1.Improvement effect and mechanism of modified Jinfeicao powder on cough-variant asthma in rats

OBJECTIVE To investigate the improvement effect and mechanism of modified Jinfeicao powder (MJCP) on cough-variant asthma (CVA) in rats.METHODS Network pharmacology was employed to identify the key targets and signaling pathways involved in the therapeutic action of MJCP against CVA. Rats were divided into normal control group, model group, positive control group (prednisone acetate, 0.9 mg/kg), and MJCP low-, medium-, and high-dose groups (3.5, 7, 14 g/kg), with six rats in each group. Except for the normal control group, CVA models were established in other groups. From day 14 of the experiment, rats in each group were administered corresponding drugs or normal saline intragastrically once daily for 14 consecutive days. After the final administration, cough frequency, serum levels of immune-inflammatory and oxidative stress-related indicators, pulmonary histopathological changes, and expression of pathway-related proteins were detected and observed.RESULTS Network pharmacology identified five key targets, including tumor necrosis factor-α (TNF-α), interleukin-6(IL-6), and protein kinase B(AKT), as well as signaling pathways such as phosphatidylinositol 3-kinase (PI3K)/AKT/mammalian target of rapamycin (mTOR) and hypoxia-inducible factor-1α (HIF-1α). Animal experimental results showed that, compared with the model group, cough frequency was significantly reduced in all treatment groups ( P <0.05). Serum levels of immunoglobulin E (IgE), IL-6, TNF-α, Src family of protein tyrosine kinase, epidermal growth factor receptor, and malondialdehyde (MDA) (except for IgE and MDA in the MJCP low-dose group), mean pathological scores, the ratio of total airway wall area to basement membrane perimeter, and the expression levels of α -smooth muscle actin, Collagen-Ⅰ, phosphorylated PI3K, phosphorylated AKT (except in the MJCP low-dose group), phosphorylated mTOR, and HIF-1α protein in lung tissues were all significantly decreased ( P <0.05). Glutathione (except in the MJCP low-dose group) and superoxide dismutase levels in serum were significantly increased( P <0.05).CONCLUSIONS MJCP may alleviate airway inflammation, airway remodeling, and oxidative stress by regulating the PI3K/AKT/mTOR and HIF-1α signaling pathways, thereby exerting its improvement effects on CVA.
2.The Mechanisms of Quercetin in Improving Alzheimer’s Disease

Alzheimer’s disease (AD) is a prevalent neurodegenerative condition characterized by progressive cognitive decline and memory loss. As the incidence of AD continues to rise annually, researchers have shown keen interest in the active components found in natural plants and their neuroprotective effects against AD. Quercetin, a flavonol widely present in fruits and vegetables, has multiple biological effects including anticancer, anti-inflammatory, and antioxidant. Oxidative stress plays a central role in the pathogenesis of AD, and the antioxidant properties of quercetin are essential for its neuroprotective function. Quercetin can modulate multiple signaling pathways related to AD, such as Nrf2-ARE, JNK, p38 MAPK, PON2, PI3K/Akt, and PKC, all of which are closely related to oxidative stress. Furthermore, quercetin is capable of inhibiting the aggregation of β‑amyloid protein (Aβ) and the phosphorylation of tau protein, as well as the activity of β‑secretase 1 and acetylcholinesterase, thus slowing down the progression of the disease.The review also provides insights into the pharmacokinetic properties of quercetin, including its absorption, metabolism, and excretion, as well as its bioavailability challenges and clinical applications. To improve the bioavailability and enhance the targeting of quercetin, the potential of quercetin nanomedicine delivery systems in the treatment of AD is also discussed. In summary, the multifaceted mechanisms of quercetin against AD provide a new perspective for drug development. However, translating these findings into clinical practice requires overcoming current limitations and ongoing research. In this way, its therapeutic potential in the treatment of AD can be fully utilized.
3.Material Basis and Its Distribution in vivo of Qili Qiangxin Capsules Analyzed by UPLC-Q-Orbitrap-MS

