1.Predictive value of dual-energy CT quantitative parameters in differential diagnosis and pathological aggressiveness of lung adenocarcinoma

Objective To investigate the predictive value of dual-energy computed tomography(DECT)quantitative parameters in the diagnosis and pathological aggressiveness of lung adenocarcinoma.Methods A total of 174 patients with lung cancer were pro-spectively selected.All patients underwent DECT examination,and the types of lung cancer were determined according to the patho-logical tissue.According to the pathological classification,they were divided into lung adenocarcinoma group(n=99),lung squamous cell carcinoma group(n=47)and small cell lung cancer group(n=28).According to whether the lung adenocarcinoma patients had pathological aggressiveness,they were divided into invasive group(n=34)and non-invasive group(n=65).Receiver operating char-acteristic(ROC)curve was used to diagnose the predictive value of DECT quantitative parameters in the diagnosis and pathological aggressiveness of lung adenocarcinoma.Results The normalized iodine concentration(NIC)and k in venous phase of lung adenocar-cinoma group were higher than those in lung squamous cell carcinoma group and small cell lung cancer group(P<0.05).ROC curve analysis showed that the combined diagnosis value of NIC and k in venous phase was higher(P<0.05).The lobular proportion,vac-uolar proportion,pleural indentation proportion,vascular cluster proportion,effective atomic number(Eff-Z),NIC and k in arterial phase,and NIC and k in venous phase in invasion group were higher than those in non-invasion group(P<0.05).Logistic regression analysis showed that pleural indentation,NIC in arterial phase and NIC in venous phase were risk factors for pathological aggerssive-ness in lung adenocarcinoma patients(P<0.05).ROC curve analysis showed that the combination of NIC in arterial phase and NIC in venous phase had higher value in predicting the pathological aggressiveness of lung adenocarcinoma(P<0.05).Conclusion The combination of NIC and k in venous phase is effective in the diagnosis of lung adenocarcinoma,while NIC in arterial phase and NIC in venous phase have high predictive values for the aggressiveness of lung adenocarcinoma.
2.Alginate lyase immobilized Chlamydomonas algae microrobots: minimally invasive therapy for biofilm penetration and eradication.

Bacterial biofilms can make traditional antibiotics impenetrable and even promote the development of antibiotic-resistant strains. Therefore, non-antibiotic strategies to effectively penetrate and eradicate the formed biofilms are urgently needed. Here, we demonstrate the development of self-propelled biohybrid microrobots that can enhance the degradation and penetration effects for Pseudomonas aeruginosa biofilms in minimally invasive strategy. The biohybrid microrobots (CR@Alg) are constructed by surface modification of Chlamydomonas reinhardtii (CR) microalgae with alginate lyase (Alg) via biological orthogonal reaction. By degrading the biofilm components, the number of CR@Alg microrobots with fast-moving capability penetrating the biofilm increases by around 2.4-fold compared to that of microalgae. Massive reactive oxygen species are subsequently generated under laser irradiation due to the presence of chlorophyll, inherent photosensitizers of microalgae, thus triggering photodynamic therapy (PDT) to combat bacteria. Our algae-based microrobots with superior biocompatibility eliminate biofilm-infections efficiently and tend to suppress the inflammatory response in vivo, showing huge promise for the active treatment of biofilm-associated infections.
3.Promotion of Nd:YAP laser biostimulation on the proliferation and osteogenic differentiation of human periodontal ligament cells through WNT/β-catenin signaling pathway

Objective·To study the effect of neodymium-doped yttrium aluminum perovskite(Nd:YAP)laser biostimulation on the proliferation and osteogenic differentiation of human periodontal ligament cells(hPDLCs)and its possible mechanism.Methods·Five premolars removed for orthodontic reasons were collected from Changzhou Stomatological Hospital,and the periodontal ligament tissues from the middle 1/3 of the roots were taken to culture hPDLCs in vitro.The cells were irradiated with the biostimulation function[G(-)mode]of the Nd:YAP laser.According to the irradiation time,the cells were divided into a control group(without laser irradiation),and groups irradiated for 5 s,10 s,15 s,20 s and 30 s.The cell counting kit-8(CCK-8)method was used to detect the proliferation of hPDLCs in each group.After osteogenic differentiation was induced,the alkaline phosphatase(ALP)content and activity level of the cells were detected using an ALP staining kit and an ALP activity detection kit.The calcium salt level of the cells was evaluated by alizarin red S staining and calcium quantitative analysis.The expression of genes and proteins related to the WNT/β-catenin signaling pathway,including dickkopf-related protein 1(DKK-1),β-catenin,and runt-related transcription factor 2(RUNX2),was analyzed by quantitative real-time PCR(qPCR)and Western blotting.Results·The results of CCK-8 showed that the proliferation level of cells in the 10 s,15 s,20 s,and 30 s groups was enhanced from 3 d after irradiation(all P<0.05).After induction of osteogenic differentiation,ALP content,activity,and calcium salt level in the laser irradiation groups increased with the extension of irradiation time(all P<0.05).The results of qPCR and Western blotting analysis showed that the expression levels of the DKK-1 gene and protein in the laser irradiation groups decreased with the extension of irradiation time.However,the expression levels of β-catenin and RUNX2 genes and proteins increased significantly with the extension of irradiation time;there were statistically significant differences between the 15 s,20 s,and 30 s groups and the control group(all P<0.05).Conclusion·Nd:YAP laser biostimulation may promote proliferation and osteogenic differentiation of hPDLCs through WNT/β-catenin signaling pathway.

