1.Effect of Silencing BHLHE41 on Angiotensin Ⅱ-induced Hypertrophy and Apoptosis of H9C2 Cardiomyocytes

Objective To investigate the effects of basic helix-loop-helix family member e41(BHLHE41)on angiotensin Ⅱ(Ang Ⅱ)-induced hypertrophy and apoptosis of H9C2 cardiomyocytes.Methods The expression level of BHLHE41 stimulated by Ang Ⅱ was investigated,and the cells were divided into normal group and Ang Ⅱ group.To investigate the effect of silencing BHLHE41(siBHLHE41)on cardiomyocyte hypertrophy and apoptosis,the cells were divided into 4groups:blank group[transfected with small in-terfering RNA(siRNA)+phosphate buffered saline(PBS)],siBHLHE41 group[transfected with siBHLHE41+PBS],model group(transfected with siRNA+Ang Ⅱ)and experimental group(transfected with siBHLHE41+Ang Ⅱ).The mRNA levels of BHLHE41,β-myosin heavy chain(β-MHC),atrial natriuretic peptide(ANP),B-type natriuretic peptide(BNP),poly ADP-ribose polymer-ase(PARP),B-cell lymphoma-2(Bcl-2),B-cell lymphoma-2-associated X protein(Bax),and p53 were detected by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR).The protein expression levels of BHLHE41,Bax,Bcl-2,PARP and p53 were detected by Western blot.Phalloidin staining was used to observe the surface area of cardiomyocytes,TUNEL staining was used to observe cardiomyocytes apoptosis.Results Compared with the normal group,the mRNA and protein expression levels of BHL-HE41 was higher in the Ang Ⅱ group.Compared with blank group,the mRNA expression levels of β-MHC,ANP,BNP in the model group were upregulated,and the protein and mRNA expression levels of PARP,Bax and p53 in model group were significantly increased,the expression level of Bcl-2 was significantly decreased,the surface area of cardiomyocytes was significantly increased,and the apopto-sis rate of cardiomyocytes was significantly increased(P<0.05).Compared with model group,the mRNA expression levels of β-MHC,ANP and BNP in the experimental group were upregulated,and the protein and mRNA expression levels of PARP,Bax and p53 in experi-mental group were significantly increased,the expression level of Bcl-2 was significantly decreased,the surface area of cardiomyocytes was increased,and the apoptosis rate of cardiomyocytes was significantly increased(P<0.05).Conclusion Silencing BHLHE41 can aggravate Ang Ⅱ-induced hypertrophy and apoptosis of H9C2 cardiomyocytes,which may be related to the p53 activation of the apoptotic signaling pathway.

3.Silencing YTHDF2 improves Ang Ⅱ-induced hypertrophy and apoptosis of primary neonatal rat cardiomyocytes

Objective To investigate the effect of YTH domain family protein 2(YTHDF2)on an-giotensin Ⅱ(Ang Ⅱ)-induced hypertrophy and apoptosis of primary neonatal rat cardiomyocytes.Methods The expression level of YTHDF2 was detected in the primary neonatal rat cardiomyo-cytes with or without Ang Ⅱ stimulation(AngⅡ group and normal group).The cells were divid-ed into blank group(transfected with siRNA+PBS),siYTHDF2 group(transfected with siYTHDF2+PBS),model group(siRNA+Ang Ⅱ)and experimental group(siYTHDF2+Ang Ⅱ)to investi-gate the effects of silencing YTHDF2 on the hypertrophy and apoptosis of cardiomyocytes.West-ern blotting and RT-qPCR were used to detect the expression of YTHDF2 at protein and mRNA levels,and RT-qPCR was employed to measure the mRNA levels of myocardial hypertrophic related genes atrial natriuretic peptide(ANP),brain natriuretic peptide(BNP)and beta-myosin heavy chain(β-MHC),and cardiomyocyte apoptosis related genes Bax and B lymphocytoma 2 gene(Bcl-2).The surface area of cardiomyocytes was observed by α-actin immunofluorescence staining.Cardiomyocyte apoptosis was observed by TUNEL staining,and the binding relationship between YTHDF2 and Bcl-2 was verified by immunoprecipitation.Results The expression of YTHDF2 at protein and mRNA levels were significantly higher in the AngⅡ group than the nor-mal group(1.49±0.03 vs 0.97±0.09,1.50±0.08 vs 1.00±0.07,P<0.05).Compared with the blank group,the surface area of cardiomyocytes was notably enlarged,apoptotic rate was obvi-ously increased,the mRNA levels of ANP,BNP,β-MHC and Bax were significantly increased,and that of Bcl-2 was remarkably decreased in the model group(P<0.05).The experimental group obtained decreased surface area and apoptotic rate of cardiomyocytes,lower mRNA levels of ANP,BNP,β-MHC and Bax,and increased mRNA expression of Bcl-2(P<0.05).Conclusion Silencing YTHDF2 can alleviate Ang Ⅱ-induced hypertrophy and apoptosis in primary neonatal rat cardiomyocyte,and YTHDF2 inhibits the expression of Bcl-2 by binding to it.

