1.Regulation of white adipose tissue in mice by immunization with recombinant Bacillus Calmette-Gue?rin with c-di-AMP adjuvant

This study assessed the role and mechanism of the recombinant Bacillus Calmette-Gue?rin vaccine(rBCG)with c-di-AMP adjuvant in regulating metabolism and immunity in epididymal white adipose(eWAT)in mice.Male C57BL/6 mice were intravenously immunized with BCG and rBCG,and their body weights were monitored.eWAT was isolated from the mice,and the stromal vascular fractions(SVFs)cell number was counted with a hemocytometer.Sections of mouse adipose tissue were prepared,and the size,number,and morphology of eWAT adipocytes and crown-like structure(CLS)formation were compared under a microscope after HE staining.The transcription levels of lipid metabolism-associated factors,cytokines and aging-associated genes in each group were determined with qRT-PCR.The body weights of mice gradually increased after immunization with BCG and rBCG.The proportions of eWAT increased,and the SVFs cell number decreased,in rBCG immunized mice.HE staining indicated that BCG immunization promoted hyperplasia,whereas rBCG immunization promoted hypertrophy of eWAT adipocytes;moreover,both BCG and rBCG immunization induced CLS formation in eWAT.The qRT-PCR results indicated that rBCG immunization inhibited the expression of genes associated with lipolysis and energy expenditure in eWAT.BCG immunization had little effect on cytokine transcription,whereas rBCG significantly induced the transcription of IFN-γ and IL-1Ra,and inhibited that of IL-15 and IL-2,but did not induce the expression of aging-associated genes.Thus,rBCG immunization induced eWAT adipocyte hypertrophy,which was associated with the inhibition of eWAT lipolysis and the regulation of cytokine expression.
2.Comparison of the toxicity and safety of protein derivatives from novel fusion strains of Mycobacterium tuberculosis

The objective of this study was to evaluate the toxicity and safety of novel Mycobacterium tuberculosis fusion strain protein derivatives,referred to as B/R strain active proteins.In cellular experiments,RAW264.7 cells were treated with each vaccine preparation,and apoptosis rates were measured.In subsequent animal experiments,C57BL/6 mice were immunized via subcutaneous injection,and their survival and body weight changes were monitored and recorded at 2,4,8,12,and 16 weeks.The lungs and spleens were harvested to calculate organ coefficients,and pathological examinations were conducted.At the eighth week of immunization,the mice were infected with high concentrations of BCG,and pathological changes in the lungs and spleens were observed 4 weeks post-infection.The apoptosis rate at 6 hours was significantly higher in the experimental group than the PBS group(P<0.05).At 12 and 24 hours,the apoptosis rate in the experimental group remained higher than that in the PBS group,although this difference was not statistically significant.After immunization,mice in all four groups exhibited normal growth patterns,as indicated by stable body weight changes.At 4 and 12 weeks post-immunization,the lung coefficients in the protein group were significantly higher than those in the PBS group at the same time points.Additionally,the lung coefficients in the BCG group were significantly elevated across all time periods(P<0.05).The spleen coefficients in the protein and BCG groups were significantly higher than those in the PBS group at 2,4,8,12,and 16 weeks,whereas the ICD B/R group showed higher spleen coefficients than the PBS group only at week 8(P<0.05).Pathological examination revealed normal lung and spleen tissues in the PBS group.However,during the 2-8 weeks immunization period,lung and spleen tissues in all experimental groups exhibited varying degrees of damage,which gradually diminished by 12-16 weeks.Notably,no tuberculosis nodules were observed in any experimental group.After infection with high concentrations of BCG,no overt pathological changes were observed on the surfaces of the lungs and spleens in any group.Microscopic examination revealed less severe pathological changes in the lungs and spleens of mice in the experimental groups than the PBS group.Furthermore,no statistically significant differences were observed between the protein group and the BCG group.Our findings suggested that the B/R strain active proteins'toxicity and safety profiles were comparable to those of BCG,and showed immunoprotective effects.This study provides an experimental foundation for the development of a novel tuberculosis vaccine.
3.Application of predictive nursing based on root cause analysis in cesarean section patients

