In the context of"Artificial Intelligence(AI)+Education"era,this study introduces the teaching reform and devel-opment path of the course"Fundamentals of Immunology and Pathogenic Biology"by Professor Lu Fangguo's team at Hunan University of Traditional Chinese Medicine.After twenty years of exploration,the course has successfully transitioned from traditional teaching to intelligent teaching,achieving a comprehensive upgrade in educational concepts,teaching methods,and resources.The reform process is divided into three stages:Early exploration,comprehensive reform,and deepening development.It encompasses the construction of a smart teaching platform,the development and promotion of new forms of digital teaching resources,and the deep integration of ideological and political education.This reform has significantly enhanced teaching quality and students'overall competencies,showcasing the innovative spirit of educators.It has gained nationwide recognition and promotion,providing valuable references for the innovation of medical education in the new era.
2.The impact of RAB2B on pancreatic cancer proliferation and metastasis via the NF-κB pathway

Objective To investigate the effects of Ras-related protein Rab-2B(RAB2B)on the biological behaviors of pancreatic cancer cells and elucidate its underlying mechanism.Methods PANC-1 cells,which exhibit relatively high RAB2B expression,and BXPC-3 cells,which display relatively low RAB2B expression,were selected from five pancreatic cancer cell lines.RAB2B-siRNA and pcDNA3.1-RAB2B plasmids were transfected into PANC-1 and BXPC-3 cells using a cell transfection technique.The CCK-8 assay was employed to evaluate the prolif-erative capacity of pancreatic cancer cells following RAB2B intervention.Wound healing and Transwell chamber assays were utilized to assess the migratory and invasive capabilities of pancreatic cancer cells.Additionally,the mRNA and protein expression levels of RAB2B,NF-κB,and Fibronectin 1(FN1)were analyzed by qRT-PCR and Western blot(WB),respectively.Results RAB2B mRNA and protein expression levels were significantly down-regulated in PANC-1 cells following transfection(P<0.05).CCK-8 assay results demonstrated that the proliferative capacity of PANC-1 cells was markedly reduced(P<0.05),and the wound-healing ability was substantially impaired(P<0.01)upon RAB2B knockdown.Transwell assays revealed a significant decrease in cell migration(P<0.01),while Western blot analysis indicated that the expression levels of phosphorylated p65 and FN1 were notably diminished(P<0.01).Conversely,overexpression of RAB2B reversed these aforementioned alterations.Conclusions Knockdown of RAB2B in PANC-1 cells significantly suppresses cell proliferation and migration,whereas overexpression of RAB2B in BXPC-3 cells markedly promotes these processes.This effect is likely mediated through the activation of the NF-κB signaling pathway and the subsequent regulation of FN1 expression.
3.Clinical analysis of severe Mycoplasma pneumoniae pneumonia in children with two different imaging manifestations

Objective:To compare the clinical characteristics of severe Mycoplasma pneumoniae pneumonia(SMPP)with bronchiolitis and consolidation to raise awareness.Methods:Retrospective analysis was conducted on children hospitalized at the first affiliated hospital of Xiamen University diagnosed with(SMPP)from February 2023 to January 2024.The children were divided into bronchiolitis group and large consolidation group based on their high-resolution CT imaging findings.Data on basic information,clinical features,auxiliary examination results,and bronchoscopy findings were analyzed.Results:This study included 415 pediatric patients,including 203 with bronchiolitis and 212 with large consolidation.The proportion of males,atopy rate,and first-degree relative atopy rate in the bronchiolitis group were higher,and the age of the bronchiolitis group was younger( P<0.05).The fever peak,incidence of fever,and low breath sound in the bronchiolitis group were lower,and the incidence of hypoxemia,wheezing,shortness of breath,inspiratory tri-concave signs,moist rales were higher than those in the consolidation group( P<0.05).The white blood cell counts,platelet counts,and lactate dehydrogenase were higher and the percentage of neutrophils,C-reactive protein,ferritin,D-dimer,and fibrinogen degradation products were lower in the bronchiolitis group than those in the consolidation group( P<0.05).The rate of 2 or more bronchoscopy procedures,and the proportion of longitudinal plica,mucus plug,and plastic in the consolidation group were higher than those in the bronchiolitis group( P<0.05).After three months of follow-up,there were 8 cases of bronchiolitis obliterans,including 7 cases in the bronchiolitis group and 1 case in the large consolidation group. Conclusion:If atopy,wheezing,and dyspnea are present in young children and preschool children with confirmed SMPP,they should be closely monitored for signs of bronchiolitis SMPP.Children with significant consolidation on imaging should have the possibility of various intrapulmonary and extrapulmonary problems taken into account.Appropriate treatment options should be implemented during the acute phase based on the imaging findings.
4.Research progress on regulation of inflammatory response by glucose metabolic reprogramming

