This study investigates the neuroprotective potential of extracellular vesicles (EVs) delivering quercetin-3-O-β-d-glucuronic acid (QG-EVs) in cerebral ischemia-reperfusion injury (CIRI). Targeted brain delivery of QG-EVs was confirmed, with neuron cells identified as pivotal in modulating CIRI through single-cell RNA sequencing (scRNA-seq). Activating transcription factor 4 (Atf4) was highlighted as a critical regulatory factor, and in vitro studies revealed that silencing Atf4 diminished the neuroprotective effects of QG-EVs, increasing oxidative stress levels and neuronal apoptosis. In a CIRI mouse model, the knockdown of Atf4 attenuated the protective outcomes provided by QG-EVs, further affirming the role of Atf4 in mediating neuroprotection. Behavioral assessments and protein analysis showed that QG-EVs significantly reduced neuronal damage and pro-apoptotic markers, while improving neurological function via Atf4 upregulation. The outcomes hint at the potential of QG-EVs as a beneficial therapeutic modality to mitigate neuronal damage in CIRI by enhancing Atf4 expression, highlighting its potential for improving ischemic stroke outcomes.

The amount of data generated by the human experience in medical cases of classic formulas is large and the research value is high, but how to extract valuable information from these massive data is an important problem of researches on classical formulas. It is concluded that there are many problems in the current study of human experience in classical formulas, such as large amount of data, uneven quality, weak extrapolation and clinical application, which seriously affected the evidence strength of human experience in classical formulas and the contribution to experience accumulation and theoretical support. Therefore, from the structural perspective of the research questions, this paper proposed the research ideas of human experience in medical cases of classic formulas: firstly, according to the characteristics of traditional Chinese medicine (TCM) cases, based on the PICO model, which refers parti-cipants (P), intervention (I), comparisons (C), and outcomes (O), a TCM medical record evaluation tool in line with the evidence-based concept was constructed to evaluate the quality of medical cases; secondly, constructing a standardized database of human experience in classical formulas and using data mining algorithms to explore the deep relationship hidden in human experience; finally, the clinical value evaluation is carried out to verify the effectiveness of the diagnosis and treatment rules of classical formulas to achieve clinical transformation.

Objective To investigate current situation of disaster medicine courses among medical schools in China and perception of disaster management among medical students. Methods The survey was conducted among medical students from 14 medical schools in China.A questionnaire which included self-evaluation of ability to practice disaster management, status of disaster medicine courses and self-interest in learning disaster management was used. Results The survey showed that only a few of medical schools offered disaster medicine courses and the quality of current courses was not satisfying.Medical students were generally lack of systematic perception of disaster management, but they were interested in studying related knowledge. Conclusions It is necessary to design disaster medicine courses of high quality and promote them among medical schools in China. It can not only meet the requirements of medical students, but also effectively improve the response capacity of medical students and doctors when facing catastrophes.

Aim To study the apoptosis effect of Sir-aitia grosvenorii extract on human lung cancer cells A549 and its mechanisms. Methods MTT assay was applied to determine A549 cell proliferation.Hoechst 33258 staining was applied to investigate morphological changes in A549 cells. To find out the cause of cell growth inhibition, several experiments on cell cycle distribution and apoptosis were performed by flow cy-tometry analysis. The expression of p21 and Bcl-2 was determined by Western blot. Results Flow cytometry analysis showed that treatment with mogrol arrested A549 cells in the G0 /G1 phase and induced apoptosis. After treatment with Siraitia grosvenorii extract,West-ern blot experiment showed cell cycle regulator p21 was up-regulaed,while the apoptosis inhibitor Bcl-2 was down-regulated.Conclusion Treatment with Siraitia grosvenorii extract arrests the A549 cells at G0 /G1 phase and induces apoptosis that may contribute to the anti-proliferation activity of mogrol through the regula-tion of p21 and Bcl-2 expression.
