1.A longitudinal study of relationship between psychological capital and meaning in life among college students

Objective:To investigate the longitudinal mutual predictive relationship between college students' psychological capital and meaning in life across time.Methods:A total of 604 college students were selected for a one-year two-stage longitudinal tracking(T1 and T2),using the Psychological Capital Questionnaire for Adolescent Students(PC-QAS)and Chinese Meaning in Life Questionnaire(C-MLQ)for measurement.Results:The T1 PC-QAS scores were positively correlated with the T1 C-MLQ scores and T2 PC-QAS scores(r=0.61,0.51,Ps<0.001).The T2 C-MLQ scores were positively correlated with the T2 PC-QAS scores and T1 C-MLQ scores(r=0.58,0.57,Ps<0.001).The results of cross lagged regression analysis indicated that the T1 PC-QAS scores posi-tively predicted the T2 C-MLQ scores(β=0.15,P<0.001),and T1 C-MLQ scores positively predicted T2 PC-QAS scores(β=0.19,P<0.01).Conclusion:The psychological capital and meaning in life of college students have a certain degree of stability,and the two could predict each other.
2.Molecular Mechanism of Astragali Radix and Hedyotis diffusa Regulating Sp1-miR-582-3p-p27 Axis to Inhibit Lung Adenocarcinoma Proliferation

ObjectiveTo investigate the effects of excessive activation of the Specific protein 1 (Sp1)-MicroRNA-582-3p (miR-582-3p)-cyclin-dependent kinase inhibitor (p27) axis on the cell cycle and proliferation of A549 lung adenocarcinoma cells at the cellular level, thereby investigating the possible mechanisms by which Astragali Radix and Hedyotis diffusa(A-H) regulate the axis to inhibit A549 cells proliferation. MethodsLentiviral plasmid transfection technology was used to obtain four stable A549 transgenic cell lines: shRNA-Sp1+LV-miR-582-3p mimic, shRNA-Sp1+LV-mNC, shRNA-NC+LV-miR-582-3p mimic, and shRNA-NC+LV-mNC. Real-time PCR was used to detect the effect of Sp1 on miR-582-3p expression in A549 lung adenocarcinoma cells. CCK-8 and colony formation assay were used to detect the effect of Sp1 on cell proliferation through miR-582-3p, while flow cytometry was used to detect the effect of Sp1 on cell cycle via miR-582-3p. Western blot was used to detect the effect of Sp1 on cyclin in A549 cells through miR-582-3p. CCK-8 was employed to detect the effects of different concentrations (5%, 10%, 20%, 40%, 80%) of A-H on A549 cell viability, and the optimal concentration was selected for subsequent mechanism exploration. Stable transgenic strains oe-Sp1 and control oe-Vector were constructed using the above-mentioned transfection techniques. Real-time PCR was then used to investigate the effect of A-H on miR-582-3p expression in A549 cells, while Western blot was employed to detect the effect of A-H on Sp1 and p27 expression in A549 lung adenocarcinoma cells. ResultsCompared with the shRNA-NC+LV-mNC group, the expression of miR-582-3p was significantly inhibited in the shRNA-Sp1+LV-mNC group (P<0.01). Compared with the shRNA-Sp1+LV-mNC group, the shRNA-Sp1+LV-miR-582-3p mimic group significantly reversed miR-582-3p expression (P<0.01), indicating that Sp1 has a significant regulatory effect on miR-582-3p. Compared with the shRNA-Sp1+LV-mNC group, the shRNA-Sp1+LV-miR-582-3p mimic group significantly reversed cell proliferation (P<0.01), suggesting that miR-582-3p can partially reverse the proliferative effect of Sp1 on A549 lung adenocarcinoma. Compared with the shRNA-Sp1+LV-mNC group, the shRNA-Sp1+LV-miR-582-3p mimic group showed a significant decrease in G0/G1 proportions, while the G2/M and S phase proportions increased significantly (P<0.01), indicating that miR-582-3p can partially reverse the effect of Sp1 on the A549 cell cycle. Compared with the shRNA-Sp1+LV-mNC group, the shRNA-Sp1+LV-miR-582-3p mimic group significantly reversed the expression of various proteins with the decrease of p27 and the increase of Cyclin D1, CDK4, Cyclin E, CDK2, p-Rb, and E2F3 (P<0.01), indicating that miR-582-3p can partially reverse the effect of Sp1 on the cell cycle of A549 lung adenocarcinoma. Compared with the blank group, the serum containing A-H significantly inhibited A549 cell viability in a concentration-dependent manner. Therefore, 10% A-H was selected for intervention of subsequent mechanism experiments. Compared with A-H+oe-Vector, A-H+oe-Sp1 significantly reversed the downregulation of Sp1, miR-582-3p, and the upregulation of p27 (P<0.01). This suggests that Sp1 can partially reverse the effects of A-H on miR-582-3p and p27 levels in A549 lung adenocarcinoma cells. ConclusionOveractivation of the Sp1-miR-582-3p-p27 axis significantly promotes the proliferation of A549 lung adenocarcinoma cells, and the potential mechanism of the inhibitory effect of A-H on the proliferation of A549 cells may be related to its inhibition on the overactivation of the Sp1-miR-582-3p-p27 axis.
3.Polyphyllin Ⅶ promotes hepatic stellate cell ferroptosis via the HIC1/CX3CL1/GPX4 axis

