Objective: To investigate the expression and functional role of FK506 binding protein 10 (FKBP10) in oral squamous cell carcinoma (OSCC), and to provide a research basis for the estimated prognosis and targeted therapy of OSCC. Methods: A total of 284 OSCC samples and 19 normal samples were selected from the Cancer Genome Atlas (TCGA) database, and diagnostic analysis was performed to determine mRNA expression. Survival analysis for FKBP10 and OSCC was conducted on a gene expression profile interaction analysis website. Real-time fluorescence quantitative PCR and Western Blot were used to detect the mRNA and protein expression of FKBP10 in four OSCC cell lines and SAS and SCC9 cells transfected with siRNA. The cell proliferation ability of FKBP10-silenced cells was detected using the CCK8 method, and the cell cycle distribution and apoptosis were detected by flow cytometry. Cell migration and invasion ability were detected through wound healing and invasion experiments. The expression changes of total protein and phosphatidylinositol 3-kinase (PI3K)-serine/threonine kinase (AKT) after FKBP10 silencing were analyzed by proteomics and Western Blot. Results: According to the analysis of gene expression levels, the mRNA expression level of FKBP10 in OSCC was significantly higher than that in normal tissues (P < 0.001). In terms of diagnosis, the expression level of FKBP10 has unique diagnostic value for OSCC (P < 0.05). The survival analysis of FKBP10 and OSCC showed that a high expression of FKBP10 led to a decrease in patient survival and poor prognosis (P < 0.05). The expression of FKBP10 mRNA and protein in OSCC cell lines was higher than that in normal oral keratinocytes (P < 0.001). Silencing FKBP10 can reduce the proliferation, invasion, and migration ability of SAS and SCC9 (P < 0.001), and also block their cell cycle in the G0/G1 phase (P < 0.001), with a significant increase in apoptosis (P < 0.05). Protein mass spectrometry and Western blot analysis revealed that FKBP10 silencing significantly downregulated the expression of multiple proteins in the RAP1 signaling pathway, mainly RAP guanine nucleotide exchange factor 1 (RAPGEF1) (P < 0.05) and the phosphorylation of PI3K-AKT proteins (P < 0.05). Conclusion FKBP10 is highly expressed in OSCC, leading to poor prognosis for patients. Downregulated FKBP10 expression can inhibit the proliferation, migration, and invasion ability of OSCC cells, hinder cell cycle progression, and promote apoptosis via the RAP1-PI3K-AKT axis. FKBP10 is a potential therapeutic target and prognostic biomarker for OSCC.

The synthetic PDCoV N protein gene was optimized and cloned into the pET-30a vector to obtain the pET-30a-N plasmid.Thenthe recombinant plasmid was transformed into three strains of BL21 E.coli using heat-shock to explore protein expression conditions.The expressed proteins was purified using Ni Focurose 6FF(IMAC)and used as antigen to immunize the New Zealand White rabbit to prepare the polyclonal antibody against the PDCoV N protein.The antibody titer was measured by indirect ELISA method.The specificity for the antibody was identified by West-ern blot and indirect immunofluorescence(IFA).The results showed that the pET-30a-N plasmid showed high expression level in BL21 StarTM(DE 3).The optimal expression condition was 37 ℃ 4 h.The purity of the target protein could reach 90.3％after purification.Indirect ELISA showed that the antibody titers was up to 1∶204 800.Western blot and IFA showed that the produced rabbit polyclonal antibody exhibited good specificity.In conclusion,the polyclonal antibody was prepared which specifically recognized the PDCoV N proteins.The results provided some references for the subsequent exploration of PDCoV N protein function and laid a foundation for establishing a diag-nostic method for PDCoV.

