1.Interpretation of "Cancer statistics, 2025": A comparative study on cancer epidemiological characteristics and long-term trends between China and the United States

In 2025, the American Cancer Society published "Cancer statistics, 2025", which projected cancer data for the upcoming year based on incidence data collected by central cancer registries (through 2021) and mortality data obtained from the National Center for Health Statistics (through 2022). Similarly, the National Cancer Center of China released "Cancer incidence and mortality in China, 2022" in December 2024, analyzing data from 22 cancer registries across the country. This study provides a comparative analysis of cancer incidence and mortality trends in China and the United States during the same period, with a focus on sex- and age-specific distributions and long-term changes in cancer patterns. Long-term trends indicate that lung and liver cancer mortality rates in China have declined, primarily due to tobacco control measures and hepatitis B vaccination programs. However, the burden of gastric and esophageal cancers remains substantial. In the United States, mortality rates for colorectal and lung cancers have continued to decline, largely attributed to widespread screening programs and advances in immunotherapy. As economic growth and social development, China’s cancer profile is gradually shifting towards patterns observed in countries with high human development index. However, the prevention and control of upper gastrointestinal cancers remains a critical public health challenge that requires further attention.
2.Differential expression of circRNAs in anterior lens capsules of high myopic patients with cataract.

OBJECTIVES:
To analyze the differential expression and biological functions of circRNAs in the anterior lens capsules of high myopic patients with cataract and their pathogenic roles in the development of this condition.
METHODS:
Anterior lens capsule specimens were collected intraoperatively from 36 patients with age-related cataract (ARC) and 36 high myopic patients with cataract. Among these, 18 specimens from each group were selected for whole transcriptome sequencing and biological analysis, and the remaining 36 specimens were used for validation of circPDGFRA, circFOXJ3, hsa_circ_0004767, hsa_circ_0007528, ciCRIM1, circMAN1A2, circSLC5A3, and circPTK2 expressions using RT-qPCR. hsa_circ_0007528 was selected for cell experiments to examine its effects on proliferation, migration, and apoptosis of lens epithelial cells (LECs).
RESULTS:
A total of 16 192 circRNAs were detected in the specimens from both groups, among which 62 circRNAs were differentially expressed (29 upregulated and 33 downregulated). GO and KEGG analyses revealed that the differentially expressed circRNAs were primarily localized in the cytoplasm, nucleoplasm, and endoplasmic reticulum, and were involved in signaling pathways associated with Gap junction and the PI3K-Akt, NF-κB, Jak-STAT, HIF-1, and MAPK signaling pathways. The ceRNA network predicted multiple target genes. RT-qPCR validation results were consistent with the sequencing data. In the LECs, upregulation of hsa_circ_0007528 significantly inhibited cell proliferation and migration and obviously promoted cell apoptosis.
CONCLUSIONS
The expression profile of circRNAs in the anterior lens capsule of high myopic patients with cataract differs from that of ARC patients. Upregulation of hsa_circ_0007528 inhibits LEC proliferation and migration and promotes cell apoptosis.

3.Screening analyses of tuberculosis infection among primary and secondary school students in Cixi City of Zhejiang Province from 2024 to 2025

