1.CiteSpace bibliometric analysis of safety and risk studies of antibody drugs in China

Objective To analyze the domestic antibody drug safety and risk research status,the latest research hotspots and frontiers in the current ten years.Methods CiteSpace 6.2.R2 software was used to analyze all literature related to the safety and risk of antibody drugs in CNKI,WanFang data and Vip database from 2012 to 2022.Results A total of 2 773 pieces of literature were obtained from the three databases,which were imported into CiteSpace after deduplication,and finally,1 870 pieces were included in the analysis.In the past decade,the number of articles published in the field of antibody drugs safety and risk research has remained at about 100 articles per year from 2012 to 2019,since 2020,the number of articles published has started to increase,and the annual number of articles published has increased to around 300 articles from 2021 to 2022.The network graph of domestic institutional cooperation showed that there was a lack of cooperation among the research institutions of antibody drug safety and risk research,mainly due to the fact that hospitals were conducting research in this field,and the types of research subjects were relatively single.The author collaboration network graph showed that the core teams in this research field,such as Li Bo,Yang Yanwei,and Lin Zhi,had the closest collaboration,while there was less collaboration among high-yield authors,additionally,some experts and scholars conducted research on their own as individuals or small groups,the research focused on adverse reactions,safety,bevacizumab,Rituximab,Meta-analysis,etc.Conclusion In the past decade,domestic research has mainly focused on the clinical efficacy and safety of antibody drugs,with few scholars exploring the risk of antibody drugs,therefore,in the future,it is necessary to pay attention to the research on the risks of antibody drugs.
2.Relationship between preoperative use of anti-tumor necrosis factor-α and postoperative infectious complications in Crohn′s disease

Objective:To investigate the impact of preoperative anti-tumor necrosis factor (anti-TNF) -α monoclonal antibody therapy on postoperative infectious complications in patients with Crohn′s disease (CD) after intestinal resection with primary anastomosis.Methods:The clinical data of CD patients who underwent intestinal resection with primary anastomosis at Jinling Hospital from January 2017 to December 2021 were retrospectively analyzed. Propensity score matching was used to match patients who did not receive or received anti-TNF treatment within 12 weeks before surgery at a ratio of 1∶2. The relationship between preoperative anti-TNF treatment and postoperative infectious complications was analyzed, and the related risk factors of postoperative infectious complications were analyzed by logistic regression analysis.Results:A total of 501 patients with CD who underwent resection and primary anastomosis were collected. After propensity score matching, 135 patients with CD were included in the analysis, 45 in the treatment group and 90 in the control group. There were no significant differences in the incidence of overall postoperative complications (24.4% vs. 25.6%, P = 0.889) and infectious complications (13.3% vs. 14.4%, P = 0.861) between the two groups. Previous history of intestinal resection due to CD and surgical site of colon were independent risk factors for infectious complications within 30 days after surgery. Conclusion:Previous history of intestinal resection of CD and colon surgery are associated with infectious complications after intestinal resection with primary anastomosis.

4.Effects of exercises on the lipid metabolism and expression of PPAR-α mRNA in skeletal muscles of ApoE knockout mice

Objective To observe the influence of exercises on the mRNA expression of peroxisome proliferator-activated receptor (PPAR-α) and its target genes of acyl-CoA oxidase (ACO),Enoyl-CoA-hydratase and 3-hydroxyacyl-CoA-dehydrogenase (EHHADH) in the skeletal muscles in insulin-resistance mice to develop a way to improve the lipid metabolism.Methods Twenty male ApoE knockout mice were randomly divided into two groups,the high-fat diet group (group HFD) and the exercise training group (group Ex).The HFD group were fed with highfat diet,while the Ex group were fed in the same way,with additionally swimming training.And ten healthy male C57BL/6j mice were chosen as the control group(group ND).After 12 weeks of intervention,the serum lipid,blood glucose and insulin levels were determined,and homeostasis model assessment insulin resistance index (Homa-IRI) was calculated.The bilateral gastrocnemiuses were cut to be observed under a transmission electron microscope,and the mRNA expression of PPAR-α,ACO and EHHADH in skeletal muscle were measured using reverse transcription polymerase chain reaction(RT-PCR).Results The transmission electron microscope showed that the sarcolemma edema,mitochondrial swelling,as well as focal myocytolysis and edema within myofibrils were observed in the HFD group.The total cholesterol,low-density lipoprotein cholesterol,free fatty acid,fasting glucose,insulin and HomaIRI of the HFD group were significantly higher than the control group (P < 0.05),while the mRNA expression of PPAR-α,ACO and EHHADH was significantly deceased than the latter(P < 0.05).After swimming,the abovementioned pathological changes disappeared.The serum lipid of the Ex group were significantly lower (P < 0.05),while HDL was significantly higher (P < 0.05).And fasting insulin,glucose and HOMA-IR of the Ex group were significantly lower (P < 0.05),while the mRNA expression of the above in the Ex group were significantly increased (P <0.05).Conclusion Swimming training could improve insulin resistance and metabolic disorder of lipid of ApoE knockout mice.The possible mechanisms may be through up-regulating the expression of PPAR-α,which in turn stimulates the expression of ACO and EHHADH mRNA to strengthen fatty acid β-oxidation.