Objective To explore the relationship between serum levels of glycoprotein fibroglycan(SRGN)and secretory CD146(sCD146)and clinicopathological characteristics and prognosis in elderly pa-tients with breast cancer(BC).Methods A total of 128 elderly BC patients treated in the hospital from April 2019 to April 2021 were retrospectively selected as the BC group,and another 70 healthy elderly women were selected as the control group.The serum SRGN and sCD146 levels in BC group and control group,and patients with different clinical and pathological characteristics in BC group were compared.Kaplan-Meier curve and COX regression were used to analyze the effect of serum levels of SRGN and sCD146 on the prognosis of eld-erly BC patients.Receiver operating characteristic curve was used to analyze the predictive value of serum lev-els of SRGN and sCD146 for the prognosis of elderly BC patients.Results The levels of serum SRGN and sCD146 were(13.17±3.35)μg/L,(118.23±20.51)ng/L in the BC group,which were higher than(2.52±0.41)μg/L,(20.03±4.16)ng/L in the control group(t=26.460,39.572,both P<0.001).Compared with elderly BC patients with TNM stage Ⅰ-Ⅱ and histological grade Ⅰ-Ⅱ,serum SRGN and sCD146 levels in elderly BC patients with TNM stage Ⅲ and histological grade Ⅲ were higher(P<0.05).The 3-year progres-sion free survival rates in SRGN high and low expression groups were 66.13%(41/62)and 92.42%(61/66),respectively,and the 3-year progression free survival rate in SRGN high expression group was lower than that in SRGN low expression group(Log Rank χ2=14.180,P<0.001).The 3-year progression free survival rates in sCD146 high and low expression groups were 68.85%(42/61)and 89.55%(60/67),respectively,and the 3-year progression free survival rate in sCD146 high expression group was lower than that in sCD146 low ex-pression group(Log Rank χ2=8.614,P=0.003).TNM stage Ⅲ,histological grade Ⅲ,serum SRGN and sCD146 were risk factors for the poor prognosis of elderly BC patients.The area under the curve(AUC)of the combination of serum SRGN and sCD146 for predicting the prognosis of elderly BC patients was 0.850,which was larger than that of SRGN(AUC=0.798)and sCD146(AUC=0.771)alone(Z=2.057,2.217,P=0.042,0.029).Conclusion Serum levels of SRGN and sCD146 are elevated in elderly BC patients,and both are in-volved in the disease progression of BC.The combination of the two factors has high predictive value for the prognosis of elderly BC patients.

Objective:To investigate the risk factors for detecting clinically significant prostate cancer (CSPCa) in patients with Prostate Imaging Reporting and Data System (PI-RADS) score≤3 on multi-parameter magnetic resonance imaging (mpMRI).Methods:Retrospective analysis was performed on the case data of 335 patients with suspected prostate cancer and PI-RADS score ≤3 who were admitted to Beijing Friendship Hospital, Capital Medical University from January 2013 to October 2022. All patients underwent 24-needle prostate biopsy. Clinical data such as age, body mass index, past medical history, serological laboratory indicators, and mpMRI imaging data were collected. The patients were grouped according to whether the puncture pathology was CSPCa or not, and the differences in clinical data between the two groups were analyzed by t-test, rank sum test and Chi-test. Multivariate Logistic regression analysis was further used to determine independent risk factors for MRI invisible prostate cancer, and receiver operating characteristics (ROC) curves were drawn. At the same time, further subgroup analysis was conducted based on whether prostate-specific antigen (PSA) was positive before puncture and PI-RADS score, respectively, and the same statistical method was used to further determine the influence of different serological indicators and PI-RADS score on the analysis results of risk factors. Results:Among all patients, 81 were CSPCa patients and 254 were non-CSPCa patients. Multivariate Logistic regression analysis showed that prostate-specific antigen density (PSAD) and PI-RADS score of 3 were independent risk factors for MRI invisible prostate cancer. At the same time, compared with suspected lesions located only in the transitional zone, the incidence of CSPCa in patients with suspected lesions located in the peripheral zone would increase, and the incidence of CSPCa would further increase when suspected lesions were found in both the transitional zone and the peripheral zone. In PSA-negative patients, only suspected lesion location was an independent risk factor for MRI invisible prostate cancer, while in PSA-positive patients, prostate volume, PSAD, and PI-RADS scores were independent risk factors. In subgroup analysis with different PI-RADS scores, suspicious lesions in both the transitional zone and peripheral zone indicate a higher likelihood of CSPCa. For patients with PI-RADS scores of 1 to 2, suspicious lesions in the peripheral zone alone may also indicated CSPCa, while for patients with PI-RADS scores of 3, the lower free prostate-specific antigen/total prostate-specific anti-principle was more accurate in predicting CSPCa.Conclusions:For patients who are clinically suspected of prostate cancer but whose PI-RADS score is less than or equal to 3 points indicated by mpMRI, it is necessary to further focus on the results of different serological indicators according to whether their PSA is positive and PI-RADS score respectively to judge whether patients should receive systemic prostate puncture, instead of using PSA level as a single indication for puncture. At the same time, clinicians should also pay full attention to the location of suspected lesions, when they are located in the peripheral zone, or there are suspected lesions in both the peripheral zone and the transitional zone, the possibility of CSPCa should be fully considered.

