1.Expression of serum LOXL4 and PTPN3 in patients with colorectal can-cer and their relationship with postoperative liver metastasis

Objective:To analyze the expression of serum lysyl oxidase like protein 4(LOXL4)and protein tyrosine phosphatase non receptor type 3(PTPN3)in patients with colorectal cancer and their relationship with postoperative liver metastasis.Methods:Totally 137 patients with colorectal cancer who visited Qingdao Central Hospital,University of Health and Rehabilitation Science from April 2017 to December 2019 were recruited as the cancer group.Complying with whether liver metastasis occurred during the follow-up period of 5 years,the patients were classified into the liver metastasis group(27 cases)and the non liver metastasis group(110 cases).Another 137 healthy individuals who underwent physical check ups were recruited as the control group.The qRT-PCR method was used to detect the ex-pression levels of serum LOXL4 and PTPN3 mRNAs.The serum LOXL4 and PTPN3 mRNAs were compared under dif-ferent clinical pathological data,and the pathological data of liver metastasis group and non liver metastasis group were compared.Cox regression analysis was used to explore the influencing factors of postoperative liver metastasis in pa-tients with colorectal cancer.ROC curve was used to explore the predictive value of serum LOXL4 and PTPN3 for postoperative liver metastasis in patients with colorectal cancer.Results:Compared with the control group,the can-cer group had higher levels of serum LOXL4 and PTPN3 mRNAs expression(P<0.05).Compared with patients with medium to high differentiation,infiltration depth≤1/2 muscle layer,and TNM stage I-II,patients with low differentia-tion,infiltration depth>1/2 muscle layer,and TNM stage III had higher levels of serum LOXL4 and PTPN3 mRNAs ex-pression(P<0.05).Compared with the non liver metastasis group,the liver metastasis group had higher levels of se-rum LOXL4 and PTPN3 mRNAs expression(P<0.05).High expression of serum LOXL4 and PTPN3 mRNAs were risk factors for liver metastasis after colorectal cancer surgery(P<0.05).Compared with the single prediction of serum LOXL4 and PTPN3 mRNAs,the AUC of serum LOXL4 combined with PTPN3 in predicting postoperative liver metasta-sis in colorectal cancer patients was higher(P<0.05).Conclusion:Serum LOXL4 and PTPN3 mRNAs expression lev-els are abnormally elevated in patients with colorectal cancer,which is related to the deterioration of pathological data such as differentiation degree,infiltration depth,and TNM staging.Elevated serum LOXL4 and PTPN3 mRNAs expres-sion levels may increase the risk of postoperative liver metastasis in patients,and the combination has higher predic-tive value for postoperative liver metastasis.
2.Retrospective Analysis of Rehabilitation Outcomes and Complications after Total Knee Arthroplasty with Different BMI Grades

Objective:To investigate the effects of different BMI classifications on rehabilitation outcomes and complications after total knee arthroplasty(TKA).Methods:Clinical data of 88 patients who underwent TKA in our hospital from January 2022 to June 2024 were retrospectively analyzed.According to the World Health Organization(WHO)BMI standards,patients were divided into normal weight group(18.5 ≤BMI<24.9 kg/m2),overweight group(25.0≤BMI<29.9 kg/m2),mild obesity group(30.0≤BMI<34.9 kg/m2),and severe obesity group(BMI≥35.0 kg/m2).Baseline data,operation time,intraoperative blood loss,hospital stay,postoperative knee function scores,pain scores,range of motion(ROM),and incidence of complications were compared among groups.Results:With the increase of BMI,operation time was prolonged,intraoperative blood loss increased,and hospital stay extended,with statistically significant differences(P<0.05).At 3-month and 6-month follow-ups,Knee Society Score(KSS)and Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC)showed that functional recovery decreased with increasing BMI(P<0.05).Regarding postoperative complications,the incidence of poor incision healing,infection,and deep vein thrombosis(DVT)in the obese groups was significantly higher than in the normal weight and overweight groups(P<0.05).Conclusion:BMI is an important factor affecting rehabilitation outcomes and complications after TKA.Patients with high BMI have slower functional recovery and higher complication rates.For patients with high BMI,preoperative weight loss and individualized rehabilitation programs should be considered to improve TKA prognosis.