Ferroptosis has been shown to mediate the development of fibrosis.Polyphyllin Ⅶ(PP7),a bioactive component of Paris polyphylla,exhibits potent anti-inflammatory activity and can significantly alleviate liver fibrosis.In this study,treatment with PP7 significantly inhibited the proliferation and activation of hepatic stellate cells(HSCs),which could be suppressed by a ferroptosis inhibitor.In addition,it promoted HSC ferroptosis by suppressing glutathione(GSH)peroxidase 4(GPX4)and enhanced the expression of CX3C chemokine ligand 1(CX3CL1).Depletion of CX3CL1 attenuated the effects of PP7 on the activation and ferroptosis of HSCs and the expression of GPX4.Notably,CX3CL1 directly interacted with GPX4,triggering HSC ferroptosis.The transcription factor hypermethylated in cancer 1(Hic1),which binds to the Cx3cl1 promoter,increased the expression of CX3CL1.Its absence resulted in downregulation of CX3CL1,sup-pressing the GPX4-dependent ferroptosis of PP7-treated HSCs and promoting their activation.HIC1 was found to directly interact with PP7 at the GLY164 site.Co-culture experiments showed that PP7-induced HSC ferroptosis attenuated macrophage recruitment by regulating inflammation-related genes.HSC-specific inhibition of HIC1 counteracted PP7-induced collagen depletion and HSC ferroptosis in vivo.These findings suggest that PP7 induces HSC ferroptosis through the HIC1/CX3CL1/GPX4 axis.

6.Research progress of perioperative immunotherapy for esophageal cancer

Esophageal cancer is one of the common malignant tumors in the digestive system, with high morbidity and mortality rates and poor prognosis. Currently, the treatment modality of esophageal cancer has gradually transitioned to surgery-based comprehensive treatment. For resec-table esophageal cancer, the combination with perioperative treatment can significantly improve the efficacy and prognosis of patients. Surgery combined with neoadjuvant chemoradiotherapy is the first choice for patients with locally advanced esophageal cancer, but many patients still suffer from recurrence and metastasis with serious adverse effects. In recent years, the emergence of immuno-therapeutic drugs such as immune checkpoint inhibitors (ICIs) has created a new era of tumor treat-ment. The application of ICIs in the perioperative period of esophageal cancer has also gradually attracted extensive attention from researchers. The authors introduce the current research progress of perioperative immunotherapy for esophageal cancer from the molecular mechanism of immuno-therapy for esophageal cancer, current perioperative treatment modalities, and potential biomarkers, aiming to provide important references for further research and clinical practice.

8.Comparison of ultrasound volume navigation and O-arm navigation in percutaneous pedicle screw fixation for thoracolumbar vertebral fractures

