1.Retrospective analysis of clinical efficacy of Nuangong Waifu formula in preventing intrauterine re-adhesion

Objective The Nuangong Waifu formula (NGWFF) is a traditional Chinese medicine prescription that has been used in gynecology of traditional Chinese medicine in our hospital for many years. It has a certain effect on preventing postoperative intrauterine re-adhesion. To further retrospectively analyze the clinical efficacy of NGWFF. Methods A total of 200 patients who were diagnosed with intrauterine adhesions and underwent intrauterine adhesion separation from January 2018 to December 2020 were retrospectively included. They were divided into control group and observation group according to different drug use for postoperative prevention of re-adhesion, with 100 cases in each group. All patients were given oral estrogen and progesterone (ethinyl estradiol tablets 0.037 5 mg, q12 h, or estradiol valerate tablets 3 mg, q12 h, a total of 21 days, 7 days after estrogen therapy plus dydrogesterone 20 mg, qd or progesterone capsules 200 mg, qd) to promote endometrial growth. In the control group, 100 patients only used estrogen and progesterone after operation. In the observation group, 100 patients were treated with NGWFF at Guanyuan acupoint (four fingers under the navel), once a day. Both groups were evaluated for the degree of intrauterine adhesions under hysteroscopy and the effective rate after 3-5 menstrual cycles of drug treatment. Results Compared with using estrogen and progesterone alone, combination use of NGWFF significantly decreased in the scores of intrauterine adhesions under hysteroscopy (2.41±1.19 vs 3.31±1.18, P=0.00), and the effective rate was also significantly higher than that in the control group ( 86 % vs 47 %, P<0.000). Conclusion The combination use of NGWFF was more effective than using estrogen and progesterone alone in preventing re-adhesion after intrauterine adhesions, which provided a scientific basis for the clinical application of NGWFF.
2.Expression of lncRNA DGCR5 in esophageal squamous cell carcinoma tissues and its clinical significance

[Abstract] Objective: To investigate the expression of long non-coding RNA (lncRNA) DiGeorge syndrome critical region gene 5 (DGCR5) in esophageal squamous cell carcinoma (ESCC) tissues, and to analyze its relationship with clinicopathological features and prognosis of ESCC patients. Methods: The expression of DGCR5 in ESCC data set from TCGA database was analyzed by bioinformatics method. Sixty pairs of ESCC tissues and para-cancerous tissues resected at the Fourth Hospital of Hebei Medical University from August 2016 to March 2017 were collected for this study. The expression of DGCR5 in ESCC tissues was detected by qPCR. The correlation between the expression of DGCR5 and the clinicopathological features and prognosis of ESCC patients was analyzed. Results: TCGAdatabase analysis showed that the expression of DGCR5 in ESCC tissues was significantly higher than that in normal esophageal tissues (P<0.01). The expression of DGCR5 in ESCC tissues was significantly higher than that in para-cancerous tissues (P<0.01). The expression level of DGCR5 was significantly correlated with TNM staging and lymph node metastasis in ESCC patients (all P<0.05). Kaplan-Meier univariate analysis showed that the 2-year survival rate of ESCC patients with high DGCR5 expression was significantly lower than that of patients with low expression (P<0.05). Conclusion: DGCR5 is highly expressed in ESCC tissues and is closely related to TNM staging, lymph node metastasis and poor prognosis, which may serve as a molecular marker for early diagnosis and prognosis prediction of ESCC.
3.Value of αL-fucosidase and its correlation with total bile acids in pregnant women with gestational diabetes mellitus

Objective To observe the detection value of α-L-fucosidase (AFU) in gestational diabetes mellitus (GDM) and the correlation of AFU with total bile acids (TBA).Methods Anthropometric parameters and serum biochemical indexes were recorded and 75 g OGTT was performed then according to OGTT result,patients with GDM were confirmed and matched the control group by age and BMI.Results The level of AFU in GDM group was higher than that that of normal glucose tolerance (NGT) group and associated with 1 hour postprandial glucose.After correcting ALT and HbA1c,AFU correlated with TBA.Conclusions During pregnancy,AFU has reference value for the diagnosis of GDM and 1 hour postprandial glucose is its independent factor.The rise AFU is combined with the decrease of.DBIL but the increase of TBA and ALP,which indicates bile duct system in patients with GDM is in an active state.
4.Validity and Reliability of the Chinese version of Psycho-Educational Profile for Children with Autism

