1.Spermine Synthase : A Potential Prognostic Marker for Lower-Grade Gliomas

Objective:
: The objective of this study was to assess the relationship between spermine synthase (SMS) expression, tumor occurrence, and prognosis in lower-grade gliomas (LGGs).
Methods:
: A total of 523 LGG patients and 1152 normal brain tissues were included as controls. Mann-Whitney U test was performed to evaluate SMS expression in the LGG group. Functional annotation analysis was conducted to explore the biological processes associated with high SMS expression. Immune cell infiltration analysis was performed to examine the correlation between SMS expression and immune cell types. The association between SMS expression and clinical and pathological features was assessed using Spearman correlation analysis. In vitro experiments were conducted to investigate the effects of overexpressing or downregulating SMS on cell proliferation, apoptosis, migration, invasion, and key proteins in the protein kinase B (AKT)/epithelialmesenchymal transition signaling pathway.
Results:
: The study revealed a significant upregulation of SMS expression in LGGs compared to normal brain tissues. High SMS expression was associated with certain clinical and pathological features, including older age, astrocytoma, higher World Health Organization grade, poor disease-specific survival, disease progression, non-1p/19q codeletion, and wild-type isocitrate dehydrogenase. Cox regression analysis identified SMS as a risk factor for overall survival. Bioinformatics analysis showed enrichment of eosinophils, T cells, and macrophages in LGG samples, while proportions of dendritic (DC) cells, plasmacytoid DC (pDC) cells, and CD8+ T cells were decreased.
Conclusion
: High SMS expression in LGGs may promote tumor occurrence through cellular proliferation and modulation of immune cell infiltration. These findings suggest the prognostic value of SMS in predicting clinical outcomes for LGG patients.

5.GPX7 regulates proliferation and senescence of hepatocellular carcinoma cells through the PI3K-AKT pathway

Objective To investigate the effects of glutathione peroxidase 7(GPX7)on the proliferation and senescence of hepatocellular carcinoma(HCC)cells and its potential value as a therapeutic target.Methods Clinical and transcriptomic data from HCC patients in The Cancer Genome Atlas(TCGA)database were analyzed to evaluate the relationship between GPX7 expression and prognosis.Western blotting,qRT-PCR,CCK-8 assay,EdU staining,and SA-β-Gal staining were employed to compare the expression and function of GPX7 in HCC cell lines and normal liver cells.A stable GPX7-knockdown Hep3B cell line was established,and transcriptomic changes were analyzed using RNA-seq.The PI3K inhibitor LY294002 and its activator 740 Y-P were utilized to determine the role of GPX7 in regulating cell senescence via the PI3K-AKT pathway.An orthotopic xenograft model was constructed using 18 male nude mice(6 weeks old,18~22 g)to observe tumor growth.Tumor and adjacent tissue samples from 56 HCC patients(obtained from the Eighth Medical Center of Chinese PLA General Hospital between January 2012 and December 2022)were analyzed to assess the relationship between GPX7 expression and clinicopathological features.Results TCGA data analysis revealed that GPX7 was significantly highly expressed in HCC tumor tissues(P<0.01),and the patients with high GPX7 expression had obviously shorter overall survival(OS)(P=0.018).Western blotting and qRT-PCR showed that the expression level of GPX7 was notably higher in the HCC cell lines than the normal liver cells(P<0.01).After GPX7 knockdown,the CCK-8 absorbance values of HCC cells were significantly reduced(P<0.01),the proportion of EdU-positive cells was obviously decreased(P<0.01),and the proportion of SA-β-Gal positive cells was notably increased(P<0.01).RNA-seq analysis revealed that differentially expressed genes were significantly enriched in the PI3K-AKT signaling pathway(P<0.05).The PI3K inhibitor LY294002 significantly enhanced the GPX7 knockdown-induced cell senescence(P<0.01),while the activator 740 Y-P partially reversed this effect(P<0.05).In the xenograft model,GPX7 knockdown significantly inhibited tumor growth in nude mice(P<0.01),and the PI3K activator 740 Y-P partially reversed this effect(P<0.01).Clinical sample analysis displayed that high GPX7 expression was negatively correlated with tumor differentiation(P<0.05),and its high expression was associated with shortened OS(HR=2.61,95%CI:1.26~5.39,P=0.005).Conclusion GPX7 regulates cell proliferation and senescence through the PI3K-AKT pathway,and may serve as a potential therapeutic target for HCC treatment.
6.Clinical application of double incision in phacoemulsification combined with trabeculectomy

Objective Conducting phacoemulsification combined with trabeculectomy through double incision to treat glaucoma and cat-aract,and observing the visual acuity,intraocular pressure before and after the operation and complications of this therapy. Methods Totally 60 patients (78 eyes) with glaucoma and cataract from January 2010 to December 2013 in our hospital were observed. 32 patients (40 eyes) who were given phacoemulsification combined with trabeculectomy through double incision were regarded as the observation group,and the other 28 patients (38 eyes) who were merely given phacoemulsification were regarded as the control group. The visual acuity,intraocular pres-sure and anterior chamber of the two groups were observed and recorded,and they were followed up for 36 months. Results Visual acuity and intraocular pressure of the two groups were improved after operation,but the observation group achieved a better result which showed a significant difference (P< 0. 05). Anterior chamber inflammation occurred in patients of the observation group,and 2 cases were of anterior chamber exudation. The control group,by contrast,has less anterior chamber inflammation. There was no hyphema,shallow anterior chamber, choroidal detachment occurred in both of the two groups. Conclusion Using phacoemulsification combined with trabeculectomy through double incision to treat glaucoma and cataract could receive a good recovery of visual acuity and intraocular pressure. Although,more severe anterior chamber inflammation occurred after operation compared with the control group,it can be alleviated soon after medication. This meth-od is a good therapy which can reduce pain and economic burden of patients from multiple operations. But it requires a strict selection of ap-propriate case.