Objective:To investigate the effectiveness of predictive nursing based on root cause analysis in patients undergoing cesarean section.Methods:A convenience sampling method was used to select 180 women who underwent cesarean section under combined spinal-epidural anesthesia in the Affiliated Hospital of Jining Medical University from September 2021 to October 2022. According to the random number table method, they were divided into a control group ( n=90) and an observation group ( n=90). The control group received routine nursing care, while the observation group received predictive nursing based on root cause analysis. Compared the pain intensity at 24 hours after cesarean section and the incidence of postoperative complications between the two groups of parturients. Results:The Visual Analog Scale scores at 24 hours post-cesarean section and the overall incidence of postoperative complications were lower in the observation group than those in the control group, and the differences were statistically significant ( P<0.05) . Conclusions:Predictive nursing based on root cause analysis can effectively relieve postoperative pain and reduce the incidence of complications in patients undergoing cesarean section with combined spinal-epidural anesthesia.

5.Research progress on regulatory mechanism of AQP4 polarization distribution in glymphatic system

The glymphatic system(GS)is a unique toxic sub-stance clearance system in brain,which is very important for maintaining the microenvironment stability of the central nervous system.The polarization distribution of aquaporin 4(AQP4)lo-cated in the terminal foot of astrocytes affects the function of GS and participates in the pathological progress of many neurodegen-erative diseases,but the detailed regulation mechanism of AQP4 polarization distribution has not been systematically summarized.Therefore,this paper systematically combs the mechanism of reg-ulating the polarization distribution of AQP4 from the perspective of the composition integrity of dystrophin-glycoprotein complex(DGC)and basement membrane foot complex,and summarizes the potential drug and non-drug therapies for targeted regulation of AQP4 polarization distribution at present,aiming at providing new target reference and theoretical basis for targeted regulation of AQP4 polarization to prevent and treat neurodegenerative dis-eases.
6.Tympanoplasty and eustachian tube balloon dilatation in the treatment of adhesive otitis media

Objective To compare the clinical effect of tympanoplasty with and without eustachian tube bal-loon dilatation(ETBD)in the treatment of adhesive otitis media(AOM).Methods A total of 43 patients with AOM and eustachian tube dysfunction(ETD)were retrospectively analyzed,and were divided into the experimental group(n=22)who underwent tympanoplasty combined with ETBD and the control group(n=21)who underwent tympanoplasty alone.The changes of average air conduction hearing thresholds,pure-tone air-bone gap(ABG)and eustachian tube score(ETS)were compared between the two groups before and after surgery at 6 months and 2 years.Results There were no significant differences between the experimental group and the control group in air conduction hearing thresholds,ABG and ETS before operation(P>0.05).Comparison within both the two groups after operation:air conduction hearing thresholds and ABG in both groups were significantly reduced(P<0.05),and ETS in the experimental group was significantly higher than that before surgery(P<0.05).Comparison be-tween two groups after operation:there were no statistically significant differences in the air conduction hearing thresholds and ABG at 6 months after surgery(P>0.05),while there was statistically significant difference in ETS(P<0.05).At 2 years,there were statistically significant differences in the air conduction hearing thresholds,ABG and ETS between the two groups(P<0.05)with the experimental group showing better outcomes.Conclusion Tympanoplasty can significantly improve the postoperative hearing of AOM.Tympanoplasty combined with ETBD not only effectively improves the eustachian tube function,but also shows better long-term follow-up hearing im-provement,making it an effective treatment for AOM patients.
7.Application effect of information platform-based chain management model on in postoperative chemotherapy patients with breast cancer

Objective:To investigate the application effect of an information platform-based chain management model in postoperative chemotherapy patients with breast cancer.Methods:A total of 126 patients with breast cancer who underwent chemotherapy after surgery were selected from Sep 2021 to Apr 2024 and randomly divided into two groups by random sampling method,with 63 cases in each group.They all received at least 3 cycles of chemotherapy.The control group received routine nursing care,while the observation group received the information platform-based chain management model based on information platform in addition to routine nursing.The compliance with chemotherapy,incidence of adverse events related to peripherally inserted central catheter(PICC)was compared between the two groups.Strategies used by people to promote health(SUPPH),Self-care Ability of PICC(CPPSM),and functional assessment of cancer therapy-breast(FACT-B)were used to compare the self-management efficacy,self-care ability and quality of life between the two groups.Results:The compliance rate of chemotherapy in the observation group was higher than that in the control group(P<0.05).After the first and third chemotherapy,the scores of SUPPH,CPPSM,and FACT-B in the observation group were higher than those in the control group(P<0.05).The incidence of adverse events related to PICC in the observation group was lower than that in the control group(P<0.05).Conclusion:The information platform-based chain management model can improve the chemotherapy compliance,enhance self-care capacity and self-management efficacy,reduce PICC-related adverse events,and ultimately elevate quality of life in postoperative breast cancer patients.
8.Effects of Epothilone D on Social Defeat Stress-induced Changes in Microtubule-related and Endoplasmic Reticulum Stress Protein Expression

