1.Rapid Qualitative Analysis Methods and Their Application in Implementation Science

Implementation science (IS) aims to systematically analyze and address the real-world gaps from evidence to practice and the influencing factors of the context. It is necessary to carry out qualitative research to gather relevant implementation outcomes. Nevertheless, traditional qualitative analysis has issues such as consuming a great deal of time and energy, and it is unable to promptly provide the crucial data required for implementation science research. The Rapid Qualitative Analysis (RQA) method, through semi-structured interviews and the adoption of techniques such as immediate data condensation and matrix analysis, can effectively shorten the cycle of qualitative data collection and data processing. RQA can promptly identify social determinants of health such as structural barriers, facilitators, and the behavioral characteristics of target groups. It provides a real-time basis for public health decision-making, the interpretation of complex social phenomena, and the process and effectiveness evaluation of research projects. Although RQA is difficult to conduct in-depth theoretical analysis based on grounded theory, its efficiency and flexibility make it the preferred tool for large-scale and time-sensitive research. Thus, it has been widely applied in implementation science research. This paper sorts out the core concepts and commonly used technical methods of RQA, as well as the differences between RQA and traditional qualitative analysis. It also explores the applications of RQA in intervention optimization, process evaluation, and implementation outcome evaluation. By integrating specific cases, this paper clarifies its application value in the field of implementation science. In the future, it is advisable to explore the integration of RQA with technologies such as artificial intelligence and big data, in order to bridge the gap between the transformation of scientific research achievements into practice. Under circumstances of limited resources or tight time constraints, RQA can be used to efficiently conduct implementation science research, providing convenient and scientific methodological and technical support for accelerating evidence-based practice.
2.Chinese Medicine in Regulating Ferroptosis for Treatment of Malignant Tumors: A Review

Malignant tumors are one of the major causes of death in the population. Owing to limited clinical treatments, susceptibility to drug resistance, and generally low cure rates of conventional therapies, new treatment strategies need to be explored. Compared with existing therapies, traditional Chinese medicine (TCM) has unique advantages, such as low side effects, in the treatment of malignant tumors. Ferroptosis is a recently characterized form of regulated cell death associated with iron metabolism imbalance, lipid peroxidation, antioxidant system malfunction and other aspects. Studies have shown that TCM regulates Fe3+, Fe2+, glutathione, glutathione peroxidase 4 and other substances related to ferroptosis, thereby affecting lipid peroxidation and antioxidant processes, and then inducing ferroptosis. Through these mechanisms, TCM plays a key role in inhibiting the growth and spread of tumor cells and is involved in multiple stages of malignant tumor progression. In this study, we systematically retrieved the literature indexed in PbuMed and China National Knowledge Infrastructure (CNKI) with the keywords TCM, ferroptosis, and malignant tumors. We outlined the mechanisms of ferroptosis and its association with malignant tumors, and summarized the research progress on the prevention and treatment of malignant tumors through the modulation of ferroptosis by TCM monomers, single herbs, and compounds. The study aims to provide new perspectives for the prevention and treatment of malignant tumors by TCM.
3.Research progress on high adaptability of pathogenic fungi to drugs

Fungal infections have emerged as a critical public health issue endangering human health. However, the existing arsenal of antifungal agents is limited in diversity and is commonly plagued by drawbacks including narrow antimicrobial spectrums and the frequent emergence of drug resistance, which severely compromises the efficacy of clinical treatments. Pathogenic fungi can develop extensive adaptability to currently available drugs through multiple mechanisms, which are mainly manifested in three aspects: drug resistance, tolerance and persistence. The molecular mechanisms and regulatory pathways underlying drug resistance, tolerance and persistence in pathogenic fungi were systematically summarized in this review, and the counteractive strategies such as combination therapy and the development of novel antifungal agents were further discussed, which aimed to provide theoretical basis and practical reference for the precision treatment of fungal infections.

