1.Role of mitophagy induced by the PTEN-induced kinase 1/Parkin signaling pathway in metabolic associated fatty liver disease and related advances in targeted therapies

Metabolic associated fatty liver disease(MAFLD)has a complex pathogenesis,and mitophagy is involved in the development and progression of MAFLD and plays a key role in liver metabolic pathways and signaling networks.Mitophagy is regulated by a variety of pathways,and the PTEN-induced kinase 1(PINK1)/Parkin pathway is considered the main pathway for regulating mitophagy.Mitophagy mediated by the PINK1/Parkin pathway can regulate lipid metabolism,inflammation,and fibrosis and delay the progression of MAFLD.This article reviews the role of mitophagy mediated by the PINK1/Parkin pathway in MAFLD and the research advances in targeted therapy,in order to provide theoretical bases and ideas for the prevention and treatment of MAFLD.
2.Analysis on Clinical Trial Registration of Acupuncture and Moxibustion for Constipation Based on ChiCTR and ClinicalTrials.gov

Objective To analyze the current situation of clinical trial registration of acupuncture and moxibustion in the prevention and treatment of constipation;To provide references for clinical research in this field.Methods The clinical trials related to acupuncture and moxibustion in the prevention and treatment of constipation were retrieved from ChiCTR and ClinicalTrials.gov from the establishment of the databases to September 20,2024.The registration time,regions and institutions,funding sources,research types,randomized and blind methods,intervention measures,sample size,outcome indicators,etc.of the included studies were analyzed.Results A total of 73 relevant clinical trials were included,mainly from China(62),the United States(3),Pakistan(2)and Egypt(2);the registered projects in China involved 16 provincial-level administrative regions,mainly distributed in Beijing(11),Shanghai(7),Sichuan(7),Shandong(7),etc.;the main type of research was intervention research(69),and most of them were randomized parallel controlled trials(42);44 clinical trials were blinded;the study was mainly based on single center,and the total number of recruited samples was 9 175,involving a variety of acupuncture intervention measures.Conclusion The clinical trials related to acupuncture and moxibustion for the prevention and treatment of constipation are showing an increasing trend year by year.The overall number of registrations is small.There are some problems,such as uneven registration areas and nonstandard filling of registration information.The quality of research needs to be improved.

Nonalcoholic steatohepatitis(NASH)is a chronic metabolic disease characterized by hepatocyte fatty degeneration and ballooning degeneration,and it plays an important role in the progression of hepatic steatosis.Recent studies have shown that immune homeostasis imbalance between T helper 17(Th17)and regulatory T(Treg)cells are closely associated with the pathological process of NASH.Transforming growth factor-β1(TGF-β1)is a key cytokine for regulating the differentiation and proliferation of Th17/Treg cells,and TGF-β1 binds to its receptor and activates the Smad signaling pathway,thereby regulating the immune balance of Th17/Treg cells and the expression of inflammatory factors and participating in the repair of liver inflammation.This article systematically reviews the molecular mechanism of the TGF-β1/Smad signaling pathway in affecting NASH by regulating the immune balance of Th17/Treg cells,in order to provide a theoretical basis for the research on the pathogenesis of NASH and related treatment strategies.

5.Construction and application of pediatric diagnosis and treatment island in a tertiary hospital

Establishing a one-stop diagnosis and treatment mode centered on patients and linked by diseases is of great significance for optimizing the medical process and improving the medical experience. In March 2024, a tertiary hospital integrated pediatric outpatient and emergency resources, established a pediatric diagnosis and treatment island through reasonable department settings, strengthened talent team construction, optimized diagnosis and treatment processes, and established supporting guarantee mechanisms. It was officially put into use in June of the same year, providing one-stop diagnosis and treatment services for children and achieving the goal of " not leaving the island for minor illnesses and not leaving the hospital for major illnesses". Before the operation of island (January May 2024), the complaint rate and waiting time for pediatric outpatient and emergency department were 39 cases per 100 000 patients and 15 to 30 minutes, respectively; After operation (June August 2024), the complaint rate and waiting time decreased to 17 cases per 100 000 people and 10 to 20 minutes respectively; The average monthly comprehensive income of outpatient and emergency departments increased by 33%. The pediatric diagnosis and treatment island mode could assist in the sustainable high-quality development of pediatrics in hospital, and provide references for optimizing outpatient and emergency department management in other tertiary public hospitals. In the future, we should further enrich the service content of the island and strengthen information technology construction.
6.Relationship between nonalcoholic fatty liver disease and long-term prognosis in patients with acute myocardial infarction

