1.Differential expression of retinal mRNA in experimental autoimmune uveitis mice based on transcriptome sequencing

Objective:To screen differentially expressed genes in the retinas of mice with experimental autoimmune uveitis (EAU) through transcriptome sequencing.Methods:Twenty 8-week-old SPF grade male C57BL/6J mice were randomly divided into EAU group and control group by random number table method, with 10 mice in each group.In the EAU group, an emulsion of interphotoreceptor retinoid-binding protein 1-20 and complete Freund adjuvant was injected subcutaneously into the inner side of the hind limbs and intravenously into the tail vein of the mice, and pertussis toxin was injected intraperitoneally to establish the EAU model.In the control group, mice were subcutaneously injected with the corresponding volume of phosphate buffer solution.On the 21st day after model establishment, the anterior segment inflammation of the mouse eyes was examined using a slit-lamp microscope.Retinal blood vessels were observed by fundus photography and clinical inflammation was scored.Hematoxylin-eosin staining was used to observe the morphological changes in the retinal tissues of the two groups.Retinal tissues were collected from two groups of mice.RNA was extracted and cDNA libraries were constructed for transcriptome sequencing.The clusterProfiler package in R programming language was used to screen for differentially expressed genes in EAU, and to conduct GO and KEGG enrichment analyses.The expression of core differentially expressed genes in the retinal tissues was then verified using real-time fluorescent quantitative PCR.The breeding and use of experimental animals were in compliance with the relevant regulations of the Animal Ethics Committee of Anhui Medical University.The study protocol was approved by the Ethics Committee of Anhui Medical University (No.LLSC20221208).Results:On the 21st day after model establishment, the clinical score of the EAU group was 2.83±0.94, significantly higher than 1.89±0.93 of the control group, and the difference was statistically significant ( t=2.290, P<0.05). The retinal blood vessels of mice in the EAU group showed tortuosity and dilation.Histopathological examination revealed a large number of inflammatory cell infiltrations in the retina, indicating successful establishment of the EAU mouse model.Transcriptome sequencing of the mouse retinal tissues identified a total of 32 473 genes, and 154 differentially expressed genes were screened out.Among them, 79 genes were up-regulated with Gm38574, Tmem81, Ptpn7 being top three and 75 genes were down-regulated with Gm19802, Fga, Cyp3a11 being top three.GO enrichment analysis showed that the differentially expressed genes were mostly related to immune response and cytokine action.KEGG pathway enrichment analysis showed that the differentially expressed genes were mainly enriched in the Toll-like pathway and cytokine signaling pathway.Compared with the control group, the relative expression levels of Gm38574, Tmem81, and Ptpn7 in the retinal tissues of the EAU group were up-regulated, while the expression levels of Gm19802, Fga, and Cyp3a11 were down-regulated, and the differences were statistically significant ( t=9.755, 5.358, 6.289, 4.312, 6.577, 6.118; all P<0.05). Conclusions:There are significant differences in the key genes Gm38574, Tmem81, Ptpn7, Gm19802, Fga, and Cyp3a11 in the retinal tissues of EAU mice, mainly corelated with the cytokine interaction pathway, Toll-like receptor signaling pathway.
2.The impact of lesions in different nuclei of the basal ganglia on speech processing function in patients with post-stroke dysarthria

Objective:To investigate the impact of lesions in different nuclei of the subcortical basal ganglia on speech processing functions in patients with post-stroke dysarthria.Methods:From July 2022 to September 2023, a total of 20 patients with post-stroke dysarthria (patient group) and 22 healthy individuals (control group) were recruited. Brain imaging data, including structural magnetic resonance imaging(MRI) and diffusion-weighted imaging (DWI), as well as behavioral data of speech fluency task and picture association task were collected. Structural MRI data was analyzed using SPM12 software to perform voxel-based morphometry (VBM), measuring cortical thickness and gray matter volume (VGM) in specific nuclei of the basal ganglia. Behavioral metrics, such as reaction time (RT) and the number of valid responses were extracted for each task. Statistical analysis was conducted using SPSS 27.0 and R 4.0 softwares. Spearman correlation analysis was applied to examine relationships between neuroimaging parameters and behavioral performance indicators.Results:The patient group exhibited significantly smaller gray matter volumes in both left and right caudate nuclei((2.69±0.92)mm 3, (3.17±0.91)mm 3 ) and putamen (3.31±1.08)mm 3, (3.66±0.91)mm 3) compared to the control group (caudate nuclei (3.19±0.36)mm 3, (3.49±0.52)mm 3 putamen (4.52±0.54)mm 3, (4.72±0.64)mm 3), with statistically significant differences ( t=-2.83, 1.68; t=-3.59, 3.52, both P<0.05). Behavioral experiments revealed that the patient group exhibited significantly prolonged reaction time during picture association and naming tasks (1 910.50(1 214.25, 3 806.75) ms, 1 362.00(978.00, 2 297.00) ms) compared to the control group (1 618.00(1 162.75, 2 401.75) ms, 1 224.00(984.25, 1 661.50) ms; Z=-5.20, -4.61, both P<0.05). Gray matter volumes in the left caudate nucleus and left putamen exhibited negative correlations with reaction times during the picture naming task ( r=-0.52, -0.54, both P<0.05). Additionally, the gray matter volume of the left putamen demonstrated a positive correlation with the number of valid responses in speech fluency task-T2 ( r=0.46, P<0.05), whereas the left globus pallidus volume showed a negative correlation with speech fluency task-T1 ( r=-0.51, P<0.05) with the same measure. Conclusion:Lesions in the left subnuclei of the basal ganglia directly impair early-stage speech functions, including conceptual preparation and lexical selection, whereas right-side lesions exert less pronounced effects on linguistic performance compared to their left counterparts. Furthermore, the basal ganglia's involvement in higher-order linguistic processing may represent an indirect consequence of cognitive decline.

