Objective To analyze the overall characteristics, research hotspots, and development trends of immune cell-related studies in radiotherapy for rectal cancer. Methods A systematic search was conducted by using Web of Science Core Collection to retrieve articles related to radiation therapy and immune cells in rectal cancer published from 1991 to 2024. Advanced bibliometric tools, such as VOSviewer and CiteSpace, were utilized to facilitate analysis and describe publication trends, geographic contributions, institutional affiliations, journal prominence, author collaboration, and prominent keywords. Results The annual number of relevant publications has increased steadily, showing remarkable growth over the past five years. Studies focusing on adaptive immune cells (T and B cells) account for the largest proportion of works and form the most extensive collaboration networks, reflecting the central role of T cell–mediated antitumor immunity in radiotherapy. Meanwhile, innate immune cells, including myeloid-derived suppressor cells, macrophages, monocytes, and neutrophils, are increasingly investigated for their regulatory roles in shaping the tumor immune microenvironment and influencing therapeutic responses. Among countries, China and the United States have contributed the highest number of publications and demonstrated strong academic influence. While several stable research groups have been formed, intergroup collaboration remains limited. Keyword analysis revealed that radiotherapy-induced immune modulation, tumor immune microenvironment remodeling, and treatment response have emerged as major research hotspots. Conclusion Research on immune cells in rectal cancer radiotherapy has progressed rapidly in recent years. The emphasis of this field has gradually shifted from single-treatment approaches to mechanistic studies investigating the interaction between radiotherapy and the tumor immune microenvironment. The field still demonstrates considerable potential for future research and clinical translational applications.

Objectives:
. This study aimed to investigate the regulatory functions of gamma-aminobutyric acid (GABA)ergic neural circuits from the inferior colliculus (IC) to the medial geniculate body (MGB) in salicylate-induced tinnitus.
Methods:
. Mice were treated with salicylate to induce tinnitus, and tinnitus-like behaviors were evaluated via gap prepulse inhibition of acoustic startle. Using combined viral tracing methodologies, we identified and mapped the pathways and connections from the IC to the MGB. Furthermore, we employed Gq-coupled human M3 designer receptors exclusively activated by designer drugs (DREADDs) and Gi-coupled human M4 DREADDs to achieve targeted excitation or suppression of GABAergic neurons in the IC and MGB. Following the administration of clozapine N-oxide, which binds to these receptors, we modulated these neural circuits to assess their impact on tinnitus severity in a mouse model.
Results:
. Our findings demonstrated that mice exposed to salicylate exhibited tinnitus-like behaviors. GABAergic neurons projecting retrogradely from the MGB to the IC were primarily concentrated in the external nucleus of the IC. After clozapine N-oxide administration, chemogenetic activation of IC-MGB GABAergic neurons aggravated salicylate-induced tinnitus. Additionally, activation of GABAergic neurons between the IC and MGB induced the perception of tinnitus even without salicylate. However, chemogenetic inhibition of the IC-MGB GABAergic circuit did not reverse salicylate-induced tinnitus.
Conclusion
. These findings suggest that activation of the IC-MGB GABAergic neural circuit may contribute to tinnitus generation through a mechanism distinct from that of salicylate-induced tinnitus. This study provides novel insights into the mechanisms underlying tinnitus.
3.Study on the inhibitory effects of polysaccharide from Plantago depressa on colitis-associated colorectal cancer in mice by regulating TNF-α/NF-κB signaling pathway and intestinal flora
China Pharmacy 2025;36(21):2662-2667
OBJECTIVE To study the inhibitory effects of polysaccharide from Plantago depressa (PLP) on colitis-associated colorectal cancer (CAC) in mice and its mechanism. METHODS The mice were randomly divided into control group, model group, PLP low-, medium- and high-dose groups (100, 200, 400 mg/kg), positive control group (sulfasalazine, 455 mg/kg), with 8 mice in each group. Except for control group, the remaining groups utilized azomethane oxide + dextran sulfate sodium to induce CAC model. At the same time, mice in each group were administered the corresponding medicinal solution via gavage once daily for 12 consecutive weeks. After the last medication, disease activity index (DAI), colon length and tumor number were detected; the histopathological morphology of the colon tissue was observed. The levels of inflammatory cytokines [interleukin-1β (IL-1β), IL-6, interferon-γ (IFN-γ), and tumor necrosis factor-α (TNF-α)] in the serum were measured. Additionally, the expression levels of proteins related to the TNF-α/nuclear factor-κB (NF-κB) signaling pathway [NF-κB p65, NF-κB inhibitor protein α (IκBα), NF-κB-inducing kinase (NIK), tumor necrosis factor receptor 1 (TNFR1), and tumor necrosis factor receptor-associated factor 2 (TRAF2)] in the colon tissue were detected. In addition, the feces of mice were collected to analyze the changes in intestinal flora composition by 16S rRNA high-throughput sequencing. RESULTS Compared with model group, DAI score, tumor number, serum levels of IL-1β, IL-6 (except for PLP low-dose group), IFN-γ and TNF-α, as well as phosphorylation levels of IκBα and NF-κB p65 and protein expressions of NIK, TNFR1 and TRAF2 (except for PLP low-dose group) in colon tissue, were all decreased significantly (P<0.05); the colon length was significantly increased (P<0.05). The colon tissue exhibited normal glandular structures, with a reduced degree of inflammatory E-mail:Ryantang0715@163.com cell infiltration, and no adenocarcinoma formation was observed. The results of gut microbiota analysis revealed that, compared with model group, the relative abundances of Firmicutes, Fusobacteria, and Megamonas in the PLP high-dose group were significantly decreased (P<0.05), while the relative abundances of Bacteradetes and Akkermansia, as well as the Shannon and Chao1 indices, were significantly increased (P<0.05). The relevant pathways through which PLP regulated intestinal flora function primarily included the TNF signaling pathway, Toll- like receptor signaling pathway, and inflammatory bowel disease signaling pathway, among others. CONCLUSIONS PLP can exert inhibitory effects on CAC by inhibiting TNF-α/NF-κB signaling pathway, and regulating intestinal flora.

