1.Factors affecting and identification of key environmental determinants of the Oncomelania hupensis snail density in the Yangtze River Delta based on machine learning models

Objective To identify factors affecting and key environmental factors of the Oncomelania hupensis snail density in the Yangtze River Delta region using machine learning methods. Methods Administrative village-level O. hupensis snail survey data in the Yangtze River Delta (including Shanghai Municipality, Jiangsu Province, Zhejiang Province and Anhui Province) from 2011 to 2021 were retrieved from the Information Management System for Parasitic Disease Control of Chinese Center for Disease Control and Prevention. Environmental factor data were captured from the Google Earth Engine platform, including elevation, slope, terrain, normalized difference vegetation index (NDVI), vegetation type, soil type, total petroleum hydrocarbon (TPH), ammonium nitrogen, inorganic nitrogen, dissolved oxygen, pH of water, chemical oxygen demand (COD) and inorganic phosphorus, and climatic factor data in the study region were retrieved from the Copernicus Climate Data Store, including annual precipitation, aridity index and annual mean temperature (AMT). O. hupensis snail survey data in the Yangtze River Delta region from 2011 to 2021 were randomly divided into a training set (70%) and a test set (30%), and five machine learning models were selected for machine learning model construction and comparative analysis of the O. hupensis snail density using the software R 4.3.0, including random forest (RF), eXtreme gradient boosting (XGBoost), support vector machine (SVM), gradient boosting machine (GBM) and neural network (NN). The XGBoost model was employed to construct a predictive model for the O. hupensis snail density, and the impact of each environmental factor on O. hupensis snail distribution was quantified. The SHapley Additive exPlanations (SHAPs) values were calculated to estimate the average contribution of each variable to the model prediction, and the core environmental factors affecting the O. hupensis snail population density were screened. Results Among the five machine learning models, the XGBoost model exhibited the optimal comprehensive performance, with the coefficient of determination (R2) of 0.855, mean squared error (MSE) of 0.188, root mean squared error (RMSE) of 0.434 and mean absolute error (MAE) of 0.155, respectively. Analysis of factors affecting the O. hupensis snail density with the XGBoost model showed that among the 16 environmental factors, the top four high-impact factors ranked by SHAPs values included annual precipitation, elevation, aridity index and NDVI, with cumulative SHAPs contributions of 75%, which was higher than that of other environmental factors. If NDVI was higher than 0.6, the O. hupensis snail density increased with NDVI and peaked if NDVI was 0.8 (1.60 snails/0.1 m2). The O. hupensis snail density increased with elevation if the elevation ranged from 14 to 40 m, and slowly rose if the annual precipitation ranged from 900 to 1 300 mm, and then increased rapidly to the peak (1.52 snails/0.1 m2) if the annual precipitation ranged from 1 300 to 1 500 mm. In addition, the O. hupensis snail density increased rapidly to the maximum (1.60 snails/0.1 m2) if the aridity index ranged from 0.8 to 1.1, and decreased gradually if the aridity index exceeded 1.1. Conclusions The XGBoost model shows excellent performance in prediction of the O. hupensis snail density and identification of key environmental factors in the Yangtze River Delta region. Annual precipitation, elevation, aridity index and NDVI are key environmental factors affecting the distribution and density of O. hupensis snails in the Yangtze River Delta region.
