1.Expression and clinical significance of CLDN6,TRIM59 and CMTM6 in nasopharyngeal carcinoma

Objective To investigate the expression and clinical significance of Claudin-6(CLDN6),tripartite motif-containing protein 59(TRIM59)and chemokine-like factor-like MARVEL transmembrane domain containing member 6(CMTM6)in nasopharyngeal carcinoma(NPC)tissue.Methods A total of 135 NPC patients were selected as the study objects,and cancer tissue(observation group)and para-cancer tissue(control group)of all patients were collected.All patients were followed up for 3 years.According to the follow-up results,93 surviving patients were included in the survival group and 42 dead patients were included in the death group.The mRNA expressions of CLDN6,TRIM59 and CMTM6 were determined by fluorescence quantitative PCR.Multivariate Cox regression was used to analyze the influencing factors of death in NPC patients.Kaplan-Meier method was used to analyze the relationship between the expression levels of CLDN6,TRIM59,CMTM6 and the prognosis of NPC patients.Results Compared with the control group,mRNA expressions of CLDN6,TRIM59 and CMTM6 were increased in the observation group(P<0.05).There were no significant differences in age,sex,body mass index,TNM stage,bone metastasis,smoking history,drinking history and hypertension history between the survival group and the death group.Compared with the survival group,the proportion of NPC family history and the mRNA expression of CLDN6,TRIM59 and CMTM6 in cancer tissue were increased in the death group(P<0.05).Multivariate Cox regression analysis showed that the increased levels of CLDN6,TRIM59 and CMTM6 in cancer tissue were influential factors for death of NPC patients(P<0.05).According to the mean expression levels of CLDN6,TRIM59 and CMTM6 mRNA in cancer tissue,patients were divided into the low expression group and the high expression group.The 3-year survival rate of the high expression group was significantly lower than that of the low expression group(P<0.05).Conclusion The mRNA expressions of CLDN6,TRIM59 and CMTM6 in NPC tissue are significantly increased,which is a risk factor for death in NPC patients,and the mRNA expressions of CLDN6,TRIM59 and CMTM6 are correlated with the prognosis of patients.

3.Role and mechanism of endoplasmic reticulum stress and NLRP3 inflammasome in acute kidney injury

Acute kidney injury(AKI)is a common critical condition in clinical practice,characterized by a rapid decline in renal function within a short period.The pathogenesis of AKI is complex and has not been fully elucidated.In recent years,studies have found that the activation of endoplasmic reticulum stress(ERS)and the Nod-like receptor family pyrin domain containing 3(NLRP3)inflammasome are closely related to the occurrence of AKI.When the kidneys is damaged,the internal environment of the kidney cells is disrupted,leading to the activation of ERS.Excessive ERS can induce apoptosis of renal cells,leading to the occurrence of AKI.Additionally,the NLRP3 inflammasome can mediate the recognition of endogenous and exogenous danger signal molecules by the host,subsequently activating caspase-1,pro-inflammatory cytokines such as IL-1β and IL-18,inducing inflammatory responses,and promoting apoptosis of renal cells.In animal models of AKI,the upregulation of ERS markers is often accompanied by increased expression levels of NLRP3 inflammasome-related proteins,indicating that ERS can regulate the activation process of the NLRP3 inflammasome.Clarifying the role and mechanism of ERS and NLRP3 inflammasome in AKI is expected to provide new insights for the prevention and treatment of AKI.
4.Research progress on animal models of contrast-induced acute kidney injury.

Contrast-induced acute kidney injury (CI-AKI) refers to acute kidney injury that occurs after intravascular contrast media is applied. It is the third most common cause for acute renal failure in hospitalized patients and can cause severe renal impairment and adverse cardiovascular outcomes. In severe cases, it can even lead to the death of the patient. Due to its complicated pathogenesis, the pathogenesis of CI-AKI has not yet been elucidated. Therefore, it is of great significance to further study the pathogenesis for the prevention of CI-AKI. Moreover, a good animal model of CI-AKI is an important tool for in-depth research on the pathogenesis of acute kidney injury induced by contrast agents.

5.Application of non-coding RNAs in early diagnosis of colorectal cancer

Non-coding RNAs (ncRNAs) is a kind of RNA without protein coding function. A large number of studies in recent years have shown that the occurrence and development of colorectal cancer (CRC) are closely related to the aberrant expression of ncRNAs. ncRNAs can be used as non-invasive biomarkers for early diagnosis of CRC. We review the research progress of ncRNAs in early diagnosis of CRC, and summarize the potential clinical value of ncRNAs detection, which can provide a theoretical basis for improving the early diagnosis rate of CRC.
6.DNA typing of HLA-I antigens with medium resolution method by DNA chip technique

Objective:To establish a DNA typing method for HLA-I antigens with medium resolution method by DNA chip technique.Methods:The chip was made with specific medium distinguish-typing probes designed according to gene frequency of HLA-I alleles from Northern Chinese. Unsymmetrical PCR was used to amplify HLA-I exon2,3,and then the PCR products labeled and hybridized with probes on the chip.Typing of HLA-I was certified by scanning the hybridizing signals of through a set of computer software.Results:HLA-I alleles were successfully typed in 30 clinical samples .This medium-distinguishing probes were able to discern 57 HLA-I alleles accurately.Conclusion:DNA typing of HLA-I by chip has been proven to be a high-resolution and high-specific method. It is able to check out the new alleles that can not be distinguished by other methods with the same resolution., and it is more intuitional and more suitable for clinical application .