ObjectiveBased on ultra-performance liquid chromatography-quadrupole-electrostatic field orbitrap high resolution mass spectrometry(UPLC-Q-Orbitrap-MS), the chemical constituents of Qili Qiangxin capsules was identified, and their distribution in vivo was analyzed. MethodsUPLC-Q-Orbitrap-MS was used to detect the sample solution of Qili Qiangxin capsules, as well as the serum, brain, heart, lung, spleen, liver and kidney tissues of mice after oral administration. Using the Thermo Xcalibur 2.2 software, the compound information database was constructed, and the molecular formulas of compounds corresponding to the quasi-molecular ions were fitted. Based on the information of retention time, accurate relative molecular mass and fragments, the compounds and their distribution in vivo were analyzed by comparing with the data of reference substances and literature. ResultsA total of 233 compounds, including 70 terpenoids, 60 flavonoids, 23 organic acids, 17 alkaloids, 20 steroids, 7 coumarins and 36 others, were identified or predicted from Qili Qiangxin capsules, 73 of which were identified matching with standard substances. Tissue distribution results showed that 71, 17, 38, 33, 32, 58 and 43 migrating components were detected in blood, brain, heart, lung, spleen, liver and kidney, respectively. Thirty-seven components were absorbed into the blood and heart, including quinic acid, benzoylaconitine benzoylmesaconine and so on. Fourteen components were absorbed into the blood and six tissues, including calycosin, methylnissolin, formononetin, alisol B, alisol A and so on. ConclusionThis study comprehensively analyzes the chemical components of Qili Qiangxin capsules and their distribution in vivo. Among them, astragaloside Ⅳ, salvianolic acid B, ginsenoside Rb1, ginsenoside Rb3, ginsenoside Rd, ginsenoside Rg3, calycosin-7-glucoside, and sinapine may be the important components for the treatment of heart failure, which can provide useful reference for its quality control and research on pharmacodynamic material basis.
4.The Predictive Value of Murray's Law-based Quantitative Flow Ratio in Side Branches for Long-term Prognosis in Patients With Non-left Main Bifurcation Lesions After Simple Main Branch Stent Implantation

Objectives:To investigate the predictive value of Murray's law-based quantitative flow ratio(μQFR)in side branches for long-term clinical prognosis in patients with non-left main bifurcation lesions who underwent simple main branch stenting,and to provide a potential functional assessment standard for intervention decision-making on coronary bifurcation lesions.Methods:A retrospective analysis was conducted in 408 patients with non-left main bifurcation lesions who underwent simple main branch stenting at Lianyungang First People's Hospital and Nanjing First Hospital between July 2018 and January 2021.The study utilized third-generation QFR software to analyze pre-and post-procedure anatomical and functional parameters of the target lesion's main branch and key branches.The primary endpoint was target vessel failure(TVF)events during the 3-year follow-up.Patients were stratified into TVF and non-TVF groups.Baseline characteristics,procedural data,and pre-/post-procedural parameters of target vessels were compared between groups.Multivariable Cox regression was performed to identify predictors of TVF.Diagnostic efficacy of predictors was evaluated using area under the receiver operating characteristic(ROC)curve(AUC)with DeLong's method for comparison.Patients were dichotomized based on the optimal cutoffof post-procedural side branch μQFR,with TVF incidence rates compared via Cox regression and Kaplan-Meier analysis.Results:During 3-year follow-up,54 patients(13.2%)experienced TVF(TVF group),data were compared with 354 patients(86.76%)without TVF(non-TVF group).The TVF group showed higher post-procedural side branch diameter stenosis([32.93±17.80]%vs.[22.62±11.96]%,P<0.001)and lower μQFR(0.80±0.10 vs.0.89±0.07,P<0.001).Multivariate Cox regression identified higher post-procedural side branch μQFR as an independent protective factor against 3-year TVF(per 0.01 increase:HR=0.903,95%CI:0.850-0.959,P<0.001).ROC curves indicated that post-procedural side branch μQFR had moderate diagnostic efficacy for predicting 3-year TVF(AUC=0.769,95%CI:0.678-0.861,P<0.001),with a significantly higher AUC value than post-operative side branch area stenosis and minimal lumen diameter(both P<0.001),the optimal cutoffvalue was 0.84.Multivariate Cox regression and Kaplan-Meier survival analysis revealed markedly higher 3-year TVF rates in patients with μQFR≤0.84 compared to patients with μQFR>0.84(HR=4.007,95%CI:2.342-6.855,P<0.001;28.3%vs.7.9%,log-rank P<0.001).Conclusions:For patients with bifurcation lesions not involving the left main,the immediate post-procedural side branch μQFR could better predict 3-year TVF than anatomical indices.Maintaining post-stenting side branch μQFR>0.84 may optimize clinical outcomes when using a single-stent strategy.