ObjectiveTo determine the prevalence and determinants of Mycobacterium tuberculosis latent infection among primary and secondary school students in Cixi City, Zhejiang Province, so as to provide references for the prevention and control of tuberculosis in school settings. MethodsInterferon-γ release assay (IGRA) testing was performed to fourth-grade primary school students , as well as to those grade 7 and grade 10 students of the academic year 2024‒2025 in Cixi City. Individuals tested positive for IGRA were subsequently subjected to chest X-ray examination and sputum tests (including three smear microscopy examinations and one mycobaterial culture), and epidemiological investigations were carried out for confirmed cases. Infection rates were compared across student categories by χ² tests, while factors influencing infection were analysed through multivariate logistic regression. ResultsA total of 36 214 students completed tuberculosis screening, with an infection rate of 0.72% (260/36 214). The tuberculosis infection rates among fourth-grade primary school students, non boarding grade 7 students, boarding grade 7 students, grade 10 students of senior high school and of vocational high school were 0.68% (89/13 139), 0.75% (86/11 501), 0.51% (13/2 553), 0.76% (52/6 819), and 0.91% (20/2 202), respectively. Multivariate logistic regression analyses indicated that students with a history of close contact with tuberculosis patients (OR=21.435, P<0.001) had a higher risk of tuberculosis infection, students with a geographic origin outside Zhejiang Province (OR=1.485, P=0.002) had a higher risk of infection than those within Zhejiang Province. Furthermore, students from ethnic minority classes (OR=4.232, P<0.001) might be high-risk groups for tuberculosis infection in high school settings. One IGRA-positive student was confirmed as bacteriologically positive pulmonary tuberculosis by liquid culture of sputum collected one month later. ConclusionSchools should prioritize tuberculosis screening. Students with a history of close contact, those with a geographic origin outside Zhejiang Province, and those enrolled in minority classes should be taken as priority targets for future tuberculosis screening programmes. A tracking and follow-up system must be established for IGRA-positive students to prevent persistent transmission of Mycobacterium tuberculosis within the school settings.
4.Construction of the quality evaluation scale of specification of management for humanistic caring in outpatients and its reliability and validity testing

Objective:To construct the quality evaluation scale of specification of management for humanistic caring in outpatients and test its reliability and validity.Methods:Referring to the group standards in Specification of Management for Humanistic Caring in Outpatients released by the China Association for Life Care,as well as relevant guidelines and literature,a pool of items for the quality evaluation scale of specification of management for humanistic caring in outpatients was formed.After expert consultation and expert argumentation,a quality evaluation scale of specification of management for humanistic caring in outpatients was constructed.From January to February 2024,243 hospital managers from 5 hospitals in Zhengzhou were selected as survey subjects to conduct item analysis,and reliability and validity testing on the scale.Results:Two rounds of expert inquiry and two rounds of expert argumentation were conducted,with questionnaire response rates of 92.00%and 100.00%,respectively,and expert authority coefficients of 0.952.In the second round of the expert inquiry scale,the mean importance score of the first-level indicators was 4.80 to 5.00,the full score ratio was 88.00%to 100.00%,the coefficient of variation was 0.04 to 0.17,and Kendall's coefficient of concordance was 0.857(P<0.001);the mean importance score of the second-level indicators was 4.60 to 5.00,the full score ratio was 80.00%to 100.00%,the coefficient of variation was 0.00 to 0.21,and Kendall's coefficient of concordance was 0.775(P<0.001);the mean importance score of the third-level indicators was 4.60 to 5.00,the full score ratio was 76.00%to 100.00%,the coefficient of variation was 0.00 to 0.21,and Kendall's coefficient of concordance was 0.830(P<0.001).Finally,a quality evaluation scale of specification of management for humanistic caring in outpatients was formed,including 5 first-level indicators,25 second-level indicators,and 58 third-level indicators.Exploratory factor analysis produced 5 common factors with a cumulative variance contribution rate of 74.628%.The Pearson correlation coefficients between the five-factor scores ranged from 0.648 to 0.798,and the correlation coefficients between the factor scores and the total score of the scale ranged from 0.784 to 0.938.The scale-level content validity index(S-CVI)of the scale was 0.945,the item-content validity index(I-CVI)was 0.725 to 1.000,the Cronbach's alpha coefficient of the total scale was 0.973,and the retest reliability coefficient was 0.934.Conclusion:The constructed quality evaluation scale of specification of management for humanistic caring in outpatients has good scientific validity and reliability,and can be used as an evaluation tool for specification of management for humanistic caring in outpatients.
5.Clinical trial of budesonide inhalation aerosol in the treatment of patients with AECOPD complicated with type Ⅱ respiratory failure