4.Role of acetylation modification in the occurrence and development of thyroid cancer and its potential clinical application value

Thyroid cancer is the most prevalent malignant tumor of the endocrine system,and its incidence has shown a significant upward trend in recent years.Acetylation,an important post-translational protein modification,is involved in regulating gene transcription,cell cycle progression,and invasion ability in various types of thyroid cancers.Additionally,histone deacetylase(HDAC)inhibitors have shown potential in the treatment of thyroid cancer.This article systematically reviews the biological functions and regulatory mechanisms of acetylation in the development and progression of thyroid cancer based on recent literature.Furthermore,it discusses the clinical application prospects of HDAC inhibitors,providing a solid theoretical basis and feasible therapeutic strategies for targeted therapy of thyroid cancer.
5.Analysis of Abnormal Hemoglobin Phenotypes and Gene Mutation in Diabetes Screening Population in Xi'an Region

Objective To investigate the mutational spectrum and phenotype of abnormal hemoglobin in the diabetes screening population in Xi'an Region. Methods A total of 30356 patients from the Second Affiliated Hospital of Xi'an Jiaotong University were collected and measured for HbA1c by high-performance liquid chromatography (HPLC) from January 2022 to December 2022. Both complete blood count and hemoglobin analysis were carried out when HPLC suggested an abnormal peak. Abnormal hemoglobins were identified by Sanger sequencing. Results In this study,the rate of abnormal hemoglobin in the diabetes screening population of Xi'an region was 0.096% (28/30356). A total of 7 types of abnormal hemoglobins were detected:Hb E (n=10),G-Coushatta (n=6),Hb G-Taibei(n=6),Hb Q-Thailand (n=2),Hb North Manchester (n=2),Hb Ottawa (n=1) and Hb Queens (n=1). One case was Hb G-Taipei combined with IVS Ⅱ-659_664 (-GCAATA),the first report in the world. The abnormal hemoglobin phenotypes were normal except for seven cases of Hb E. Except for Hb G-Coushatta and Hb Ottawa,which did not interfere with the glycation test,the other five abnormal hemoglobins affected the glycation test results. Conclusion There is a certain prevalence rate of abnormal hemoglobin in the diabetes screaning population of Xi'an,mainly Hb E,Hb G-Taibei and Hb G-Coushatta. Most abnormal hemoglobins present normal phenotypes,but they could interfere with glycation test results.
6.Extraction and culture of enteric glial cells from C57BL/6 newborn neonatal mice

BACKGROUND:The pathogenesis of inflammatory bowel disease involves inflammation,immune activation,visceral hypersensitivity,and dysbiosis of the gut microbiota.Inflammation promotes the release of inflammatory mediators by immune cells,damaging the enteric nervous system.Enteric glial cells are an important component of the intestinal nervous system and are excellent cells for studying intestinal neuroinflammation.Primary enteric glial cells play a crucial role in exploring cell therapies for intestinal nervous system diseases.Currently,the methods for obtaining these cells are mostly cumbersome.Therefore,finding a convenient and fast method for extracting this cell is crucial.OBJECTIVE:To establish a method for optimizing the isolation,culture,and identification of mouse enteric glial cells.METHODS:0-7-day-old C57BL/6 neonatal mice were euthanized by excessive inhalation of isoflurane.After soaking in 75%alcohol for disinfection,the duodenum(1 cm below the pylorus to 1 cm above the Qu's ligament)was removed by laparotomy at the midline of the abdomen.A 1 mL syringe was filled with DPBS and the intestinal contents were repeatedly rinsed until the intestine became translucent,and the mesentery and blood vessels were peeled off.The duodenum was cut to a size of 1 mm and digested in 0.25%EDTA trypsin for 20 minutes.Then an equal amount of DMEM/F12 complete culture medium was added to terminate digestion.The liquid was filtered through a 100 μm cell filter,centrifuged,and the cells were resuspended in 1 mL of DMEM/F12 complete culture medium.When the cell adhesion growth density reached 80%,cells were digested for subculture.When cells were cultured to the third generation,glial fibrillary acid protein labeled with enteric glial cells was used for identification by immunofluorescence method.RESULTS AND CONCLUSION:The isolated and cultured cells were full of colloids,with protrusions extending outward and passable.Glial fibrillary acid protein staining was positive.This method can successfully isolate and culture enteric glial cells and is easy to operate,providing a stable model for the study of the pathophysiology of the enteric nervous system.
7.Analysis and suggestions for the FDA drug labeling rules on cardiac safety risk warnings

