1.Torso Simulation Research on Electromagnetic Compatibility of Implantable Neurostimulator.

In the YY 0989.3-2023 standard, clause 27.106 specifies the protection test against electromagnetic interference, but it only briefly describes the test level for electromagnetic exposure, and does not detail the parameters of the torso. This study aims to explore the internal field distribution for different torso parameters under electromagnetic exposure, and explore the patterns of field distribution through modeling and simulation. The results indicate that the parameters of the torso significantly affect the internal field distribution. The findings of this study provide a basis and reference for the electromagnetic compatibility test for implantable neurostimulator products.

2.Inhibitory effects of sinomenine hydrochloride in T-cell acute lymphoblastic leukemia CEM cells and transcriptomic analysis

BACKGROUND:Sinomenine hydrochloride has anti-tumor effects,but it is rarely reported in T-cell acute lymphoblastic leukemia.OBJECTIVE:To investigate the inhibitory effect of sinomenine hydrochloride on CEM cells in acute T lymphoblastic leukemia.METHODS:Different concentrations(0.5,1,2 and 4 mmol/L)of sinomenine hydrochloride were used to act on CEM cells.CCK-8 assay was used to detect the inhibition rate of cell proliferation and calculate the IC50.Inverted microscope and Giemsa staining were used to observe the changes of CEM cells.The RNA sequencing was performed to analyze the differential gene expression and biological information.Combined with transcriptome sequencing analysis results,flow cytometry was used to detect the apoptosis rate of CEM cells after treatment with of different concentrations(1,2,and 4 mmol/L)of sinomenine hydrochloride.Western blot assay was used to detect the expression of Bcl-2,Bax,and Caspase-9 proteins in CEM cells after treatment with of different concentrations(1,2,and 4 mmol/L)of sinomenine hydrochloride.RESULTS AND CONCLUSION:(1)Sinomenine hydrochloride inhibited the growth of CEM cells in dose and time-dependent manner.(2)After dosing,the number of CEM cells decreased and pyknosis appeared.(3)RNA sequencing revealed 53 differential expressed genes.Gene Ontology was significantly enriched in cellular process,cellular anatomical entities,and binding.Signaling pathway analysis related to tumor was apoptosis.(4)Sinomenine hydrochloride induced the apoptosis of CEM cell in a concentration-dependent manner.(5)Sinomenine hydrochloride could promote the expressions of Bax and Caspase-9,but restrain the expression of Bcl-2 in CEM cells.Therefore,sinomenine hydrochloride can induce apoptosis in CEM cells and suppress cell proliferation,maybe via up-regulation of the protein levels of Bax and caspase-9 and down-regulation of the protein level of Bcl-2.
3.Association of sperm aneuploidy with unexplained spontaneous recurrent abortion

Objective To investigate the correlation of sperm aneuploidy with unexplained recurrent spontaneous abortion(URSA).Methods Thirty-four male partners of couples diagnosed with URSA in the Center of Reproductive Medicine of Northwest Women's and Children's Hospital from November 2021 to December 2023 were enrolled as the experimental group,after excluding chromosomal structural or numerical abnormalities in either partner of the couples,female age≥37 years,and other known female-related causes of miscarriage.Meanwhile,30 men,who had given birth to healthy children,or were infertile only due to female factors and had normal semen parameters,were selected as control group.Fluorescence in situ hybridization(FISH)was used to detect sperm aneuploidy after routine semen analysis for the males of the two groups.Results There were no significant differences in male age,semen volume,sperm concentration,percentage of progressively motile sperm,and percentage of sperm with normal morphology between the two groups(all P>0.05).However,in the experimental group,the total nullisomy rate(2.10±1.93)% versus(0.54±0.27)%,total dip-loidy rate(0.25±0.27)% versus(0.05±0.05)%,and disomy rate of chromosome 18(0.77±1.15)% versus(0.18±0.21)%,sex chromosomes(2.79±2.38)% versus(0.78±0.52)%,chromosome 13(0.79±0.99)% versus(0.21±0.22)%,and chromosome 21(1.24±1.68)% versus(0.22±0.19)% were significantly higher than those in the control group(P<0.05).When the the mean aneu-ploidy rate in the control group(x+2s)was set as the upper limit of the normal reference value,the proportions of disomy rates of chro-mosomes 18,13,21 and sex chromosomes,as well as the total diploidy rate exceeding the threshold in the experimental group were 41.18%,35.29%,41.18%,55.88%,and 47.06%,respectively,with the proportion of abnormal disomy rate of sex chromosomes be-ing the highest.Conclusion The aneuploidy rate of sperm may be a related factor leading to unexplained recurrent spontaneous abor-tion in females,which has certain clinical significance for the assessment of male fertility and the diagnosis of male infertility.
4.Rabbit bone marrow mesenchymal stem cells transfected with recombinant lentivirus and decalcified bone matrix to construct transgenic tissue engineering materials