Objective To observe the clinical efficacy and safety of budesonide inhalation aerosol combined with non-invasive positive pressure ventilation(NIPPV)in the treatment of acute exacerbation of chronic obstructive pulmonary disease(AECOPD)combined with typeⅡ respiratory failure.Methods According to cohort method,patients with AECOPD and type Ⅱ respiratory failure were divided into treatment group and control group.The control group was given NIPPV(4 h/once,tid),while treatment group was given budesonide inhalation aerosol(344 μg/once,bid)on basis of control group.All patients were treated for 4 weeks.The clinical curative effect,disease severity,respiratory,pulmonary function indexes[peak expiratory flow(PEF),forced expiratory volume in one second(FEV1),vital capacity(VC)],serum inflammatory factors[interleukin(IL)-17,IL-6,soluble intercellular adhesion molecule(sICAM)-1]were compared,and evaluated the safety.Results There were 38 cases in control group and 42 cases in treatment group.After treatment,total response rates in treatment group and control group were 92.86%(39 cases/42 cases)and 76.32%(29 cases/38 cases),and the difference was significant(P<0.05).After treatment,scores of COPD assessment test(CAT)in treatment group and control group were(9.61±1.86)and(15.23±2.45)points;scores of modified british medical research council respiratory questionnaire(mMRC)were(1.74±0.31)and(2.43±0.35)points;PEF were(374.63±30.42)and(345.82±26.34)L·min-1;FEV,were(1.59±0.40)and(1.30±0.33)L;VC were(2.64±0.64)and(2.07±0.56)L;levels of serum IL-17 were(67.03±8.21)and(80.79±10.49)μg·L-1;IL-6 levels were(14.86±3.75)and(20.42±4.39)ng·L-1;sICAM-1 levels were(118.63±17.72)and(156.42±24.38)ng·mL-1,and the differences were statistically significant(all P<0.05).In treatment group,adverse drug reactions were mainly on palpitation,dizziness and dry mouth,while which were mainly on dizziness,dry mouth and nausea in control group.There was no significant difference in total incidence of adverse drug reactions between treatment group and control group[9.52%(4 cases/42 cases)vs 7.89%(3 cases/38 cases),P>0.05].Conclusion Budesonide inhalation aerosol combined with NNIPPV can effectively control disease severity,relieve dyspnea,improve pulmonary function and alleviate inflammatory response in patients with AECOPD and type Ⅱ respiratory failure,which has good safety.
6.Single-cell transcriptomic sequencing coupled with Mendelian randomization analysis elucidates the pivotal role of CTSC in chronic rhinosinusitis

Objective:To investigate the molecular mechanisms of chronic rhinosinusitis (CRS), to identify key cell subgroups and genes, to construct effective diagnostic models, and to screen for potential therapeutic drugs.Methods:Key cell subgroups in CRS were identified through single-cell transcriptomic sequencing data. Essential genes associated with CRS were selected and diagnostic models were constructed by hdWGCNA (high dimensional weighted gene co-expression network analysis) and various machine learning algorithms. Causal inference analysis was performed using Mendelian randomization and colocalization analysis. Potential therapeutic drugs were identified using molecular docking technology, and the results of bioinformatics analysis were validated by immunofluorescence staining. Graphpad Prism, R, Python, and Adobe Illustrator software were used for data and image processing.Results:An increased proportion of basal and suprabasal cells was observed in CRS, especially in eosinophilic CRS with nasal polyps (ECRSwNP), with P=0.001. hdWGCNA revealed that the "yellow module" was closely related to basal and suprabasal cells in CRS. Univariate logistic regression and LASSO algorithm selected 13 key genes ( CTSC, LAMB3, CYP2S1, TRPV4, ARHGAP21, PTHLH, CDH26, MRPS6, TENM4, FAM110C, NCKAP5, SAMD3, and PTCHD4). Based on these 13 genes, an effective CRS diagnostic model was developed using various machine learning algorithms (AUC=0.958). Mendelian randomization analysis indicated a causal relationship between CTSC and CRS (inverse variance weighted: OR=1.06, P=0.006), and colocalization analysis confirmed shared genetic variants between CTSC and CRS (PPH4/PPH3>2). Molecular docking results showed that acetaminophen binded well with CTSC (binding energy:-5.638 kcal/mol). Immunofluorescence staining experiments indicated an increase in CTSC +cells in CRS. Conclusion:This study integrates various bioinformatics methods to identify key cell types and genes in CRS, constructs an effective diagnostic model, underscores the critical role of the CTSC gene in CRS pathogenesis, and provides new targets for the treatment of CRS.
7.Colorimetric-Switch Sensing System Based on Cobalt,Nitrogen and Chlorine-doped Carbon Dots for Detection of Acid Phosphatase Activity

