1.Research on the improvement of cognitive impairment,endoplasmic reticulum stress and neuroinflammation in Alzheimer's disease by emodin

Objective·To explore the effects and potential mechanisms of emodin on Alzheimer's disease(AD).Methods·Wild-type C57BL/6J mice and 3×Tg-AD mice were divided into 6 groups:Control group(C57BL/6J mice),AD group(3×Tg-AD mice),Emodin 25 mg/kg group(3×Tg-AD mice+Emodin 25 mg/kg),Emodin 50 mg/kg group(3×Tg-AD mice+Emodin 50 mg/kg),Emodin 100 mg/kg group(3×Tg-AD mice+Emodin 100 mg/kg)and Donepezil group(3×Tg-AD mice+Donepezil 3 mg/kg).The Morris water maze test was used to evaluate the learning and memory abilities of mice.The expression of glial fibrillary acidic protein(GFAP),glucose-regulated protein 78kDa(GRP78),and inositol-requiring enzyme 1α(IRE1α)was detected by immunohistochemistry.The levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and IL-6 in brain tissue were measured by enzyme-linked immunosorbent assay(ELISA).Western blotting was used to detect the expression of NF-κB p65,p-NF-κB p65,p38,and p-p38 proteins.Results·Compared with the control group,mice in the AD group showed impaired cognition,increased GFAP expression,elevated levels of TNF-α,IL-1β and IL-6,and increased expression of GRP78 and IRE1α,along with enhanced phosphorylation of NF-κB p65 and p38.Compared with the AD group,emodin improved cognitive impairment of AD mice,inhibited astrocyte overactivation and neuroinflammation,and decreased the expression of GRP78,IRE1α,phosphorylated NF-κB p65,and phosphorylated p38 in brain tissue.Conclusion·Emodin can effectively improve cognitive impairment in AD mice,which may be related to the inhibition of endoplasmic reticulum stress-mediated neuroinflammation in astrocytes.
2.Efficacy and safety of transdermal delivery of compound glycyrrhizin injection as an adjunctive treatment for erythematotelangiectatic rosacea

Objective:To evaluate the efficacy and safety of transdermal delivery of compound glycyrrhizin injection as an adjunctive treatment for erythematotelangiectatic rosacea (ETR).Methods:This was a randomized controlled trial conducted from March to October 2024. At Sichuan Provincial People′s Hospital, 60 patients with newly diagnosed ETR were prospectively enrolled and randomized by a random number table into study group [ n=30; 6 male and 24 female; aged 18-60 (38.9±9.8) years] and control group [ n=30; 4 male and 26 female; aged 18-60 (35.7±10.1) years]. The study group received transdermal delivery of compound glycyrrhizin injection by a medium-frequency drug-delivery therapeutic apparatus together with oral azithromycin and hydroxychloroquine sulfate, whereas the control group received oral azithromycin and hydroxychloroquine sulfate. The efficacy evaluations were conducted at baseline and at weeks 2, 4, 6, and 8 post-treatment. Outcome measures included percentage of erythema area, stratum corneum hydration, transepidermal water loss (TEWL), clinician′s erythema assessment (CEA), erythema and telangiectasia scores, dermatology life quality index (DLQI), and efficacy rate. Adverse reactions during treatment were also recorded. Results:Compared with baseline, both groups exhibited significant reductions in percentage of erythema area, TEWL, CEA, erythema and telangiectasia score, and DLQI, and significant increases in stratum corneum hydration at each post-treatment time point (all P<0.05). After 2, 4, 6, and 8 weeks of treatment, the percentage of erythema area, erythema and telangiectasia scores in the study group were all lower than those in the control group, while the stratum corneum hydration level in the study group was higher than that in the control group (all P<0.05). After 4, 6, and 8 weeks of treatment, TEWL, CEA, and DLQI in the study group were all lower than those in the control group (all P<0.05). After 4 weeks of treatment, the efficacy rate in the study group was 56.7% (17/30), which was higher than that of the control group at 23.3% (7/30, P=0.046). After 6 weeks of treatment, the efficacy rate in the study group was 83.3% (25/30), higher than that of the control group at 50.0% (15/30, P=0.020). After 8 weeks of treatment, the efficacy rate in the study group was 86.7% (26/30), higher than that of the control group at 66.7% (20/30, P<0.001). No severe adverse reactions were observed in either group. Conclusion:Transdermal delivery of compound glycyrrhizin injection as an adjunctive treatment for ETR demonstrates favorable efficacy and good safety.

