1.XU Jingfan's Experience in Treating Spleen and Stomach Diseases with Shichangpu (Rhizoma Acori Tatarinowii):Based on the Theory of 'Aromatics Acting on the Spleen'
Journal of Traditional Chinese Medicine 2026;67(12):1258-1261
This article summarized the clinical experience of professor XU Jingfan,a traditional Chinese medicine (TCM) master, in treating spleen and stomach diseases with aromatic herb Shichangpu (Rhizoma Acori Tatarinowii), based on the theory of 'aromatics acting on the spleen'. Shichangpu is commonly combined with Peilan (Herba Eupatorii) to aromatically awaken the spleen, and self-made Xingpi Kaiwei Formula (醒脾开胃方) can be used for postprandial distress syndrome of the spleen-stomach disharmony type. It is paired with Diyu (Radix Sanguisorbae) to resolve turbidity and promote wound healing, with a self-made Changyu Decoction (菖榆煎) for retention enema in chronic colitis and inflammatory bowel disease. When paired with Yujin (Radix Curcumae), it can be used to promote qi and resolve constraint, and can be combined with modified Sini Powder (四逆散) for epigastric pain of liver-stomach disharmony type. Paired with Fangfeng (Radix Saposhnikoviae) to dispel wind and resolve dampness, and together with Tongxie Important Formula (痛泻要方), it is used to treat diarrhea-predominant irritable bowel syndrome of liver constraint and spleen deficiency type. It is paired with Yizhiren (Fructus Alpiniae Oxyphyllae) for its warming and astringing properties, and together with modified Linggui Zhugan Decoction (苓桂术甘汤) for phlegm-rheum due to spleen-stomach yang deficiency type. Clinically, great importance is also attached to tongue inspection related to Shichangpu, closely focusing on the pathogenesis of internal accumulation of damp turbidity in the middle jiao (焦). Moreover, when treating spleen and stomach diseases, the dosage should be light and flexible.

BACKGROUND:miRNAs expression has been reported to be associated with hepatic and renal fibrosis, and dermal fibrogenesis. Moreover,a targeted regulatory relationship between miR-192-5p and epidermal regulators has been demonstrated in gouty arthritis.OBJECTIVE:To investigate the expression and regulatory role of miR-192-5p in hypertrophic scar and to verify whether there is a targeted regulatory relationship between miR-192-5p and epidermal regulators. METHODS:(1) Six cases of hypertrophic scar tissue and six cases of normal skin tissue were collected from the First Affiliated Hospital of Xinjiang Medical University. And miR-192-5p and epidermal regulator mRNA expression were detected by qRT-PCR. (2) The primary hypertrophic scar fibroblasts were obtained using tissue explant method and cultured to 3-6 generations for subsequent experiments. There were three groups in the experiment:negative control group,miR-192-5p mimic group and miR-192-5p inhibitor group. The latter two groups were transfected with the corresponding sequences. Cell proliferation viability was detected by the cell counting kit-8 assay and EdU kit;and the migration ability was detected by the cell scratch test. Cell apoptosis was detected by flow cytometry. The gene and protein expressions of epidermal regulator, type Ⅰ collagen, type Ⅲ collagen and α-smooth muscle actin were detected by qRT-PCR and western blot, respectively. miR-192-5p targets were predicted by a bioinformatics website, and target binding was validated by dual luciferase assay. RESULTS AND CONCLUSION:(1) Compared with normal skin tissues and their fibroblasts,miR-192-5p and epidermal regulator were highly expressed in hypertrophic scar and hypertrophic scar fibroblasts (P<0.05 or P<0.01). (2) After overexpression of miR-192-5p,cell proliferation was enhanced (P<0.05) and EdU positive cell rate increased (P<0.01) when compared with the negative control group;after inhibition of miR-192-5p,cell viability (P<0.05) and EdU positive rate decreased (P<0.05). (3) At 24 hours after overexpression of miR-192-5p,compared with the negative control group, the area between cell scratches and apoptosis rate decreased in the miR-192-5p mimic group (P<0.05) but increased in the miR-192-5p inhibitor group (P<0.01). (4) At 48 hours after transfection, the mRNA and protein levels of epidermal regulator were significantly decreased in the miR-192-5p mimic group, while the mRNA and protein levels of type Ⅰ collagen, type Ⅲ collagen and α-smooth muscle actin were significantly increased (P<0.05 or P<0.01). The miR-192-5p inhibitor group showed opposite changes in the above four indicators (P<0.05 or P<0.01). (5) The Targetscan website predicted that epidermal regulator had a potential binding site for miR-192-5p. (6) Dual luciferase assays showed that miR-192-5p could bind to epidermal regulator in a targeted manner. To conclude, overexpression of miR-192-5p can decrease the expression of epidermal regulator, and the two may be negatively regulated, suggesting that regulation of epidermal regulator may play a role in inhibiting the proliferation of hypertrophic scar fibroblasts.

