1.Clinical evaluation of endoscopic super-hemorrhoidal banding for 112 cases of internal hemorrhoids with prolapse (with video)

Objective:To evaluate the safety and efficacy of endoscopic super-hemorrhoidal banding (SHB) in the treatment of internal hemorrhoids with prolapse.Methods:From June 2019 to June 2020, a total of 112 patients with grade Ⅱ and Ⅲ internal hemorrhoids underwent endoscopic SHB at People′s Hospital of Qiandongnan Miao and Dong Autonomous Prefecture. The efficacy of SHB, intraoperative and postoperative complications associated with the operation and patients′ satisfaction were prospectively studied. The follow-up time was over six months.Results:A total of 112 patients received SHB for internal hemorrhoids, 74 patients (66.1%) with grade Ⅱ hemorrhoids and 38 patients (33.9%) with grade Ⅲ hemorrhoids. The operations were successfully performed in all patients. No anal pain or bleeding was reported during the operation. All patients experienced various degrees of anal bulge after the operation, which lasted for 2-24 hours before spontaneous disappearance. Five patients with grade Ⅲ internal hemorrhoids showed dysuria, which was relieved with local heat on the bladder. One patient with grade Ⅲ internal hemorrhoid had bloody stool on day 14. The bleeding was stopped with another banding at the site of bleeding ulcer. All patients completed the follow-up. No complications such as bleeding, pain, infection or stenosis occurred in one month after operation. Eighteen patients with grade Ⅲ had partial prolapse after operation without bleeding or other symptoms. Six of them received additional SHB treatment and prolapse disappeared. No postoperative complications occurred in the six months of follow-up in all patients. The satisfaction rate was 100.0% according to the survey.Conclusion:Endoscopic SHB is an effective, safe and simple technique for internal hemorrhoids with prolapse.
2.Application of empowerment psychological nursing model combined with hope theory in patients with intracranial aneurysms

Objective:To explore the application effect of empowerment psychological nursing model combined with hope theory in patients with intracranial aneurysms.Methods:A total of 90 patients with intracranial aneurysm who were admitted to Ruian People's Hospital in Zhejiang Province from August 2015 to January 2019 were randomly selected and they were divided into the observation group and the control group according to the random number table method, with 45 patients in each group. The control group received routine nursing, while the observation group received nursing based on empowerment psychological nursing model combined with hope theory. Before the intervention and 2 months after the intervention, Herth Hope Inventory (HHI) , Self-Perceived Burden Scale (SPBS) , Psychological Capital Questionnaire (PCQ) and Short Form 36-item Health Survey (SF-36) were used to evaluate the hope level, self-perceived burden, psychological capital and quality of life of patients in two groups. The Glasgow Outcome Scale (GOS) was used to compare the prognosis of the two groups after 2 months of intervention.Results:After 2 months of intervention, the HHI, PCQ, and SF-36 scores of the observation group were higher than those before the intervention, and higher than those in the control group, and the differences were statistically significant ( P<0.05) . SPBS scores of patients in both groups were lower than those before intervention, and the score in the observation group was (27.28±5.06) , lower than (32.64±4.12) in the control group. The differences were statistically significant ( P<0.05) . After 2 months of intervention, the excellent and good rate of GOS classification was 91.11% (41/45) in the observation group and 80.00% (36/45) in the control group, but the difference between the two groups was not statistically significant ( P>0.05) . Conclusions:Empowerment psychological nursing model combined with hope theory can effectively improve the hope level and psychological capital of patients amd reduce the self-perceived burden, thus improving the quality of life.
3.Analysis of genomic DNA methylation level in foxtail millet by Methylation Sensitive Amplified Polymorphism.

DNA methylation is an important type of epigenetic modification in eukaryotes. In order to research genome-wide methylation levels and patterns in foxtail millet (Setaria italica), the Methylation Sensitive Amplified Polymorphism (MSAP) analysis (employing double digestion with EcoR I and Hpa II/Msp I) was established and applied in two foxtail millet cultivars (Chaogu 58 and Yugu 1). The results showed that 32 pairs of MSAP primers were selected from 100 MSAP primers, and 1 615 and 1 482 clearly distinguishable and reproducible bands were amplified from Chaogu 58 and Yugu 1 respectively, including 3 types of methylation patterns. Cytosine methylation levels of CCGG context in Chaogu 58 and Yugu 1 were characterized as 6.93% and 8.77% respectively. Such different genomic DNA methylation levels between two foxtail millet varieties may provide a preliminary reference for the cultivation of this crop from a novel epigenetic viewpoint.

4.Comparison of MITE transposons mPing in different rice subspecies.

The mPing family is the first active MITE TE family identified in rice genome. In order to compare the compositions and distributions of mPing family in the genomes of two rice subspecies japonica (cv. Nipponbare) and indica (cv. 93-11), we initially estimated the copy numbers of mPing family in those two subspecies using Southern blot and then confirmed the results by searching homologous copies in each reference genome using Blastn program, which turned out to have 52 and 14 mPing copies in corresponding reference genome, respectively. All mPing members in Nipponbare genome belong to mPing-1, while there are 3 mPing-1 and 11 mPing-2 copies in 93-11 genome. By further investigating the 5-kb flanking sequences of those mPing copies, it was found that 23 and 3 protein-coding genes in Nipponbare and 93-11 genome are residing adjacent to those mPing copies respectively. These results establish the preliminary theoretical foundation for further dissecting the genetic differences of japonica and indica rice in terms of the diversities and distributions of their component mPing.

5.Effects of the expression of KAI1 mRNA on patients with gastric cancer

Objective To investigate the relationship between the expression of KAI1 mRNA and the clinicopathological parameters and prognostic outcome of patients with gastric Callcer. Methods The expressions of KAI1 mRNA in 70 samples of gastric cancer tissue and 18 samples of benign gastric lesion were detected by in situ hybridization.Results The positive rates of KAI1 mRNA expression in benign gastric lesion and gastric cancer tissue were 94%(17/18)and 31%(22/70),respectively.The expression of KAI1 mRNA was influenced by tumor difierentiation degree and the state of lymph node metastasis.The positive rate of KAI1 mRNA expression was gradually decreased as the increase of invasion depth and the tumor clinical stages.The 5-year survival rate of patients with positive expression of KAI1 mRNA WaS higher than these with negative expression of KAI1 mRNA.Conclmions The less and down-regulation of KAI1 gene expression may be involved in the occurrence and development of gastric cancer.KAI1 gene might be a valuable indicator for early diagnosis and predicting the malignancy and metastasis potential of gastric cancer and the prognosis of patients with gastric cancer.