5.Procoagulant role,mechanism and potential applications of inorganic polyphosphate

Inorganic polyphosphate(polyP)is wide spread in biology and exhibits striking procoagulant ef-fects in vivo.Long-chain polyphosphate(such as those present in infectious microorganisms)is a potent,natural patho-physiologic activator of the contact pathway of blood clotting and promotes thrombosis and inflammation,which suggest that antagonizing polyP function in vivo may be an attractive approach for identifying novel antithrombotic agents,perhaps with reduced bleeding side effects compared with conventional anticoagulant drugs.Short-chain polyphosphate(secreted from activated human platelets)remarkably accelerates factor V activation,absolutely abrogates the anticoagulant function of tissue factor pathway inhibitor,increase fibrin clot structure,and accelerates factor XI activation by thrombin.So polyP may be used as hemostatic agent.In this paper,the procoagulant role of polyP and the mechanism underlying polyP modu-lates blood clotting and the strategy of screening polyP inhibitors are described in detail.
6.Expression levels of serum KLK5, IL-17 and IL-18 in patients with rosacea and their clinical significance

Objective:To investigate the expression levels of serum KLK5, IL-17 and IL-18 in patients with rosacea and their clinical significance.Methods:A prospective study included 65 patients with rosacea from the Department of Dermatology, Sichuan Provincial People's Hospital, from May to October 2022. There were 8 males and 57 females with an average age of (30.9±9.8) years, including 47 cases of erythematotelangiectatic type and 18 cases of papulopustular type. Twenty healthy individuals from the same hospital's physical examination during the same period were included as control group, consisting of 3 males and 17 females with an average age of (33.8±10.0) years. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression levels of serum KLK5, IL-17, and IL-18. Investigator's global assessment (IGA) score was used to assess the severity of papules and pustules, and clinical erythema assessment (CEA) score was used to assess the severity of persistent erythema. The expression levels of serum KLK5, IL-17, and IL-18 were compared between the two groups, and the correlation between the severity of the disease and the expression levels of KLK5, IL-17, and IL-18 was analyzed. Additionally, the expression levels of KLK5, IL-17, and IL-18 in different subtypes of rosacea were compared.Results:The expression levels of serum KLK5, IL-17, and IL-18 in the rosacea group were (3134.01±448.82), (154.78±37.64), (236.24±38.81) pg/ml, respectively, all of which were higher than those in the healthy control group (2421.54±340.07), (95.36±24.94), (181.61±23.88) pg/ml, with statistically significant differences (all P<0.05). The levels of serum KLK5, IL-17, and IL-18 in patients with rosacea were positively correlated with IGA scores ( r=0.534, 0.767, 0.417) and CEA scores ( r=0.852, 0.459, 0.309) (all P<0.05). There were no statistically significant differences in the expression levels of serum KLK5, IL-17, and IL-18 between the erythematotelangiectatic and papulopustular subtypes of rosacea (all P>0.05). Conclusions:The expression levels of serum KLK5, IL-17 and IL-18 in patients with rosacea are higher than those in the healthy control group and are positively correlated with the severity of rosacea.
7.Progress in research and development of biodegradable metallic vascular stents.

Vascular stents are an essential tool in cardiovascular interventional therapy, and their demand is growing with the increasing incidence of cardiovascular diseases. Compared with permanent stents, which are prone to in-stent restenosis, and drug-eluting stents, which may cause late stent thrombosis, biodegradable stents offer advantages. After providing early radial support to prevent elastic recoil, biodegradable stents gradually degrade, allowing the vessel to regain its natural physiological contractility and undergo positive remodeling. A review of the current mainstream biodegradable metal stents, magnesium-based, iron-based, and zinc-based alloys, shows promising findings in both preclinical and clinical research. Magnesium-based stents exhibit good operability and low thrombosis rates, but their limitations include rapid degradation, hydrogen evolution, and significant pH changes in the microenvironment. Iron-based stents demonstrate excellent mechanical strength, formability, biocompatibility, and hemocompatibility, but their slow corrosion rate hampers broader clinical application; accelerating degradation remains key. Zinc-based alloys have a moderate degradation rate but relatively low mechanical strength; enhancing stent strength by alloying with other elements is the main improvement direction for zinc-based stents.

8.Effects of daily mean temperature and other meteorological variables on bacillary dysentery in Beijing-Tianjin-Hebei region, China.

BACKGROUND:
Although previous studies have shown that meteorological factors such as temperature are related to the incidence of bacillary dysentery (BD), researches about the non-linear and interaction effect among meteorological variables remain limited. The objective of this study was to analyze the effects of temperature and other meteorological variables on BD in Beijing-Tianjin-Hebei region, which is a high-risk area for BD distribution.
METHODS:
Our study was based on the daily-scale data of BD cases and meteorological variables from 2014 to 2019, using generalized additive model (GAM) to explore the relationship between meteorological variables and BD cases and distributed lag non-linear model (DLNM) to analyze the lag and cumulative effects. The interaction effects and stratified analysis were developed by the GAM.
RESULTS:
A total of 147,001 cases were reported from 2014 to 2019. The relationship between temperature and BD was approximately liner above 0 °C, but the turning point of total temperature effect was 10 °C. Results of DLNM indicated that the effect of high temperature was significant on lag 5d and lag 6d, and the lag effect showed that each 5 °C rise caused a 3% [Relative risk (RR) = 1.03, 95% Confidence interval (CI): 1.02-1.05] increase in BD cases. The cumulative BD cases delayed by 7 days increased by 31% for each 5 °C rise in temperature above 10 °C (RR = 1.31, 95% CI: 1.30-1.33). The interaction effects and stratified analysis manifested that the incidence of BD was highest in hot and humid climates.
CONCLUSIONS
This study suggests that temperature can significantly affect the incidence of BD, and its effect can be enhanced by humidity and precipitation, which means that the hot and humid environment positively increases the incidence of BD.

