1.Expert consensus on the combination of anti-EGFR monoclonal antibodies and immune checkpoint inhibitors in the treatment of head and neck squamous cell carcinoma(2025 edition)

Head and neck squamous cell carcinoma(HNSCC)is the most common type of head and neck cancer,and its treatment prognosis remains suboptimal.Although the survival of HNSCC patients has improved with the widespread use of anti-epidermal growth factor receptor(EGFR)monoclonal antibodies and immune checkpoint inhibitors(ICIs),there is still significant room for further improvement.Recent studies have suggested that the combination of anti-EGFR monoclonal antibodies and ICIs offers promising efficacy and safety,earning recommendations from authoritative guidelines such as the National Comprehensive Cancer Network(NCCN)and the Chinese Society of Clinical Oncology(CSCO).However,the application of this combination therapy is still in the early exploratory stage,numerous questions regarding the standardized clinical use of this combination therapy remain unanswered,including the mechanisms of synergy associated with the combination treatment,its therapeutic value in different patient populations,and safety considerations.Committee of Head and Neck Cancer,Chinese Society of Clinical Oncology organized an expert panel to develop"Expert consensus on the combination of anti-EGFR monoclonal antibodies and immune checkpoint inhibitors in the treatment of head and neck squamous cell carcinoma(2025 edition)",based on evidence-based medicine and clinical practice exploration through multiple rounds of discussions.This consensus addressed the mechanisms of the combination of anti-EGFR monoclonal antibodies and ICIs,stratified treatment approaches,applications in special populations,and safety considerations.The consensus has been registered on Practice guideline REgistration for transPAREncy(PREPARE)with the registration number PREPARE-2025CN666.It is hoped that this consensus will provide clearer and more practical guidance for clinicians,further promoting the rational application of this combination therapy in clinical practice and offering more treatment options for HNSCC patients.
2.Practice and exploration of horizontal scientific research contract information management based on risk prevention and control

Objective:This study aims to improve the management level and quality of horizontal scientific research contracts in hospitals, reduce the risk of contract management, and explore an efficient mode of information management of horizontal scientific research contract management.Methods:Taking a Tertiary Grade A hospital as an example, this study described and summarized the management status of horizontal scientific research contracts from 2018 to 2022, sorted out the potential risks and prevention and control nodes of the review, signing, performance, and archiving of horizontal scientific research contracts in the whole cycle management process, and designed an information management model according to the relevant elements of risk prevention and control and the standardized requirements of contract management, thereby developing information platform related functions.Results:The horizontal research contract management function of the clinical research platform based on the integration concept was available to meet the requirements of risk prevention and control of horizontal research contracts, and the contract review and signing cycle of the information management mode was shortened by 33.33% and 31.95% respectively compared with the traditional management mode. The platform realized a standardized whole process management of contract submission, review, repair and revision, signing, performance, and filing.Conclusions:Information management can effectively reduce the risk of horizontal scientific research contract management. It is necessary to strengthen the docking and interaction function with other information platforms and further explore the information auxiliary function to improve the refined management effect of horizontal scientific research contracts.

5.Neoadjuvant therapy for locally advanced oral cavity cancer

Oral cavity cancer is the most common type of head and neck squamous cell carcinoma,and patients with locally advanced can-cer are mainly treated via surgical resection,followed by adjuvant radiotherapy.However,over the past decade,therapeutic progress in loc-ally advanced oral cavity cancer has been limited,with the overall survival remaining below 50%.Although many studies have reported high tumor response rates with neoadjuvant chemotherapy and targeted therapy,these findings have not translated into enhanced patient sur-vival.Neoadjuvant immunotherapy has become a research hotspot owing to the success of immunotherapy with immune checkpoint inhib-itors for relapsed and metastatic head and neck squamous cell carcinoma cases,with many ongoing randomized controlled studies focusing on drug discovery.In this review,we discuss the relevant clinical studies and highlight the therapeutic advancements in neoadjuvant immun-otherapy for locally advanced oral cavity cancer.
6.The construction of employer's satisfaction evaluation index towards the post competency of residents