Inflammatory response is a common feature of many acute and chronic diseases,which involves in activation of various cell types.Similar to tumor cells,inflammatory response is regulated by glucose metabolic reprogramming.Cells involved in pro-inflammatory response,such as M1 macrophages,Th1 and Th17 lymphocytes,must generate energy rapidly through glycolysis transformed by glucose metabolic reprogramming to promote inflammation,while cells involved in immune regulation and anti-inflam-matory response,such as regulatory T cells and M2 macrophages,preferentially use fatty acid oxidation and oxidative phosphorylation as a main source for energy production.This article systematically reviews the role of glucose metabolic reprogramming in different stages of inflammatory response,as well as the various molecular mechanisms of glucose metabolic reprogramming in inflammatory response,including activation or inhibition of signaling pathways,epigenetic regulation,post-transcriptional regulation and post-translational modification,which aims to provide a reference for exploring the specific regulatory mechanisms of glucose metabolic reprogramming on inflammatory response and the therapeutic targets of inflammatory-related diseases.

7.Study on the Effect and Mechanism of Ziyin Mingmu Formula in Inhibiting Laser-Induced Choroidal Neovascularization in Mice Based on Network Pharmacology

OBJECTIVE To investigate the effect and potential mechanism of Ziyin Mingmu Formula in treating age-related mac-ular degeneration(AMD)by combining network pharmacology with animal model validation.METHODS Active ingredients of Ziyin Mingmu Formula were obtained from the TCMSP and BATMAN databases,and their targets were searched using the PharmMapper da-tabase.AMD disease targets were identified using the GeneCards,DrugBank,OMIM,and TTD databases.Venny analysis was per-formed to identify the intersection of active ingredient and disease targets.A protein-protein interaction(PPI)network was constructed using the String database,and core targets were screened using Cytoscape 3.9.0 software.KEGG pathway enrichment analysis was per-formed using the DAVID database.Molecular docking of key active ingredients with core targets was performed using Autodock software.A laser-induced mouse choroidal neovascularization(CNV)model was used.Optical coherence tomography angiography(OCTA)was used to assess CNV area in vivo,immunofluorescence staining was used to assess CNV area on choroidal flat mounts,and Western blot analysis was used to examine the expression of proteins involved in the vascular endothelial growth factor(VEGF)pathway.RESULTS Network pharmacology analysis identified 221 active ingredients in the Ziyin Mingmu Formula.PPI analysis i-dentified 29 core targets,including SRC,protein kinase B(AKT1),mitogen-activated protein kinase 1(MAPK1),and heat shock protein 90α family class A member 1(HSP90AA1).KEGG analysis revealed that the VEGF signaling pathway was the most highly en-riched.Molecular docking revealed that the core targets SRC,AKT1,MAPK1,and HSP90AA1 had good binding affinity with the main active ingredients,diosmetin,catechin,vestitol,and licochalcone A.Animal experiments showed that the Ziyin Mingmu Formula significantly reduced CNV area in model mice,downregulated VEGF protein expression,decreased VEGFR2,p38,and ERK1/2 pro-tein phosphorylation levels,and inhibited the VEGF signaling pathway.CONCLUSION Ziyin Mingmu Formula may inhibit CNV for-mation by regulating the VEGF signaling pathway.

9.Value of microbiological rapid on-site evaluation in treatment of patients with severe community-acquired pneumonia

OBJECTIVE To explore the value of microbiological rapid on-site evaluation(M-ROSE)technique in treatment of the patients with severe community-acquired pneumonia(SCAP).METHODS A total of 124 patients with SCAP who were treated in the department of respiratory and critical care medicine of The Fourth Medical Center of Chinese PLA General Hospital from Sep.2023 to Dec.2024 were enrolled in the study and were random-ly divided into the M-ROSE group and the control group in a 1∶1 ratio based on the status of M-ROSE for bron-choalveolar lavage fluid(BALF).The M-ROSE test and conventional etiological test[metagenomic next genera-tion sequencing(mNGS),smear,culture]were performed for the M-ROSE group,and the conventional etiologi-cal test was only carried out for the control group.The baseline data,symptoms and signs,C-reactive protein lev-el,treatment status and outcomes were observed and compared between the two groups of patients.RESULTS A-mong the 62 patients for whom the BALF specimens were detected with M-ROSE,45(72.58%)patients showed the consistent test result for fungi with mNGS,47(75.81%)patients showed the same test result for cocci with mNGS,and 30(48.39%)patients showed the same test result for bacilli with mNGS.The duration of the M-ROSE test was 1.50(1.50,2.00)h,shorter than that of the smear,culture and mNGS(P<0.05).The body temperature returning to the normal and the property,volume of sputum were improved more early in days in the M-ROSE group than in the control group after the anti-infection treatment(P<0.05);the level of inflammatory factor CRP declined more quickly in the M-ROSE group than in the control group(P<0.05);the effective rates of treatment of the M-ROSE group were higher than those of the control group after the hospitalization for 3,5 and 7 days(P<0.05).CONCLUSION The M-ROSE test for BALF may facilitate the rapid etiological diagnosis for the patients with SCAP in early stage,provide guidance for the anti-infection treatment strategies,and accelerate the improvement of symptoms and inflammatory factors;it has certain clinical application value.