Element doping is one of the strategies for increasing the activity of enzyme-like materials.In this study,cobalt,nitrogen and chlorine-doped carbon dots(Co,N,Cl-CDs)were prepared by one-step hydrothermal synthesis utilizing CoCl2 and glutamate as raw ingredients.In weakly acidic environments,Co,N,Cl-CDs exhibited high peroxidase-like activity and could catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine(TMB)by hydrogen peroxide into oxided TMB(oxTMB),resulting in a distinct color change.Free radical scavenging and electron spin resonance techniques were used to further investigate the catalytic reaction mechanism of Co,N,Cl-CDs,and the reactive intermediates produced during the reaction were identified as hydroxyl radical.Acid phosphatase(ACP)could catalyze the hydrolysis of sodium ascorbyl phosphate to release ascorbic acid,which could reduce the blue oxTMB into colorless TMB.Based on this principle,a colorimetric-switching sensing system for detection of ACP was developed by coupling Co,N,Cl-CDs and ACP.The sensing system demonstrated good linearity in ACP activity range of 0.5-20 U/L,with a detection limit of 0.17 U/L.The developed sensing system was used to detection of ACP in real serum samples with good recoveries(97.1%-105.7%)and reproducibility(RSD<5%).The sensing system developed here based on Co,N,Cl-CDs had many advantages such as low detection cost,process visualization,and high sensitivity,providing reference for detection of ACP.
8.Assessment of grassroots Party branches in a tertiary public hospital

Objective This study aims to optimize the evaluation index system for the Party branch work of a hospital,grounded on Party regulations,and employ diversified assessment methods to integrate Party branch development with the advance-ment of medical,educational,and research endeavors.Methods This study utilized literature review and textual analysis to identi-fy relevant documents.Through qualitative interviews and convenience sampling,25 questionnaires were distributed to Party repre-sentatives at all levels within the hospital,yielding 25 valid responses.The 5-point Likert scale was applied to rate and assess the importance of 74 selected assessment indicators.Results A comprehensive evaluation system was established,which includes 5 pri-mary indicators,14 secondary indicators,and 53 tertiary indicators.Conclusion The existing comprehensive quantitative assess-ment system for Party branches has been optimized.The adoption of diversified evaluation methods has led to a clearer,more explic-it,and standardized objectives for Party branch construction,thereby enhancing its guiding and motivational role.
9.Role of pseudogenes in proliferation,invasion and metastasis of hepatoma cells

Pseudogenes have long been regarded as fossilized,nonfunctional genomes that are evidence of human evolution.However,in recent years,pseudogenes have been found to play an important role in both physiological and pathological processes,especially in tumors,regulating occurrence and development of tumors in parental gene dependence and parental gene independence,including liver cancer.In this paper,we summarized research progress of pseudogenes and their roles in proliferation,apoptosis,inva-sion and metastasis of liver cancer cells,providing new ideas for treatment of liver cancer.

The cell cycle is the series of events by which a cell divides into two daughter cells and is guided by a unique system of cell cycle regulation.The disorder of cell cycle regulation system is the fundamental cause of tumorigenesis.This paper will focus on the key regulatory points of cell cycle and discuss the research progress of tumor cell cycle regulation,so as to provide theore-tical basis for the development of new therapeutic strategies for tumors.