5.Influencing factors for condom use among men who have sex with men

Objective:
To investigate the status of condom use and its influencing factors among men who have sex with men (MSM), so as to provide a basis for improving condom utilization rates and AIDS prevention and control in this population.
Methods:
From May to October 2024, a snowball sampling method was employed to recruit MSM in Songjiang District, Shanghai Municipality. Self-administered questionnaires were used to collect data on demographic characteristics, AIDS-related knowledge, sexual behaviors, pre-exposure prophylaxis (PrEP) and post-exposure prophylaxis (PEP), and condom use in the past six months. Multivariable logistic regression model was used to analyze the influencing factors for consistent condom use.
Results:
A total of 921 MSM were surveyed, with a median age of 29.00 (interquartile range, 9.00) years. Among them, 697 (75.68%) were aware of AIDS-related knowledge, 826 (89.69%) expressed willingness to use PrEP, and 835 (90.66%) were willing to use PEP. Additionally, 787 (85.45%) MSM reported their age at first homosexual intercourse as ≥18 years, while 519 (56.35%) reported consistent condom use in the past six months. Multivariable logistic regression analysis revealed that MSM who were aware of AIDS-related knowledge (OR=0.582, 95% CI: 0.423-0.801), willing to use PrEP (OR =0.611, 95% CI: 0.385-0.969), and whose age at first homosexual intercourse was <18 years (OR=0.480, 95% CI: 0.330-0.700) were less likely to consistent use condoms.
Conclusion
The proportion of consistent condom use among the MSM remains relatively low, which is primarily associated with AIDS-related knowledge, willingness to use PrEP, and the age at first homosexual intercourse.
6.Effects of safflower polysaccharide on ischemic stroke in rats by regulating the Gas6/Axl signaling pathway

OBJECTIVE To investigate the effects of safflower polysaccharide (SPS) on ischemic stroke (IS) in rats by regulating the growth arrest-specific protein 6 (Gas6)/Axl receptor tyrosine kinase (Axl) signaling pathway. METHODS The modified suture-occluded method was employed to establish a rat model of middle cerebral artery occlusion (MCAO) for IS. The rats were then randomly divided into the model (Model) group, SPS low- and high-dose (SPS-L, SPS-H, 50 and 100 mg/kg) groups, and integrated traditional Chinese and Western medicine (SPS-H+Nim, SPS 100 mg/kg+Nim 15 mg/kg) group, with 10 rats in each group. Another 10 rats were selected as the sham operation (Sham) group. Rats in each group were administered corresponding doses of medication or an equal volume of normal saline intragastrically, with continuous intervention for 14 days. The neurological function of rats in each group was evaluated 24 hours after drug administration. The morphological changes in hippocampal tissue were observed. The contents of interleukin-6 (IL-6), IL-1β, and tumor necrosis factor-α (TNF-α) in serum, the percentage of cerebral infarction area, the neuronal apoptosis rate, as well as the contents of malondialdehyde (MDA), superoxide dismutase (SOD), and brain-derived neurotrophic factor (BDNF) in hippocampal tissue, and the relative protein expression levels of Gas6, phosphorylated Axl (p-Axl), Axl, B-cell lymphoma-2 (Bcl-2), and Bcl-2 associated X protein (Bax) were measured. RESULTS Compared with Model group, the neurological function of rats in SPS-L group, SPS-H group and SPS-H+Nim group improved significantly, while serum contents of IL-6, TNF-α, and IL-1β, the percentage of cerebral infarction area, the apoptotic rate of neurons as well as MDA content and relative protein expression of p-Axl and Bax were decreased significantly ( P <0.05 or P <0.01). The contents of SOD and BDNF, as well as the relative protein expression of Gas6, Axl, and Bcl-2 in hippocampal tissue, were significantly increased ( P <0.05 or P <0.01), and the ischemic injury in hippocampal tissue imp roved to varying degrees. CONCLUSIONS SPS can reduce neuroinflammation, oxidative stress, and neuronal apoptosis in IS rats, alleviate nerve injury, and improve neurological function, which may be achieved by activating the Gas6/Axl signaling pathway.
7.Regulation of Notch1/Hes1 signaling axis by total flavonoids of Drynariae Rhizoma for promoting chondrocyte autophagy and inhibiting apoptosis:a mechanistic study

