1.Effects of CREB and miR-132-3p on aluminum-induced abnormal phosphorylation of tau protein in rat hippocampus

Background Cyclic AMP response element binding protein (CREB) and miR-132-3p have been proved to be related to many neurodegenerative diseases. Our research group previously has demostrated that the neurotoxicity of aluminum is relevant to abnormal phosphorylation of tau protein, but whether aluminum affects the abnormal phosphorylation of tau protein through GREB and miR-132-3p has not been reported yet . Objective To investigate the effect of aluminum on CREB and miR-132-3p during abnormal phosphorylation of tau protein in rat hippocampus. Methods Twenty-eight two-month-old SD rats with comparable weigh, were randomly assigned to four groups: control group (saline) and low, middle, and high dose exposure groups [10, 20, and 40 μmol·kg−1 Al(mal)3] with each group containing 7 rats, and the exposure period was 3 months by intraperitoneal injection every other day. After rats’ exposure to aluminum, Morris water maze was employed to assess their capabilities of learning and memory. The miR-132-3p gene expression level was detected by quantitative real-time PCR (qRT-PCR). The levels of CREB, phosphorylated CREB (p-CREB) (Ser133), RAS p21 protein activator 1 (RASA1) tau, and p-tau (Ser396) proteins were determined by Western blot. Results The results of Morris water maze showed that in the navigation experiment (from first day to the fifth day), the average escape latency of the rats exposed to three doses of aluminum was longer than that of the control rats (P<0.05). The middle dose group and the high dose group demonstrated shorter duration and lower frequency of platform traversal in the designated quadrant when compared to the control group and the low dose group (P<0.05). Moreover, the duration in the target quadrant of the rats exposed to high dose aluminum was shorter than that of the rats exposed to medium dose aluminum (P<0.05). The results of Morris water maze suggested that aluminum could damage the learning and memory ability of rats. The qRT-PCR findings indicated a decline in miR-132-3p gene expression in rat hippocampus correlating with higher Al(mal)3 dose (P<0.05). The Western blot test showed that the protein expressions of CREB and p-CREB (Ser133) were reduced in both the middle dose group and the high dose group (P<0.05) when compared to the control group and the low dose group, and likewise, compared to the control group, the group receiving low dose exhibited lower level of p-CREB (Ser133) protein expression (P<0.05). It was found that the further increase of aluminum exposure dose would lead to the further decrease of CREB and p-CREB (Ser133) protein expression levels (F=36.429, P<0.001; F=78.672, P<0.001), aluminum exposure dose was negatively correlated with the expression levels of the two proteins (r=−0.848, P<0.001; r=−0.928, P<0.001). The expression levels of RASA1 protein and tau protein in the aluminum exposure groups surpassed those in the control group (P<0.05). The tau protein phosphorylation level was higher in the middle dose group than in the control group (P<0.05), while the high dose group showed elevated phosphorylation level relative to the control group, the low dose group, and the middle dose group (P<0.05). Conclusion Aluminum may promote abnormal phosphorylation of tau protein by affecting CREB and miR-132-3p, which eventually leads to the impairment of learning and memory ability.
2.Application of nursing cooperation method in pulmonary nodule localization using indocyanine green combined with methylene blue

Objective To discuss the nursing cooperation method in CT-guided pulmonary nodule localization using indocyanine green combined with methylene blue.Methods A total of 81 patients,who needed to receive pulmonary nodule localization before thoracoscopic resection,were enrolled in this study.The nursing cooperation measures,including the preparation and injection of indocyanine green combined with methylene blue,breathing mode coordination,position management,precise localization coordination,and psychological care,were analyzed.Results Successful nodule localization was accomplished in all patients.No statistically significant difference in the incidence of adverse events existed between the patients with different clinical characteristics(P>0.05).The gender,age,lesion site,nodule size,nodule-pleura distance,posture during puncturing and needle path route were not the factors influencing the occurrence of adverse reactions in patients receiving CT-guided pulmonary nodule localization using indocyanine green combined with methylene blue.Conclusion In performing CT-guided pulmonary nodule localization using indocyanine green combined with methylene blue,the nursing cooperation method carries generality and stability,which is suitable for patients with different clinical characteristics.
4.Knowledge, attitude and practice and training needs of posture management of premature infants among nurses in Neonatal Intensive Care Unit

