1.Analysis of influencing factors of time prolongation of time up and go test in maintenance hemodialysis patients

Objective To explore the influence factors of timed up and go test(TUG)in patients with maintenance hemodialysis(MHD).Methods Cross-sectional investigation method was used to select patients who underwent regular hemodialysis in Beijing Luhe Hospital,Capital Medical University from June 1,2023 to January 31,2024,with complete data and cooperation.The patients were respectively treated with a simple mental state checklist and 5 times sit to stand test(STS-5)and TUG scale were used to assess cognitive function,lower limb muscle strength,balance and gait function.Grouping was carried out according to the results of TUG.If TUG 12s,it is judged as the TUG normal group;if TUG>12s,it is judged as the TUG prolonged group.Multi-factor Logistic regression was used to analyze the influencing factors in the TUG prolonged group,and the predictive value of influencing factors to TUG prolonged patients was analyzed by receiver operating characteristic(ROC)curve.Results A total of 146 patients with MHD were included in this study,including 86 patients in TUG prolonged group and 60 patients in TUG normal group.Gender,age,combined hypertension,combined diabetes,blood glucose,education level,Minimum Mental State Examination(MMSE)score,C-reactive protein were compared between groups.Serum albumin,serum creatinine,serum sodium,whole parathyroid hormone,low density lipoprotein-cholesterol,serum uric acid,STS-5 results,and single urea clearance index(Kt/V)had statistical differences(P<0.05).Multivariate Logistic regression showed that old age,female,low MMSE score,low Kt/V,and STS-5 duration were independent risk factors for TUG prolonged(P<0.05).ROC curve showed that the area under the curve(AUC)of age,gender,total MMSE score and STS-5 time in predicting TUG prolongation in MHD patients were 0.825,0.678,0.777 and 0.836,respectively(P<0.01),which had high predictive value.The AUC of Kt/V in predicting TUG prolongation was 0.602(P=0.037),has a certain predictive value;AUC of TUG prolongation in MHD patients predicted by the above 5 indicators as a global model was 0.923(P<0.01).Conclusions The age,gender,MMSE score,STS-5 prolongation and Kt/V level of MHD patients are independent risk factors for TUG prolongation,and may be biological indicators for predicting TUG prolongation.

3.Research Advance of Chinese Medicine in Treating Atherosclerosis: Focus on Lipoprotein-Associated Phospholipase A2.

As a serious cardiovascular disease, atherosclerosis (AS) causes chronic inflammation and oxidative stress in the body and poses a threat to human health. Lipoprotein-associated phospholipase A2 (Lp-PLA2) is a member of the phospholipase A2 (PLA2) family, and its elevated levels have been shown to contribute to AS. Lp-PLA2 is closely related to a variety of lipoproteins, and its role in promoting inflammatory responses and oxidative stress in AS is mainly achieved by hydrolyzing oxidized phosphatidylcholine (oxPC) to produce lysophosphatidylcholine (lysoPC). Moreover, macrophage apoptosis within plaque is promoted by localized Lp-PLA2 which also promotes plaque instability. This paper reviews those researches of Chinese medicine in treating AS via reducing Lp-PLA2 levels to guide future experimental studies and clinical applications related to AS.

4.Exploration of IRES Elements within the ORF of the Coxsackievirus B3 Genome.

Objective:
This study aimed to identify internal ribosome entry sites (IRESs) in the open reading frame (ORF) of the Coxsackievirus B3 (CVB3) genome.
Methods:
The sequences of P1, P2, or P3 of the CVB3 genome or the truncated sequences from each antithymocyte globulin (ATG) to the end of the P1, P2, or P3 gene were inserted into the pEGFP-N1 vector. After transfection, possible IRES-dependent green fluorescent protein (GFP)-fused proteins were detected by anti-GFP western blotting. The sequences of possible IRESs were inserted into specific Fluc/Rluc bicistronic vectors, in which the potential IRESs were determined according to the Fluc/Rluc activity ratio. Expression of Fluc and Rluc mRNA of the bicistronic vector was detected by RT-qPCR.
Results:
After transfection of full length or truncated sequences of the P1, P2, or P3 plasmids, six GFP-fused protein bands in P1, six bands in P2 and nine bands in P3 were detected through western blotting. Two IRESs in VP2 (1461-1646 nt) and VP1 (2784-2983 nt) of P1; one IRES in 2C (4119-4564 nt) of P2; and two IRESs in 3C (5634-5834 nt) and 3D (6870-7087 nt) of P3 were identified according to Fluc/Rluc activity ratio. The cryptic promoter was also excluded by RT-qPCR.
Conclusion
Five IRESs are present in the CVB3 coding region.