Objective:To develop employer's satisfaction evaluation index system towards post competency of residents.Methods:Using Delphi method, the employer's satisfaction evaluation index system towards post competency of residents was formulated through a two-round expert consultation among 19 experts. SPSS 20.0 was used for statistical analysis to calculate the positive coefficient, authority coefficient and coordination degree of experts.Results:For the two rounds of the expert consultation, the questionnaire recovery rate was 95.0% and 100% respectively. The experts' authority coefficient was 0.92, and the system was ultimately determined, including 5 first-level indicators, and 32 secondary indicators. The coefficient of variation (CV) of first-level indicators was 0.00, and the Kendall's W of secondary indicators was 0.663. Conclusions:Employer's satisfaction evaluation index system towards post competency of residents is scientific and reliable, which could provide employer with systematic and objective tool to evaluate residents' post competency.
7.Risk assessment and screening of common malignant tumors among female urban residents in Deyang

Objective To analyze the screening results and risk assessment of common malignant tumors among female residents in Deyang from 2017 to 2019, so as to provide basis for cancer prevention and control. Methods Women who underwent physical examinations and completed the survey of common malignant tumors related risk factors from 2017 to 2019 were selected. The high risk rate, screening rate and detection rate of common malignant tumor among female urban residents in Deyang were analyzed. Results From 2017 to 2019, a total of 49,871 female urban residents completed the survey of common malignant tumor related risk factors. The risk rates of breast cancer, lung cancer, cervical cancer and colorectal cancer were 13.24%, 7.65%, 12.92% and 5.53%, respectively. Among all the age groups, women at the age of 50~59 years old were at the highest risk of breast cancer, lung cancer, cervical cancer, colorectal cancer; The screening rates of breast cancer, lung cancer, cervical cancer and colorectal cancer were 24.68%, 11.72%, 34.62% and 14.15%, respectively; The screening rates of breast cancer, lung cancer, cervical cancer and colorectal cancer among women with different educational levels showed significant difference (P<0.05). The screen rates of breast cancer, lung cancer, cervical cancer and colorectal cancer among high risk groups were 24.68%, 11.75%, 26.56% and 14.35%, respectively; The detection rates of breast cancer, lung cancer, cervical cancer and colorectal cancer among the screening groups were 9.33%, 2.68%, 9.82%, and 2.02%, respectively (P<0.05). Conclusion Female residents in Deyang from 2017 to 2019 have high risk and detection rates of breast cancer and cervical cancer, so active screening of malignant tumors among urban female residents can effectively improve the residents' quality of life, and is of vital importance for the early diagnosis and treatment of malignant tumors.
8.Combinational expression of geranylgeranyl diphosphate synthase and taxadiene synthase in Coprinopsis cinerea.

Taxa-4(5),11(12)-diene is the precursor for paclitaxel biosynthesis. The diterpenoid paclitaxel (marketed as Taxol), a plant secondary metabolite isolated from yew, is an effective drug widely used in the treatment of numerous cancers. However, further application of taxol has been restricted due to its low yield in plants and the difficulties in extraction. To increase the intact isoprene flux, we constructed the fusion gene plasmid pBgGGTS and individual cassette plasmid pBgGGgTS to enhance the expression levels of geranylgeranyl diphosphate synthase gene (ggpps) and a taxadiene synthase gene (ts) in Coprinopsis cinerea. These two plasmids were separately transformed into C. cinerea LT2 strain, resulting in several putative transformants. Putative transformants were determined by PCR technique, indicating that 5 out of 13 putative transformants transformed by pBgGGTS and 6 out of 13 putative transformants transformed by pBgGGgTS, respectively. Additionally, the Southern blotting analysis of these 10 transformants confirmed that both ggpps and ts gene were stably integrated into the genome of C. cinerea. Crude extracts from each of the transformants were analyzed. There is no difference in the mycelium extracts among the wild-type LT2 and two types of transformants. However, analysis of culture filtrates indicated that an additional GC peak was found at the retention time of 16.762 min which was absent in the wild type control. The mass fragmentation pattern of this peak had the same diagnostic ions with taxa-4(5),11(12)-diene. According to peak area, the amounts of taxa-4(5),11(12)-diene in each fermented broth were 44 ng/L (transformed with pBgGGgTS) and 30 ng/L (transformed with pBgGGTS), respectively. In conclusion, co-expression of the ggpps and ts gene could increase the taxadiene production in C. cinerea.