BACKGROUND:The use of autologous or allogeneic transplantation materials for tissue repair and reconstruction is limited and carries limitations in clinical applications.Transgenic stem cells and tissue engineering materials offer new therapeutic possibilities.OBJECTIVE:To investigate the in vitro biological characteristics of enhanced green fluorescent protein recombinant lentivirus transfected into rabbit bone marrow mesenchymal stem cells and the biommineralization characteristics of transgenic tissue engineering materials constructed using decalcified bone matrix.METHODS:Rabbit bone marrow mesenchymal stem cells were obtained through adherent culture and density gradient centrifugation.The fifth-generation bone marrow mesenchymal stem cells were transfected with enhanced green fluorescent protein recombinant lentivirus at the optimal multiplicity of infection(MOI=100).The differences in cell proliferation capacity,cell phenotype,cell cycle,alkaline phosphatase expression,osteogenic transcription factor(Runx2)expression,and osteocalcin gene expression after osteogenesis were observed between the transfected cells and non-transfected cells in vitro.The micromorphology and elemental energy spectrum analysis of the transgenic tissue engineering materials constructed from cells and decalcified bone matrix scaffold were also observed.RESULTS AND CONCLUSION:Bone marrow mesenchymal stem cells transfected with enhanced green fluorescent protein recombinant lentivirus showed that cell proliferation at 24 and 48 hours after transfection was slower than that of untransfected cells(P<0.05).After 72 hours,the cell phenotype remained unchanged,and the expressions of alkaline phosphatase,Runx2,and osteocalcin after osteogenic induction were not significantly different from those of untransfected bone marrow mesenchymal stem cells(P>0.05).The transgenic tissue engineering materials constructed in vitro using enhanced green fluorescent protein recombinant lentivirus transfected bone marrow mesenchymal stem cells and decalcified bone matrix showed good biocompatibility on the decalcified bone matrix scaffold.Based on fluorescence expression intensity,it was estimated that the target gene would exert its maximum biological function in about 2 weeks,and calcium phosphate deposits would continue to appear,demonstrating superior biomineralization characteristics.

6.Clinical application of Endosize intelligent surgical planning software in guiding the treatment of Stanford type B aortic dissection with Castor stent

Objective To explore the technical characteristics and clinical application value of Endosize intelligent surgical planning software in guiding the treatment of Stanford type B aortic dissection(TBAD)with single-branch Castor stent.Methods A retrospective analysis was conducted on the clinical data of 37 patients with TBAD who were manually measured to have insufficient proximal anchoring zones.Preoperative computed tomography angiography(CTA)images of the patients were analyzed using Endosize software for precise endovascular measurements and three-dimensional visual reconstruction of the aortic anatomy in the interventional treatment area.The C-arm angle was preset,and the stent type,size,and surgical pathway were precisely planned.Intraoperatively,digital subtraction angiography(DSA)was used to verify the consistency between the preoperative plan and the actual surgical plan,and the postoperative outcomes and perioperative complications were analyzed.Comprehensive evaluation was performed in combination with early follow-up results.Results The measurement results of the Endosize software were highly consistent with DSA results.Among the 37 patients,3 had sufficient proximal anchoring zones,and 34 had insufficient proximal anchoring zones.The consistency rate between manual measurements and software measurements was 91.89%.All 34 patients with insufficient proximal anchoring zones successfully underwent treatment with single-branch Castor stent.The stent type and size planned preoperatively were completely consistent with those actu-ally used,and the technical success rate was 100%.No adverse events occurred during the perioperative period.The mean follow-up duration was(6.09±4.42)months,with no deaths during this period.All branch stents remained patent,no endoleaks occurred,thrombosis occurred in all of the dissection false lumen,and aortic morphology was remodeled.Conclusion Endosize intelligent surgical planning software can provide detailed preoperative information for endovascular treatment,and it demonstrates significant clinical application value in guiding the treatment of TBAD with insuffi-cient proximal anchoring zones using single-branch Castor stent.