Objective:To explore the teaching reform of the Food Analysis experiment course under the concept of employment-oriented education,and to improve students'practical ability and employment competitiveness.Methods:Taking students majoring in Food Quality and Safety at a certain university as the research objects,the control group adopted traditional teaching,while the experimental group carried out reforms by optimizing the curriculum system,innovating the teaching mode,and establishing a diversified evaluation system.The total scores,teaching effectiveness,satisfaction with the teaching mode,and the evaluation of graduates by employers between the two groups were compared.Results:The average total score of the experimental group was significantly higher than that of the control group(P<0.05).The teaching effect evaluations of the experimental group in improving experimental operation skills and enhancing employment position cognition were all higher than those of the control group(P<0.05).The satisfaction of the experimental group with the teaching mode was better than that of the control group(P<0.05).In the evaluation results of employers on graduates,the excellent rate of the experimental group was significantly higher than that of the control group(P<0.01).Conclusion:The reform measures effectively improve the teaching quality and students'comprehensive literacy,and deliver high-quality talents with both practical ability and innovative thinking to the food industry.
7.Clinical trial of budesonide and formoterol fumarate powder for inhalation in the treatment of elderly patients with cough variant asthma

Objective To compare the effects of different doses of budesonide and formoterol fumarate powder for inhalation combined with montelukast sodium tablet in the treatment of cough variant asthma(CVA)and the improvement of airway function and inflammatory factors.Methods Elderly patients with cough variant asthma were randomly divided into group A and group B.Both groups of patients received budesonide and formoterol fumarate powder for inhalation combined with montelukast sodium tablet.Group A was given budesonide and formoterol fumarate powder for inhalation(Ⅱ),2 inhalation per time,twice a day;Group B was given budesonide and formoterol fumarate powder for inhalation,4 inhalation per time,twice a day;budesonide fumatrol inhalation powder mist for continuous treatment for 6 months,and montelukast sodium tablet 10 mg once a day for at least 3 months.The nighttime cough scores of the two groups were compared before treatment and after treatment.The percentage of forced expiratory volume in one second(FEV1)in the predicted value,the maximum mid expiratory flow(MMEF),the fractional exhaled nitric oxide(FeNO),interleukin-5(IL-5)and eosinophils were compared between the two groups.The incidence of adverse drug reactions and the recurrence rate within 1 year were compared between the two groups.Results A total of 45 cases were enrolled in both the group A and the group B.At 9 months after treatment,the nocturnal cough scores of the group A and the group B were(0.93±0.42)and(0.65±0.29)points,respectively;the percentage of FEV1 in the predicted value were(97.75±9.67)%and(100.93±11.06)%,respectively;the MMEF values were(2.81±1.04)and(3.08±1.09)L·s-1,respectively;the FeNO values were(18.94±9.75)and(15.94±7.96)ppb,respectively;the IL-5 levels were(10.88±7.06)and(8.11±5.56)pg·mL-1,respectively.The above indicators in group B showed statistically significant differences compared to group A(all P<0.05).The total incidence of adverse drug reactions in group A and group B were 8.89%(5 cases/45 cases)and 13.33%(6 cases/45 cases),respectively.The recurrence rates was 15.56%(7 cases/45 cases)and 13.33%(6 cases/45 cases),respectively.There was no statistically significant difference in the above indicators between group B and group A(all P>0.05).Conclusion For elderly patients with CVA,higher dose of budesonide and formoterol fumarate powder for inhalation combined with montelukast sodium tablet can better improve cough symptoms,reduce the level of airway hyperresponsiveness and inflammatory factors,reduce the recurrence rate,and the patients are well tolerated.
8.Efficacy analysis of facial melanocytic nevus excision along the pigmented contours

Objective:To evaluate the cosmetic effect of facial melanocytic nevus excision along the pigmented contours.Methods:A total of 318 patients with facial melanocytic nevi were collected from the Department of Burns and Plastic Surgery, the Affiliated Yongchuan Hospital of Chongqing Medical University from October 2018 to August 2020. The patients collected after October 2019 underwent contoured excision of facial melanocytic nevi (observation group), while those collected before October 2019 underwent fusiform excision of lesions (control group). Recurrence rates, wound lengths, and scar conditions were compared between the two groups one year after surgery. Comparisons of measurement data between the two groups were conducted using the t test. Results:The observation group included 159 patients (372 melanocytic nevi), comprising 24 males and 135 females, aged from 15 to 45 (25.4 ± 6.5) years; the short diameters of the melanocytic nevi ranged from 2 to 6 (3.9 ± 0.6) mm, and the long diameters ranged from 2 to 8.5 (4.3 ± 0.7) mm. The control group consisted of 159 patients (390 melanocytic nevi), including 29 males and 130 females, aged from 16 to 48 (26.3 ± 7.3) years; the short diameters of the melanocytic nevi ranged from 2 to 6 (3.7 ± 0.8) mm, and the long diameters ranged from 2.2 to 8.4 (4.5 ± 0.5) mm. There were no significant differences in age, gender composition, or short and long diameters of the melanocytic nevi between the two groups (all P > 0.05). After one year of follow-up, 4 patients experienced recurrence in the observation group, and 3 experienced relapse in the control group; the scar scores were 0.64 ± 0.24 points in the observation group and 0.58 ± 0.19 points in the control group, and there were no significant differences in the recurrence rates or scar scores between the two groups (both P > 0.05) ; the increase in wound length was significantly lower in the observation group (27.44% ± 8.46%) than in the control group (48.42% ± 38.84%, t = 10.19, P = 0.004) . Conclusion:Compared with the fusiform excision technique, the pigmented contoured excision technique showed similar efficacy for the treatment of facial melanocytic nevi, but resulted in shorter wound lengths.
9.Bioinformatics analysis of genes associated with poor prognosis in colorectal cancer

