1.Association of adverse childhood experiences with psychopathology, sleep, and cognitive function in children: the mediating role of putamen volume

BackgroundAdverse childhood experiences (ACEs) are strong predictors of psychopathology across the lifespan, exerting extensive adverse effects on physical and mental health during early development. While ACEs increase the risk of childhood psychopathology, sleep quality and impair cognitive function, the early neurobiological mechanisms mediating ACE-induced cognitive impairment remain unclear. ObjectiveTo explore the association between ACEs exposure and the risk of psychopathology, sleep disorders, and cognitive function, and to examine the mediating role of the putamen volume in the relationship between ACEs and cognitive function from a neurodevelopment perspective. MethodsData were obtained from the Adolescent Brain Cognitive Development (ABCD) study, comprising 10 572 children aged 9 to 10. Participants were categorized into five groups based on the cumulative score of ACEs reported by their parents and the children themselves: no-exposure group (0 types of ACEs), the one-exposure group, the two-exposure group, the three-exposure group, and the high-risk exposure group (≥ 4 types of ACEs). Logistic regression analysis was employed to evaluate the association between ACEs and psychopathological symptoms [T score of the Child Behavior Checklist (CBCL) ≥ 65]. Multiple linear regression analysis was ultilized to analyze the correlations between ACEs and the scores of the Sleep Disturbance Scale for Children (SDSC), the NIH Toolbox Cognition Battery (NIHTB-CB), as well as the multiple structures in the cerebral cortex and subcortex. The Bootstrap method was applied to test the mediating effect of the putamen volume between ACEs and neurocognitive function. ResultsAfter adjusting for confounders, ACEs cumulative score was correlated with children's psychopathological symptoms in a dose-response manner. Compared with the no-exposed group, the high-risk exposure group had a 2.08-fold (OR=2.080, 95% CI: 1.401–3.088) increased risk of internalizing problems and a 3.13-fold (OR=3.132, 95% CI: 1.703–5.760) increased risk of externalizing problems, and the risk of conduct problems was 4.68 times that of the no-exposed group (OR=4.681, 95% CI: 2.528–8.669). ACEs cumulative score positively predicted the total score of SDSC (β=0.080, 95% CI: 0.059–0.102), with relatively stronger predictive effects on sleep-wake transition disorder (β=0.086, 95% CI: 0.063–0.109) and sleep onset and maintenance disorder (β=0.066, 95% CI: 0.043–0.089). In terms of neurocognition and brain structure, ACEs cumulative score negatively predicted the overall cognitive composite score (β=-0.025, 95% CI: -0.050–-0.001), the crystallized cognitive composite score (β=-0.035, 95% CI: -0.060–-0.010), the volume of the left putamen nucleus (β=-0.025, 95% CI: -0.046– 0.003), and the volume of the right putamen nucleus (β=-0.023, 95% CI: -0.044– 0.003). The total putamen volume played a partial mediating role between ACEs and the overall cognitive composite score, the effect value was -0.002 (95% CI: -0.004–-0.001), accounting for 7.49% of the total effect. ConclusionACEs exposure may increase the risk of psychopathological and sleep disturbances in children and may impair neurocognitive function. Reduced putamen volume may serve a potential neurobiological pathway underlying the impact of ACEs on childhood cognition. 2.Clinical research of Eccentric mixing in correcting EDTA-dependent pseudothrombocytopenia

Objective:Evaluate To evaluate the efficacy of eccentric mixing in correcting EDTA-dependent pseudothrombocytopenia (EDTA-PTCP) and platelet (PLT) count.Methods:A case-control study was conducted using EDTA-PTCP patient samples collected from the First Affiliated Hospital of Harbin Medical University from April 1st, 2024, to December 27th, 2024, including 82 cases with mean age 57.98±17.28 years old. EDTA-PTCP was defined as PLT≥3 under microscopy. Samples underwent eccentric mixing at 500 r/min or 3000 r/min for durations ranging from 0.5 to 13 minutes to determine the optimal PLT disaggregation protocol. Repeated-measures ANOVA and paired t-tests were used to analyze PLT results. While chi-square tests were used to compare effective disaggregation rates under the optimal eccentric mixing method and sodium citrate anticoagulation. Statistical significance was defined as P<0.05. Results:Eccentric mixing at 3000 r/min for 10 minutes significantly elevated PLT counts to (135.6±63.91)×10 9/L, compared with untreated group (47.72±25.47)×10 9/L ( t=5.74, P<0.001). Blood analyzer showed that 3000 r/min for 10 minutes method did not alter red blood cell (4.32±0.76 vs. 4.27±0.75)×10 12/L nor white blood cell (6.17±2.22 vs. 6.36±2.14)×10 9/L counts compared with untreated group, in which red blood cell (4.27±0.75)×10 12/L ( t=0.40, P=0.690) and white blood cell(6.36±2.14)×10 9/L ( t=0.48, P=0.630). The eccentric mixing achieved a disaggregation rate of 92.68% while sodium citrate anticoagulation 63.41%, with significant differenct (χ2=20.5, P<0.001). Conclusion:Eccentric mixing at 3000 r/min for 10 minutes effectively disaggregates platelets in EDTA-PTCP, correcting PLT counts without affecting other blood cells. This technique outperforms anticoagulant replacement strategies, offering a more efficient solution for clinical use as an alternative for changing anticoagulant.
3.Endocardial myocardial biopsy and ultrastructural characteristics in heart failure patients with reduced ejection fraction

