1.Expression and significance of ChREBP network members and TUG1 in intrauterine adhesion

Objective To investigate the expression and significance of carbohydrate response element-binding protein(ChREBP)network members(ChREBP,Mlx,Max,Mxd1,HDAC1)and taurine-upregulated gene 1(TUG1)in intrauterine adhesion(IUA).Methods Ten female SD rats with 8 weeks old were selected as the study objects.The mechanical curettage plus lipopolysaccharide infection were used to construct the IUA rat model as the IUA group(n=5)and the uterine cavity of the control group did not perform any oper-ation(n=5);HE and Masson staining were respectively used to assess the number of endometrial glands and the degree of fibrosis;Western blot was used to detect the expression level of ChREBP network members in the nucleus and cytoplasm;RT-qRCR was used to detect the expression level of TUG1.Results Compared with the control group,the number of endometrial glands in the IUA group was decreased(P<0.05),the percentage of the fibrotic area was increased(P<0.05),the protein expression level of ChREBP network members in the nucleus was down-regulated,the protein expression level of ChREBP network members in the cytoplasm was up-regulated and the mRNA relative expression level of TUG1 was up-regulated(P<0.05).Conclusion Blocked nuclear translocation of ChREBP and reduced the recruitment of its downstream network mem-bers may lead to attenuate the repressive effect on TUG1 transcription,thereby promoting IUA occurrence.
2.Study on the quality standard and application of polyoxyl(15)hydroxystearate

Objective:To study and establish the quality standard of Polyoxyl 15 hydroxystearate(HS15),a phar-maceutical excipient,and systematically evaluate its functionality-related characteristics and safety.HS15 was applied to the preparation of docetaxel(DTX)injection to further investigate the safety and pharmacokinetic char-acteristics of the injection in vitro and in vivo.Methods:Based on the general USP-NF2024,EP11.0 and the fourth general rules of the Chinese Pharmacopoeia 2020 edition,the quality standards of HS15 were studied.Combined with the functional properties of surfactants,the critical micelle concentration of HS15 was investigated,and its safety was investigated by hemolysis test and vascular irritation test in vitro.HS15 was further applied to the preparation of DTX injection,and the safety and efficacy of the preparation were comprehensively evaluated by in vitro cytotoxicity test and in vivo pharmacokinetic study.Results:According to the experimental results and the pharmacopoeia of various countries,the quality standard of HS15 was preliminarily formulated.When the concen-tration of HS15 was 1 mg·mL-1,the hemolysis rate was about0.2%,the vascular irritation was small,and the DTX injection was safe in vitro and in vivo.The pharmacokinetic behavior was in line with expectations.Conclusion:This study successfully established the quality standard of HS15,and its functional correlation index research and safety evaluation strategy can provide reference for the quality control of similar excipients.The appli-cation of HS15 in the preparation of DTX injection provides a theoretical and experimental basis for its application in the development of insoluble antitumor drug injection.
3.Effects of lncRNA ZFAS1 on hippocampal neuron damage andcognitive function in diabetic encephalopathy mice

AIM:To investigate the expression profile and biological significance of long noncoding RNA(lncRNA)zinc finger antisense 1(ZFAS1)in the brains of mice with diabetic encephalopathy(DE).METHODS:Ten db/m mice served as the normal control group,while twenty 22-week-old db/db mice were used to establish the DE model and randomly divided into two subgroups:ten as the db/db model control and the remaining ten receiving ZFAS1 gene knockdown(db/db+sh-ZFAS1)via lentiviral transfection.Weekly measurements of body weight and blood glucose levels were performed.Brain tissues were collected for Nissl staining to evaluate neuronal damage,TUNEL assay to detect apop-tosis,and immunofluorescence staining to examine neural biomarker expression.Serum levels of tumor necrosis factor-α(TNF-α)and oxidative stress markers,including reactive oxygen species(ROS),malondialdehyde(MDA),superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px),were determined.Western blot was conducted to quantify the protein expression levels of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax),p38 mitogen-activated protein kinase and phosphorylated p38(p-p38)in brain tissues.The expression levels of ZFAS1 and caspase-3 mRNA were determined by RT-qPCR.RESULTS:Knockdown of ZFAS1 in db/db mice significantly improved cognitive function,alleviated hippocampal neuronal damage,and reduced body weight and blood glucose levels(P<0.01).More-over,oxidative stress was mitigated,as evidenced by decreased MDA and ROS levels(P<0.01)and increased activity of antioxidant enzymes,GSH-Px,SOD and CAT(P<0.01 or P<0.05).Meanwhile,ZFAS1 silencing down-regulated Bax and p-p38/p38 protein expression(P<0.01 or P<0.05)while up-regulating Bcl-2(P<0.01).Consistently,RT-qPCR confirmed significant down-regulation of ZFAS1 and caspase-3 mRNA levels(P<0.01).CONCLUSION:lncRNA ZFAS1 is highly expressed in the hippocampus of DE mice.Down-regulation of ZFAS1 expression enhances cognitive func-tion,suppresses oxidative stress,and inhibits neuronal apoptosis,thereby attenuating neural damage in DE.
4.Identification of porcine-derived circ_PIK3C2A and its effect on PEDV replication in host cells after infection