Objective:To investigate the relationship between nonalcoholic fatty liver disease (NAFLD) and the long-term prognosis in patients with acute myocardial infarction (AMI).Methods:It was a retrospective cohort study. A total of 712 patients diagnosed with AMI who were admitted to the People′s Hospital of Xinjiang Uygur Autonomous Region from January 2018 to December 2019 were continuously included as subjects. The fatty liver index (FLI) was used to evaluate the degree of hepatic steatosis. Subjects were divided into No NAFLD group (FLI?30), grade 1 NAFLD group (30≤FLI?60), and grade 2 NAFLD group (FLI≥60). The endpoint event was defined as the occurrence of major cardiovascular adverse events (MACEs). The Cox proportional risk model was used to assess the risk of MACEs. The Kaplan-Meier curve was used to analyze the survival differences between the groups, and subgroup analysis was performed for age, gender, and complicity with hypertension, diabetes, dyslipidemia, and abdominal obesity.Results:During the follow-up period, the incidence of MACEs was 10.92% (26/238) in the No NAFLD group, it was 18.01% (47/261) in the grade 1 NAFLD group, and 24.41% (52/213) in the grade 2 NAFLD group, there was significant difference in the incidence of adverse events among the groups ( χ2=9.136, P?0.05). After adjusting for confounding factors, compared with the No NAFLD group, the risk of MACEs in grade 1 NAFLD group was increased by 69.0%( HR=1.690, 95% CI: 1.026-2.783, P=0.039) and 131.2%( HR=2.312, 95% CI: 1.415-3.773, P=0.001), respectively. Kaplan-Meier curve analysis indicated that the cumulative incidence of MACEs was significantly increased and the prognosis was worse in grade 2 NAFLD group (Log-rank test χ2=13.500, P=0.001). The results of subgroup analysis suggested that grade 2 NAFLD could significantly increase the risk of MACEs in all age groups, women, dyslipidemia, normal body type, and people with or without hypertension or diabetes. In particular, it had higher predictive value in women and normal-size people (interaction P?0.05). Conclusion:The degree of NAFLD is closely related to the long-term prognosis of AMI patients, the more severe the NAFLD, the higher the risk of adverse events in AMI patients.
7.Research Progress of Active Components of Chinese Materia Medica in the Treatment of Non-alcoholic Fatty Liver Disease by Regulating SREBPs Signaling Pathway

Non-alcoholic fatty liver disease(NAFLD)is characterized by excessive fat accumulation in the liver,which progresses to steatosis,inflammation and fibrosis with the progression of the disease,involving multiple cellular pathways and molecular mechanisms.Sterol regulatory element-binding proteins(SREBPs)have become key regulators of lipid metabolism in NAFLD,providing potential therapeutic targets for NAFLD.This article summarized molecular mechanism of SREBPs in NAFLD,and concluded the active components of Chinese materia medica targeted to regulate the SREBPs signaling pathway.It was found that the active components of Chinese materia medica(glycosides,flavonoids,polyphenols,polysaccharides,alkaloids,etc.)could inhibit the activation of SREBPs,inhibit de novo lipogenesis in the liver,and promote the β oxidation of free fatty acids and cholesterol metabolism,thus delaying or preventing the progress of NAFLD,which provided a reference for the treatment of NAFLD.
8.Effects of scaffold materials combined with biological factors on biological characteristics of dental follicle cell proliferation and osteogenic differentiation

BACKGROUND:Dental follicle cells are widely used in periodontal tissue regeneration engineering because of their excellent characteristics.With the development of biological scaffold materials,their relationship with periodontal tissue regeneration technology is increasingly close.OBJECTIVE:To review the performance of ivory follicle cells under the influence of internal and external biological factors by different experiments,and analyze their effects on the biological characteristics of dental follicle cells with scaffold materials.METHODS:Using"dental follicle cell,scaffolds,material,periodontal tissue regeneration,tissue engineering,review"as English and Chinese key words,the articles published in PubMed,Sciencedirect,and CNKI from 2013 to 2023 were searched,and finally 95 articles were included for analysis and discussion.RESULTS AND CONCLUSION:(1)Dental follicle cells originate from dental follicle tissue,which has certain stem cell differentiation potential.Because of its excellent performance,it is actively used in periodontal tissue regeneration engineering research.(2)The proliferation and osteogenic differentiation of dental follicle cells are affected by many biological factors,and both endogenous and exogenous factors can promote the proliferation and osteogenic differentiation of dental follicle cells to a certain extent.(3)3D printing technology and nanotechnology enable researchers to manufacture more suitable scaffold materials.(4)Polymer materials show us their flexibility and plasticity in periodontal tissue regeneration.We can manufacture targeted scaffold materials according to different defect sites to achieve efficient tissue regeneration.The good biocompatibility of inorganic materials makes them widely used in periodontal tissue regeneration engineering.By adjusting the content of nanoscale inorganic materials or improving the performance of scaffolds,scaffolds with better biocompatibility can be prepared.(5)There are many new synthetic(composite)materials,which show us excellent characteristics.However,because the mechanism of biological factors in scaffold materials on dental follicle cells is complicated,and the research on dental follicle cells is mostly concentrated on in vitro culture,so how to make scaffold materials more suitable for the growth and development of dental follicle cells and apply them safely and effectively in clinical treatment is the future research direction.
9.Advances in Lipidomics Research on Metabolic Associated(Non-alcoholic)Steatohepatitis

Metabolic associated(non-alcoholic)steatohepatitis(MASH/NASH)is a progressive form of metabolic associated(non-alcoholic)fatty liver disease(MAFLD/NAFLD),characterized by hepatic fat accumulation,inflammation,with or without fibrosis.It can further progress to liver cirrhosis or hepatocellular carcinoma,and it is a common and frequently-occurring liver disease.In recent years,due to its unique advantages in analyzing lipid metabolism disorders,lipidomics has become an important tool in the research of MASH/NASH.This article reviews the role of lipidomics in elucidating the pathological mechanisms of MASH/NASH,discovering biomarkers,and exploring therapeutic targets,aiming to provide a theoretical basis for basic research and new drug development for MASH/NASH.