Objective To screen genes associated with poor prognosis of colorectal cancer(CRC)through bioinformatics analysis.Methods The gene expression profiles of GSE74602,GSE110223,GSE113513 and GSE141174 were obtained from Gene Expression Omnibus(GEO)database,including samples from 65 CRC tissues and 65 normal tissues.Differentially expressed genes(DEGs)between CRC tissues and normal tissues were screened out by GEO2R tool and Venn software,and the consistent genes were extracted from DEGs by Venn software.A protein-protein interaction(PPI)network was constructed by the STRING database to identify key genes,which were analyzed by Kaplan-Meier Plotter and GEPIA.Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis was conducted on the 13 selected genes.Results There were 171 DEGs obtained from the four datasets,including 148 up-regulated genes and 23 down-regulated genes.Up-regulated DEGs were enriched in the redox processes,bicarbonate transport,digestion,ion trans-membrane transport,and one-carbon metabolism;and down-regulated DEGs were enriched in the positive regulation of cell proliferation.The survival curve analysis showed that 30 of the 87 genes were significantly associated with poor survival prognosis.GEPIA showed that 13 of the 30 genes were highly expressed in CRC tissues compared to normal tissues,among which MYC and FGFR3 markedly enriched in the CRC pathway.Conclusion This study identifies that MYC and FGFR3 are significantly up-regulated in CRC and closely associated with poor prognosis,suggesting that they may serve as potential prognostic markers and therapeutic targets for CRC patients.
10.Association between maternal periconceptional supplementation of folic acid or multiple micronutrients containing folic acid and perinatal mortality rate

Objective:To describe the prevalence of perinatal death in Tongzhou District of Beijing,and to estimate the association between maternal periconceptional supplementation of folic acid or multiple micronutrients containing folic acid and perinatal mortality rate.Methods:A retrospective cohort study was conducted based on the maternal and child care system in Tongzhou District of Beijing.The subjects were 94 490 perinatal who were born during January 2013 to December 2018.The information on perina-tal outcomes and maternal periconceptional supplementation of folic acid or multiple micronutrients contai-ning folic acid were collected.The Poisson log-linear model was used to estimate the association between maternal periconceptional folic acid or multiple micronutrients supplementation and perinatal mortality rate.Results:The overall perinatal mortality rate was 2.71‰.The perinatal mortality rates for maternal nutrients supplementation containing folic acid and no supplementation during periconceptional period were 2.63‰ and 3.43‰,respectively,and the difference in rates was not statistically significant[crude risk ratio(cRR)=0.77,95%CI:0.54-1.14].After adjusting for potential confounding factors in-cluding ethnicity,age,education level,occupation,household registration,parity,numbers of fetuses,gestational age,pregnant with assisted reproductive technology,delivery year and pre-pregnancy body mass index,the rates remained not statistically significant[adjusted risk ratio(aRR)=0.93,95%CI:0.77-1.13].The perinatal mortality rates were 2.23‰ and 2.99‰ for pure folic acid and multi-nutrients supplements containing folic acid,respectively,and the difference in rates was statistically sig-nificant(cRR=1.34,95%CI:1.02-1.76).The rates difference remained statistically significant after adjusting for potential confounders(aRR=1.31,95%CI:1.06-1.62).Additionally,the peri-natal mortality rates differences among the non-supplementation group and the supplementation group with variate timing of initiation(pre-conception or post-conception)or frequency of supplementation(low-frequency or high-frequency)were not statistically significant,regardless of adjusting for confounders.Conclusion:The overall perinatal mortality rate was lower than the national average level in Tongzhou District of Beijing.Maternal periconceptional supplementation of pure folic acid or micronutrients contai-ning folic acid had no impact on perinatal mortality.The association between maternal periconceptional supplementation of multiple micronutrients containing folic acid and perinatal mortality rate need further research.