Circular RNA(circRNA),as a kind of non-coding RNA,regulates a variety of biological functions.To explore the effect of circRNA on PEDV replication in the host porcine intestinal epi-thelial cells,this study screened and analyzed the differentially expressed circRNAs by bioinforma-tic software in African Green Monkey renal cells(Vero-E6 cells)infected by porcine epidemic di-arrhea virus(PEDV),the differentially expressed circRNA ssc_circ_PIK3C2A was identified and the secondary structure was analyzed.PCR was used to identify the ssc_circ_PIK3C2A circRNA structure,the model of PEDV-infected IPEC-J2 cells was constructed,the TCID50 test was used to validate the viral titer of PEDV.The expression of circ_PIK3C2A was detected by qRT-PCR in IPEC-J2 infected by PEDV.circ_PIK3C2A qRT-PCR was performed to detect the expression of N gene of PEDV when ssc_circ_PIK3C2A was over-expressed in IPEC-J2 cells.The results showed that ssc_circ_PIK3C2 A is a porcine circular RNA with a typical circular structure,the virus titer of PEDV reached 10-6/mL after PEDV infected IPEC-J2 cells for 48 h,the expression of circ_PIK3C2A increased extremely(P<0.01)at 6 h after PEDV-infection,with the extension of infec-tion time,its expression gradually decreased,and the expression was the lowest at 24 h,but there was no time-dependent trend.The expression of PEDV N gene decreased significantly when ssc_circ_PIK3C2A was over-expressed in IPEC-J2 cells.In conclusion,when PEDV infects IPEC-J2 cells,the expression of porcine circ_PIK3C2A decreases,and replication of PEDV increases signifi-cantly in IPEC-J2 cells.our result provides a basis for further study of the mechanism of circular RNA on PEDV replication and its physiological activities in host cells in the future.

8.Affecting factors of the selection for electronically-delivered cognitive behavioral therapy among patients with insomnia

Objective:To explore the factors influencing electronically-delivered cognitive behavioral therapy for insomnia (e-CBTI) selection among patients with insomnia, and to provide empirical evidence for the clinical promotion of e-CBTI.Methods:Between November 2022 and July 2023, a convenience sampling method was employed to collect samples from 38 public hospitals and their surrounding communities. A total of 756 insomnia patients aged 18-65 years were included in the study. Based on their choice to participate in e-CBTI treatment, patients were divided into two groups: 605 patients (605/756,80.0%) chose to participate in e-CBTI treatment, while 151 patients (151/756,20.0%) did not. Rater-rated structured clinical interviews were applied in the current study, assessing individual′s insomnia symptoms, mental disorders, other sleep disorders, and suicidality and self-injury. The chi-square test and logistic regression were applied in data analyses.Results:Compared to the control group, the e-CBTI group had a higher proportion of females (χ 2=4.29) and a higher educational level (χ 2=5.76), while the proportion of living in rural areas was lower (χ 2=6.87), all with P<0.05. Additionally, the e-CBTI group exhibited a higher proportion of daytime symptoms compared to the control group, including decreased attention or memory (χ 2=16.42), impaired function or performance (χ 2=6.75), mood changes (χ 2=4.71), and reduced enthusiasm, energy, or motivation (χ 2=4.77), all with P<0.05. The e-CBTI group also showed a higher tendency for delayed sleep phase (χ 2=6.61), generalized anxiety disorder (χ 2=6.22), suicidal ideation (χ 2=10.45), and non-suicidal self-injury (χ 2=5.19), all with statistically significant differences ( P<0.05). Multivariate logistic regression analysis revealed that the type of residence ( OR=0.53, 95 %CI=0.29-0.95), educational level ( OR=2.42, 95 %CI=1.34-4.37), decreased attention or memory ( OR=1.95, 95 %CI=1.11-3.42), impaired function or performance ( OR=1.80, 95 %CI=1.15-2.82), and suicidal ideation ( OR=3.12, 95 %CI=1.29-7.58) remained significant influential factors in the selection of e-CBTI. Insomnia′s nighttime symptoms, including difficulty falling asleep, difficulty maintaining sleep, and early morning awakenings, were not associated with the inclination to choose e-CBTI. Conclusions:Daytime symptoms of insomnia and suicidal ideation are significant factors that prompt insomnia patients to choose e-CBTI.
9.Efficacy and safety of chimeric antigen receptor T-cell therapy followed by allogeneic hematopoietic stem cell transplantation in 21 patients with Ph-like acute lymphoblastic leukemia

