1.Relationship between serum KNG1 and TIMP3 levels and coronary artery lesions in children with Kawasaki disease at the acute phase

Objective To investigate the relationship between serum kininogen-1(KNG1)and tissue inhib-itor of metalloproteinases-3(TIMP3)levels and coronary artery lesions(CAL)in children with Kawasaki dis-ease(KD)at the acute phase.Methods A total of 132 children with KD at the acute phase admitted to the hospital from October 2020 to February 2024 were selected as the KD group,and were divided into CAL group(66 cases)and non CAL group(66 cases)according to whether they were combined with CAL.In addition,66 healthy children during the same period were selected as the control group.Serum KNG1 and TIMP3 levels were measured using enzyme-linked immunosorbent assay.A multivariate unconditional Logistic regression model with CAL as the dependent variable was established to identify independent factors influencing CAL in children with KD at the acute phase.Receiver operating characteristic(ROC)curve was plotted to evaluate the predictive value of serum N-terminal pro-brain natriuretic peptide(NT-proBNP),KNG1,and TIMP3 levels for CAL in children with KD at the acute phase.Results Compared to the control group,serum KNG1 level was elevated in the KD group(P<0.05),and TIMP3 level was reduced(P<0.05).In the CAL group,serum KNG1 level was higher(P<0.05),and TIMP3 level was lower compared to the non CAL group(P<0.05).Independent risk factors for CAL in children with KD at the acute phase included immunoglobulin resistance(OR=5.618,95%CI:1.203-26.248),increased NT-proBNP level(OR=1.003,95%CI:1.000-1.005),and increased KNG1 level(OR=1.648,95%CI:1.251-2.173),while increased TIMP3 level was identified as an independent protective factor(OR=0.966,95%CI:0.949-0.983,P<0.05).The combination of serum NT-proBNP,KNG1,and TIMP3 levels yielded an area under the curve of 0.930 for predicting CAL in children with KD at the acute phase,which was larger than 0.785,0.801,0.792 of NT-proBNP,KNG1,and TIMP3 a-lone(P<0.05).Conclusion increased level of serum KNG1 and decreased level of TIMP3 are independently associated with the occurrence of CAL in children with KD at the acute phase.The combined prediction of ser-um NT-proBNP,KNG1,and TIMP3 levels has high value in predicting CAL in children with KD at the acute phase.
2.Analysis of the endoscopic screening results of Beijing-Tianjin-Hebei Region Gastrointestinal Endoscopy Medical Association from 2016 to 2020

Objective:To evaluate the development and application of gastrointestinal endoscopy technology in Beijing-Tianjin-Hebei (BTH) region from 2016 to 2020, and the impact of the corona virus disease 2019 (COVID-19) epidemic on gastrointestinal endoscopy screening and lesion detection rate of medical institutions.Methods:Data of gastroscopy and colonoscopy cases from 26 cooperative institutions in BTH Region Gastrointestinal Endoscopy Medical Association from January 2016 to December 2020 were collected by questionnaire. The number of gastrointestinal endoscopy, the detection of main lesions (including upper gastrointestinal malignant tumors, early gastric cancer and colon cancer), and the number of endoscopic treatment were retrospectively analyzed by year.Results:From 2016 to 2019, the number of gastroscopy and colonoscopy showed a yearly increasing trend with an annual growth rate of over 10%. Compared with 2019, the number of gastroscopy and colonoscopy decreased by 10.86% and 8.29%, respectively, in 2020 due to the impact of the epidemic. The annual detection rates of upper gastrointestinal malignant tumors, early gastric cancer and colon cancer were on a rise, from 7.22%, 1.49% and 8.98% in 2016 to 9.87%, 2.71% and 12.04% in 2020, respectively. The number of gastroscopic mucosal resection, submucosal dissection and colonoscopic endoscopic submucosal dissection increased yearly, from 2 132, 300 and 217 cases in 2016 to 5 466, 872 and 560 cases in 2020, respectively.Conclusion:The Medical Association has promoted the expansion of endoscopic screening and the application of endoscopic treatment techniques, resulting in a continuous increase in the endoscopy detection rate and early cancer diagnosis rate in the BTH region. The sharp decrease of gastrointestinal endoscopy procedures and the increase in the lesion detection rate in 2020 reflect the impact of epidemic COVID-19 on detection of gastrointestinal cancers.
3.Effects of agmatine on permeability of blood-brain barrier and expression of aquaporin 4 and matrix metalloproteinase 9

Objective To investigate the protection of agmatine on blood brain barrier (BBB) permeability and the effect on the expression of aquaporin 4 (AQP4) and matrix metalloproteinase 9 (MMP9) in ischemic reperfusion injury rats.Methods Sixty healthy male Sprague-Dawley rats were randomly divided into normal control group (NC),model group and agmatine group,and there were 20 rats in each group.Normal control group was treated with intraperitoneal injection of saline in 2 hours after incision and suture of skin.Model group was treated with intraperitoneal injection of saline in 2 hours after the establishment of middle cerebral artery occulation (MCAO) model.Agmatine group was treated with intraperitoneal injection of agmatine (AGM) in 2 hours after the establishment of MCAO model.The damage of blood brain barrier was detected by measuring the permeability of blood brain barrier.The infarct size of brain was observed with TTC staining.The morphological changes of neurons were observed with electron microscope.The expressions of AQP4 and MMP9 were detected using immunohistochemical method.Results The permeability of BBB of agmatine treatment group (0.31±0.10) decreased significantly compared with the model group (0.46±0.09) (P<0.05),but was still significantly higher than the normal control group (0.24±0.12) (P<0.05).There was no infarction area in normal control group and the infarction areas of agmatine group decreased obviously compared with the model group.Compared with model group,the number of neurons with morphological changes reduced significantly and the degree of pathological changes of neurons was obviously improved in agmatine group.Compared with normal control group,the expression of AQP4 and MMP9 in ischemic penumbra of left cerebral hemisphere parietal cortex in the rats of model group and agmatine group increased significantly(P<0.05).And the expression of AQP4 and MMP9 in model group was significantly higher than that of agmatine group(P<0.05).Conclusion Agmatine has a protective effect on BBB with ischemia-reperfusion injury due to its down-regulation the expression of AQP-4 and MMP-9.
4.EFFECT OF TAURINE ON IRON BIOAVAILABILITY IN RATS AND ITS POSSIBLE MECHANISM

Objective To investigate the effect of taurine on iron bioavailability in rats and its possible mechanism. Method SD rats were randomly divided into three groups:normal group (NG),taurine treated group (TG),experimental control group (EG). TG and EG were induced to be anemic experimentally for 4w before the study. Then the rats were fed iron supplements for 25 d,meanwhile TG was treated with taurine following hemoglobin(Hb) repletion bioassay. Regression analysis of TG and AG were measured following Hb repletion bioassay using iron supplements. And the relative biological value of TG following Hb repletion were calculated. The serum iron,unsaturated iron-binding capacity(UIBC),total iron-binding capacity(TIBC),transferin saturation(TS)and the expression of hepcidin were detected. Results The iron bioavailability of TG was higher than EG,and the RBV of TG was 126%. Other hematological variables of TG were close to NG,and better than EG. The expression of hepcidin was increased compared with NG,but decreased compared with EG. Conclusion Taurine can improve iron bioavailability in rats via suppressing hepcidin expression in rats.