Cancer is the main cause of death, and drug therapy has greatly improved the effectiveness of anti-tumor treatment. However, there are problems such as high adverse reactions and the risk of developing drug resistance after long-term use. There is an urgent need to seek new drug targets.Human antigen R(HuR),as an RNA binding protein, promotes the whole process of tumor occurrence, development and metastasis through post transcriptional regulation of mRNA stability, and HuR is general-ly highly expressed in tumor tissue, making it a new target for an-ti-tumor therapy and a standard for prognosis evaluation.Tradi-tional Chinese medicine formulas and their various chemical components can inhibit tumor proliferation, induce tumor cell ap-optosis,inhibit angiogenesis, suppress immune escape, and re-verse tumor drug resistance by regulating HuR activity. This re-view summarizes the importance of HuR in regulation of tumor progression,as well as analyzes the mechanisms of the antitumor effects through active ingredients of Chinese medicine with the regulation of HuR.It is expected to provide new ideas for tumor therapy and guidance for the development of HuR-targeted anti-tumor drugs.

Objective:To evaluate the efficacy and safety of nebulized inhalation of recombinant human interferon (IFN) α1b injection in the treatment of respiratory syncytial virus (RSV) associated lower respiratory tract infections (pneumonia and bronchiolitis) in children. Methods:A randomized, double-blind, parallel, placebo-controlled add-on design was used.Children with pneumonia or bronchiolitis aged 2 months to 5 years who tested positive for RSV antigen within 72 hours of onset from 30 clinical trial sites including Beijing Children′s Hospital, Capital Medical University between February 2021 and December 2022 were included in this study and randomly divided into 2 groups at a ratio of 1∶1 based on a stratified-block method. Both groups received basic treatments such as cough control, asthma relieving, expectorant treatment, fever reduction, oxygen therapy, etc.The experimental group received additional nebulized inhalation of IFN α1b injection at a dose of 2.0 μg/(kg·time), twice a day.The control group received nebulized inhalation of placebo twice a day.Clinical efficacy was evaluated based on indicators such as the duration of clinical symptoms and signs, and the Kaplan-Meier method was used to calculate the median and 95% CI of the duration of clinical symptoms and signs. The Log-rank test was used to compared data between groups.Safety was assessed through the incidence of adverse reactions and laboratory tests, and the Chi-square test was used to analyze the difference between groups. Results:There were 123 children in the experimental group and 122 children in the control group. The median durations of all the 5 clinical symptoms and signs [including shortness of breath, wheezing, dyspnea (visible retractions), decreased transcutaneous oxygen saturation, and abnormal mental state] in the experimental group after treatment were slightly shortened than those in the control group [2.7 d(95% CI: 1.9-3.0 d)] vs.[2.9 d(95% CI: 2.6-3.6 d), P=0.027].The improvement in dyspnea (retractions) was especially pronounced in the experimental group, with a relief rate of 50.0% (0, 100%) on the first day of administration[compared with 0 (0, 50.0%) in the control group ( Z=2.002, P=0.025)].The median duration of dyspnea in the experimental group was nearly 1 day shorter than that in the control group [1.0 d(95% CI: 0.7-1.7 d) vs.1.8 d(95% CI: 1.0-2.5 d), P=0.046].There were no significant difference in hospital stay [6.0(5.0, 8.0) d vs.6.5(5.0, 8.0) d, Z=0.675, P=0.500], oxygen therapy duration [32.0(14.0, 96.3) h vs.39.0 (24.0, 83.2) h, Z=0.094, P=0.925], the recovery rate from clinical symptoms during treatment [(105/106, 99.1%) vs.(96/101, 95.0%)], and recurrence rate [(0/106, 0) vs.(2/101, 2.0%)] between the 2 groups (all P>0.05).However, the above-mentioned four indicators in the experimental group showed a trend of clinical benefits. The quantitative virus detection results showed that the RSV viral load in both groups decreased after treatment compared to before treatment. After 2 days of treatment, the decline rate of RSV viral load from the baseline was 0.90 lg copies/(mL·d) in the experimental group and 0.25 lg copies/(mL·d)in the control group, with a statistically significant difference ( P<0.05).Furthermore, there was no statistically significant difference in the incidence of adverse reactions between the 2 groups ( P>0.05).Importantly, no drug-related serious adverse reactions occurred in both groups. Conclusions:The nebulized inhalation therapy of IFN α1b demonstrates efficacy and safety in treating pediatric RSV associated lower respiratory tract infections. It particularly offers outstanding clinical therapeutic value for severe children.
