). Conclusions: There is a relationship between mammographic features and molecular subtypes of invasive breast cancer.
3.Analysis of microsatellite polymorphism in Candida albicans strains isolated from patients with vulvo-vaginal candidiasis in Guizhou region

Objective To investigate the genotype distribution of microsatellite locus CAI among Candida albicans ( C.albicans ) strains and to evaluate its relationship with the epidemic of vulvovaginal candidiasis ( VVC) in Guizhou region .Methods Ninety independent C.albicans strains isolated from pa-tients with VVC in Guizhou were investigated based on single-strand conformation polymorphisms ( SSCP ) and GeneScan analysis .The genotypes of C.albicans strains were identified by microsatellite locus CAI pol-ymorphism analysis .The gene polymorphism and the cluster of C.albicans strains were analyzed by using software SPSS 19.0.A logistic regression model was used to analyze the relationship between genotype distri -bution of CAI microsatellite among C.albicans strains and VVC infection .Results Twenty-seven distinct CAI genotypes with various patterns were identified from 90 C.albicans strains by GeneScan analysis .Clus-ter analysis showed that the C.albicans strains were classified into three clusters ( ClusterⅠto Cluster Ⅲ) . Three predominant genotypes including 30-45, 32-46 and 30-46 and other 7 highly similar genotypes be-longed to clusterⅡthat accounted for 70.0%(63 strains) in all strains.The odds ratio for the predominant genotypes associated with VVC infection was 4.3.Conclusion The predominant distribution of genotypes was observed among the isolated C.albicans strains.The predominant genotypes of C.albicans were highly associated with the occurrence of VVC .
4.In vitro study of antisense oligodeoxynucleotides to inhibit HBV expression and induce apoptosis

Objective To study the effect and mechanism of antisense oligodeoxynucleotides (ASONs) inhibiting hepatitis B virus (HBV) expression. Methods We designed and synthesized antisense oligodeoxynucleotides directly against HBV PreS 2 gene and noncomplementary sequence control. 2.2.15 cells were chosen as cell model. Inhibitory effect of ASONs on HBV gene expression were assayed by ELISA. Cell apoptosis and proliferation were detected by Fascan Flow Cell Cytometer. Effect of ASONs on cell metabolism was detected by radioimmunoassay (RIA) and MTT assay. Results ASONs were able to effectively inhibit HBV expression. Their inhibitory rates of HBsAg and HBeAg were 66% and 91%, respectively. Noncomplementary sequence control group (both inhibitory rates were 11%) was not able to inhibit HBV expression. ASON might induce host cell apoptosis. Cell apoptosis rates on 3rd and 6th day were 6.10% and 6.43%, respectively. Proliferation index were 37% and 36%, respectively. Results of RIA and MTT showed that ASON had not cytoxicity on host cell. Conclusions Not only are ASON able to inhibit HBV gene expression with sequence specific but also clear HBV in the way of apoptosis.