Objective:To explore the independent risk factors for chemoresistance during gemcitabine plus cisplatin (GC) adjuvant chemotherapy in patients with locally advanced bladder cancer after radical cystectomy and to construct a related predictive model.Methods:The clinical data of 228 patients with locally advanced bladder cancer who received GC chemotherapy after radical cystectomy at Beijing Friendship Hospital, Capital Medical University, from January 2013 to June 2024 were retrospectively analyzed. Among them, 184 were males, and 44 were females, with an average age of (68.8±10.6)years and an average body mass index (BMI) of (24.2±3.6)kg/m 2. According to tumor progression during chemotherapy, patients were divided into a chemotherapy-resistant(CR) group ( n=59) and a non-chemotherapy-resistant(NCR) group ( n=169). Independent sample t-test, chi-square test, and non-parametric test were used to compare general clinical characteristics and relevant examination results during chemotherapy between the two groups. Multivariate linear regression analysis was used to identify independent risk factors for GC chemoresistance. Propensity score matching (PSM) was used to match the TNM stage data between the two groups, and Kaplan-Meier and log-rank tests were used to compare overall survival(OS)after matching. Results:The median number of chemotherapy cycles was 3 in the CR group and 4 in the NCR group. Compared with the NCR group, CR patients were younger [(66.3±9.4) years vs.(69.7±10.9)years], had a higher proportion of kidney transplantation history[6.8%(4/59) vs. 0.6%(1/169)], hypertension [50.8%(30/59) vs. 36.1%(61/169)], coronary heart disease[23.7%(14/59) vs.9.5% (16/169)], and hydronephrosis [13.6%(8/59) vs. 4.1%(7/169)](all P<0.05). CR patients had a higher proportion of T 4 stage [20.3% (12/59) vs. 5.9% (10/169)], N 2 stage [42.4% (25/59) vs. 8.3% (14/169)], multifocal tumors at initial diagnosis [59.3% (35/59) vs. 26.6% (45/169)], and larger maximum tumor diameter [2.5 (1.5, 3.4) cm vs. 1.6 (1.2, 2.5) cm] (all P < 0.05). The CR group showed higher proportions of long-term urinary tract infection (UTI) [90.1% (53/59) vs. 7.7% (15/169)], higher systemic immune-inflammation index (SII) [991.6 (451.0, 1577.9) vs. 462.8 (309.0, 766.7)], absolute neutrophil count [6.5(4.1, 7.8)× 10 9/L vs. 3.9 (2.9, 5.1)× 10 9/L], and platelet count [(220.0 ± 96.2)× 10 9/L vs. (191.0 ± 64.8)× 10 9/L], but lower albumin levels [(34.3 ± 4.2) g/L vs. (39.9 ± 3.8) g/L] and albumin-to-globulin ratio (A/G) [(1.2 ± 0.3) vs. (1.3 ± 0.2)] (all P < 0.05). Multivariate linear regression analysis identified only T stage and long-term UTI as independent risk factors for GC chemoresistance( P<0.05).The probability of GC chemoresistance in bladder cancer patients was calculated as: P(Chemoresistance)=[0.155×T stage+ 0.624×(long-term UTI)]×100%(long-term UTI = 1 if present during chemotherapy, otherwise=0). After PSM, survival analysis showed that the median OS was significantly higher in the NCR group (55 months) than that in the CR group (30 months) ( P=0.020). Conclusions:This study demonstrates that advanced T stage and persistent UTI are independent risk factors for GC chemotherapy resistance in locally advanced bladder cancer patients. Based on these findings, a predictive model for chemotherapy resistance probability was constructed using multivariate linear regression analysis.

Objective:To design a systematic emotion management training (EMT) targeted the characteristics of coping strategies based on rehabilitation assistant and explore the feasibility of online EMT in improving cognitive coping strategies, mental health, and social support of patients with coronary heart disease(CHD).Methods:From June 2021 to December 2022, a total of 106 CHD patients were screened from cardiovascular units of a hospital in Zhengzhou. All participants were divided into the intervention group ( n=53) and control group ( n=53) using the coin toss method. The patients in intervention group received 7 weeks emotion management training on the basis of routine health education, the patients in control group received 7 weeks routine health education. The scores of cognitive coping strategies, anxiety, depression, and social support were measured at baseline and post-intervention. Statistical analysis was conducted using SPSS 24.0 software.The cognitive coping strategies, mental health and social support scores between two groups were compared using independent sample t-tests or paired t-test. Pearson analysis was used to examine the correlation between cognitive coping strategies, mental health and social support. Results:The scores of adaptive coping strategies, maladaptive coping strategies, depression, anxiety, insomnia and social support in patients with CHD were (69.52±7.60), (35.22±6.15), (8.82±2.66), (8.78±1.99), (10.97±2.86), and (57.48±9.79), respectively. After intervention, the scores of maladaptive coping strategies, self-blame, rumination, catastrophizing, blaming others, putting into perspective, insomnia, anxiety, and depression in the intervention group were significantly lower than those in the control group( t=-7.742, -4.395, -4.781, -6.105, -6.256, -5.327, -6.017, -7.288, -7.749, all P<0.05). The scores of adaptive coping strategies, positive reappraisal, refocusing on planning and social support were significantly higher than those of the control group( t=7.314, 6.733, 5.874, 3.562, all P<0.05). In the intervention group, there were statistically significant differences in cognitive coping strategies, anxiety, depression, insomnia and social support scores before and after the test(all P<0.05) and they were not statistically significantly different in the control group(all P>0.05). Correlation analysis showed that maladaptive coping strategies were positively correlated with depression and anxiety scores ( r=0.421, 0.408, both P<0.05). Adaptive coping strategies were negatively correlated with depression and insomnia scores ( r=-0.225, -0.240, both P<0.05), and positively correlated with social support ( r=0.219, P=0.034) among patients with CHD. Conclusion:The online EMT based on rehabilitation assistant may be a useful intervention for patients with CHD which can promote psychological health, social support as well as positively transforming maladaptive coping strategies into adaptive coping strategies.