6.Research on Satisfaction Degree of Doctor-patient Communication Quality based on Patient Perception

Objective sampling method was used to conduct a questionnaire survey on outpatients in two hospitals in Guangdong province in order to evaluate patients’ satisfaction with the quality of medical service. This paper explored the factors that affect patients’ evaluation of medical service quality, and found that patients’ age was negatively correlated with the evaluation of medical service quality. It is suggested that the establishment of friendly medical institutions should be carried out according to the national policy. At the same time, the management mechanism of hospital should be improved, the number of medical service centers for "efficient" should be increased, and the medical service personnel should be regularly trained; carry out medical knowledge education in community, improve the popularization of personal medical knowledge and close the cognitive gap between doctors and patients.

METTL3 and METTL14 are two components that form the core heterodimer of the main RNA m6A methyltransferase complex (MTC) that installs m6A. Surprisingly, depletion of METTL3 or METTL14 displayed distinct effects on stemness maintenance of mouse embryonic stem cell (mESC). While comparable global hypo-methylation in RNA m6A was observed in Mettl3 or Mettl14 knockout mESCs, respectively. Mettl14 knockout led to a globally decreased nascent RNA synthesis, whereas Mettl3 depletion resulted in transcription upregulation, suggesting that METTL14 might possess an m6A-independent role in gene regulation. We found that METTL14 colocalizes with the repressive H3K27me3 modification. Mechanistically, METTL14, but not METTL3, binds H3K27me3 and recruits KDM6B to induce H3K27me3 demethylation independent of METTL3. Depletion of METTL14 thus led to a global increase in H3K27me3 level along with a global gene suppression. The effects of METTL14 on regulation of H3K27me3 is essential for the transition from self-renewal to differentiation of mESCs. This work reveals a regulatory mechanism on heterochromatin by METTL14 in a manner distinct from METTL3 and independently of m6A, and critically impacts transcriptional regulation, stemness maintenance, and differentiation of mESCs.

9.Investigation on the current status of the allocation of radiological diagnosis and treatment resources in Linyi

Objective To understand the present state of radiological diagnosis and treatment resources allocation and to provide data support for the health administrative department to further optimize the allocation of resources in Linyi. Methods All hospitals which conducted radiological diagnosis and treatment in the city were surveyed. The data were analyzed by using SPSS software. Results There were 305 radiological diagnosis and treatment institutions, with 943 sets of radiological diagnosis and treatment equipment in Linyi. There were 0.89 sets of diagnostic and therapeutic equipment per 10000 people, and the average number of diagnostic and therapeutic equipment per institution in urban areas is 1.48 times that in county areas. There were 2208 radiological diagnosis and treatment staff in the city, and each hospital had 7.24 radiation staff, among which the number ofin tertiary, secondary, primary and unrated hospitals was 89.63, 17.64, 3.37 and 2.77, respectively. Conclusion The allocation of Diagnostic Radiology equipment is out of balance in Linyi, and the large-scale Diagnostic Radiology equipment is less and distributed centrally. The number of diagnostic and therapeutic radiographers per 10000 people is insufficient and unevenly distributed. Radiological diagnosis and treatment resources should be allocated scientifically to promote balanced development among regions and optimize the allocation of resources.
10.Quantification of serum apolipoprotein E and patient phenotyping using isotope dilution liquid chromatography-tandem mass spectrometry

Objective To establish an isotope dilution liquid chromatography tandem mass spectrometry method (ID-LC-MS) for quantification of serum apolipoprotein E and phenotyping. Methods Method establishment. Samples underwent denaturing, alkylation and trypsin digestion with addition of internal standards as isotope labelling arginine. SB-C18 column was used for the liquid chromatographic separation and mass spectrometry positive ion mode and multiple reaction monitoring were employed for quantification and phenotyping. Precision, accuracy and linearity were investigated for method evaluation. 40 serum samples from Shanghai Dongfang Hospital during Oct. to Dec., 2018 were used for method comparison between ID-LC-MS and immunoassay. Deming regression and Bland-Altman were used for method comparison analysis and SPSS 24 for linearity. Results Target peptides reached their releasing maximum within 4 hours and SE did at 3 hours. 3 phenotyping of ApoE were observed, such as E3/E3, E2/E3 and E3 / E4. The imprecision of IQC was 5.2 % . The relative bias for low and high levels of accuracy-based samples was 7.6 % and 3.6 %, respectively. Deming regression showed the intercept with 95 % confidence interval (CI) was 6.44-11.44 (P<0.05 and the 95% confidence interval for the slopewas 0.77-0.89 (P<0.05). The coefficient was r=0.97. The mean difference was - 2.95 mg / L with 95 % CI-4.26--1.65 mg/L. The linearity covered from 16.9 to 58.5 mg/L. Conclusion ID-LC-MS can be used to quantify serum apolipoprotein E and simultaneously detect its phenotyping.