Objective:To explore the status quo of knowledge, attitude and practice in the posture management of premature infants among nurses in Neonatal Intensive Care Unit (NICU) and related training needs.Methods:This study is a cross-sectional study. From March to April 2022, 207 NICU nurses from 6 ClassⅢ Grade A hospitals in Shandong Province were selected by convenient sampling. The nurses were investigated online by using the self-developed General Information Questionnaire, Premature Infant Posture Management Knowledge, Attitude and Practice Questionnaire for NICU Nurses and Premature Infant Posture Management Training Needs Questionnaire for NICU Nurses. Multiple linear regression was used to analyze the influencing factors of NICU nurses' knowledge, attitude and practice in the posture management of premature infants. A total of 207 questionnaires were distributed, and 201 valid questionnaires were recovered, with an effective recovery rate of 97.10% (201/207) .Results:Among 201 NICU nurses, the scores of premature infant posture management knowledge, attitude, practice and total score were (59.12±7.82) , (33.10±3.09) , (54.92±7.36) and (147.14±15.15) respectively. Multiple linear regression analysis results show that the nature of the working hospital, whether they received relevant training, and the importance of participating in relevant training were the influencing factors of NICU nurses' total score of knowledge, attitude and practice in the posture management of premature infants ( P<0.05) . 78.11% (157/201) of NICU nurses accepted the training of premature infant posture management, 98.01% (197/201) of NICU nurses were willing to accept the training of premature infant posture management, and 91.04% (183/201) of NICU nurses thought it was very important to participate in the training of premature infant posture management. NICU nurses believed that the main obstacle affecting the quality of posture management of premature infants was the lack of relevant knowledge. The most desired training content was the effect of different postures on premature infants. Conclusions:NICU nurses have a high level of knowledge, attitude and practice in posture management of premature infants. Managers should strengthen the training of NICU nurses in the posture management of premature infants, improve the ability of NICU nurses in the posture management of premature infants, and promote the improvement of the nursing quality of premature infants.
5.Efficacy and Mechanism of Shenbai Jiedu Prescription Against Proliferation of HCT116 Cells

ObjectiveTo investigate the mechanism by which Shenbai Jiedu prescription (SBJDF) inhibits the proliferation of colorectal cancer (CRC) HCT116 cells. MethodAfter 48 h treatment of HCT116 cells with SBJDF (0, 0.25, 0.5, 1, 2, 4 g·L-1), the viability of HCT116 cells were determined by methyl thiazolyl tetrazolium (MTT) colorimetry. Following the classification of cells into blank control group and SBJDF (1, 2, 4 g·L-1) groups, the effect of SBJDF on HCT116 cell morphology was observed under an inverted microscope. The effects of SBJDF on the proliferation of HCT116 cells and mitochondrial membrane potential (Δψm) were detected by colony formation assay and JC-1 probe, respectively. The flow cytometry was then performed for determining cell cycle distribution and apoptosis. The effects of SBJDF on cell cycle-, apoptosis-, and nuclear factor kappa-B (NF-κB) signaling pathway-related proteins were determined by Western blot. ResultSBJDF effectively inhibited the vitality of HCT116 cells and changed their morphology in a concentration-dependent manner. Compared with the blank control group, SBJDF at 1, 2, 4 g·L-1 significantly reduced cell colony formation (P<0.05, P<0.01),and SBJDF at 2 and 4 g·L-1 arrested the HCT116 cell cycle at G0/G1 phase (P<0.05, P<0.01). Compared with the blank control group, SBJDF at 1, 2, 4 g·L-1 remarkably down-regulated the protein expression of CyclinD1 (P<0.05, P<0.01). SBJDF at 2 and 4 g·L-1 lowered the CyclinA2 and cyclin-dependent kinase 4 (CDK4) (P<0.05, P<0.01). SBJDF at 4 g·L-1 reduced the cyclin-dependent kinase 1 (CDK1) (P<0.01). Compared with the blank control group, SBJDF at 2 and 4 g·L-1 induced HCT116 cell apoptosis, down-regulated the protein expression of anti-apoptosis-related proteins Bcl-2 and Bcl-xl as well as the NF-κB signaling pathway-related proteins IκB kinase α (IKKα),inhibitor α of NF-κB (IκBα),and phospho-NF-κB p65 (p-p65) (P<0.05, P<0.01), and diminished the mitochondrial membrane potential of HCT116 cells. ConclusionSBJDF inhibits the proliferation of HCT116 cells, which may be related to its inhibition of the activation of NF-κB signaling pathway and the induction of cell cycle arrest and apoptosis.
6.Shenbai Jiedu Prescription Inhibits Proliferation of Colorectal Cancer Cells by Regulating PTEN/PI3K/Akt Signaling Pathway

