1.Altered eotaxin-1 and interleukin-34 levels in obsessive-compulsive disorder: a case-control observational study in Bangladesh

Objectives:
Obsessive-compulsive disorder (OCD) is a prevalent mental health condition that impacts daily life. It is thought to be associated with genetic, biological, and structural brain changes, serotonergic abnormalities, altered neuromodulation, and environmental factors.Limited observational studies have examined cytokines in Bangladeshi patients with OCD. This study aimed to assess the levels of eotaxin-1 and interleukin (IL)-34 in individuals with this disorder.
Methods:
This case-control observational study included 58 patients with OCD and 30 healthy controls (HCs) matched for age, sex, and body mass index. The severity of OCD was assessed using the Yale-Brown obsessive-compulsive scale (Y-BOCS). Psychiatrists evaluated participants according to the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition. Serum levels of eotaxin-1 and IL-34 were measured using enzyme-linked immunosorbent assay kits.
Results:
Patients with OCD exhibited significantly higher serum eotaxin-1 levels (121.13 ± 7.84pg/mL) than HCs (85.52 ± 9.42 pg/mL). Conversely, IL-34 levels were considerably lower in patients than in HCs (119.02 ± 14.53 pg/mL vs. 179.96 ± 27.88 pg/mL). The Cohen d values for eotaxin-1 and IL-34 were 0.55 and −0.48, respectively. Among patients with OCD, a significantpositive correlation was found between serum eotaxin-1 level and Y-BOCS score, along with a negative correlation between serum eotaxin-1 and IL-34 levels.
Conclusion
The findings suggest that altered eotaxin-1 and IL-34 levels may be associated withOCD. These chemokines and cytokines could serve as primary tools for assessing the risk of OCD, warranting further clinical investigation. This could potentially support more extensive research and the development of diagnostic and therapeutic strategies targeting these pathways.

5.Prevalence and intersecting burdens of reproductive health morbidities among female fish farmers in the coastal region of Bangladesh

Background:Female fish farmers in coastal Bangladesh face significant reproductive health(RH)challenges due to occupational,nutritional,and sociocultural factors.However,their health burdens have been largely overlooked in labor and health policies.Objectives:This study aims to investigate the prevalence,severity,determinants,and emotional and relationship consequences of RH morbidities among female fish farmers in Shyamnagar,Satkhira,Bangladesh.Methods:A cross-sectional survey with structured questionnaires was conducted among 297 female fish farmers.Data were supplemented by rural appraisal activities.Descriptive statistics,chi-square tests,Poisson regression,and Structural Equation Modeling were used for analysis.Results:The study found that 59.60%(177/297)of participants suffered from moderate to severe uterine issues,remarkably menstrual irregularities(92.65%,164/177),backache(59.32%,105/177),pelvic and fatigue pain(42.37%,75/177),anemia(28.24%,50/177),with the"21-30"year age group bearing a higher physiological burden with greater relational and emotional disruptions than"31-40"year age group,and"41-50"year age group women.High exposure to occupational chemicals(81.82%,243/297)and lack of personal protective equipment(93.94%,279/297)were key contributors.Access to formal healthcare is limited due to financial hardships(85.86%,255/297),social stigma(51.52%,153/297),and inadequate health services(46.47%,138/297).Structural Equation Modeling results also revealed significant associations among RH morbidities,emotional distress,social isolation,and diminishing well-being.Conclusion:The study highlights the urgent need for integrated health policies addressing reproductive care,occupational safety,and mental health support for female fish farmers in coastal Bangladesh.Addressing these issues will improve their health,well-being,and resilience.

Background::Noncommunicable diseases (NCDs) are public health threats globally and recognized impediments to socioeconomic development. This study aimed to identify the prevalence and clustering of NCDs risk factors among Bangladeshi men and women aged 20-59 years using nationally representative data.Methods::This study was conducted in 82 rural, nonslum urban, and slum clusters across all eight administrative divisions of Bangladesh using multistage cluster sampling. A total of 4917 men and 4905 women aged 20-59 years were included in the study. Descriptive analyses were performed to report the prevalence and distribution of behavioral and clinical risk factors. Multivariable binary logistic regression was performed to identify factors associated with the coexistence of three or more NCD risk factors.Results::The prevalence of tobacco use (any form), insufficient physical activity, inadequate fruit and vegetable consumption, overweight and obesity, and central obesity were 38.3%, 13.6%, 87.1%, 42.3%, and 36.0%, respectively. Furthermore, 21.9% and 4.9% participants had hypertension and self-reported diabetes, respectively. Regarding the clustering of risk factors, 37.1% men and 50.8% women had at least three NCD risk factors. Only 3.0% men and 1.8% women reported no NCD risk factors. Age, place of residence, education, and wealth status were associated with the presence of at least three risk factors for both sexes.Conclusion::Since a large proportion of Bangladeshi 20-59 years old population had multiple risk factors, population-based programs with multisectoral approaches are essential to reduce NCDs among Bangladeshi women and men.

10.Bone marrow stem cells incubated with ellipticine regenerate articular cartilage by attenuating inflammation and cartilage degradation in rabbit model

Background:
Ellipticine (Ellip.) was recently reported to have beneficial effects on the differentiation of adipose-derived stem cells into mature chondrocyte-like cells. On the other hand, no practical results have been derived from the transplantation of bone marrow stem cells (BMSCs) in a rabbit osteoarthritis (OA) model.
Objectives:
This study examined whether autologous BMSCs incubated with ellipticine (Ellip.+BMSCs) could regenerate articular cartilage in rabbit OA, a model similar to degenerative arthritis in human beings.
Methods:
A portion of rabbit articular cartilage was surgically removed, and Ellip.+BMSCs were transplanted into the lesion area. After two and four weeks of treatment, the serum levels of proinflammatory cytokines, i.e., tumor necrosis factor α (TNF-α) and prostaglandin E2 (PGE2), were analyzed, while macroscopic and micro-computed tomography (CT) evaluations were conducted to determine the intensity of cartilage degeneration.Furthermore, immuno-blotting was performed to evaluate the mitogen-activated protein kinases, PI3K/Akt, and nuclear factor-κB (NF-κB) signaling in rabbit OA models. Histological staining was used to confirm the change in the pattern of collagen and proteoglycan in the articular cartilage matrix.
Results:
The transplantation of Ellip.+BMSCs elicited a chondroprotective effect by reducing the inflammatory factors (TNF-α, PGE2) in a time-dependent manner. Macroscopic observations, micro-CT, and histological staining revealed articular cartilage regeneration with the downregulation of matrix-metallo proteinases (MMPs), preventing articular cartilage degradation. Furthermore, histological observations confirmed a significant boost in the production of chondrocytes, collagen, and proteoglycan compared to the control group. Western blotting data revealed the downregulation of the p38, PI3K-Akt, and NF-κB inflammatory pathways to attenuate inflammation.
Conclusions
The transplantation of Ellip.+BMSCs normalized the OA condition by boosting the recovery of degenerated articular cartilage and inhibiting the catabolic signaling pathway.