Objective:
Epothilone D (EpoD), microtubule (MT) stabilizing agent, demonstrated promising results in the animal models of Alzheimer’s disease, Parkinson’s disease and schizophrenia. The present study sought to investigate preventive effects of EpoD on altered changes of MT related proteins and endoplasmic reticulum (ER) stress proteins induced by social defeat stress (SDS).
Methods:
We measured protein expression levels of -tubulin and its post-translational modifications, MT-associated protein 2, stathmin1 and 2 with their phosphorylated forms, and ER stress markers, 78-kDa glucose-regulated protein (GRP-78) and CCAAT/enhancer binding protein (C/EBP)-homologous protein (CHOP) in the prefrontal cortex (PFC) and hippocampus (HIP) of C57BL/6J strain mice treated with EpoD (2 mg/kg) or its vehicle, dimethylsulfoxide (DMSO), and exposed to SDS.
Results:
We observed lower levels of acetylated -tubulin, MAP2, p-STMN (Ser16), and GRP-78 in the PFC of the EpoD-Con group when compared to the DMSO-Con group. On the other hand, in the HIP, there were significantly higher levels of tyrosinated -tubulin and GRP-78 in the EpoD-Defeat group compared to the DMSO-Defeat group.Furthermore, the level of MAP2 in the HIP was found to be lower in the EpoD-Con group compared to the DMSO-Con group.
Conclusion
Our results suggest that EpoD exhibits a dual impact, manifesting both beneficial and detrimental effects on the aberrant changes of MT-related proteins and ER stress proteins induced by SDS, depending on the brain regions.These findings underscore the complexity of EpoD’s effects, necessitating further exploration to understand its intricate mechanisms in cellular pathways linked to SDS.

Objective:
Impact of Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5) changes on the criteria for schizophrenia (SZ) has been reported to be minimal in previous studies. However, this could be different in first-episode schizophrenia spectrum disorders (FE-SSDs). We investigated what proportion of patients with FE-SSDs was diagnosed based on the sole presence of bizarre delusions (BDs) or first rank auditory hallucinations (FRAHs). Their alternative diagnosis by the DSM-5 was established and diagnostic stability over 1-year was identified.
Methods:
This was a retrospective review study on the medical records, case report forms for the subjects with FE-SSDs (n=404) participated in the Korea Early Psychosis Study. The two Japanese sites reviewed retrospectively only medical records of the subjects with FE-SSDs (n=103). We used three different definitions of BDs (strict, narrow, and broad) and specified subtypes of Other Specified Schizophrenia spectrum and Other psychotic disorders (OSSOs). To ensure inter-rater reliability between the hospitals, regular zoom meetings were held.
Results:
Forty (7.89%) subjects out of 507 were found to be diagnosed as SSDs based on the sole presence of BDs or FRAHs. All these patients met the criteria of OSSOs and were classified as having pure delusion (n=22), delusion with attenuated auditory hallucinations (AHs) (n=5), pure AHs (n=3) and AHs with attenuated delusion (n=10). The patients with first and second subtypes (n=27) were found to have BDs. The BDs fulfilled mostly strict definitions or satisfied the next broadest definition. The diagnostic stability of FE-OSSOs and its subgroups (first and second subtypes) over 1-year was substantially high (70.27% and 84% respectively).
Conclusion
These findings suggest that more rigorous diagnostic assessment should be performed especially to differentiate OSSOs from SZ in patients with FE-SSDs and more refined classification of the subtypes for OSSOs considered in the next DSM revision.