ObjectiveTo study the mechanism of Shenbai Jiedu prescription inhibiting the proliferation of HCT116 colorectal cancer (CRC) cells by regulating the phosphatase and tensin homolog deleted on chromosome ten (PTEN)/phosphatidylinositol 3-kinase (PI3K)/ protein kinase B (Akt) signaling pathway. MethodShenbai Jiedu prescription was extracted by water extraction and alcohol precipitation to prepare freeze-dried powder. HCT116 cells were cultured in vitro, and treated with different concentrations of Shenbai Jiedu prescription (2, 4, 8, 16 g·L-1). The inhibitory effect of Shenbai Jiedu prescription on the proliferation of HCT116 cells was tested by methyl thiazolyl tetrazolium (MTT). Real-time quantitative PCR was used to detect the mRNA expression levels of PTEN, PI3K, Akt, glycogen synthase kinase-3β (GSK-3β), c-Myc, survivin and Cyclin D1. Western blot was employed to measure the protein expression levels of PTEN, phosphorylated PTEN (p-PTEN), PI3K, Akt, phosphorylated Akt (p-Akt), GSK-3β, phosphorylated GSK-3β (p-GSK-3β), c-Myc, survivin and Cyclin D1, β-catenin nuclear import was explored by immunofluorescence assay. ResultCompared with the control group, Shenbai Jiedu prescription inhibited the proliferation of HCT116 cells in a dose-dependent manner (P<0.01). Compared with the control group, the mRNA expression levels of PTEN and GSK-3β were up-regulated whereas those of PI3K, Akt, c-Myc, survivin and CyclinD1 were down-regulated after treatment with Shenbai Jiedu prescription (P<0.01). The protein expression levels of PTEN, p-PTEN and GSK-3β were up-regulated whereas those of PI3K, Akt, p-Akt, GSK-3β, p-GSK-3β, c-Myc, survivin and CyclinD1 were down-regulated (P<0.05, P<0.01). Immunofluorescence assay showed that Shenbai Jiedu prescription suppressed β-catenin nuclear import in HCT116 cells. ConclusionShenbai Jiedu prescription inhibited the proliferation of HCT116 cells via the mechanism of regulating the PTEN/PI3K/Akt signaling pathway.
7.Effect of Different Drying Methods on Volatile Components in Pseudostellariae Radix Based on GC-IMS

ObjectiveTo compare the effects of different drying methods on volatile components of Pseudostellariae Radix. MethodThe samples were dried by different methods, including air drying, sun drying, hot air drying (40, 60, 80 ℃) and vacuum freeze drying. Gas chromatography-ion mobility spectrometry (GC-IMS) was used to compare the changes of volatile components in the samples after different treatments. The samples were incubated at 80 ℃ and 500 r·min-1 for 15 min, the injection temperature was 85 ℃, the injection volume was 200 μL, the flow rate of carrier gas was from 2 mL to 150 mL during 20 min, and the temperature of IMS detector was 60 ℃. SE-54 capillary column (0.32 mm×30 m, 0.25 μm) was used, the column temperature was 60 ℃, and the analysis time was 35 min. The differential spectra of volatile components were constructed and analyzed by principal component analysis (PCA). ResultA total of 37 volatile components were identified from dried Pseudostellariae Radix. The number of compounds in descending order was ketones, aldehydes and alcohols. There were some differences in the volatile components in samples dried by different methods. And the volatile components in samples with sun drying, air drying and hot air drying at 40 ℃ were similar, compared with other drying methods, vacuum freeze drying and hot air drying at 80 ℃ had great effects on the volatile components of Pseudostellariae Radix, and the compounds in the samples with vacuum freeze drying were the least. ConclusionIn this study, GC-IMS for the detection and analysis of volatile components in Pseudostellariae Radix is established, which has the characteristics of high efficiency, nondestructive inspection and simple sample processing. This method can be used for the distinction of Pseudostellariae Radix dried by different methods. And hot air drying at 40 ℃ can effectively retain the volatile components of Pseudostellariae Radix, and achieve similar flavor to samples with sun drying and air drying.
8.A Deep Mesencephalic Nucleus Circuit Regulates Licking Behavior.