5.Angiogenesis function of astragaloside IV in rats with myocardial infarction via PKD1-HDAC5-VEGF pathway

OBJECTIVE This study aimed to investigate the role and mechanism of Astragaloside IV (AS-IV)in rats with myocardial infarction.METHODS The myocardial infarction model was established by ligation of the left anterior descending artery. The rats were randomly divided into sham, DMSO, model group, AS-IV and CID755673 groups. The rats were sacrificed 4 weeks later, and segmental heart samples were used for hematoxylin and eosin staining and masson staining. The expression of PKD1, HDAC5 and VEGF were analyzed using immunohistochemistry, reverse transcription poly-merase chain reaction and western blot. RESULTS Compared with the sham operation and DMSO groups,morphology of myocardium in model group was disordered,accompanied with necrotic myocar-dial cells and obvious collagen tissues. After treatment with AS-IV, the morphology of myocardium was obviously improved, and the number of new blood vessels increased significantly. However, after treatment with CID755673, the myocardial tissue of rats became disordered again, the necrotic cells increased, and some vessels closed. The expression levels of PKD1, HDAC5 and VEGF mRNA and protein in myocardial tissue of model group were significantly lower than the other four groups(P<0.05), whereas these levels in the AS-IV group were significantly higher than those in the other four groups (P<0.01). Additionally, the CID755673 group had significantly higher levels of PKD1, HDAC5 and VEGF mRNA and protein than the sham group, DMSO group and model group (P<0.05). CONCLUSION AS-IV may partly promote the angiogenesis of myocardial tissue in rats with myocardial infarction via the PKD1-HDAC5-VEGF pathway.
6.The diagnosis value of whole body bone imaging with coincidence SPECT/CT fusion imaging for solitary“hotspots”

Objective To evaluate the differentiating diagnosis value of coincidence SPECT/CT fusion imaging used in the soli-tary“hotspots”in the whole body bone imaging.Methods Fifty patients with malignancy were selected to have examination.the sensitivity,specificity and accuracy of these imaging methods were calculated by contrasting the pathology examination,CT and MRI examination,more than 6 months follow-up results between the cases.Results The sensitivity,specificity and accuracy of SPECT imaging were 94%,23%,and 63%,respectively,and that of CT were 47%,87%,66%;the sensitivity,specificity and accu-racy of SPECT/CT were 95%,80%,88%,respectively.The specificity and accuracy of SPECT/CT was significantly higher than SPECT(P<0.05)and its sensitivity and accuracy was higher than that of CT(P<0.05).Conclusion Coincidence SPECT/CT hy-brid imaging could raise the sensitivity,specificity and accuracy of diagnosis of solitary“hotspot”in whole bone imaging,which is of high clinical application value.
7.3D visualization for simulating percutaneous transcatheter closure of atrial septal defect :a pilot clinical study

Objective To evaluate the clinical value of 3D visualization method for simulating percutaneous transcatheter closure of atrial septal defect(ASD).Methods 3D volume render method was used for visualizing ASD and surrounding structures and 3D modeling method was applied for simulate the shape of occlusion device.The size and the distance between the lower edge of device and atria-ventricular valve of simulation occluder and actual selected atrial septal occluder (ASO) were compared in 30 patients underwent successful transcatheter closure of ASD.Results The location,geometry,size,extent of ASDs in children were displayed in 3D visualization.No significant difference was found between simulation occluder and ASO size measured from left atrium[(26.07±5.32) cm vs.(25.91±5.32) cm]and right atrium[(22.13±5.31) cm vs.(22.08±5.26) cm,all P >0.05].The distances from simulation occluder to mitral valves[(5.76±2.39) cm]and to tricuspid valves[(8.25±2.40) cm]were similar as ASO to atria-ventricular valves[(5.61±2.26) cm and (8.02±2.48) cm,respectively,all P>0.05].Conclusions The simulating pereutaneous transcatheter closure of ASD by 3D visualization technique could be a helpful noninvasive approach for ASO selection before the procedure of transcatheter occlusion of ASD.
8.Study on quality of flavone in various cultivars of Chrysanthemum morifolium for medicine.

OBJECTIVETo study on dynamic accumulation of various flavone in Chrysanthemum morifolium, in order to appraise the quality of flavone.
METHODThe extract was hydrolyzed and the flavonoid components were determined by RP-HPLC.
RESULT AND CONCLUSIONIn the samples from Tongxiang, Tezhongboju showed the highest flavonoids content with the total flavone 9.81%, quercetin 4.17 per thousand and apigenin 17.02 per thousand. In the samples from Sheyang, Hongxinju showed the highest content of luteolin with 6.84 per thousand. In the samples from Jiangning, Tezhongboju showed the highest content of total flavone with 7.59%, Yizhongdabaiju showed the highest content of quercetin with 2.78 per thousand, Sheyangxiaobaiju showed the highest content of luteolin with 4.27 per thousand, Sheyanghongxinju showed the highest content of apigenin with 11.20 per thousand. There were differences in the content of flavonoids among the different cultivars, and the same cultivars showed also different when planted in different areas.

9.Selective adsorption of multi-walled carbon nanotubes with liquiritin and isoliquiritin.

Selective adsorption of active ingredients liquiritin and isoliquiritin from Glycyrrhiza uralensis Fisch with multi-walled carbon nanotubes (MWNTs) has been studied. Distribution coefficients of liquiritin between ethanol solvent and r-MWNTs or o-MWNTs in 293K is 37.5 and 43.3, while the distribution coefficients of isoliquiritin between ethanol solvent and r-MWNTs or o-MWNTs is 717 and 1080, respectively. It was indicated that the distribution coefficient of isoliquiritin adsorbed by MWNTs was much larger than that of liquiritin, and oxidation treatment of MWNTs could obviously enhance their adsorption ability. The possible reasons that MWNTs can selectively adsorb isoliquiritin other than liquiritin were discussed on the bases of molecular structure of the active ingredients and their interaction with nanotubes.