Objective:To compare the efficacy of ultrasound volume navigation (UVN) and that of O-arm navigation in the treatment of thoracolumbar vertebral fractures with percutaneous pedicle screw fixation.Methods:A retrospective study was conducted to analyze the clinical data of 29 patients with thoracolumbar vertebral fracture who had been treated with percutaneous pedicle screw fixation at Department Ⅰ of Minimally Invasive Spinal Surgery, Luoyang Orthopedic Hospital from January 2022 to June 2023. The patients were assigned into 2 groups according to different navigation methods. In group UVN of 17 cases, there were 14 males and 3 females with a mean age of 50.0 (44.0, 53.5) years, and the pedicle screw fixation was guided by UVN; in group O-arm of 12 cases, there were 10 males and 2 females with an age of (40.6±11.1) years, and the pedicle screw fixation was guided by O-arm navigation. The 2 groups were compared in terms of insertion time per screw, total dose of intraoperative radiation, radiation dose per screw, accuracy of screw placement, rate of facet joint violation, and visual analogue scale (VAS) pain scores for lumbar pain at postoperative 1, 3, and 6 months.Results:There were no statistically significant differences in the preoperative general data between the 2 groups, indicating comparability ( P>0.05). There was no statistically significant difference in the insertion time per screw between group UVN and group O-arm [(13.2±1.3) min versus (14.1±1.3) min] ( P>0.05). The total dose of intraoperative radiation and radiation dose per screw in group UVN [(1.80±0.54) mSv and (0.41±0.10) mSv] were significantly lower than those in group O-arm [(3.52±0.33) mSv and (0.85±0.11) mSv] ( P<0.05). The accuracy of screw placement and rate of facet joint violation in group UVN [94.7% (71/75) and 17.6% (3/17)] were not significantly different from those in group O-arm [92.2% (47/51) and 8.3% (1/12)]] ( P>0.05). The VAS score at postoperative 1 month in group UVN [4.0 (3.5, 5.0) points] was significantly lower than that in group O-arm [5.0 (5.0, 6.0) points] ( P<0.05). There were no statistically significant differences between the 2 groups in the VAS pain scores at postoperative 3 and 6 months ( P>0.05). Conclusions:In the treatment of thoracolumbar vertebral fractures with percutaneous pedicle screw fixation, screw placement guided by UVN is as time-saving, intuitive, and accurate as that guided by O-arm navigation. However, UVN is better than O-arm navigation in radiation avoidance and pain relief.

10.Comparison of screw placement guided by O-arm navigation and ultrasound volume navigation in minimally invasive transforaminal lumbar interbody fusion.

OBJECTIVE:
To compare the effectiveness of O-arm navigation and ultrasound volume navigation (UVN) in guiding screw placement during minimally invasive transforaminal lumbar interbody fusion (MIS-TLIF) surgery.
METHODS:
Sixty patients who underwent MIS-TLIF surgery for lumbar disc herniation between June 2022 and June 2023 and met the selection criteria were included in the study. They were randomly assigned to group A (screw placement guided by UVN during MIS-TLIF) or group B (screw placement guided by O-arm navigation during MIS-TLIF), with 30 cases in each group. There was no significant difference in baseline data, including gender, age, body mass index, and surgical segment, between the two groups ( P>0.05). Intraoperative data, including average single screw placement time, total radiation dose, and average single screw effective radiation dose, were recorded and calculated. Postoperatively, X-ray film and CT scans were performed at 10 days to evaluate screw placement accuracy and assess facet joint violation. Pearson correlation and Spearman correlation analyses were used to observe the relationship between the studied parameters (average single screw placement time and screw placement accuracy grading) and BMI.
RESULTS:
The average single screw placement time in group B was significantly shorter than that in group A, and the total radiation dose of single segment and multi-segment and the average single screw effective radiation dose in group B were significantly higher than those in group A ( P<0.05). There was no significant difference in the total radiation dose between single segment and multiple segments in group B ( P>0.05), while the total radiation dose of multiple segments was significantly higher than that of single segment in group A ( P<0.05). No significant difference was found in the accuracy of screw implantation between the two groups ( P>0.05). In both groups, the grade 1 and grade 2 screws broke through the outer wall of the pedicle, and no screw broke through the inner wall of the pedicle. There was no significant difference in the rate of facet joint violation between the two groups ( P>0.05). In group A, both the average single screw placement time and screw placement accuracy grading were positively correlated with BMI ( r=0.677, P<0.001; r=0.222, P=0.012), while in group B, neither of them was correlated with BMI ( r=0.224, P=0.233; r=0.034, P=0.697).
CONCLUSION
UVN-guided screw placement in MIS-TLIF surgery demonstrates comparable efficiency, visualization, and accuracy to O-arm navigation, while significantly reducing radiation exposure. However, it may be influenced by factors such as obesity, which poses certain limitations.