Objective To improve and refine the relevant regulations and guiding principles of warnings on drug instructions and labels in China.Methods This paper sorted out the drug instructions of small molecule anti-tumor drugs listed by the U.S.Food and Drug Administration(FDA)from 2005 to 2022,included the drugs mentioned in the QT interval prolongation risk,analyzed the clinical research and QT research results,and sorted out the identification and warning rules of the instructions.Results A total of 35 drugs were included,4 drugs wrote the risk of QT interval prolongation in the black box warning,21 drugs were wrote in the warning and precautions position,6 drugs were wrote in the adverse reaction section,and 2 drugs were only described under clinical pharmacology section.According to the severity of the QT interval prolongation caused by the drug and whether there were serious clinical consequences,they were displayed in the warnings(black box warnings),precautions(warnings and precautions)and adverse reactions in the instructions.Conclusion The aim of this article is to provide a reference for the writing of QT risk warning information of the instructions of domestic drug production enterprises and regulatory departments.It is recommended to clarify the severity of drug safety and the location of the instructions in clinical research,and continue to carry out safety monitoring and update the instructions in time after listing.
8.Analysis of risk factors for postoperative gastrointestinal disorders in patients craniocerebral injury

Objective To analyze the clinical risk factors for gastrointestinal dysfunction in patients with traumatic brain injury.Methods A retrospective analysis was conducted on clinical data of 313 cases of traumatic brain injury in our hospital from January 2022 to October 2023.According to whether gastrointestinal dysfunction occurred at admission,the patients were divided into the occurrence group(Group A,90 cases)and the non-occurrence group(Group B,223 cases).Multivariate Logistic regression was used to analyze the risk factors of gastrointestinal dysfunction.The receiver operating characteristic(ROC)curve was used to evaluate the efficacy of each factor in predicting gastrointestinal dysfunction in patients with craniocerebral injury.Results Among 313 patients,90 cases(28.75%)experienced gastrointestinal dysfunction.Logistic analysis showed that the admission GCS score was ≤8 points(OR=3.117,95%CI:1.358-7.157,P=0.007),pulmonary infection(OR=7.129,95%CI:1.889-27.377,P=0.004),intracranial infection(OR=3.152,95%CI:1.162-8.548,P=0.024),frontal lobe injury(OR=5.430,95%CI:1.481-19.915,P=0.011),abnormal serum Na levels(OR=8.482,95%CI:1.958-36.247,P=0.004),and decreased Alb(OR=3.001,95%CI:1.137-7.918,P=0.026)and elevated hs CRP(OR=3.142,95%CI:1.159-8.522,P=0.024)were independent risk factors for gastrointestinal dysfunction in patients with traumatic brain injury.The receiver operating characteristic(ROC)curve was used to evaluate the efficacy of each factor in predicting gastrointestinal dysfunction in patients with craniocerebral injury.Conclusion Admission GCS score≤8,pulmonary infection,intracranial infection,frontal lobe injury,abnormal serum Na levels,decreased Alb,and elevated hs CRP are independent risk factors for gastrointestinal dysfunction in patients with traumatic brain injury.
9.DNA methylation age prediction model based on ovarian granulosa cells