2.Assessment of survival vulnerability of Oncomelania hupensis in Jiangxi Province under climate change

Objective To assess the survival vulnerability of Oncomelania hupensis in Jiangxi Province under future climate scenarios, and to identify low-vulnerability areas for its survival in this province. Methods Village-level O. hupensis snail survey and O. hupensis snail control with chemical treatments in Jiangxi Province from 2016 to 2024 were captured from the Parasitic Disease Prevention and Control Information Management System of China Disease Prevention and Control Information System. Climatic data were primarily sourced from the Resource and Environmental Science Data Platform, Chinese Academy of Sciences (http://www.resdc.cn/), including annual average temperature, annual average precipitation, annual accumulated temperature above 10 °C, annual accumulated temperature above 0 °C, annual maximum temperature, annual minimum temperature, and annual average relative humidity, and nineteen bioclimatic variables were downloaded from the WorldClim website (https://www.worldclim.org/), including mean diurnal range, isothermality, temperature seasonality, and so on. Elevation and normalized difference vegetation index were catprued from the Resource and Environmental Science Data Platform, Chinese Academy of Sciences (http://www.resdc.cn/), and distance to rivers was downloaded from the WorldPop website (http://www.worldpop.org), and land use and land cover (LULC) data were downloaded from the Big Earth Data Center, Chinese Academy of Sciences (https://data.casearth.cn/), and nature reserve data were obtained from the China Nature Reserve Specimen Resource Sharing Platform (http://www.papc.cn/). Three Shared Socioeconomic Pathways (SSPs) from the Beijing Climate Center-Climate System Model version 2-Medium Resolution (BCC-CSM2-MR) global climate model were employed as future climate scenarios, including SSP126, SSP245, SSP585, and the biomod2 ensemble model in R package was used to simulate suitable habitats for O. hupensis snails in Jiangxi Province in 2050 and 2070 under these scenarios. A snail survival vulnerability index was constructed based on the area of suitable snail habitats, area covered by snail control through chemical treatment, area covered by nature reserves, and changes in snail habitat fragmentation, and a map of snail survival vulnerability distribution was plotted. Results The real area of snail habitats ranged from 78 486.76 to 85 309.47 hm2, and the area of snail control with chemical treatment ranged from 10 138.98 to 13 240.16 hm2 in Jiangxi Province from 2016 to 2024. There were 429 to 531 villages detected with snails during the nine-year period, and the number of actually snail-infested villages ranged from 645 to 686. A total of 818 snail-present points and 1 996 snail-absent points were obtained from snail survey records. The best performance of the biomod2 ensemble model was achieved if a weighted mean approach was used as the ensemble strategy, with a true skill statistic value of 0.799 and an area under the receiver operating characteristic curve of 0.957, and modeling identified annual average relative humidity and annual average precipitation as two most influencing climatic variables for snail distribution. Relative to the current areas of suitable snail habitats under present climate conditions, the area of suitable snail habitats was projected to expand by 24.49% to 46.28% in Jiangxi Province under future climate scenarios, and the proportion of nature reserves areas in the areas of suitable snail habitats was projected to decrease slightly from the current 2.77% to approximately 2.52%, while the proportion of areas of snail control through chemical treatment in areas of suitable snail habitats varied from 0.64% to 19.57%, and the percentage of changes in snail habitat fragmentation ranged from 3.86% to 12.23%. Based on these four indicators, the snail survival vulnerability index was estimated to range from –1.96 to 0.62 in Jiangxi Province. The arithmetic mean of the snail survival vulnerability index differed under three SSP scenarios (SSP126, SSP245 and SSP585), with the highest mean value (–0.69) in 2070 under SSP126, and the lowest mean value (–0.78) in 2070 under SSP585. Conclusions The snail survival vulnerability index ranges from –1.96 to 0.62 in Jiangxi Province under future climate scenarios, and the suitable habitats for O. hupensis snails appear an overall tendency towards expansion. Low-vulnerability snail habitats are mainly distributed along the shores of Poyang Lake and the Yangtze River in Jiangxi Province, partially overlapping with nature reserves. Intensified surveillance of O. hupensis snails is recommended in these areas in the future.
