Background and Objectives: Metoclopramide is an antagonist of dopamine D2 receptor and is capable of alleviating chemotherapy-induced nausea and vomiting. However, its underlying mechanisms and function in improving the efficiency of chemotherapy are not fully understood. In this study, we investigated the sensitizing effect of metoclopramide on the platinum-based drugs-mediated apoptosis in human head and neck cancer cells.Subjects and Method Apoptosis was analyzed using a cell-based cytometer. The protein expression and messenger ribonucleic acid (mRNA) levels were assessed by Western blotting and real-time polymerase chain reaction, respectively. Results: Metoclopramide sensitized the platinum-based drug (cisplatin and oxaliplatin)-mediated apoptosis in AMC-HN4 cells, but not in normal cells. Mechanistically, we found that metoclopramide decreased Mcl-1 protein expression through post-translational regulation. Moreover, the overexpression of Mcl-1 prevented apoptosis by combined treatment of metoclopramide and platinum-based drugs. Conclusion Metoclopramide induced proteasome-mediated Mcl-1 downregulation, resulting in increased sensitivity to platinum-based drugs.

Background: and Purpose The Treat Stroke to Target (TST) was a randomized clinical trial involving French and Korean patients demonstrating that a lower low-density lipoprotein cholesterol (LDL-C, <70 mg/dL) target group (LT) experienced fewer cerebro-cardiovascular events than a higher target (90–110 mg/dL) group (HT). However, whether these results can be applied to Asian patients with different ischemic stroke subtypes remains unclear. Methods: Patients from 14 South Korean centers were analyzed separately. Patients with ischemic stroke or transient ischemic attack with evidence of atherosclerosis were randomized into LT and HT groups. The primary endpoint was a composite of ischemic stroke, myocardial infarction, coronary or cerebral revascularization, and cardiovascular death. Results: Among 712 enrolled patients, the mean LDL-C level was 71.0 mg/dL in 357 LT patients and 86.1 mg/dL in 355 HT patients. The primary endpoint occurred in 24 (6.7%) of LT and in 31 (8.7%) of HT group patients (adjusted hazard ratio [HR]=0.78; 95% confidence interval [CI]=0.45–1.33, P=0.353). Cardiovascular events alone occurred significantly less frequently in the LT than in the HT group (HR 0.26, 95% CI 0.09–0.80, P=0.019), whereas there were no significant differences in ischemic stroke events (HR 1.12, 95% CI 0.60–2.10, P=0.712). The benefit of LT was less apparent in patients with small vessel disease and intracranial atherosclerosis than in those with extracranial atherosclerosis. Conclusion In contrast to the French TST, the outcomes in Korean patients were neutral. Although LT was more effective in preventing cardiovascular diseases, it was not so in stroke prevention, probably attributed to the differences in stroke subtypes. Further studies are needed to elucidate the efficacy of statins and appropriate LDL-C targets in Asian patients with stroke.

BACKGROUND/OBJECTIVES: Rosa hybrida has been demonstrated to exert biological effects on several cell types. This study investigated the efficacy of the edible ethanol extract of R.hybrida (EERH) against human colorectal carcinoma cell line (HCT116) cells.MATERIALS/METHODS: HCT116 cells were cultured with different concentrations of EERH (0, 400, 600, 800, and 1,000 µg/mL) in Dulbecco’s modified Eagle medium. Cell viability was measured using the 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide and viable cell counting assays. Cell cycle pattern was observed by flow cytometry analysis. The wound-healing migration assay, invasion assay, and zymography were used to determine the migratory and invasive level of HCT116 cells treated with EERH. The protein expression and binding ability level of HCT116 cells following EERH treatment were analyzed via immunoblotting and the electrophoretic mobility shift assay. RESULTS: EERH suppressed HCT116 cell proliferation, thus arresting the G1-phase cell cycle.It also reduced cyclin-dependent kinases and cyclins, which are associated with p27KIP1 expression. Additionally, EERH differentially regulated the phosphorylation of extracellular signal-regulated kinase 1/2, c-Jun NH2-terminal kinase, p38, and protein kinase B. Moreover, EERH treatment inhibited the enzymatic activity of matrix metalloproteinase-9 (MMP-9) and MMP-2, resulting in HCT116 cell migration and invasion. The EERH-induced inhibition of MMP-9 and MMP-2 was attributed to the reduced transcriptional binding of activator protein-1, specificity protein-1, and nuclear factor-κB motifs in HCT116 cells. Kaempferol was identified as the main compound contributing to EERH's antitumor activity. CONCLUSION EERH inhibits HCT116 cell proliferation and metastatic potential. Therefore, it is potentially useful as a preventive and curative nutraceutical agent against colorectal cancer.

Excessive wear of the teeth, which causes functional, aesthetic problems, or sensitivity symptoms, is referred to as pathological wear. If this leads to the collapse of occlusion, extensive restoration is necessary to restore function and aesthetics.When restoring pathologically worn dentition, it is essential to identify the cause of the wear before determining treatment plan to get a favorable prognosis. In this case, a patient exhibited severe tooth wear with decreased vertical dimension and clinical, radiographic examination, and medical history indicated the pathological wear caused by a combination of factors. Consequently, full-mouth rehabilitation was indicated and the vertical occlusal height was determined considering the appropriate restorative space. Based on the causes of the wear, the restoration method, materials, and occlusal pattern were selected and successfully restored to normal occlusal state through full-mouth rehabilitation with zirconia full-veneered crowns.

Purpose: . The purpose of this study is to analyze the biomechanical differences in rotation angle-torque according to the type of implant and to compare the effects of contamination and sandblasting on the removal torque value (RTV) of abutment screws. Materials and methods: . For this study, an automated device for tightening and loosening implant screws was developed, and each 15 implant-abutment complexes of external and internal connection type were prepared, divided into three groups according to the surface treatment of the screw:control group (no treatment), experimental group-1 (artificial saliva contamination and chlorhexidine (CHX) rinsing), and experimental group-2 (artificial saliva contamination, CHX rinsing, and subsequent 50 µm Al 2 O 3 sandblasting). FirstRTV was measured for each group, followed by different post-treatment procedures for the screws, and then Second-RTV was measured. During the procedure, the removal torque value and rotation angle vs. time were recorded at a 20 data/s.A standardized protocol was followed for all the tightening and loosening procedures: screw was tightened with 30 Ncm torque and maintained for 5 s, and then rotated in the reverse direction until the torque value reached 0 Ncm. After 10min of rest period, the screws were loosened to initial point. Results: . Statistical analysis of measured data revealed that the internal connection type showed significantly higher First-RTV compared to the external connection type, however, no significant differences in Second-RTV were found. Both experimental groups 1and 2 showed a decrease in RTVs for both implant types, while the control group showed a decrease in RTV only for the internal type. The internal connection type required a significantly larger rotation angle during the tightening and loosening process. Conclusion . Within the limitations of this study, both artificial saliva contamination and CHX cleansing, as well as sandblasting, decreased RTV in both internal and external connection types. Internal connections were more susceptible to the effects of contaminants.