Objective:To develop a novel ovarian biological age prediction model based on DNA methylation for ovarian aging assessment.Methods:A prospective cohort study method was used. From March 2024 to January 2025, we collected 96 ovarian granulosa cell samples of infertility patients due to fallopian tube factors or male factors from the Reproductive Medicine Center of the Second Affiliated Hospital of Naval Medical University. Then we analyzed DNA methylation patterns across five age-associated gene regions ( ELOVL2, miR29B2C, TRIM59, KLF14 and FHL2) in a discovery cohort comprising 63 human ovarian granulosa cell samples. Targeted bisulfite sequencing was performed through PCR amplification followed by next-generation DNA sequencing. Leveraging elastic net regression analysis, we developed a predictive model incorporating 29 methylation sites that demonstrated strong age correlation. The model was subsequently validated using an independent cohort comprising 33 human ovarian granulosa cell samples. Results:The DNA methylation age prediction model based on ovarian granulosa cells showed the following results in the discovery cohort as follows: median absolute error (MAE) was 2.534 ( R=0.742, P<0.001). In the independent validation cohort, MAE was 3.019 ( R=0.729, P<0.001). Conclusion:In this study, we utilized human ovarian granulosa cells as experimental samples to develop a novel DNA methylation-based model for predicting ovarian biological age. By integrating multiple methylation sites across five age-related gene regions, this model serves as a robust indicator of ovarian aging status.
10.Construction and evaluation of a cell model simulating the change of testicular microenvironment mediated by hypoxic and high-pressure conditions in varicocele mice

Objective:Varicocele(VC)induces male infertility by mediating changes in the testicular microenvironment,in which testicular hypoxia and high-pressure are important pathological conditions.This study aims to compare the mouse spermatogenesis(GC-2spd)cells and Sertoli(TM4)cells of mouse testis after hypoxic modeling and hypoxic and high-pressure combined modeling,and to explore the feasibility of establishing a hypoxic and high-pressure combined cell model.Methods:On the basis of cell hypoxia induced by CoCl2,the complex model of testicular cell hypoxia and high pressure was constructed by changing the osmotic pressure of GC-2 and TM4 cell medium with a high concentration of NaCl solution.After selecting the intervention concentration of CoCl2 by MTT test and detecting the expression level of HIF-1α for the determination of the optimal osmotic pressure conditions of the cell model,the cells were divided into normal group,hypoxia model group and composite model group.And the levels of OS,programmed cell death,inflammatory factors,and the expression levels of pyroptosis-related proteins were compared between the normal group and the groups with different modeling methods.Results:The optimal intervention concentration of CoCl2 in GC-2 and TM4 cells was 150 and 250μmol/L,respectively,and the expression of HIF-1α was the highest in both cells under osmotic pressure of 500 mOsmol/kg(P<0.05).Compared with the normal group,the SOD levels of GC-2 and TM4 cells decreased(all P<0.05),CAT level decreased(all P<0.05),and MDA level increased(all P<0.01),and the OS level of GC-2 and TM4 cells was more obvious than that of the hy-poxia model group(all P<0.05).Compared with the normal group,apoptosis occurred in GC-2 and TM4 cells after composite model-ing(all P<0.05).Compared with the normal group,the mRNA expressions of IL-1β,IL-18,TNF-α and COX-2 in GC-2 and TM4 cells significantly increased(P<0.01)and higher than those in hypoxia model group(P<0.05)and induced pyroptosis(P<0.01).The expression level of GSDMD increased(P<0.05).Conclusion:The cell model with hypoxia and high pressure com-bined modeling can not only induce oxidative stress and apoptosis of cells better than that with hypoxia alone,but also further cause in-flammatory response damage and pyroptosis,which simulates the changes of testis microenvironment mediated by hypoxia and high pressure combined conditions in VC.This cell model can be used for studying the pathogenesis of VC-associated male infertility,evalu-ating drug efficacy,and exploring pharmacological mechanisms.