3.Role of serum total bile acid level in development of arrhythmia in ApoE-/-mice

Objective:To discuss the role of changes of serum total bile acid(TBA)levels induced by long-term high-fat diet in the occurrence of supraventricular arrhythmia(SVA)in the apolipoprotein E knockout(ApoE-/-)mice,and to clarify its mechanism.Methods:Twenty ApoE-/-mice were randomly divided into normal diet group and high-fat diet(HFD)group(n=10);after 20 weeks of feeding,surface electrocardiogram was used to detect cardiac electrophysiology of the mice in various groups;echocardiography was used to detect cardiac systolic function and structure in the mice in various groups;enzyme-linked immunosorbent assay(ELISA)was used to detect serum levels of blood lipids,total bile acid(TBA)and inflammatory factors in the mice in various groups;hematoxylin-eosin(HE)staining was used to detect cardiac inflammatory response in the mice in various groups;Masson staining was used to observe myocardial fibrosis degree in the mice in various groups.Results:Compared with normal diet group,4 cases of junctional premature beat(JPB)/junctional tachycardia(JT),1 case of premature atrial contraction(PAC)and 1 case of premature ventricular contraction(PVC)were found in HFD group,while only 1 case of JPB/JT and 1 case of PAC were found in normal diet group.Compared with normal diet group,the heart rate of the mice in HFD group was significantly decreased(P<0.05);the QRS and QT intervals were significantly prolonged(P<0.05);the ejection fraction(EF)and fractional shortening(FS)were significantly decreased(P<0.05);the end-diastolic volume(EDV)was increased(P<0.05),but there was no significant difference in end-systolic volume(ESV)between groups(P>0.05);the left ventricular internal diameter at end-diastole(LVIDd)and left ventricular internal diameter at end-systole(LVIDs)were significantly increased(P<0.05).There were no significant differences in plasma total cholesterol(TC),triglyceride(TG),high-density lipoprotein(HDL-c)and low-density lipoprotein(LDL-c)levels and body weight between normal diet group and HFD group(P>0.05).Compared with normal diet group,the TBA level of the mice in HFD group was significantly increased(P<0.05).There were no significant differences in interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),monocyte chemoattractant protein-1(MCP-1),and C-X-C motif chemokine ligand 1(CXCL-1)levels between HFD group and normal diet group.Compared with normal diet group,the interleukin-1β(IL-1β)level in HFD group showed an increasing trend,but there was no significant difference between groups(P>0.05).The HE staining results showed that the inflammatory cell infiltration was similar between HFD group and normal diet group.The Masson staining results showed that compared with normal diet group,the fibrosis of the mice in HFD group showed an increasing trend,but there was no significant difference in myocardial fibrosis area between groups(P>0.05).Conclusion:Long-term high-fat diet may increase serum TBA level in ApoE-/-mice,which may induce SVA.
4.Progress of drugs targeting the hypoxic microenvironment of leukemia

Leukemia is a malignant proliferative tumor that seriously endangers the quality of life of patients. In recent years, new research achievements have been made on the pathogenesis of leukemia, and some progresses of various drugs targeting the hypoxic microenvironment of leukemia have been made. This article reviews the progress of drugs targeting hypoxia microenvironment to treat leukemia from the aspects such as reactive oxygen species associated with anoxic microenvironment, hypoxia inducible factor, CXC chemokine receptor and vascular endothelial growth factor.
5.Efficacy and safety of dye-free submucosal injection solution for gastric endoscopic submucosal dissection

To evaluate the efficacy and safety of dye-free submucosal injection solution for gastric endoscopic submucosal dissection (ESD), a retrospective cohort study was performed on data of inpatients with early gastric cancer and precancerous lesions who underwent ESD at the Digestive Endoscopy Center of Jiangsu Province Hospital of Traditional Chinese Medicine from January to December 2020. Cases were divided into dye-free submucosal injection solution group (the observation group) and dye-containing solution group (the control group). A total of 108 cases met the eligibility criteria for analysis (39 VS 69). Baseline characteristics were comparable between the two groups ( P>0.05). Compared with the control group, the observation group showed similar median procedure time (30.5 min VS 35.0 min), median dissection speed (0.3 cm2/min VS 0.4 cm2/min), mean volume of injection solution used (39.2 mL VS 38.8 mL), en bloc resection rate [100.0% (39/39) VS 98.6% (68/69)], and curative resection rate [97.4% (38/39) VS 97.1% (67/69)] (all P>0.05). Postoperative stay was 3.0±0.8 days in the observation group and 3.2±0.8 days in the control group ( t=-0.908, P=0.378). Delayed bleeding occurred in 3 (7.7%) patients VS 2 (2.9%) patients ( P=0.349), and postoperative infection occurred in 3 (7.7%) patients VS 8 (11.6%) patients ( P=0.743), respectively. In gastric ESD, dye-free submucosal injection solution demonstrates efficacy comparable with dye-containing solution and does not appreciably increase the incidence of intraoperative or postoperative complications.