9.Factors influencing HIV infection among college students who are young men who have sex with men in Tianjin

Objective:
To analyze associated factors of HIV infection among college students who are young men and have sex with men (YMSM) in Tianjin, providing reference for HIV prevention and control among YMSM college students.
Methods:
During Aug. 1st, 2018 to Dec. 31st, 2018, SHENLAN recruited college students who were YMSM aged 18-24 years from gay baths, gay bars, QQ, WeChat and gay dating app BLUED. HIV infection status and associated factors (general demographic characteristics, unsafe sexual behaviors, addictive substance using, basic knowledge of HIV) was collected and analyzed.
Results:
A total of 470 college students, including 21 HIV infected (4.47%), were enrolled in this study. Univariate Logistic regression analyses indicated that age, age at first sex behavior, HIV related knowledge, tobacco use, recreational drug usage, syphilis infection was associated with HIV infection among YMSM students (P<0.05). Multivariate analysis found age of first sex (OR=21.20,95%CI=3.09-145.43), recreational drug use (OR=5.07,95%CI=1.77-14.48), lack of HIV related knowledge (OR=3.38,95%CI=1.33-8.63)were associated with HIV infection (P<0.05).
Conclusion
College students who are YMSM in Tianjin have a high rate of HIV infection, who deserves further attention. Targeted campus HIV/AIDS prevention program should be developed combined with specific characteristics of this population.
10.Application of iterative reconstruction in upper limb intravascular fistula CT angiography

Objective To assess the image quality (IQ) of an iterative reconstruction (IR) technique(sinogram affirmed iterative reconstruction,SAFIRE) for upper limb intravascular fistula CT angiography on a second generation dual-source CT scanner and determine the optimal dose level using IR with IQ comparable to filtered back projection(FBP).Methods The experiment was a prospective study.A total of 150 patients were averagely divided into 5 groups to perform upper limb intravascular fistula CT angiography.The control group was scanned with conventional tube output (120 kV,110 mAs) and images were reconstructed with FBP.The tube current of the other 4 groups (reconstructed with SAFIRE) was successively reduced to 70%,60%,50% and 40% of that of control group.Quantitative measurements of signal-to-noise ratio (SNR) and contrast-to-noise ratio (CNR) were obtained in two regions of interest (ROI) in each group.The ROIs were placed at the subclavian artery near shoulder joint,and the stoma of radial artery and cephalic vein.Analysis of variance (ANOVA) was used for comparisons of objective evaluation parameters (SNR,CNR) and radiation dose (CTDIvol,DLP,E) among 5 groups.Two radiologists graded IQ in a blinded fashion on a 4-point scale (4-excellent,3-good,2-fair and 1-poor).The Kruskal-Wallis nonparametric test was performed for detection of differences in subjective evaluation of IQ among groups.Results The radiation doses of groups 77,66,55 and 44 mAs were 70%,55%,40% and 25% of the control group respectively.The differences of SNR and CNR among groups 77,66,44 mAs and the control group were statistically significant in ROI1 (F =24.018,20.386,P < 0.05),SNR and CNR of group 77 and 66 mAs were higher than the control group while group 44 mAs was lower.The difference of SNR and CNR among group 55 mAs and the control group was not statistically significant in ROI1 (P > 0.05).The differences of SNR and CNR among groups 77,66,55 mAs and the control group were statistically significant in ROI2 (F =15.934,13.818,P < 0.05),and groups 77,66 and 55 mAs had higher SNR and CNR.The difference of SNR and CNR between group 44 mAs and the control group was not statistically significant in ROI2 (P > 0.05).Groups 77,66,55 mAs and the control group had higher scores of the subjective IQ (≥ 3) which was descended in group 44 mAs.The differences of contrast,sharpness,noise and general impression of images among the 5 groups were statistically significant by Kruskal-Wallis nonparametric test(H =10.268,14.542,15.840,11.014,P < 0.05).Using the criterion of excellent IQ(score 4),the ROC curve of dose levels and IQ acceptability established a reduction of 60% of effective dose [Group 55 mAs:(3.545 ± 1.396) mSv] as an optimum cutoff point (AUC:0.844,95% CI:0.705-0.982).Conclusions Iterative reconstruction technique could provide a dose reduction to 40% (3.545 mSv) compared with the filtered back projection in upper limb intravascular fistula CT angiography without reducing equivalent image quality.
