Objective To investigate the effects of body weight and length of SD rats on the carotid artery balloon injury-induced vascular stenosis model in order to provide a reference for replicating an ideal vascular stenosis mod-el.Methods Male rats were divided into three groups based on body weight and length. The CONQUEROR? SC PTCA balloon catheter was employed, with a fixed balloon inflation volume of 0.2 mL to induce injury in the left common carotid artery, while the right side served as a control. As soon as surgery operation, one rat from each group was selected for Evans Blue dye verification.Fourteen days later, the injured and contra lateral common ca-rotid arteries from remaining rats were harvested for HE staining to check the extent of stenosis.Based on these find-ings,six rats within the optimal range of body weight and length were selected for further validation. Results Rats with body weights ranging from 280 to 380 g(corresponding body lengths of 21.0-26.5 cm)underwent balloon catheter injury, resulting in endothelial detachment and varying degrees of stenosis in the common carotid artery. In rats weighing 280-300 g(body lengths of 21.0-22.5 cm)had severe stenosis or occlusion of the common carotid artery with thrombosis. In rats weighing 320-340 g(body lengths of 23.0-24.5 cm),the internal and external elastic plates of the common carotid artery were ruptured and the vascular morphology was abnormal.Conversely, rats weighing 360-380 g(body lengths of 25.0-26.5 cm)did not show any ruptured elastic laminae or thrombus formation in the common carotid artery, and the extent of vascular stenos in rats with a body weight of 360 g was moderate and uniform. The results of the repeated validation experiments were consistent. Conclusions Rats with a body weight range of 360 g(corresponding body length of 25.0-26.5 cm)are suitable for development of an ideal vascular stenosis model.

At present, conventional periodontal treatment cannot achieve complete and effective periodontal tissue regeneration. Cell sheet technology (CST) is a kind of cell transplantation method without scaffold material that can maintain complete extracellular matrix, important ion channels of cells, growth factor receptors, etc., and ensure the interaction between cells and the extracellular matrix. In this paper, the application and research progress of the cell sheet in the field of periodontal tissue regeneration are reviewed. Different types of seed cells can be prepared into monolayer cell sheet, multilayer cell sheet, cell sheet fragments and cell sheet polymers. Among them, the monalayer cell sheet is easily damaged and requires high deoperator; the multilayer cell sheet shows improved mechanical properties, but its thickness needs to be controlled to avoid cell necrosis. The cell sheet fragment can be used in the narrow space between the alveolar bone and root cementum to reduce the difficulty of operation and improve the mechanical properties of the cell sheet. Cell sheet polymers are three-dimensional structures that can provide strong mechanical support and improve the stability of the cell sheet, but the stability of their biological activity needs to be further improved. In methods for construction of the cell sheet, the antifibrosis and antiangiogenesis properties of the amniotic sheet have shown that this structure is suitable as the matrix of cell culture; the method of using a temperature-sensitive culture dish is simple and easy; continuous induction with vitamin C can retain some important proteins on the cell surface; and the magnetic tissue engineering method can increase cell adhesion and easily form a stable cell sheet. The above methods have their own characteristics. In clinical applications, monolayer cell sheet is mainly used for direct transplantation to the receiving site to construct periodontal tissue; multilayer cell sheet of the same or different species overlap and are then transplanted to the receiving site; and multilayer cell sheet of the same kind are wrapped with scaffold material and then transplanted to the receiving site to construct a three-dimensional structure. Overall, cell sheet technology has shown good potential in periodontal tissue regeneration.

Objective To explore the mechanism of icariin mediating migration of mesenchymal stem cells (MSC) in rat bone marrow. Methods MSC proliferation was detected by CCK-8 test. Cell apoptosis was examined with Hoechst33342 staining after the establishment of cellular oxygen and glucose deprivation model. The protein expressions of CXCR4, the receptor of SDF-1, in the surface of MSC after stimulated by icariin were detected through Western blot. The migration of MSC was observed by Transwell chemotaxis assay. Results 0.01μmol/L, 0.1μmol/L and 1μmol/L of icariin could significantly promote the proliferation of MSC, while 10μmol/L of icariin inhibited the proliferation of MSC. After treatment of oxygen and glucose deprivation in vitro, 0.1μmol/L and 1μmol/L of icariin could inhibit the apoptosis of MSC. Icariin could not only improve the expression of CXCR4 in MSC, but also increase the number of transmembrane migrated MSC. After the addition of CXCR4 antagonist AMD3100, there was no significant difference in the number of cell migration among the different groups. Conclusion Icariin with appropriate concentration can promote the proliferation, survival and migration of MSC. SDF-1/CXCR4 signaling pathway is involved in the regulation of MSC migration by icariin.