OBJECTIVE To investigate the effects of total flavonoids of Drynariae Rhizoma (TFRD) on autophagy and apoptosis in LPS-induced chondrocytes via the regulation of the Notch1/hairy and enhancer of split 1 (Notch1/Hes1) signaling axis. METHODS Human chondrocyte cell line C28/I2 cells were cultured with 5 μg/mL LPS to esta blish in vitro inflammatory injury model. The cells were separated into normal control group, model group, TFRD group (200 μg/mL), TFRD+peroxiredoxin 1 (Prdx1) small interfering RNA (si-Prdx1) group and TFRD+si-Prdx1 negative control (si-NC) group, with 6 replicate wells in each group. Cells were transfected with si-Prdx1 or si-NC for 24 hours, pretreated with TFRD for 2 hours, and then exposed to LPS, with a total culture duration of 48 hours. Apoptotic rate, the proportion of apoptotic cells, monodansylcadaverine (MDC) fluorescence intensity, as well as the contents of matrix metalloproteinase-13 (MMP-13), a disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5), and cartilage oligomeric matrix protein (COMP) were measured. Additionally, the protein expression levels of X-linked inhibitor of apoptosis protein (XIAP), poly(ADP-ribose) polymerase 1 (PARP1), Beclin-1, microtubule-associated protein 1 light chain 3 Ⅱ/Ⅰ (LC3-Ⅱ/Ⅰ), PTEN-induced putative kinase 1 (PINK1), Notch1, Hes1, and Prdx1 were assessed. RESULTS Compared with model group, the apoptotic rate, the proportion of apoptotic cells, the contents of MMP-13 and ADAMTS5 as well as protein expressions of PARP1 were significantly decreased, while MDC fluorescence intensity, COMP content, protein expressions of XIAP, Beclin-1, LC3-Ⅱ/Ⅰ, PINK1, Notch1, Hes1 and Prdx1 were significantly increased ( P <0.05). Compared with TFRD+si-NC group, the changes in the aforementioned indicators (except for Notch1 and Hes1) in the cells of the TFRD+si-Prdx1 group were significantly reversed ( P <0.05). CONCLUSIONS TFRD may activate the Notch1/Hes1 signaling axis, and up-regulate the expression of the downstream target molecule Prdx1, thereby inhibiting LPS-induced chondrocyte apoptosis, promoting protective autophagy, and consequently improving cartilage metabolic homeostasis.
8.Research progress on the mechanism of traditional Chinese medicine regulating oxidative stress to promote diabetic wound healing

The process of diabetic wound healing is highly complex, and the persistence of non-healing wounds is closely associated with sustained oxidative stress in the body . Traditional Chinese medicine (TCM) demonstrates unique therapeutic advantages in promoting diabetic wound repair by modulating oxidative stress through multiple targets and pathways. This article presents a systematic review of the mechanism of TCM regulating oxidative stress to promote diabetic wound healing. It has been found that TCM monomers (such as rutin, baicalin, lonicerin, and curcumin), extracts (including aqueous extract of Gynura divaricata , extract of Polygonatum kingianum , extract of Ginkgo biloba leaves, etc), and compound formulations (such as Badu shengji powder, Danggui sini decoction, Compound ANBP, etc) can effectively alleviate oxidative stress-induced damage in diabetic wounds by modulating related signaling pathways, including nuclear factor-erythroid 2-related factor 2, nuclear factor κB,advanced glycation end products (AGEs)/receptors of AGEs, and silencing information regulatory factor 1. The underlying mechanisms are mainly manifested as: activating the antioxidant defense system, inhibiting inflammatory response, and improving mitochondrial function, thereby synergistically promoting wound healing.
9.Inhibitory Effect of Solute Carrier Family 7 Member 5 Inhibitor JPH203 on Renal Fibrosis Induced by Unilateral Ureteral Obstruction in Mice