Objective:To evaluate the efficacy and safety of chimeric antigen receptor T-cell (CAR-T) therapy followed by allogeneic hematopoietic stem cell transplantation (allo-HSCT) in patients with Ph-like acute lymphoblastic leukemia (Ph-ALL) .Methods:Patients with Ph-ALL who underwent CAR-T therapy followed by allo-HSCT from March 2018 to August 2023 at the First Affiliated Hospital of Soochow University were included, and their clinical data were retrospectively analyzed.Results:Of the 21 patients, 14 were male and 7 were female. The median age at the time of CAR-T therapy was 22 (6-50) years. Seven patients had ABL1-like rearrangements, and 14 had JAK-STAT rearrangements. Prior to CAR-T therapy, 12 patients experienced hematologic relapse; 7 were multiparameter flow cytometry minimal residual disease (MFC-MRD) -positive and 2 were MFC-MRD-negative. CAR-T cells were derived from patients’ autologous lymphocytes. Nine patients were treated with CD19 CAR-T cells, and 12 were treated with CD19/CD22 CAR-T cells. After assessment on day 28 after CAR-T therapy, 95.2% of the patients achieved complete remission, with an MRD-negative remission rate of 75%. Nineteen patients developed grade 0–2 cytokine release syndrome (CRS) and 2 patients suffered grade 3 CRS, all cases of which resolved after treatment. All patients underwent allo-HSCT after CAR-T therapy. The median time from CAR-T therapy to allo-HSCT was 63 (38-114) days. Five patients experienced relapse after CAR-T therapy, including four with hematologic relapse and one with molecular relapse. The 3-year overall survival (OS) rates in the ABL1 and JAK-STAT groups were (83.3±15.2) % and (66.6±17.2) %, respectively ( P=0.68) . The 3-year relapse-free survival (RFS) rates were (50.0±20.4) % and (55.6±15.4) % in the ABL1 and JAK-STAT groups, respectively. There was no significant difference in 3-year OS or RFS between the two groups. Conclusions:CAR-T therapy followed by allo-HSCT leads to rapid remission in most patients with Ph-ALL and prolongs leukemia-free survival.
10.Research progress on the effect of gastrointestinal microbiota on intrahepatic cholestasis of pregnancy

Intrahepatic cholestasis of pregnancy (ICP), as an idiopathic disease, often occurs in the second and third trimester of pregnancy. It is characterized by maternal pruritus, abnormal liver function and raised serum bile acids which are resolved rapidly and spontaneously after delivery. It has been reported that abnormal intestinal microbiota and endogenous bile acids play important roles in the progression of cholestatic liver diseases. Dramatic changes in metabolic hormone levels and energy metabolism during pregnancy disrupt the microbiota composition and bile acid pool as well. Altered intestinal permeability, overaccumulation of bile acids, aberrant expression of bile acid transporters and nuclear receptors are all involved in the pathogenesis of cholestasis. This paper reviews the changes of intestinal flora and the mechanism of gastrointestinal microbiota in intrahepatic cholestasis of pregnancy, explore the potential treatments for ICP.