6.Improvement of sulfur mustard-induced acute lung injury by resveratrol in mice and the potential mechanism

OBJECTIVE To study how resveratrol(Res)mitigates acute lung injury(ALI)in mice induced by sulfur mustard(SM)and potential mechanism.METHODS Male C57BL/6N mice were randomly divided into the control group(50 μL physiological saline via nebulization),SM group(SM 5 mg·kg-1 via nebulization),SM+Res 10 or 20 mg·kg-1 groups(1 h after administration of SM 5 mg·kg-1,Res 10 or 20 mg·kg-1 was administered via nebulization).Mice in each group were weighed 0,24,48 and 72 h after SM administration.Mice were sacrificed 72 h after SM administration,and lung tissues were collected,weighed for wet and dry weights,and the wet/dry ratio was calculated.HE staining was employed to detect the pathological changes of lung tissues while real-time quantitative PCR(RT-qPCR)was used to detect the mRNA expression levels of interleukin 6(IL-6)and IL-1β.Transcriptomic changes of the SM group and the SM+Res 20 mg·kg-1 group were detected with the next-generation sequencing technology.RT-qPCR was used to detect the mRNA expression changes of adenosine 5'-monophos-phate-activated protein kinase(AMPK)and silence information regulator 1(SIRT1)in lung tissues.RESULTS 72 h after SM administration,the body mass of mice in the SM group was significantly decreased compared with normal control group,while the wet/dry ratio of the lung was significantly increased,so were the mRNA expression levels of IL-6 and IL-1β in lung tissues were also significantly increased.Pathological changes of lung tissues included alveolar cavity atrophy,marked parenchymal tissues,and diffuse infiltration of local inflammatory cells.Compared with the SM group,the body mass of mice in the SM+Res 10 and 20 mg·kg-1 groups significantly increased,the wet/dry ratio of lung tissues was significantly reduced,and expressions of IL-6 and IL-1β mRNA were significantly decreased in SM+Res 10 and 20 mg·kg-1 groups.Compared with the normal control group,the mRNA expression levels of AMPK and SIRT1 in lung tissues of the SM group were significantly decreased.Compared with the SM group,the mRNA expression levels of AMPK in lung tissues of the SM+Res 10 and 20 mg·kg-1 groups were significantly increased while the mRNA expression level of SIRT1 was significantly decreased.CONCLUSION Res can mitigate ALI in mice induced by SM,and the mechanism may be related to the inhibition of cell apoptosis by regulating the AMPK/SIRT1 signaling pathway.
7.Development, reliability, and validity of a treatment-related quality of life scale for Chinese patients with multiple myeloma

Objective:To develop a treatment-related quality of life scale for Chinese patients with multiple myeloma (MM) and to test its reliability and validity.Methods:The initial scale was constructed through a literature search, Delphi expert correspondence, and cognitive testing. This study conducted a preliminary survey of 379 patients with MM and a formal survey of 865 patients from the hematology departments of 155 hospitals nationwide from February 2024 to March 2024. The final scale was obtained after conducting item analysis and reliability and validity tests on the initial scale.Results:The constructed scale contains 36 items covering six domains: physiological, psychological, social, treatment side effects, general health, and others. In the preliminary survey, the Cronbach’s alpha coefficient of each item ranged from 0.597 to 0.939, and the test-retest reliability was 0.747 ( P<0.001). Exploratory factor analysis extracted eight common factors with a cumulative variance contribution of 60.058%. In the formal survey, the Cronbach’s alpha coefficient of each item ranged from 0.484 to 0.930, and the test-retest reliability was 0.835 ( P<0.001). Confirmatory factor analysis revealed a comparative fit index of 0.750, a root-mean-square error of approximation of 0.090, and a root-mean-square residual of 0.067. Conclusion:The treatment-related quality of life scale for Chinese patients with MM designed in this study exhibited good reliability and validity, reflecting the impact of treatment on the quality of life of patients. This scale can provide a reference to clinicians for assessing the disease status of patients.