Objective:To test the validity and reliability of Chinese version of Psycho-Educational Profile (C-PEP) for children with autism in order to develop an assessment tool suitable for Chinese autistic children.Methods:A sample of 438 children who were diagnosed as autism according to the Diagnostic and Statisti-cal Manual of Mental Disorders,Fourth Edition (DSM-IV)criteria and aged from 2 to 11 coming from four differ-ent medical or educational rehabilitation agencies of Beijing,Wuhan,Shenzhen and Changchun were selected.488 Children with 2 to 7 years old were sampled from ordinary kindergartens.This study examined the validity and reli-ability of C-PEP using autism sample.Construct validity was tested with the C-PEP by inter-subscale Pearson corre-lations and confirmative factor analysis.Criterion-related validity was tested with the Peabody Picture and Vocabula-ry Test (PPVT),Combined Raven Test (CRT),Adaptive Behavior Scale (ABS),Childhood Autism Rating Scale (CARS)and the age as criteria.Discriminate validity was obtained by comparing the differences between the typi-cal and autism samples with the total scores of C-PEP.Internal consistency and rater reliability of C-PEP were ex-amined by Cronbach α,ANOVA of rater variation and intra-class correlation coefficients.Results:The inter-sub-scale correlations of C-PEP were medium relevance,the subscale and domain correlations were relatively high.Confirmative factor analysis indicated a relatively high fitting index between the model and sample da-ta.Regarding the concurrent validity of the C-PEP,the development total score was significantly correlated with the Raven's IQ,PPVT IQ,and ADQ scores,pathological behavior total score was positively related to the CARS score.T test results showed that children with autism were significantly behind the typical children at all age lev-els.Results also revealed that the internal consistency based on coefficient alpha and the intra-class correlation coef-ficients of raters for all subtests of C-PEP were reliable.Conclusion:It suggests that the Chinese version of Psycho-Educational Profile (C-PEP)is of good validity and reliability,and it could be used as an effective tool for measur-ing children with autism.
5.Effects and mechanisms of periostin overexpression on invasion and migration of the nasopharyngeal carcinoma 6-10B cell line

Objective To explore the effects and mechanisms of periostin overexpression on migration and invasion of nasopharyngeal carcinoma ( NPC) cell line.Methods The recombinant plasmids [ pCMV-neo ( +)-periostin ] and control plasmids [pCMV-neo (+)] were transfected into 6-10B cells using lipofectamine 2000TM reagent.The expression of periostin was detected with PCR and Western blotting .Transwell chamber invasion assay was employed to assay the migration and invasion of 6-10B cells before and after transfection .A gelatin zymogram was used to detect the activity of MMP-2 and MMP-9 in cultivated supernatant of 6-10B cells before and after transfection .The expression of integrin-αvβ5 was detected by immunohistochemistry ( IHC) in 6-10Bperiostin cells, 6-10Bvector cells and 6-10B cells as well as normal nasopharyngeal mucosa ( NNM) and NPC and at the same time periostin also was detected by immumohistochemistry in NNM and NPC, and densitometry analysis using image-pro plus 6.0 software, and the correlation between periostin and integrin-αvβ5 on NPC was assayed with statistics .Results Over expression of periostin promoted cell migration and invasion.The expression levels of integrin-αvβ5 in primary NPC and 6-10Bperiostin cells were significantly higher than those in NNM and 6-10Bvector, 6-10B cells.The expression in NPC of integrin-αvβ5 showed positively correlated with the expression of periostin (r=0.682, P<0.01).Conclusion Periostin plays an important role in regulation of cell migration and invasion probably by combining with integrin-αvβ5 to improve the activities of MMPs .
6.Efficacy and safety of ibutilide for conversion of atrial fibrillation/flutter.

OBJECTIVETo investigate the efficacy and safety of intravenous ibutilide for conversion of atrial fibrillation (AF) and flutter (AFL) to sinus rhythm.
METHODSNinety-nine consecutive patients aged 18-75 y with AF/AFL were included. The duration of arrhythmia was <90 d (1 h-90 d) and ventricular rate was >60 beats/min. Patients were assigned randomly into two groups: 49 patients in ibutilide group received ibutilide 1 mg, then repeated if AF/AFL was not converted after 10 min; 50 patients in propafenone group received propafenone 70 mg, then repeated if AF/AFL persisted after 10 min. Two drugs were diluted by 50 ml of 5% glucose and injected intravenously within 10 min.
RESULTSVentricular rates were decreased in both groups. AF/AFL were converted in 34 of 49 patients (69.4 % ) in ibutilide group and in 22 of 50 patients (44.0 %) in propafenone group (P <0.05). The converting time of ibutilide was significantly shorter than that of propafenone [(16.79 ± 12.31) min compared with (36.92 ± 11.38)min, P <0.01]. The most serious adverse effect of ibutilide was non-sustained monomorphic ventricular tachycardia (3/49,6.12 %). Transient hypotension and heart pause were the main adverse events in patients who received propafenone, acute left heart failure occurred in one patient of propafenone group.
CONCLUSIONIntravenous ibutilide is a safe and effective agent for cardioversion of recent-onset AF/AFL. Furthermore,strict processing under electrocardio-monitoring is important.