Objective: To explore the quality of life and mental health status of adolescents in Zhengzhou, and to compare with HongKong and Taiwan. Methods: A total of 6 401 students from 12 primary and secondary schools in Zhengzhou City. A total of 3 642 students from HongKong and 1 547 students from Taiwan were selected by cluster sampling. And Padiatric Quality of Life Inventory Version 4.0, Self-Esteem Scale, General Self-efficacy Scale, Depression Anxiety Stress Scale and self-made general situation questionnaire were used to conduct questionnaire survey. Results: The total score of quality of life and the scores of each dimension in Zhengzhou were significantly higher than those in HongKong, while self-esteem and anxiety were lower than those of Taiwan adolescents（P<0.05). In addition to self-esteem, anxiety and stress, the scores of quality of life and mental health of adolescents of different grades and genders in Zhengzhou were statistically different(t=13.53，20.71，10.92，20.26，14.68，-16.03，21.26；6.16，3.81，-2.22，-0.33，8.76，4.16，2.71，P<0.01). The quality of life of adolescents in HongKong and Taiwan in different grades and genders were basically the same as those in Zhengzhou, and the differences of depression and stress scores in grades were the same as those in Zhengzhou. Conclusion The overall quality of life and mental health of adolescents in Zhengzhou is better than that in Hong Kong and Taiwan. It is necessary to explore the relationship between the quality of life and mental health of adolescents in order to improve their quality of life.

Objective:To explore the current situation of emotional and behavioral problems in children with hematological tumors, and the relationship with caregivers′ coping styles and family cohesion.Methods:Using cross-sectional study design and cluster sampling method, caregivers of children with hematological tumors hospitalized in hematology department of five third-class hospitals in Zhengzhou from November 2020 to January 2021 were selected as the respondents. The strengths and difficulties questionnaire (parent version), simple coping style questionnaire, family cohesion scale and self-made general information questionnaire were used for questionnaire survey. Statistical analysis was performed by SPSS 24.0 software using Spearman correlation analysis and multivariate Logistic regression analysis.Results:A total of 237 questionnaires were distributed and 214 valid questionnaires were collected. The abnormal detection rates of total difficulty score, emotional symptoms, conduct problems, hyperactivity, peer interaction problems and prosocial behavior were 17.8%, 22.9%, 10.7%, 8.4%, 39.7% and 13.6% respectively. The results of correlation analysis showed that caregivers′ positive coping was negatively correlated with the total score of difficulties, hyperactivity and peer interaction problems ( r=-0.186, -0.153, -0.174, all P<0.05), and positively correlated with the score of prosocial behavior ( r=0.214, P<0.05). Caregivers′ negative coping was positively correlated with the total score of difficulties, emotional symptoms and hyperactivity ( r=0.203, 0.204, 0.170, all P<0.05). Family cohesion was negatively correlated with the total score of difficulties, emotional symptoms, conduct problems, hyperactivity and peer interaction problems ( r=-0.254, -0.225, -0.183, -0.137, -0.195, all P<0.05), and positively correlated with prosocial behavior ( r=0.235, P<0.01). Multivariate Logistic regression analysis showed that positive coping of caregivers was a protective factor of peer interaction in children ( β=-0.050, OR=0.951, 95% CI=0.907-0.996), while family intimacy was also a protective factor for children with prosocial behavior ( β=-0.045, OR=0.956, 95% CI=0.923-0.991). Conclusion:The emotional and behavior problems of children with hematological tumors are serious, and the positive coping of caregivers and family cohesion have certain predictive value for the occurrence of negative emotional behavior problems in children, so medical staff should take corresponding measures to reduce the occurrence of emotional and behavior problems in children.