Licking behavior is important for water intake. The deep mesencephalic nucleus (DpMe) has been implicated in instinctive behaviors. However, whether the DpMe is involved in licking behavior and the precise neural circuit behind this behavior remains unknown. Here, we found that the activity of the DpMe decreased during water intake. Inhibition of vesicular glutamate transporter 2-positive (VGLUT2+) neurons in the DpMe resulted in increased water intake. Somatostatin-expressing (SST+), but not protein kinase C-δ-expressing (PKC-δ+), GABAergic neurons in the central amygdala (CeA) preferentially innervated DpMe VGLUT2+ neurons. The SST+ neurons in the CeA projecting to the DpMe were activated at the onset of licking behavior. Activation of these CeA SST+ GABAergic neurons, but not PKC-δ+ GABAergic neurons, projecting to the DpMe was sufficient to induce licking behavior and promote water intake. These findings redefine the roles of the DpMe and reveal a novel CeASST-DpMeVGLUT2 circuit that regulates licking behavior and promotes water intake.
9.Application of image similarity measure based on structure information and intuitionistic fuzzy set in radiotherapy setup verification

Objective:To propose a method of image similarity measurement based on structure information and intuitionistic fuzzy set and measure the similarity between CT image and CBCT image of radiotherapy plan positioning, aiming to objectively measure the setup errors.Methods:A total of four pre-registration images of a nasopharyngeal carcinoma patient on the cross-sectional and sagittal planes and a pelvic tumor patient on the cross-sectional and coronal planes were randomly selected. Five methods were used to quantify the setup errors, including correlation coefficient, mean square error, image joint entropy, mutual information and similarity measure method.Results:All five methods could describe the deviation to a certain extent. Compared with other methods, the similarity measure method showed a stronger upward trend with the increase of errors. After normalization, the results of five types of error increase on the cross-sectional plane of the nasopharyngeal carcinoma patient were 0.553, 0.683, 1.055, 1.995, 5.151, and 1.171, 1.618, 1.962, 1.790, 3.572 on the sagittal plane, respectively. The results of other methods were between 0 and 2 after normalization, and the results of different errors of the same method slightly changed. In addition, the method was more sensitive to the soft tissue errors.Conclusions:The image similarity measurement method based on structure information and intuitionistic fuzzy set is more consistent with human eye perception than the existing evaluation methods. The errors between bone markers and soft tissues can be objectively quantified to certain extent. The soft tissue deviation reflected by the setup errors is of significance for individualized precision radiotherapy.
10.The effect of butylphthalide injection combined with interventional therapy on patients with acute anterior circulation large vessel occlusive stroke

Objective To investigate the effect of butylphthalide and sodium chloride injection on patients who received endovascular treatment for acute anterior circulation large vessel occlusive stroke.Methods A total of 173 patients were identified from February 2015 to December 2017 in the Department of Neurology of Jingling Hospital in this retrospective observational study.Propensity score-matching analysis was performed to balance differences in baseline characteristics between patients who received butylphthalide injection (butylphthalide group) and those who did not (control group).The modified Rankin Scale scores at 90 days were compared between the butylphthalide and control groups.Results A total of 144 patients who received endovascular treatment for acute anterior circulation large vessel occlusive stroke were finally analyzed,54 cases in the butylphthalide group and 90 cases in the control group.The proportion of good functional outcome at 90 days in the butylphthalide group was higher than that in the control group (63.0% (34/54) vs 44.4% (40/90);x2=4.633,P=0.031).Thirty-six pairs were matched successfully by the propensity score matching,36 patients in the butylphthalide group and 36 in the control group.There was no statistically significant difference in the 90-day functional outcome between the two groups (66.7% (24/36) vs 44.4% (16/36);x2=3.600,P=0.058).One hundred and fifteen patients were recanalized,47 cases in the butylphthalide group and 68 cases in the control group,and after the propensity score matching,30 pairs were analyzed.The proportion of good functional outcome at 90 days in the butylphthalide group was higher than that in the control group (73.3% (22/30) vs 46.7% (14/30);x2=4.444,P=0.035).Conclusion After propensity score-matching,butylphthalide and sodium chloride injection could improve 90-day functional outcome in patients with acute anterior circulation large vessel occlusive stroke and obtained recanalization by endovascular treatment while could not before propensity score-matching.
