Objective To study on the effects and mechanisms of ganoderma lucidum polysaccharides(GLP)on enteritis induced by enterotoxigenic Escherichia coli(ETEC)in mice. Methods Twenty-four 6-to 8-weeks-old male SPF Kunming mice were selected.A mouse enteritis model was induced by intraperitoneal injection of ETEC bacterial suspension.Mice were divided into normal group,ETEC infection group,ETEC infection+high-dose GLP group, and ETEC infection+ow-dose GLP group using a random number table method, six mice in each group.Pathological changes in mice small intestine tissue were observed through HE staining. The expression of C-reactive protein(CRP),tumor necrosis factor-(TNF)-α,and interleukin(IL)-6 proteins in mice small intestine tissues were detected by Western blot method, and expression levels of Toll-like receptor 4(TLR4),nuclear factor-KB(NF-κB)p65,and myeloid differentiation factor 88(MyD88)mRNA in mice small intestine tissues were detected by RT-qPCR technique to explore the regulatory effect of GLP on ETEC-induced mice intestinal inflammation through TLR4/NF-κB signaling pathway. Results Compared with control group, mice infected with ETEC showed lethargy, reduced appetite, damaged intestinal villi, significant inflammatory infiltration in tissues, and an increasing trend in disease activity index(DAI)scores(P<0.05).The relative expression levels of TLR4,NF-κB p65,and MyD88 mRNA,as well as protein expression levels of CRP,TNF-α,and IL-6 in small intestine tissues of mice, were significantly increased(P<0.01).Compared with ETEC-infected group, mice in different dosage groups showed improvements in mental status and appetite. The relative expression levels of TLR4,NF-κB p65,and MyD88 mRNA in small intestine tissues of mice were significantly decreased in all dosage groups(P<0.01).In ETEC infection+high-dose group, protein expression levels of CRP,TNF-α,and IL-6 in small intestine tissues of mice were significantly decreased(P<0.01),while the decrease in ETEC infection+low-dose group was not significant(P>0.05).Compared with ETEC infection+low-dose group,ETEC infection+high-dose group showed significant decreases in relative expression levels of TLR4 and MyD88 mRNA,as well as in protein expression level of IL-6 in small intestine tissues of mice(P<0.01 or P<0.05).Conclusion GLP can effectively relieve intestinal inflammation induced by ETEC,and may exert its anti-inflammatory effect through regulating TLR4/NF-κB signaling pathway, which may be proportional to its concentration.

BACKGROUND:
Reports on the prevalence of psoriatic arthritis (PsA) among Chinese patients with psoriasis are very limited. This study, conducted by rheumatologists, estimated the prevalence of PsA in a large number of Chinese patients with psoriasis.
METHODS:
Consecutive patients with a confirmed diagnosis of psoriasis attending nine dermatology clinics in five hospitals were recruited. All psoriasis patients were asked to complete a questionnaire comprising 16 questions to identify possible cases of PsA. All patients with one or more positive answers to the questionnaire were evaluated by two experienced rheumatologists.
RESULTS:
A total of 2434 psoriasis patients, including 1561 males and 873 females, were enrolled. Both the questionnaire and rheumatologists' examinations were completed in the dermatology clinics. The results identified 252 patients with PsA, comprising 168 males and 84 females. The overall prevalence of PsA among psoriasis patients was 10.4% (95% confidence interval [95% CI], 9.1%-11.7%). By sex, the prevalence was 10.8% (95% CI, 9.2%-12.5%) for males and 9.6% (95% CI, 7.7%-11.9%) for females and there was no significant sex difference in the prevalence of PsA (P = 0.38). Of the 252 PsA patients, 125 (49.6%, 95% CI, 41.3%-59.1%) were newly diagnosed by rheumatologists. Consequently, the prevalence of undiagnosed PsA among psoriasis patients was 5.2% (95% CI, 4.4%-6.2%).
CONCLUSION
The prevalence of PsA in the Chinese population with psoriasis is about 10.4%, which is almost double that of previous reports in the Chinese population, but lower than that in Caucasians.

Objective To investigate the best pathway of laparoscopic surgery for choledocholithiasis. Methods According to MRCP,the cystic duct was divided into four types. The thick-straight type (n =89)and the thin-crooked type (n =65)patients randomly received the laparoscopic operations via the cystic duct or com-mon bile duct. The operation time, transit operation rate, complication rate and length of hospital stay after opera-tion were analysed. Results There were 46 cases of the thick-straight type via cystic duct,43 cases via common bile duct. The former had shorter operation time and hospital stay after operation (P <0.05),but there was no significant difference in transit operation rate and complication rate. There were 32 cases of the thin-crooked type via cystic duct and 33 cases via common bile duct and there were significant differences in the operation time, transit operation rate, complication rate and length of hospital stay after operation(P <0.05).Conclusion Ac-cording to different types,LCTBDE is suitable for the patients with thick-straight type, and LCHTD can be per-formed in thin-crooked type. Therefore,it can maximize the advantage of minimally invasive surgery.

Objective To explore how does chymase affect diabetic cardiomyopathy by investigating the gene expression of chymase and the relationship between the gene expression of chymase and the angiotensin Ⅱ in the cardiomyopathy of streptozotocin-induced diabetic hamasters. Methods Diabetic hamsters were induced by intraperitoneal injection of STZ 40 mg/kg once a day for 3 days. After stabilization of diabetic state for 18 weeks, the myocardial ultrastructure was observed with electron microscope and pathologic changes were observed by light microscopy. Immunohistochemistry was used to measure the level of expression of type Ⅰ and type Ⅲ collagen in diabetic and normal hamster hearts. Level of blood glucose, lipoprotein was determined using biocehemical methods. Apoptosis of cardiomyocyte was measured using TUNEL methods. Radioimmunuoassay method was used to determine the level of angiotensin Ⅱ. RT-PCR was used to determine chymase gene expression (corrected by?-actin). ResultsComparing with the control group, levels of serum glucose, TG, TC, LDL in DM group were much higher. Concentrations of collagen Ⅰ, Ⅲ and angiotensin Ⅱ [(95.8?16.0)?g/kg tissue vs (51.1?20.8)?g/kg tissue] in myocardial tissue in DM group were much higher than those in control group. RT-PCR result showed: Comparing with the control group, the mRNA expression of chymase in DM group was promoted significantly (0.810?0.026 vs 0.490?0.087). Conclusion In diabetic hamsters, the gene expression of chymase were much higher than thatin the control group, accompanying higher level of Angiotensin Ⅱ, higher levels of expression of collagen Ⅰ and Ⅲ. This result suggests that chymase plays an important role in the diabetic cardiomyopathy by promoting the activity of chymase.