Objective:To summarize the best evidence for safely managing home parenteral nutrition in adults.Methods:Clinical decisions, guidelines, evidence summaries, expert consensus, systematic reviews, Meta-analyses, and so on related to the safety management of home parenteral nutrition in adults were electronically retrieved on UpToDate, Guidelines International Network, Agency for Healthcare Research and Quality, Joanna Briggs Institute Evidence-Based Health Care Center Database, Cochrane Library, PubMed, Web of Science, China National Knowledge Infrastructure, WanFang Data, China Biology Medicine disc and so on. The search period was from database establishment to December 30, 2023. Two researchers who received evidence-based nursing training conducted literature quality evaluation and evidence extraction.Results:A total of 14 articles were included, including three guidelines, six expert consensus, one hospital standard, and four meta-analyses. Thirty-three pieces of evidence were summarized from four aspects: team building, transitional care, preventing and managing complications, monitoring, follow-up, and quality control.Conclusions:The best evidence for safety management of home parenteral nutrition in adults is summarized. Medical and nursing staff should select and apply evidence based on clinical scenarios and patient preferences.

6.The correlation of helper T lymphocyte 1/helper T lymphocyte 2 with clinical and image features in patients with idiopathic pulmonary fibrosis

Objective To identify whether the helper T lymphocyte 1 (Th1)/helper T lymphocyte 2 (Th2) of patients' serum and bronchoalveolar lavage fluid (BALF) at admission could represent the severity of idiopathic pulmonary fibrosis (IPF) and whether its change at six months could predict the progression of the disease.Methods Eighty-three patients with IPF were subjected to pulmonary function tests (PFFs),dyspnea scores,arterial blood gas analysis,six-minute walk test (6MWT) and high-resolution computed tomography (HRCT).The serum and BALF specimen of these patients were obtained as well as 20 control serum and 10 control BALF specimen.A total of 55 patients were followed up,and their BALF and serum levels of interferon γ (IFNγ) and IL-4 were detected by enzyme-linked immunoadsorbent assay (ELISA).The correlation between the IFNγ/IL-4 levels (at admission and the change of that at six months follow-up) and the clinical,physiological and image features in the IPF patients were analyzed.Results The baseline serum and BALF level of IFNγ/IL-4 (0.8 ± 0.3) in the IPF patients was lower than that in the control group (1.4 ± 0.2),which showed significant correlation with the course of disease,dyspnea scores,FEV1 %,FVC%,TLC%,maximum desaturation,6MWD and CT-fib (all P values < 0.05).The serum level of IFNγ/IL-4 showed positive correlation with CT-alv (r =0.340,P < 0.01).During follow-up,no statistic difference was found in the serum levels of IFNγ,IL-4 and IFNγ/IL-4 between the IPF patients with or without glucocorticoids treatment.There were significant aggravation in the dyspnea scores,FEV1%,FVC%,CT-alv,CT-fib,IFNγ and IL-4 at six months follow-up.Significant correlation had been showed between the change of the serum IFNγ/IL-4 level with the changes of the dyspnea scores,FVC%,TLC%,DLCO%,6MWD and CT-fib in the IPF patients (P <0.05).Conclusions There are disequilibrium of the Th1/Th2 in the serum and the BALF of the IPF patients.The Th1/Th2 level could represent severity of the disease,and the serum level change of Th1/Th2 in the follow-up could predict the progression of the diseases in the IPF patients.
7.Role of Head Band for Nosocomial Infection Control in Operating Room

OBJECTIVE To discuss the role of head hygiene for nosocomial infection control in operating room.METHODS The head band was weared to improve head hygiene,and enhance the consciousness in control nosocomial infection.RESULTS From Mar 2004 to Mar 2006 there were 2534 cases operated and 23 cases suffered incision infection(0.91%) when surgeon was without use of head band;but from Mar 2004 to Mar 2006 there were 3014 cases operated and the incision infection rate was 0.40%(12 cases) when surgeon was with use of head band.It was an effective method for reducing sweat-inducing contamination by wearing head band to control nosocomial infection in operating room.CONCLUSIONS To improve the head hygiene by wearing head band is arelatively simple practice,it can control nosocomial infection in operating room.

Objective To study the effect of diet containing different proportions of porphyra powder on relieving constipation, volatile organic compounds and inhibiting intestinal E. coli. Method Mice were divided into 3 groups: blank group, 2 test groups.