8.Association of thyroid autoimmunity with thyroid function and gestational diabetes mellitus among pregnant women at first-trimester

Objective:To analyze the relationship among thyroid autoimmunity (TAI), thyroid function, and gestational diabetes mellitus (GDM) in early pregnant women in Xi'an.Methods:A prospective study included pregnant women who underwent prenatal check-ups at the Northwest Women's and Children's Hospital from November 2020 to October 2021, with a gestational age of 6 to 14 weeks. Thyroid function, thyroid peroxidase antibody (TPOAb), thyroglobulin antibody (TgAb), and urinary iodine levels were measured, and the prevalence of thyroid disease and GDM was monitored. The subjects were divided into four groups: TPOAb positive only, TgAb positive only, both TPOAb and TgAb positive, and both TPOAb and TgAb negative, to compare the differences in the prevalence of thyroid disease and GDM among the groups. Statistical analysis was performed using Kruskal-Wallis rank-sum test, Bonferroni correction, Chi-square test, and a multivariate logistic regression model was used to analyze the relationship between TAI, thyroid disease, and GDM. Results:A total of 20 243 early pregnant women were included in this study, among which 1 615 (7.98%) were positive for TPOAb only; 865 (4.27%) were positive for TgAb only; 1 672 (8.26%) were positive for both TPOAb and TgAb (both positive group); and 16 091 (79.49%) were negative for both TPOAb and TgAb (both negative group). The thyroid stimulating hormone levels in the TPOAb positive only group, TgAb positive only group, and both positive group were significantly higher than those in the both negative group, respectively (Bonferroni correction, all P<0.05); the free thyroxine level in the TPOAb positive only group was significantly lower than that in the both negative group ( P<0.05). After adjusting for age, pre-pregnancy body mass index, and urinary iodine levels, multivariate logistic regression analysis showed that compared to the both negative group, the risk of developing hypothyroidism during pregnancy was significantly increased in the both positive group ( OR=11.49, 95% CI: 2.84-46.39); the risk of developing subclinical hypothyroidism during pregnancy was significantly increased in the TgAb positive only group ( OR=1.99, 95% CI: 1.05-3.76) and the both positive group ( OR=3.74, 95% CI: 2.49-5.63); the risk of developing GDM was significantly increased in the TgAb positive only group ( OR=1.43, 95% CI: 1.04-1.96) and the both positive group ( OR=1.94, 95% CI: 1.53-2.46). Among early pregnant women with normal thyroid function, after adjusting for age, pre-pregnancy body mass index, and urinary iodine levels, multivariate logistic regression analysis showed that compared to the both negative group, the risk of developing GDM was significantly increased in the TgAb positive only group ( OR=1.46, 95% CI: 1.06-2.02) and the both positive group ( OR=1.80, 95% CI: 1.40-2.32). Conclusion:TgAb positive only is a risk factor for subclinical hypothyroidism and GDM. Screening for thyroid autoantibodies, especially TgAb, during pregnancy helps in the early identification of high-risk pregnant women for thyroid dysfunction and GDM.