This consensus consolidates expert insights regarding diagnostic and treatment procedure for hand injury in microsurgery, emergency support protocols and key points of emergency workflow optimisation. It summarises the opinions of experts and puts forward standardised recommendations to guide clinical practice in microsurgical treatment process, so as to further improve the quality of treatment for hand injury in microsurgery and maximise the protection of limb function and quality of life of patients.
6.Correlation of Serum Galanin,Obestatin and Apelin-12 Expression with Clinicopathological Features and Helicobacter pylori Infection in Patients with Gastric Cancer

Objective To explore the expression changes of Galanin,Obestatin and Apelin-12 in the serum of Gastric Cancer(GC)patients and their correlation with Helicobacter pylori(Hp)infection.Methods 92 GC patients admitted to Tianmen First People's Hospital Affiliated to Wuhan University of Science and Technology from December 2021 to December 2023 were re-garded as the GC group,and a total of 76 age-and sex-matched non-GC(gastritis)patients were selected as control group.The 13C breath test method was used to detect Hp infection in patients.Basedon the test results,Hp infected GC patients were separat-ed into grade Ⅰ infection group(n=23),grade Ⅱ infection group(n=29)and grade Ⅲ infection group(n=26).Enzyme linked im-munosorbent assay(ELISA)was applied to detect serum levels of Galanin,Obestatin,Apelin-12 and inflammatory factors.Pear-son's method was used to analyze the correlation between serum levels of Galanin,Obestatin and Apelin-12 and Hp infection in GC patients.Receiver operating characteristic(ROC)curve was applied to evaluate the diagnostic value of serum Galanin,Obestatin and Apelin-12 for GC.Results The Hp positive rate in the GC group was greatly higher than that in the control group(39.47%vs 84.78%),and the differences was statistically significant(χ2=37.214,P<0.001).The serum Galanin level in GC group(19.69±7.23pg/ml)was lower than that in control group(32.73±12.45pg/ml),the level of Obestatin,apelin-12(40.46±11.78ng/ml,323.09±59.25ng/L)were higher those that of the control group(26.15±8.27ng/ml,264.72±40.96ng/l),and the differences were statistically significant(t=8.468,8.926,7.271,all P<0.05),and their levels were related to TNM staging and lymph node metastasis(t=2.963~12.070,all P<0.05).Comparison of serum Galanin,Obestatin and Apelin-12 levels between the non-infected group and theⅠ,ⅡandⅢ grade infection group showed statistically significant differences(F=26.594,38.245,13.545,all P<0.05).Pearson correnlation analysis showed that serum Galanin levels were negatively correlated with DOB values and TNF-α,IL-1β and IL-6 levels(r=-0.477~-0.387,all P<0.001),Obestatin and Apelin-12 levels were positively correlated with DOB values and TNF-α,IL-1β,IL-6 levels(r=0.389~0.453,all P<0.001).The AUC(95%CI)of serum Galanin,Obestatin and Apelin-12 levels for single and combined diagnosis of GC were 0.853(0.790~0.903),0.809(0.741~0.865),0.803(0.735~0.861)and 0.925(0.874~0.960),respectively.The combined diagnostic efficacy superior to the individual diagnosis of Galanin,Obestatin,and Apelin-12,and the differences were statistically significant(Z=2.237,3.719,3.913,all P<0.05).Conclusion The low expression of Galanin and high expression of Obestatin and Apelin-12 in serum of GC patients are closely related to Hp infection,and their combination has certain diagnostic value for GC.
7.Analysis of clinicopathological parameters and risk factors for the patients with connective tissue disease-associated interstitial lung disease complicated with EBV viraemia

Objective To investigate the clinicopathological parameters and risk factors of the patients with connective tissue disease-associated interstitial lung disease(CTD-ILD)complicated with Epstein-Barr virus(EBV)viraemia.Methods The CTD-ILD pa-tients admitted to Department of Respiratory and Critical Care Medicine,Nanjing Drum Tower Hospital from January 2023 to April 2024 were collected.Based on the detection results of EBV DNA,the patients were divided into the EBV DNA(+)group and EBV DNA(-)group.The clinicopathological parameters of the two groups were analyzed.Results Out of 162 CTD-ILD patients who underwent EBV DNA testing,a total of 28 were found to have EBV viraemia.The levels of serum albumin([32.7±4.1]g/L vs[34.8±3.8]g/L,t=2.559,P<0.05),oxygenation index([268.5±94.0]mmHg vs[323.2±120.9]mmHg,t=2.062,P<0.05),and percentages of predicted diffusing capacity for carbon monoxide([30.9±15.3]% vs[44.9±18.8]%,t=2.127,P<0.05])in the EBV DNA(+)group were significantly lower than those in the EBV DNA(-)group,while the levels of lactate dehydrogenase(LDH,[369.1±206.2]U/L vs[298.8±128.7]U/L,t=2.335,P<0.05)were significantly higher than that in the EBV DNA(-)group.The acute exacerbation of ILD in the EBV DNA(+)group was more common than that in the EBV DNA(-)group(P<0.05).Multivariate Lo-gistic analysis showed that honeycombing and low oxygenation index were independent risk factors of CTD-ILD patients complicated with EBV viraemia.Conclusion The CTD-ILD patients complicated with EBV viraemia have poorer oxygenation and are more prone to suf-fer from acute exacerbation of ILD.Honeycombing in chest HRCT and low oxygenation index are independent risk factors of CTD-ILD patients complicated with EBV viraemia.
