1.One case of VV-ECMO treatment for dimethyl sulfate poisoning

This paper analyzes the treatment process of a patient who developed severe acute respiratory distress syndrome (ARDS) following a chemical leak of dimethyl sulfate caused by factory operational error in February 2023, and explores the efficacy of extracorporeal membrane oxygenation (ECMO) in patient management. The patient developed severe respiratory symptoms five hours after chemical exposure and received sequential treatment with nasal high-flow oxygen therapy and invasive mechanical ventilation upon admission. When the oxygenation index dropped below 100 mmHg, veno-venous (VV) ECMO was initiated alongside 12-hour daily prone position ventilation to improve oxygenation and lung compliance. The patient recovered and was discharged following treatment. For patients with dimethyl sulfate poisoning complicated by severe ARDS, early implementation of VV-ECMO significantly improves treatment outcomes and prognosis.
2.Distribution of street rabies virus in salivary glands of dogs and mice following ex-perimental infection

The rabies virus(RABV)that causes rabies mainly attacks the peripheral and central nervous systems.In the later stages of infection,it is scattered in the salivary glands and transmit-ted to other susceptible animals through infectious saliva.To study dispersion of the RABV in the three pairs of salivary gland tissues,the street strain PB4 of the RABV was inoculated into 21-day-old female mice through the hind limb muscles.During the moribund stage of the mice,the sublin-gual gland,submandibular gland and parotid gland were collected,respectively.The TCID50 titer of RABV in the three kinds of glands of the mice and the copy number of the RABV N gene were de-tected,and RABV in different salivary glands was observed by immunofluorescence.The results showed that PB4 was dispersed in all three kinds of salivary glands of the mice,with the largest a-mounts in the parotid gland,followed by the submandibular gland,and the lowest amount in the sublingual gland.Three-month-old dogs were inoculated with PB4 through the cranial cavity,and saliva were collected every 12 h after inoculation.The saliva samples were detected by TCID50 and RT-qPCR.And during the moribund stage of the dogs when the disease occurred,the three pairs of salivary glands were collected.Through the determination of the TCID50 titer,RT-qPCR and immu-nofluorescence detection,it was demonstrated that among the three different salivary glands of the dogs,the largest amount of PB4 was found in the parotid gland and the lowest in the sublingual gland.Our results in mice and dogs clearly proved that the parotid gland was consistently found to exhibit the highest content of street RABV among the three major salivary glands,which could en-rich experimental data for analyzing the dispersion of RABV in the salivary glands and interpreta-tion of the intermittent secretion of saliva in clinically rabid dogs.
3.Function of ubiquitin-specific protease 47 in regulating rabies virus infection

This study aims to inquire about the fluctuations of ubiquitin-specific protease 47 on neu-roblastoma cells(Neuro-2a,N2a)infected by rabies virus(RABV).USP47 expression levels were detected after RABV infection in N2a cells through RT-qPCR,protein immunoblotting,and virus titer determination.The levels of RABV nucleoprotein and phosphoprotein gene and protein,and RABV titers in supernatants were analyzed during overexpression and knockdown of USP47.The results showed that RABV infection increased USP47 gene level in N2a cells.When overexpression of USP47,the levels of RABV N and P were increased,and the virus titers were also improved.Mo-reover,the level of interleukin-6(IL-6)genes decreased.Knocking down USP47 expression reduced levels of RABV N and P genes and proteins,lowered the virus titer,and elevated the IL-6 gene lev-el.The results suggest that USP47 promotes RABV infection and suppresses IL-6 expression.This finding lays the foundation for further investigation into the molecular mechanisms by which USP47 regulates RABV infection.

7.Impact of goal-directed fluid therapy based on pulse pressure variation on cognitive function in patients undergoing robot-assisted radical resection of gynecological malignancies