9.Xuefu Zhuyu Mixture Ameliorates Myocardial Injury in Coronary Heart Disease Rats by Modulating TLR4/PI3K/AKT/mTOR Signaling Pathway

Objective To investigate the therapeutic effects and mechanisms of Xuefu Zhuyu Mixture(XFZY)in rats with coronary heart disease(CHD).Methods A CHD rat model was established.Successfully modeled rats were randomly divided into five groups(n=12 each):model group,Lipitor group,low-dose XFZY group(7.02 g·kg-1·d-1),high-dose XFZY group(14.04 g·kg-1·d-1),and high-dose XFZY+RS09[Toll-like receptor 4(TLR4)agonist]group(14.04 g·kg-1·d-1 XFZY+intraperitoneal RS09),with an additional normal control group(n=12).After treatment,following treatment completion,all rats underwent echocardiography to assess left ventricular end-diastolic diameter(LVDd),end-systolic diameter(LVDs),ejection fraction(LVEF),and fractional shortening(FS).Myocardial histopathology and vascular remodeling indices were evaluated via hematoxylin-eosin(HE)staining,while myocardial fibrosis was examined using Masson's trichrome staining.Enzyme-linked immunosorbent assay(ELISA)was employed to quantify inflammatory cytokine levels[tumor necrosis factorα(TNF-α),interleukin(IL)-1β,and IL-6]in myocardial tissues.Immunohistochemical analysis determined protein expression of B-cell lymphoma-2(Bc12)and Caspase 3 in myocardial tissues.Western Blot was performed to measure myocardial protein expression levels of TLR4,phosphatidylinositol 3-kinase(PI3K),phosphorylated protein kinase B(p-AKT),total AKT,phosphorylated mammalian target of rapamycin(p-mTOR),and total mTOR.Results Compared with the normal group,rats in the model group exhibited severe myocardial tissue damage,along with significantly elevated collagen volume fraction,LVDs,LVDd,myocardial levels of IL-6,TNF-α and IL-1β,cardiomyocyte apoptosis ratio,aortic media thickness,and protein expression of Caspase 3,TLR4,PI3K,p-AKT/AKT and p-mTOR/mTOR in myocardial tissue(P<0.05).Conversely,LVEF,FS,aortic luminal diameter,and Bcl-2 protein content were significantly reduced,the difference being statistically significant(P<0.05).Compared to the model group,both low-and high-dose XFZY groups as well as the Lipitor group demonstrated attenuated myocardial lesions,with significant reductions in collagen volume fraction,LVDs,LVDd,myocardial IL-6,TNF-α,IL-1β levels,cardiomyocyte apoptosis ratio,aortic media thickness,and protein expression of Caspase 3,TLR4,PI3K,p-AKT/AKT and p-mTOR/mTOR(P<0.05).Concurrently,LVEF,FS,aortic luminal diameter,and Bcl-2 protein content were significantly increased,the difference being statistically significant(P<0.05).No statistically significant differences were observed between the high-dose XFZY group and the Lipitor group for any aforementioned parameters(P>0.05).However,rats treated with high-dose XFZY plus RS09 showed aggravated myocardial injury with manifested interstitial fibrosis,alongside significantly intensified inflammatory response,cardiomyocyte apoptosis,and vascular remodeling compared to the high-dose XFZY alone group(all P<0.05).Conclusion XFZY alleviates CHD symptoms by suppressing TLR4/PI3K/AKT/mTOR pathway activation,thereby reducing myocardial inflammation,fibrosis,apoptosis,and vascular remodeling.
10.Research progress of LSD1 in regulating tumor immune mechanisms

Lysine-specific demethylase 1(LSD1)plays a regulatory role in tumor immunity.LSD1 affects CD8+T cell infiltration by regulating AGO2 protein level,and it is required for normal B cell proliferation and differentiation via reducing local chromatin accessibility.LSD1 inhibition promotes NK cell cytotoxicity of cancer cells through restoring the expression of ULBPs.LSD1 affects the resistance of M1 macrophages to oxidative stress by regulating the level of SOD2;In addition,regulating the expression of LSD1 affects the anti-tumor function of CD 19 CAR-T cells and immune checkpoint blockers.The development of drugs targeting LSD1 will provide new ideas for immunotherapy of tumors.