Objective:To investigate the endocardial myocardial biopsy and ultrastructural features of heart failure patients with reduced ejection fraction, to determine their histopathologic phenotype, and to explore the diagnostic utility of endomyocardial biopsy in such patients.Methods:A total of 35 patients with heart failure with reduced ejection fraction diagnosed at Beijing Anzhen Hospital and underwent endomyocardial biopsy were collected between January 2022 and December 2023. The clinical features, histopathological, and ultrastructural characteristics were analyzed and compared with 11 patients with heart failure with preserved ejection fraction.Results:The age ranged from 35-58 years, with median age of 51 years; there were 26 males and 9 females. Myocardial fibrosis and myocardial fiber disorders were the most common histopathologic changes [97.1% (34/35) and 74.3% (26/35), respectively]. Myocardial fibrosis was not statistically different between the heart failure with reduced ejection fraction group and the heart failure with preserved ejection fraction group [13.3% (5.7%-21.4%) vs. 13.2% (9.3%-34.2%), P=0.279]. Significant ultrastructural changes were dense mitochondrial proliferation, vacuolar degeneration, and disorganized arrangement of myocardial fibers with localized lysis and fracture. After endomyocardial biopsy, the etiology was identified in 11 patients (31.4%,11/35), with a prevalence of cardiac amyloidosis of 17.1% (6/35). Conclusions:Endomyocardial biopsy is useful for early diagnosis and precise treatment in patients presenting with heart failure with reduced ejection fraction. Histopathological and ultrastructural analyses can uncover potential treatments, and predict and improve prognosis by providing relevant information for understanding the pathogenesis and clinical evolution.

6.Clinicopathological characteristics and molecular alterations of primary cardiac leiomyosarcoma: report of five cases

Objective:To investigate the clinical, pathologic and radiologic features and molecular alterations in patients with primary cardiac leiomyosarcoma (PCLMS).Methods:Five cases of PCLMS were collected in Beijing Anzhen Hospital from January 2016 to December 2020. The clinical, pathologic and radiologic data, and molecular alterations were analyzed, and the patients were followed up.Results:All five patients were female, and had no history of leiomyosarcoma in other parts of the body. The age of patients ranged from 37 to 62 years (median 47 years). The main clinical symptoms were chest pain and dyspnea, one also presented with palpitation and lower limb weakness and one with dizziness. Two tumors were located in the left atrium, two in the right atrium, and one in the right ventricle, and they maximal diameter ranged from 2.5 to 14.0 cm (mean 6.2 cm). The neoplasms presented as medium-echo masses with a broad base in the echocardiography, and as a low-density, solid mass when detected by contrast-enhanced CT. Histologically, two tumors were well-differentiated and three were moderately and poorly differentiated, and two included extensive, loose myxoid stroma. Immunohistochemical staining showed that PCLMS was positive for SMA, desmin, MDM2, and epidermal growth factor receptor. Fluorescence in situ hybridization showed ALK gene rearrangement in two cases, and COL1A1-PDGFB fusion in three cases. All cases received surgical excision and two cases received chemotherapy. Three patients died within 0-11 months (mean survival of 7.7 months) and two patients were alive.Conclusions:PCLMS is a malignant tumor with a high recurrence rate and poor prognosis. These cases may provide useful information to improve the diagnosis and management of PCLMS.
7.Analysis on serotype and antimicrobial resistance of invasive non-typhoidal Salmonella

Objective:To investigate the serotypes and antimicrobial resistance of seven invasive non-typhoidal Salmonella (iNTS) isolates. Methods:For 7 iNTS strains collected, serotype identification, antimicrobial susceptibility testing and whole genome sequencing were performed. We identified, annotated and analyzed the serotypes, MLST types, and antimicrobial resistance genes.Results:Among the 7 tested iNTS isolates, we found one Salmonella Typhimurium strain and two Salmonella Ⅰ 4, [5], 12: i:- strains whose MLST types were ST34, two Salmonella Enteritidis strains, one Salmonella Corvallis strain and one strain of unknown serotype with the antigenic formulae of Ⅰ 4, [5], 12: d:- (ST279 type). Six of seven strains were monophasic and the deletion or pseudogenization of Salmonella Flagellum gene might contribute to the enhancement of Salmonella invasiveness. None was found to be resistant to tigarcycline, aztreonam, amikacin, cephalosporins and carbapenem and one Salmonella Typhimurium strain was found to be co-resistant to eight classes of antimicrobials at the same time. Resistance genes were generally in accord with relative resistant phenotypes. Conclusion:The iNTS strains could show high level multi-drug resistance, indicating that close attention should be paid to the resistance of iNTS though the overall resistance might be relatively not high.
8.Physical activity level in physical education class of elementary school in Xi an