Chinese Journal of Biotechnology 2025;41(2):753-770
Lateral organ boundaries (LOB) domain (LBD) genes encode a family of transcription factors ubiquitous in higher plants, playing crucial roles in the growth, development, and stress responses. Hippophae rhamnoides, known for its drought, cold, and saline-alkali tolerance, offers significant economic benefits and ecological values. Utilizing the whole genome data and bioinformatics approaches, this study identified and analyzed the LBD gene family in H. rhamnoides. Additionally, we examined the expression pattern of HrLBD genes by integrating the transcriptome data from male and female flower buds in development. Eleven LBD genes were identified in H. rhamnoides, and these genes were distributed on five chromosomes. The HrLBD proteins showed the lengths ranging from 159 aa to 302 aa, the molecular weights between 18 249.91 Da and 33 202.01 Da, and the subcellular localization in the nucleus or chloroplasts. LBD protein domains and gene structures were highly conserved, featuring similar motifs. The phylogenetic analysis of HrLBD genes and the LBD genes in Arabidopsis thaliana and Hordeum vulgare revealed that HrLBD genes falled into two major categories: Class Ⅰ and Class Ⅱ. The transcriptome data and RT-qPCR showed that HrLBD genes were highly expressed in male flower buds, with up-regulated expression levels throughout bud development, indicating a role in the specific stage of male flower bud development. This study lays a theoretical foundation for exploring the roles of HrLBD genes in the growth, development, and sex differentiation of H. rhamnoides flower buds.

Chinese Journal of Preventive Medicine 2024;58(9):1432-1437
Immune-mediated neuropathies (IMN) are a heterogenous group of disorders affecting the peripheral nervous system, due to dysregulation of the immune system. It mainly includes Guillain-Barre syndrome, chronic inflammatory demyelinating polyradiculoneuropathy, multifocal motor neuropathy and so on. Most of these diseases can be clinically improved by appropriate immunotherapy, but some patients still have unsatisfactory results. Therefore, studying the pathophysiology of the occurrence and development of diseases can reveal the nature of diseases and provide a theoretical basis for the prevention, diagnosis and treatment of diseases. In this paper, the pathophysiological mechanism of various IMNs is described in detail, with emphasis on immunological mechanism, and the progress of diagnosis and treatment of various IMNs is briefly introduced.

BACKGROUND:Hypoxic exercise can promote the degradation of body fat,and changes in the external environment can affect the circadian rhythm of animals, but the mechanisms by which changes in circadian rhythm regulate adipose tissue browning and fat degradation are unclear. OBJECTIVE:To elucidate the mechanism of clock gene regulation on epididymal adipose tissue Browning in obese rats undergoing hypoxia exercise. METHODS:Forty obese rats were randomly selected and divided into four groups(n=10 per group):normoxic sedentary group, hypoxic sedentary group, normoxic exercise group, and hypoxic exercise group for 4 weeks of intervention. The rats in the sedentary groups were not intervened, while those in the hypoxic groups lived in a hypoxic chamber with an oxygen concentration of 13.6%for the whole day.In the exercise groups, adaptive training was performed in the 1st week, and the speed and length of training remained unchanged for the last 3 weeks. The body mass, body length and perirenal fat mass of obese rats were measured.Serum levels of triacylglycerol, total cholesterol,low-density lipoprotein cholesterol, and high-density lipoprotein cholesterol in obese rats were detected by a biochemical assay kit.Liver fat content was observed by oil red O staining.Hematoxylin-eosin staining was used to evaluate the browning of epididymal adipose tissue of rats in different groups.RNA sequencing combined with bioinformatics analysis was used to analyze transcriptome changes in adipose tissue. The mRNA expressions of PGC-1α,Beclin 1,KLF 2 and Perilipin 1 in epididymal adipose tissue were detected by RT-PCR. RESULTS AND CONCLUSION:Hypoxic exercise intervention significantly decreased body mass, body fat percentage,Lee's index, serum triacylglycerol, total cholesterol, and low-density lipoprotein cholesterol levels(P<0.01),and significantly increased high-density lipoprotein cholesterol level(P<0.01).Oil red O staining and hematoxylin-eosin staining results showed that hypoxic exercise was more effective in promoting fat mobilization in liver tissue and promoting the browning of parepididymal adipose tissue compared with normoxic sedentary group, hypoxic sedentary group, and normoxic exercise group.RNA-seq results showed that hypoxic exercise significantly upregulated the expression of clock genes Dbp,Nr1d1,Sik1 and adipose tissue browning gene Ppargc1a(PGC-1α)and downregulated the expression of Arntl(Bmal1),accompanied by the enhanced expression of genes related to substance metabolism.qRT-PCR indicated that hypoxic exercise significantly increased the mRNA expression levels of PGC-1α and Perilipin1(P<0.01).Therefore,these findings indicate that clock genes play an important role in promoting adipose tissue browning during hypoxic exercise.