ObjectiveTo investigate the effect of solute carrier family 7 member 5 (SLC7A5) inhibitor JPH203 on renal fibrosis induced by unilateral ureteral obstruction in mice. MethodsSixteen SPF male C57BL/6 mice were randomly divided into the control group and the experimental group, with 8 mice in each group. The mouse model of renal fibrosis was established by unilateral ureteral obstruction. From the third day after surgery, the mice in the control group were intraperitoneally injected with phosphate-buffered saline (PBS) for 11 consecutive days, and the injection dose was 200 μL/d. Mice in the experimental group received intraperitoneal injection of JPH203 (50 mg/kg) every day for 11 days. On day 14, the mice were euthanized, then the kidney tissues were obtained. Hematoxylin and eosin (HE) staining was used to assess renal tissue damage, Masson staining was used to evaluate collagen fiber deposition in the extracellular matrix, and immunohistochemistry was used to detect the levels of fibroblast activation markers α-smooth muscle actin (α-SMA) and collagen type Ⅰ (COL-Ⅰ) in kidney tissues. Western blotting was further performed to measure the expression levels of SLC7A5 and transforming growth factor-β1 (TGF-β1), as well as the phosphorylation levels of mammalian target of rapamycin complex 1 (mTORC1) signaling pathway-related molecules. Real-time quantitative PCR was used to verify changes in the mRNA levels of SLC7A5, α-SMA, and COL-Ⅰ in kidney tissues. ResultsCompared with the control group, the experimental group showed reduced destruction of renal tissue structure and a significantly lower pathological injury score (P<0.05). Additionally, collagen deposition in the extracellular matrix was decreased, and the percentage of collagen fiber area was significantly reduced (P<0.001) in the experimental group. The levels of fibroblast activation markers α-SMA and COL-Ⅰ were significantly lower in the experimental group (both P<0.001). The expression levels of SLC7A5 and TGF-β1 were also significantly decreased (P<0.001), and the phosphorylation levels of mTORC1 signaling pathway-related proteins 4E-BP1 and mTORC1 were significantly reduced (P<0.001). Real-time quantitative PCR confirmed that the mRNA levels of SLC7A5, α- SMA, and COL-Ⅰ in kidney tissues were significantly lower in the experimental group (P<0.001). ConclusionJPH203 may inhibit the progression of renal fibrosis in mice by suppressing SLC7A5 expression, regulating the mTORC1 signaling pathway, and altering fibroblast activation status.
10.Clinical characteristics and genetic analysis of 22 Chinese pedigrees affected with Neurofibromatosis type I.

OBJECTIVE:
To explore the genetic variants and phenotypic characteristics of patients with Neurofibromatosis type I (NF1).
METHODS:
Twenty two NF1 patients who presented at Enze Medical (Center) Group in Taizhou between 2018 and 2024 were selected as the study subjects. Clinical phenotype and family history were collected for the patients. Whole exome sequencing (WES) was carried out for the 22 probands to screen the variants of NF1 gene. Candidate variants were verified by Sanger sequencing of their family members. This study was approved by the Medical Ethics Committee of the Hospital (Ethics No.: K20230902).
RESULTS:
The 22 probands were diagnosed between the age of 5 months to 47 years old, and have all shown cafe au lait spots on their skin. Seventeen patients exhibited the phenotype at birth, and 11 had various degrees of neurofibromatosis. Among them, probands 1 and 13 underwent surgical resection of the tumor but had recurred, while proband 12 had amputation due to the huge size and serious impact of the neurofibroma and had no recurrence. Five patients had various degrees of scoliosis. In total 22 germline mutations and one somatic mutation were identified among the 22 families, with 5 variants unreported previously, including 1 nonsense mutation c.1603C>T (Q535*), 3 frameshift mutations [c.7268_7269delCA (Thr2423fs), c.2293del (Arg765Alafs*26), and c.5433_5438delinsGC (Phe1812ArgfsTer50)], and 1 deletion involving exons 41-44 of the NF1 gene and adjacent introns. Proband 13 was found to harbor germline mutation c.6796C>T (Gln2266Ter) and somatic mutation c.1019_1020del (Ser340Cysfs Ter12) in the peripheral blood and tumor tissue, respectively. Among the 22 NF1 probands, 6 had received treatment due to severe illness. Proband 1 had tumor resection in the right upper limb, but was found to have malignant lung tumor and died during follow-up. Proband 12 had multiple recurrence of neurofibroma in the left ring finger. Proband 4 underwent spinal correction surgery due to severe scoliosis. Proband 11 had died due to a central nervous system disease. Among the 22 germline mutations, 6 had led to the occurrence of truncated proteins, which may have a more severe impact on the phenotype.
CONCLUSION
This study investigated the genetic variants and clinical phenotypes of 22 NF1 families and identified 5 novel variants of the NF1 gene, which has expanded the genotypic and phenotypic spectra of the NF1. Preliminary studies have identified an association between truncated mutations, young age, and severe phenotypes, which may provide important clues for prognosis evaluation. For the clinical diagnosis and treatment of NF1, it is necessary to consider the phenotypic characteristics and genetic testing in combination with genetic counseling and long-term follow-up.