Objective:
To assess physical activity level and characteristics in elementary physical education (P.E.) class among different weight status and grades students.
Methods:
ActiGraphGT3X was used to measure physical activity of 60 students in grade 1, 2, and 4 during three P.E. classes. BMI was calculated according to the student s height and weight in the latest physical fitness test results, and the weight status was divided according to the national Physical Fitness Standards for Students in different grades of elementary schools. Independent sample t test and analysis of variance were used to explore the difference of physical activity.
Results:
The proportion of moderate to high intensity physical activity (MVPA) in elementary school P.E. class was 21.9% (8.8 min). The proportion of MVPA in P.E. classes and the number of steps in normal weight students were 25.4% and (1 247.0± 326.3 ), respectively, which were significantly higher than those of underweight students, but not significantly different from those of overweight students. The MVPA of male and female PE classes in the second grade (27.0%, 26.4%) was significantly higher than that in the first grade, but there was no significant difference compared with that in the fourth grade ( P >0.05).
Conclusion
Moderate to high intensity physical activity is low in P.E. class in elementary school. Improving teachers awareness of promoting physical activity in PE class will benefit the MVPA in students.
9.Prevalence and influencing factors of colorectal polyps in Lanxi residents

Objective:
To investigate the epidemic status and influencing factors of colorectal polyps among urban residents in Lanxi.
Methods:
A representative urban community was selected from Lanxi. A total of 935 local residents aged 18-80 years received questionnaire survey,physical examination,laboratory tests and colonoscopy. Multivariate Logistic regression analysis was applied to exam the potential influencing factors for colorectal polyps.
Results :
Eventually 880 residents were involved into the statistical analysis. Colorectal polyps were detected in 167 participants with a crude prevalence of 18.98% and adjusted prevalence for age and gender of 13.28%. The prevalence of colorectal polyps in men(28.70%)was significantly higher than that in women(12.92%,P<0.05). The results of logistic regression analysis showed that age(OR=1.038,95%CI:1.018-1.057),males(OR=1.919,95%CI:1.303-2.826),the middle school level of education(OR=2.443,95%CI:1.507-3.961)and constitution of Yin-Asthenia(OR=0.203,95%CI:0.057-0.720)were influencing factors for colorectal polyps;among male participants,hypertension(OR=1.721,95%CI:1.018-2.908)was a risk factor for colorectal polyps;among female participants,age(OR=1.076,95%CI:1.042-1.111),BMI(OR=1.099,95%CI:1.001-1.207),the middle school level of education(OR= 3.507,95%CI:1.794-6.856)and constitution of Yin-Asthenia(OR=0.160,95%CI:0.033-0.770)were influencing factors for colorectal polyps.
Conclusion
Age,sex,education level and constitution of Yin-Asthenia were associated with the risk of colorectal polyps among residents in Lanxi;hypertension was associated with the risk of colorectal polyps in males,while age,BMI,education level and constitution of Yin-Asthenia were associated with the risk of colorectal polyps in females.
10.Effect of Electroacupuncture on the Behavior of APP/PS1 Transgenic Mice of Different Age

Objective To study the effect of electroacupuncture on the behavior of APP/PS1 transgenic mice of different age, and to explore the optimal intervention time of electroacupuncture in treating Alzheimer's disease (AD). Method APP/PS1 double-transgenic mice of 4 months, 6 months and 9 months old, 20 in each age group, were randomized into a model group and an electroacupuncture group, and ten C57BL/6 wild-type mice were taken as a control group; after 6-week electroacupuncture treatment, the Morris water maze was adopted for spatial memory and behavioral test, and the changes of behavior in each group were observed.Result Of the 5-month-old mice, there were no significant between-group differences (P>0.05), while the time factor (day) produced a statistical significance (P<0.01); of the 7-month-old mice, there were significant differences considering the different groups, time factor and interaction (P<0.01); of the 10-month-old mice, there were significant differences considering the different groups, time factor and interaction (P<0.05). The spatial probe test showed that there were significant between-group differences in comparing the platform crossings and swimming distance in platform quadrant in each age group (P<0.05).Conclusion Electroacupuncture can improve the learning and memory of APP/PS1 double-transgenic mice, the age of 6-7 months old is possibly the optimal intervention time of electroacupuncture for AD, but there still requires further mechanism studies.