Objective To investigate the effects of goal-directed fluid therapy(GDFT)based on pulse pressure variation(PPV)on postoperative cognitive function in patients undergoing Da Vinci robot-assisted radical resection of gynecological malignancies.Methods A total of 60 patients who underwent robot-assisted radical resection of gynecological malignancies were randomly divided into control group and observation group using a random number table method,with 30 patients in each group.The control group received conventional fluid replacement regimen,while the observation group received GDFT regimen based on PPV.The surgical conditions,hemodynamic indices[heart rate(HR),mean arterial pressure(MAP),PPV],cerebral oxygen saturation(rSO2),gastrointesti-nal function,renal function,cognitive function[Mini-Mental State Examination(MMSE)score],and postoperative conditions of the two groups were compared.Results The amount of crystalloid fluid and urine output in the observation group were less than those in the control group,while the amount of colloid fluid was more than that in the control group,with statistically significant differences(P<0.05).Five minutes after anesthesia induction,the levels of MAP,HR and rSO2 in the observation group were higher than those in the control group,with statistically significant differences(P<0.05).Five minutes after anesthesia induction,1 hour after the Trendelenburg position,and imme-diately after the end of surgery,the PPV levels in the observation group were lower than those in the control group,with statistically significant differences(P<0.05).After surgery,the levels of lactic acid and urea nitrogen in both groupswere higher than those before surgery,but those in the observa-tion group were lower than those in the control group,with statistically significant differences(P<0.05).After surgery,the creatinine levels in both groups were lower than those before surgery,but those in the observation group were higher than those in the control group,with statistically signifi-cant differences(P<0.05).The time to first flatus in the observation group was earlier than that in the control group,with a statistically significant difference(P<0.05).On days 3,7 and 30 after surgery,the MMSE scores in the observation group were higher than those in the control group,with statistically significant differences(P<0.05).The drainage tube indwelling time and hospital stay in the observation group were shorter than those in the control group,with statistically significant differences(P<0.05).There was no statistically significant difference in the overall incidence of postoperative complications between the two groups(P>0.05).Conclusion GDFT based on PPV can stabilize the hemodynamic status and rSO2 in patients undergoing Da Vinci robot-assisted radical resection of gynecological malignancies and reduce postoperative cognitive impairment.
8.Effect of online and offline high definition transcranial direct current stimulation on finger motor skill learning in healthy adults:a randomized controlled trial

Objective To investigate the effect of high definition transcranial direct current stimulation(HD-tDCS)at different stim-ulation timings(online and offline)on finger motor skill learning in healthy adults. Methods From March to November,2022,31 right-handed male participants were recruited from Shenyang Sport Uni-versity and randomly assigned to three groups,who received online,offline or sham HD-tDCS respectively dur-ing the three-day sequential visual isometric pinch task learning process.The daily motor skill learning gains,to-tal motor skill learning gains and speed-accuracy function(SAF)were observed. Results The daily motor skill learning gains increased in all the groups,and it was the best in the online group(F=24.692,P<0.001);the learning gains were better in the online group than in the offline group(t=4.732,P<0.001)and the sham group(t=3.839,P=0.004)on the third day.The total motor skill learning gains were ranked as the online group=the offline group>the sham group.SAF data indicated that the online group outper-formed both the offline and sham groups. Conclusion HD-tDCS can promote the finger motor skill learning in healthy adults,and online HD-tDCS is more effec-tive.
9.Differential expression profile in Neuro-2a cells infected by rabies virus

Rabies is a zoonotic disease that poses a global public health threat.Rabies virus(RABV)is neurotropic and can cause severe neurological disorders and behavioral abnormalities in host,with a fatality rate nearly 100%.In order to identify the key genes for RABV affecting neuronal cell function,we established and analyzed the mRNA expression profile of Neuro-2a(N2a)cell in-fected with challenge virus standard(CVS)-11 by RNA sequencing(RNA-seq).Biological func-tions of differentially expressed genes(DEGs)were determined by GO and KEGG enrichment a-nalysis.The results showed that there were 415 differentially expressed genes in N2a infected with CVS-11 strain,of which 89 were up-regulated and 326 were down-regulated.These genes were re-lated to a variety of biological processes,such as axon guidance pathway,cholesterol metabolism pathway,nitrogen metabolism pathway,and glycosphingolipid biosynthesis pathway,many of them have been shown to be closely associated with RABV infection.A total of 12 DEGs related to axon conduction,antigen processing and presentation pathways were selected and detected by real-time PCR,and their expression trends were consistent with the RNA-seq results.The genomic tran-scriptomic data on N2a cell under RABV infection will provide new clues for probing the mecha-nisms of RABV infection and transmission in the nervous system in the future.
10.Effects of group mindfulness yoga training on sleep quality in college students

ObjectiveTo explore the effectiveness of mindfulness yoga training on sleep quality in college students, so as to provide references for the intervention of sleep problems in college students. MethodsFrom March 28, 2021 to April 20, 2021, A sample of 45 college students with Pittsburgh Sleep Quality Index (PSQI) score ≥8 was recruited. Participants were randomly divided into study group (n=22) and control group (n=23). Participants in the intervention group received mindfulness yoga training twice a week for 4 weeks. The control group did not receive mindfulness yoga training during the trial, but the same training would be given after the trial. Before and after the intervention, PSQI and the N-back training tasks were used to evaluate the sleep quality and working memory ability of the two groups. ResultsThe main effects of time on the PSQI total score, the accuracy and reaction time of N-back training task were statistically significant (F=37.240, 6.898, 10.775, P<0.05 or 0.01). The main effects of grouping factors on PSQI total score and the accuracy of N-back training task were statistically significant (F=8.057, 11.093, P<0.05 or 0.01). The interaction of time and group had statistical significance on PSQI total score and the response time of the N-back training task(F=32.730, 17.578, P<0.01). After intervention, the PSQI total score of the study group was lower than that before intervention (F=69.898, P<0.01), and the response time of N-back training task was shorter than that before intervention (F=27.939, P<0.01), the differences were statistically significant. ConclusionGroup mindfulness yoga training may help improve the sleep quality of college students.
